        Measurement of three-dimensional flame structure by combined laser diagnostics

        To investigate three-dimensional flame structures of turbulent premixed flame experimentally, dual-plane planar laser induced fluorescence (PLIF) of CH radical has been developed. This dual-plane CH PLIF system consists of two independent conventional CH PLIF measurement systems and laser beam from each laser system are led to parallel optical pass using the difference of polarization, and CH PLIF is conducted in two parallel two-dimensional cross sections. The newly-developed dual-plane CH PLIF is combined with single-plane OH PLIF and stereoscopic particle image velocimetry (PIV) to clarify the relation between flame geometry and turbulence characteristics. The laser sheets for single-plane OH PLIF and stereoscopic PIV measurement are located at the center of two planes for CH PLIF. The separation between these two planes is selected to 500 μm. The measurement was conducted in relatively high Reynolds number methane-air turbulent jet premixed flame. The experimental results show that various three-dimensional flame structures such as the handgrip structure, which has been shown by three-dimensional direct numerical simulations (DNS), are included in high Reynolds number turbulent premixed flame. It was shown that the simultaneous measurement containing newly-developed dual-plane CH PLIF is useful for investigating the three-dimensional flame structures. To analyze the flame structures quantitatively, the flame curvature was estimated by using the CH and OH PLIF images, and the probability density function (pdf) of the curvatures was compared with the results of DNS. It was revealed that the minimum radius of curvature of the flame front coincides with Kolmogorov length. However, the feature of pdf of the flame curvature is slightly different from result of DNS, if the curvature was estimated from experimental results in two-dimensional cross section. On the other hand, the feature of pdf of mean curvature that calculated from triple-plane PLIF results is similar to that obtained from three-dimensional DNS.

        消費税の課税ㆍ非課税区分をめぐる法的ㆍ実務的問題点

        일본 국내에서 행해지는 자산의 양도 또는 보세지역에서 인도되는 외국 화물 가운데 일정한 요건에 해당되는 것은 소비세가 부과되지 않는 비과세거래로 된다. 자산의 양도에 있어서 과세ㆍ비과세의 구분은 소비세에 있어서 중요한 ① 과세표준액의 계산, ② 매입세액공제액의 계산, ③ 기준기간의 과세매출액의 계산 등의 계산에 영향을 미친다. 또한 소비세는 본래 소비자에게 전가되는 것이 예정된 세이기 때문에 과세되는 것을 비과세로 판단한 거래를 한 경우에는 과소신고뿐만이 아니라 거래상대방에 대한 전가가 가능하지 않으므로 소비세 상당액만큼 가격 인하를 한 것과 같은 결과가 되어 버린다. 따라서, 과세ㆍ비과세 구분을 둘러싼 판정은 거래의 당사자에 있어서도 매우 중요한 문제이며, 명확하고 구체적인 규정을 마련하는 것이 불가결하다. 그런데 현실적으로는 조문의 규정은 매우 간결하고 명확히 해석될 수 있는 것 같이 보여도 실제로 적용하려면 예상치 못한 의문점이나 문제점이 발생하고 있다. 이에 국세청은 비과세에 관한 많은 기본통칙이나 질의 응답사례를 공표하여 대응하고 있다. 그렇지만 통칙이나 질의응답사례에 너무 의지하게 되면 법령의 해석에 대한 과세관청의 판단에 크게 의존하게 되고, 과세관청의 통칙이나 질의응답사례를 보지 않으면 납세자 스스로 세액계산을 할 수 없게 만든다. 조세법률주의 원칙을 훼손되는 결과를 가져오게 된다.

        Comparison of the Efficiencies of Different Affinity Tags in the Purification of a Recombinant Secretory Protein Expressed in Silkworm Larval Hemolymph

        Silkworms are useful bioreactors for heterologous protein expression when used in conjunction with the Bombyx mori nucleopolyhedrovirus (BmNPV) bacmid system. However, purification from silkworm hemolymph is difficult since it contains various kinds of proteins. In this study, we investigated an effective single-step method for the purification of affinity-tagged single-chain antibody variable region fragment (scFv) from silkworm larval hemolymph. A 5-fold higher expression level was obtained when scFv was fused with the His tag than when it was fused with the Strep II or GST tags. However, the His tag was inadequate for single-step purification since it led to the nonspecific binding of contaminants. The purification recoveries of GST-, Strep II-, and His-tagged scFvs were 91.8%, 43.7%, and 27.2%, respectively. The specific amount of single-step purified GST-tagged scFv was 2.2~2.7 fold higher than the amounts of the His- and Strep II-tagged constructs. The purities of Strep II- and GST-tagged scFvs in the eluent were 98.4% and 83.0%, respectively.         Early Improvement and Marriage Are Determinants of the 12-Month Treatment Outcome of Paroxetine in Outpatients with Panic Disorder

        Objective: In this study, we investigated the determinants of remission and discontinuation of paroxetine pharmacotherapy in outpatients with panic disorder (PD). Methods: Subjects were 79 outpatients diagnosed with PD who took 10-40 mg/day of paroxetine for 12 months. The candidate therapeutic determinants included the serotonin transporter gene-linked polymorphic region and the −1019C/G promoter polymorphism of the serotonin receptor 1A as genetic factors, educational background and marital status as environmental factors, and early improvement (EI) at 2 weeks as a clinical factor were assessed. The Clinical Global Impression scale was used to assess the therapeutic effects of the pharmacotherapy. Results: Cox proportional hazards regression was performed to investigate the significant predictive factors of remission and discontinuation. EI was only a significant predictive factor of remission. EI was a significant predictive factor of remission (hazard ratio [HR], 2.709; 95% confidence interval [CI], 1.177-6.235). Otherwise, EI and marital status were significant predictive factors of the discontinuation. EI (HR, 0.266; 95% CI, 0.115-0.617) and being married (HR, 0.437; 95% CI, 0.204-0.939) were considered to reduce the risk of treatment discontinuation. In married subjects, EI was a significant predictive factor of the discontinuation (HR, 0.160; 95% CI, 0.045-0.565). However, in unmarried subjects, EI was not a significantly predictive factor for the discontinuation. Conclusion: EI achievement appears to be a determinant of PD remission in paroxetine treatment. In married PD patients, EI achievement also appears to reduce a risk of discontinuation of paroxetine treatment

        Genetic Polymorphism of 1019C/G (rs6295) Promoter of Serotonin 1A Receptor and Catechol-O-Methyltransferase in Panic Disorder

        Objective-Family and twin studies have suggested genetic liability for panic disorder (PD) and therefore we sought to determine the role of noradrenergic and serotonergic candidate genes for susceptibility for PD in a Japanese population. Methods-In this age-and gender-matched case-control study involving 119 PD patients and 119 healthy controls, we examined the genotype distributions and allele frequencies of the serotonin transporter gene linked polymorphic region (5-HTTLPR), -1019C/G (rs6295) promoter polymorphism of the serotonin receptor 1A (5-HT1A), and catechol-O-methyltransferase (COMT) gene polymorphism (rs4680) and their association with PD. Results-No significant differences were evident in the allele frequencies or genotype distributions of the COMT (rs4680), 5-HTTLPR polymorphisms or the -1019C/G (rs6295) promoter polymorphism of 5-HT1A between PD patients and controls. Although there were no significant associations of these polymorphisms with in subgroups of PD patients differentiated by gender or in subgroup comorbid with agoraphobia (AP), significant difference was observed in genotype distributions of the -1019C/G (rs6295) promoter polymorphism of 5-HT1A between PD patients without AP and controls (p=0.047). Conclusion-In this association study, the 1019C/G (rs6295) promoter polymorphism of the 5-HT1A receptor G/G genotype was associated with PD without AP in a Japanese population.

        Rapid On-Site Evaluation by Endosonographers during Endoscopic Ultrasonography-Guided Fine-Needle Aspiration for Diagnosis of Gastrointestinal Stromal Tumors

        Background/Aims: Endoscopic ultrasonography-guided fine-needle aspiration (EUS-FNA) has been used to diagnose gastrointestinal submucosal tumors (SMTs). Although rapid on-site evaluation (ROSE) has been reported to improve the diagnostic accuracy of EUS-FNA for pancreatic lesions, on-site cytopathologists are not routinely available. Given this background, the usefulness of ROSE by endosonographers themselves for pancreatic tumors has also been reported. However, ROSE by endosonographers for diagnosis of SMT has not been reported. The aim of this study was to evaluate the diagnostic accuracy of EUS-FNA with ROSE by endosonographers for SMT, focusing on diagnosis of gastrointestinal stromal tumor (GIST), compared with that of EUS-FNA alone. Methods: Twenty-two consecutive patients who underwent EUS-FNA with ROSE by endosonographers for SMT followed by surgical resection were identified. Ten historical control subjects who underwent EUS-FNA without ROSE were used for comparison. Results: The overall diagnostic accuracy for SMT was significantly higher in cases with than without ROSE (100% vs. 80%, p=0.03). The number of needle passes by FNA with ROSE by endosonographers tended to be fewer, although accuracy was increased (3.3±1.3 vs. 5.9±3.8, p=0.06). Conclusions: ROSE by endosonographers during EUS-FNA for SMT is useful for definitive diagnosis, particularly for GIST.

        Genomic Profiling Shows Increased Glucose Metabolism in Luminal B Breast Cancer

        We had previously reported a close association between pathological response and the maximum tumor standardized uptake value (SUVmax) measured by 18F-fluorodeoxyglucose positron emission tomography prior to chemotherapy in estrogen receptor (ER)-positive breast cancer. We hypothesized that glucose hypermetabolism by luminal B tumors may result in chemotherapy responsiveness. Using a single-gene expression assay, TargetPrint® (Agendia) and a 70-gene expression classifier,MammaPrint® (Agendia), we divided 20 patients with ERpositive primary breast cancer into luminal A and luminal B subtypes and compared the tumor SUVmax value between the two groups. A significantly higher SUVmax was measured for luminal B tumors (n=10; mean±SD, 7.6±5.6) than for luminal A tumors (n=10; mean±SD, 2.6±1.2; p=0.01). Glucose hypermetabolism could help predict intrinsic subtyping and chemotherapy responsiveness as a supplement to ER, progesterone receptor, HER2,and Ki-67 histochemical scores.

        Study on laser assisted milling of ferrous based consolidated material

        This study deals with the laser beam assisted milling to improve the machinability of a consolidated structure formed by layered manufacturing processes. The laser beam used is a continuous CO2 laser with a maximum output power of 100 W. The metal powder for layered manufacturing is a ferrous-based mixture. To investigate the influence of laser conditions on the machinability, specific cutting force,tool wear and tool temperature of the flank face are measured. The results showed that the specific cutting force of the consolidated structure was decreased by the effect of energy input on the structure surface. The progress of wear on the flank face assisted by the laser beam was 20% slower due to the softening of the consolidated structure and the melting of the powder which remained on the structure surface. The dimensional accuracy and surface finishing of the consolidated structure were influenced by the rise of the tool temperature on the flank face.

        NDR1-Dependent Regulation of Kindlin-3 Controls High-Affinity LFA-1 Binding and Immune Synapse Organization

        <P><B>ABSTRACT</B></P><P>Antigen-specific adhesion between T cells and antigen-presenting cells (APC) during the formation of the immunological synapse (IS) is mediated by LFA-1 and ICAM-1. Here, LFA-1–ICAM-1 interactions were measured at the single-molecule level on supported lipid bilayers. High-affinity binding was detected at low frequencies in the inner peripheral supramolecular activation cluster (SMAC) zone that contained high levels of activated Rap1 and kindlin-3. Rap1 was essential for T cell attachment, whereas deficiencies of ste20-like kinases, Mst1/Mst2, diminished high-affinity binding and abrogated central SMAC (cSMAC) formation with mislocalized kindlin-3 and vesicle transport regulators involved in T cell receptor recycling/releasing machineries, resulting in impaired T cell-APC interactions. We found that NDR1 kinase, activated by the Rap1 signaling cascade through RAPL and Mst1/Mst2, associated with and recruited kindlin-3 to the IS, which was required for high-affinity LFA-1/ICAM-1 binding and cSMAC formation. Our findings reveal crucial roles for Rap1 signaling via NDR1 for recruitment of kindlin-3 and IS organization.</P>

        A Study on Aspects of Attrition Wear of Cutting Grains in Grinding Process

        In this study, the influences caused by the wear of grinding wheel are investigated experimentally. The infrared radiation pyrometer with an optical fiber is developed and applied for the temperature measurement in the surface grinding process of carbon steels. Experimental data such as the temperature, the number and the size of effective cutting grains and the grinding forces are obtained over the wheel life, and they are used for the calculation of the thermal energy fraction to the workpiece, the grinding wheel and the chips. Thermal damage begins to be caused on the ground surface, as the attrition wear of grains decreases the number of effective cutting grains and degrades the efficiency of grinding wheel. The measuring of radiation pulses from the cutting grains makes it possible to detect the degradation of grinding wheel at an early stage of wheel life and prevent the severe damages on work surface.
