Effect of hemin and zinc on experimental colon carcinogenesis in mice

Excessive intake of red meat has been associated with colon carcinogenesis. The effect of hemin and zinc on colon carcinogenesis was investigated in male ICR mice. After acclimation for 1 week, five-week-old mice received three s.c. injections (0-2nd weeks of the experiment) of azoxymethane (AOM; 10 mg/kg b.w.) weekly and were treated by 2% dextran sodium sulfate (DSS) in drinking water for the next 1 week to induce aberrant crypt foci (ACF). Mice were fed on AIN-76A purified rodent diet for 6 weeks. There were three experimental groups; control, hemin, and hemin + zinc groups. The daily oral doses of hemin and zinc were 534 mg/kg and 55 mg/kg b.w., respectively. After staining colonic mucosa with 0.2% methylene blue, the ACF, aberrant crypts (AC), and polyps were counted. Lipid peroxidation in liver was evaluated by thiobarbituric acid-reactive substances (TBARS) assay. The numbers of AC, ACF, large ACF (i.e., ≥4 AC/ACF), and polyps in the hemin group were higher than those in the control group. In hemin + zinc group, the numbers of AC, ACF, large ACF were significantly lower compared to the hemin or control groups (p < 0.05), and the number of polyps was also significantly lower compared to that in the hemin group (p < 0.05). The TBARS level in the livers of the hemin + zinc group was significantly lower than that of the hemin group (p < 0.05). These results suggest that hemin enhances colon carcinogenesis and that co-treatment with zinc can protect against the induction of colon carcinogenesis.

A methanol extract from seeds of Paeonia lactiflora (Paeoniaceae, peony) was found to possess different antiproliferative activities against four different human cancer cell lines: Hela, MCF-7, HepG2 and HT-29. Furthermore, five different methanol (20, 40, 60, 80 and 100% MeOH) fractions obtained by fractionation of the methanol extract of the seeds on a Diaion HP-20 column exhibited differential antiproliferative effects against the above four cancer cell lines. Among five fractions, the 60% MeOH fraction showed relatively lower antiproliferative activity on MCF-7 estrogen-sensitive breast cancer cell than the other cancer cell lines. Systematic separation of 60% the MeOH fraction by silica gel and Sephadex LH-20 columns led to the isolation of four known stilbenes, trans-resveratrol (1), trans-(+)- ε-viniferin (2), gnetin H (3) and suffruticosol B (4). The four stilbenes (1~4) exerted differential biphasic effects on cell proliferation of MCF-7 cells in a similar manner as genistein, a soybean isoflavone used as a positive reference, in the concentration range from 1.0 to 200 μM. Three stilbenes (1~3) weakly stimulated the proliferation of MCF-7 cells at doses below 10 μM. However, strong antiproliferative effects on MCF-7 cell were exerted by extract 1 at a dose of 200 μM, and by 2 and 3 at doses above 25 μM. In contrast, 4 inhibited the proliferation of MCF- 7 cell at a dose below 25 μM, but stimulated cell proliferation at concentrations of 50 and 100 μM. All four stilbenes (1~4) stimulated the proliferation of ROS 17/2.8 osteoblast-like cells in the range of 10^(-10)~10^(-1) μM. Compound 1 exhibited especially potent proliferative activity, although its activity was weaker than that of genistein. Additionally, three resveratrol oligomers (2~4) also exhibited concentration-dependently moderate proliferative activity, but less than that of 1. These results suggest that resveratrol, and its dimer and trimers from the seeds of Paeonia lactiflora may act as a phytoestrogen, but in a somewhat different manner from that of genistein.

Obesity and related metabolic diseases are major problems worldwide. Some probiotics are currently considered potential therapeutic strategies for obesity. We aimed to investigate the anti-obesity efficacy of Latilactobacillus sakei WIKIM31 in obese mice induced by a high fat diet. The administration of a high-fat diet with L. sakei WIKIM31 reduced body weight gain, epididymal fat mass, triglyceride and total cholesterol levels in the blood, and remarkably decreased the expression of lipogenesis-related genes in the epididymal adipose tissue and liver. Interestingly, intake of L. sakei WIKIM31 improved gut barrier function by increasing the gene expression of tight junction proteins and suppressing the inflammatory responses. Additionally, L. sakei WIKIM31 enhanced the production of short-chain fatty acids, such as butyrate and propionate, in the intestinal tract. These results showed that L. sakei WIKIM31 can be used as a potential therapeutic probiotic for obesity.

PURPOSE: This study aimed to examine the effects of resistance and balance training on physical function and postural control in individuals with Down syndrome (DS). METHODS: Ten adults with DS constituted the experimental group (EX) and attended an 8-week resistance and balance training program. The results were compared with those of the control group (CON), which consisted of 10 patients who did not undergo any physical training. Body composition, postural stability, and physical fitness were measured before and after the training program. Resistance and balance training were performed for 60 minutes, three times a week for 8 weeks. RESULTS: Body weight, body mass index, body fat percentage, and waist circumference decreased significantly in the EX group after completing the 8-week resistance and balance training program. The center of pressure and the difference between standing foot pressure on the left and right were significantly improved in the EX group. Physical fitness including sit-to-stand repetitions and 10 m shuttle duration significantly improved in the EX and CON groups. CONCLUSIONS: These results suggest that resistance and balance training to improve musculoskeletal problems is an effective strategy to prevent injury, fatigue, and falls during exercise and to improve general health in individuals with DS.
A case of colovesical fistula complicated by sigmoid diverticulitis

Colovesical fistula is a rare complication of inflammatory or neoplastic disease, and traumatic or iatrogenic injuries. Among them, sigmoid diverticulitis is the most frequent cause of colovesical fistula. There are few reported cases of the colovesical fistula in Korean literature, that is probably because the left colonic diverticulum is not common in Korea. We present here a case of patient with colovesical fistula complicated by sigmoid diverticulitis. A 66-year-old male patient was transferred to our hospital due to complaining of abdominal pain and mucoid stool. An abdominal and pelvic CT scan revealed diverticulitis and peridiverticular abscess in sigmoid colon. The patient was admitted and treated with intravenous antibiotic agents for four weeks, followed by oral antibiotic therapy. Six weeks after being discharged from the hospital, he was admitted again with complaints of pneumaturia, fecaluria, and dysuria. Follow up CT scan demonstrated fistula between sigmoid colon and bladder dome portion. Under the diagnosis of colovesical fistula due to diverticulitis of sigmoid colon, low anterior resection and partial cystectomy with cystostomy were performed.
Oral bisphosphonates induced osteonecrosis of the mandible

Bisphosphonates have been approved for Paget’s disease, cancer - related hypercalcemia, bone involvement in multiple myeloma or solid tumors and osteoporosis. Although, underlying pathophysiological mechanisms remain unclear, it seems that bisphosphonates inhibit osteoclast precursor cells, modulate migratory and adhesive characteristics and induce apoptosis of osteoclasts. Furthermore impacts on angiogenesis, microenvironment and signal transduction between osteoclasts and osteoblasts. In this report, we present a case of oral bisphosphonates induced osteonecrosis of the mandible in a 84-year-old patient who received for two years. Two tapered screw vent implants(Zimmer, USA) were placed in the area of first and second molar. Two weeks later after crowns restored, some inflammatory signs and symptoms were observed on the second molar area. Sequestrum was formed and the sequestrum was removed with the implant. Frequent follow-up checks and oral hygiene maintenances were done and the first molar implant was restored. There is insufficient evidence suggests that duration of oral bisphosphonate therapy correlates with the development and severity of osteonecrosis. Therefore, dentists should not overlook the possibility of development of bisphosphonate induced osteonecrosis in patients who have taken oral forms of medication for less than three years.
Design of Binary LDPCA Codes with Source Revealing Rate-Adaptation

<P>In this letter, we consider binary low density parity check accumulate (LDPCA) codes with source revealing rate-adaptation for distributed source coding (DSC) applications. We propose an effective algorithm for selecting source symbols to the source revealing. Numerical results demonstrate that the proposed scheme significantly outperforms the conventional LDPCA codes in the high compression rate region in asymmetric DSC systems.</P>
