<P><B>Abstract</B></P><P>Selective retrieval of sequence-verified oligonucleotides (oligos) from next-generation sequencing (NGS) flow cells, termed megacloning, promises accurate and reliable gene synthesis. However, gene assembly requires a complete collection of overlapping sense and nonsense oligos, and megacloning does not typically guarantee the complete production of sequence-verified oligos. Therefore, missing oligos must be provided <I>via</I> repetitive rounds of megacloning, which introduces a bottleneck for scaled-up efforts at gene assembly. Here, we introduce the concept of high-depth tiled oligo design to successfully utilize megacloned oligos for gene synthesis. Using acquired oligos from a single round of the megacloning process, we assembled 72 of 81 target Cas9-coding gene variants. We further validated 62 of these <I>cas9</I> constructs, and deposited the plasmids to Addgene for subsequent functional characterization by the scientific community. This study demonstrates the utility of using sequence-verified oligos for DNA assembly and provides a practical and reliable optimized method for high-throughput gene synthesis.</P>
Development of bacteria-based microrobot using biocompatible poly(ethylene glycol).

<P>For the development of bacteria-based biomedical microrobot, we propose the fabrication method of biocompatible poly(ethylene glycol) (PEG) microbeads using a cross-junction microfluidic channel. PEG droplets were polymerized by ultraviolet (UV) irradiation to form PEG microbeads of 8.18 3.4 μm diameter in a microfluidic channel. Generally, the bacteria did not attach to the surface of the PEG microbeads because of their hydrophilicity. We modified the selective surface of the PEG microbeads using poly-L-lysine (PLL), promoting attenuated Salmonella typhimurium adhesion using the submerging property of PEG microbeads on agarose gel: the bacteria could thus be attached to the PLL-coated surface region of the PEG microbeads. The selectively PLL-coated PEG microbeads group showed enhanced motility compared with the PLL-uncoated and completely PLL-coated PEG microbeads groups. The selectively PLL-coated PEG microbeads group showed 12.33 and 7.40 times higher average velocities than the PLL-uncoated and completely PLL-coated PEG microbeads groups, respectively. This study verified the successful fabrication of bacteria-based microrobots using PEG microbeads, and the enhanced motility of the microrobots by selective bacteria patterning using agarose gel and PLL.</P>

<P>Many multicomponent protein complexes mediating diverse cellular processes are assembled through scaffolds with specialized protein interaction modules. The multi-tRNA synthetase complex (MSC), consisting of nine different aminoacyl-tRNA synthetases and three non-enzymatic factors (AIMP1–3), serves as a hub for many signaling pathways in addition to its role in protein synthesis. However, the assembly process and structural arrangement of the MSC components are not well understood. Here we show the heterotetrameric complex structure of the glutathione transferase (GST) domains shared among the four MSC components, methionyl-tRNA synthetase (MRS), glutaminyl-prolyl-tRNA synthetase (EPRS), AIMP2 and AIMP3. The MRS-AIMP3 and EPRS-AIMP2 using interface 1 are bridged via interface 2 of AIMP3 and EPRS to generate a unique linear complex of MRS-AIMP3:EPRS-AIMP2 at the molar ratio of (1:1):(1:1). Interestingly, the affinity at interface 2 of AIMP3:EPRS can be varied depending on the occupancy of interface 1, suggesting the dynamic nature of the linear GST tetramer. The four components are optimally arranged for maximal accommodation of additional domains and proteins. These characteristics suggest the GST tetramer as a unique and dynamic structural platform from which the MSC components are assembled. Considering prevalence of the GST-like domains, this tetramer can also provide a tool for the communication of the MSC with other GST-containing cellular factors.</P>

In future ubiquitous generation, users will use more services than now. And the advertisements will periodically send to users for advertising the services. The service must be provided the appropriate it that is user's preference on ubiquitous. And it has to provide at wanted time and wanted location. Similarly advertisement will be provided the customized it to users. To serve the customized service, first it needs the user's interest and preference. Consequently the demand about user profile or preference information will become larger than. However the security problems may be caused by these exchanged data. In this paper, we will provide the method for resolving the security problem and propose the technique to allow the restricted advertisements.

The one-dimensional gratings and two-dimensional crossed gratings on 4 inch silicon wafer in subwavelength scale have been fabricated by holographic lithography system, which was constructed using He-Cd laser (λ=441.6㎚). For the good surface relief profile, the holographic lithography process was modulated with the exposure energy and developing time. After measuring the surface profiles of nano-structures, master structures on photoresist have been used to fabricate metallic nano-stampers using electroforming process for UV molding.

<P>We demonstrated nonvolatile 8 × 8 array organic memory devices utilizing a PC-interface memory cell tester. The organic memory devices composed of a Ag/poly(3-hexylthiophene-2,5-diyl) (P3HT)/p<SUP>+</SUP> poly-Si structure exhibited excellent memory performance properties, including stable switching behavior, proper statistical distribution, and long retention time. We succeeded in independently addressing and reading the data in the memory cell array using the PC-interface memory cell tester, opening an avenue toward more realistic organic memory device applications.</P>

현재의 많은 추천 시스템들은 사용자와 아이템에 기반하여 적절한 추천을 생성 및 제공하고 있다. 그러나 실제 생활 환경에서의 아이템 사용 내역은 추천에서 배제되고 있는 상황이다. 실제적인 사용 현황이 추천 시스템에 적용된다면 보다 정확한 추천이 가능할 것이다. 따라서 본 논문에서는 생활 환경 속에서 센서를 통한 정보 수집을 가정하고 이를 처리하여 생성한 상황 정 보른 기반으로 추천을 생성하는 기법을 제안하고자 한다.

현재의 웹 검색은 단순히 키워드 매칭만을 통해 필요한 정보들을 검색하기 때문에 그 결과가 사용자가 원하는 정보와는 의미적으로 상이한 결과들을 많이 포함하고 있다. 사용자가 원하는 정보와 의미적으로 정확히 일치하는 정보들을 추출하기 위해서는 웹 자원에 대한 정확한 의미 부여와 웹 자원들 사이의 의미적 연관성을 기술할 수 있는 지식 표현 수단인 온톨로지가 필요하다. 웹 기술 표준화 단체인 W3C에서는 이와 같은 웹 자원에 대한 의미 표현 기술로 OWL(Web Ontology Language)이라는 웹 온톨로지 언어를 발표하였으나 아직 이를 효과적으로 저작, 편집할 수 있는 전용 도구의 개발은 아직 미비한 실정이다. 따라서 본 논문은 OWL의 생성 및 편집을 효과적으로 제공할 수 있는 저작 시스템을 설계하고 구현하였다. Current web search includes a lot of different results with information that user does not want, because it searches information using keyword mapping. Ontology can describe the correct meaning of web resource and relationships between web resources. And we can extract suitable information that user wants using Ontology. Accordingly, we need the ontology to represent knowledge. W3C announced OWL(Web Ontology Language), meaning description technology for such web resource. However, the development of a special tool that can effectively compose and edit OWL is inactive. In this paper, we designed and developed an OWL authoring system that can effectively provide the generation and edit about OWL.
Direct Observation of Ag Filamentary Paths in Organic Resistive Memory Devices

<P><B>Abstract</B></P><P>We demonstrate bipolar switching of organic resistive memory devices consisting of Ag/polymer/heavily‐doped p‐type poly Si junctions in an 8 × 8 cross‐bar array structure. The bistable switching mechanism appears to be related to the formation and rupture of highly conductive paths, as shown by a direct observation of Ag metallic bridges using transmission electron microscopy and energy‐dispersive X‐ray spectroscopy. Current images of high‐ and low‐conducting states acquired by conducting atomic force microscopy also support this filamentary switching mechanism. The filamentary formation can be described by an electrochemical redox reaction model of Ag. Our results may also be applied to other kinds of organic materials presenting similar switching properties, contributing to the optimization of device scaling or memory performance improvement.</P>
