        Genome-wide investigation of a Korean synthetic breed, Woori-Heukdon using the Illumina PorcineSNP60K BeadChip

        Background: Woori-Heukdon (KWH) is a Korean synthetic pig breed generated using Chookjin-Duroc (KCD), Chookjin-Chamdon (KCC), and their crossbreds. Currently, there is a severe lack of studies investigating the Korean breed populations including wild boars (KWB) throughout the genome. Objective: This study was performed to investigate the genetic characteristics of Korean pig populations at the genome-wide level. Methods: Using the SNP dataset derived from genotyped and downloaded datasets using the Illumina PorcineSNP60K BeadChip, we compared the genomes of 532 individuals derived from 23 pig breeds to assess the genetic diversity, inbreeding coefficient, genetic differentiation, and population structure. Results: KWB showed the lowest average expected heterozygosity (HE = 0.1904), while KWH showed the highest genetic diversity (HE = 0.02859) among Korean populations. We verified that the genetic composition of KWH, showing USD of 74.8% and KCC of 25.2% in ADMIXTURE analysis. In population structure analyses, KCC was consistently shown to be separated from other pig populations. In addition, we observed gene flow from Western pigs to a part of Chinese populations. Conclusion: This study showed that Korean native pigs, KCC have genetic differences in comparison with Chinese and Western pigs; despite some historical records and recent genetic studies, we could not find any clear evidence that KCC was significantly influenced by Chinese or Western breeds in this study. We also verified the theoretical genomic composition of KWH at the molecular level in structure analyses. To our knowledge, this is the first genomic study to investigate the genomic characteristics of KWH and KCC.

        The admittance or impedance spectroscopy is one of the useful tools to study the equivalent circuit model of organic thin film devices and electrical properties of the film, such as dielectric relaxation and dielectric loss. We have investigated the frequency dependent characteristics of indium tin oxide/TPD/aluminum single layer device. The conductance increases with the applied DC bias in the low frequency region. This indicates that the electrical conductivity increases with the applied bias. The capacitance is nearly constant with the bias in the measured frequency region, which indicates the device acts as a parallel plate capacitor. The Cole-Cole plot of the admittances takes a one-semicircle shape, which means that the device can be modeled as a parallel resistor-capacitor equivalent circuit. We have also studied the complex permittivity of the device with the DC bias and frequency. The imaginary part of the permittivity shows the dielectric dispersion at the low frequencies and the dielectric absorption in the high frequency domain. The dispersion is a proportional to a reciprocal of the frequency. The dielectric absorption represents the relaxation peak and the dielectric relaxation time is about 2.65 μs.

        고리 및 영광 논토양에 대한 ^137Cs의 모의 사고 침적시 토양-벼 전이계수

        고리 및 영광 원전 반경 5km 내 11곳의 논토양을 원상대로 재배상자에 채취하여 실험 온실로 운반한 후 벼의 이식 전과 생육중에 ^137Cs을 담수의 표면에 처리하였다. 이식전 처리시 ^137Cs의 토양-작물체 전이계수(m²kg^-1-dry plant)는 토양에 따라 8배 정도의 변이를 보였고 평균은 지역간 차이가 거의 없이 쌀알의 경우 1.1 x 10^-4 , 볏짚의 경우 2.7 x 10^-4 이었다. 분얼초기 처리와 수잉기 처리시 전이계수는 이식전 처리에 비해 대체로 2∼3 배 및 15∼30 배 정도 높았다. 토양 유기물 함량과 전이계수 간에는 정의 상관이 있었다. 본 실험결과에 입각하여 우리나라 논에 대한 일회성 사고침적시 적용할 수 있는 ^137Cs 전이계수의 대표치를 침적시기별로 제안하였다. Undisturbed soil blocks were taken into culture boxes from 11 paddy fields within 5 km radii of Kori and Yonggwang NPPs and carried to a greenhouse where ^137Cs was applied to the surface of standing water before rice transplanting and during its growth. Transfer factors (m²kg^-1-dry plant) of ^137Cs applied before transplanting varied with soils by a factor of about 8. The mean was 1.1 x 10^-4 for hulled rice seeds and 2.7 x 10^-4 for straw without significant regional differences. The ^137Cs applications at the early tillering stage and booting stage resulted in, on the whole, 2∼3 and 15∼30 times, respectively, higher transfer factors than the application before transplanting. A positive correlation was found between soil organic matter and transfer factor. Generic values of transfer factors were proposed for an acute accidental deposition of ^137Cs onto Korean paddy fields in different times of the year.

      • 분자 배향성 제어를 이용한 유기전기발광소자 개발에 관한 연구

        The organic electroluminescent devices based on small molecules or conjugated polymers have been greater interest due to their possible applications for flat panel displays. They are attractive because of the capability of multicolor emission, low operating voltage, low power consumption, and competitive cost. The high mobilities allow larger current densities to be reached under operation in the space charge limited conduction region, hence facilitate access to higher luminance as required for passive matrix devices. In this study, we have investigated the molecular orientation control effect of organic electroluminscent devices. We aligned the p-sexiphenyl, which is a blue emitting material, using simple rubbing treatment. We obtained the anisotropic optical propery by UV/visible absorption measurement. In the current density-voltage relationship, we observed that the current density of the rubbing treated devices is larger than that of the non-rubbed device. This indicates that there is a more efficient carrier injection from the anode in the rubbed p-sexiphenyl devices.

      • 위기 유가족을 위한 단기 집단상담 프로그램 개발

        본 연구는 재해 및 위기상황이 발생한 후 이로 인해 가까운 사람을 잃은 유가족들을 위한 단기 집단상담의 한 모형을 개발제시한 것이다. 기존에 나와 있는 위기상담 프로그램들을 비교하였고 이를 기반으로 인지행동치료적 접근에서 이들이 자신의 삶을 좀 더 효과적으로 통제할 수 있는 프로그램을 개발하고자 하였다. 지금까지의 재해 및 위기상황을 주제로 하는 우리나라의 논문들을 고찰해본 결과 의료적, 물질적, 재정적 지원체계는 어느 정도 갖춰져 있으나 심리적, 정서적 지원체계는 거의 없는 실정이다. 최근 발생한 대구지하철참사의 경우만 보더라도 이에 대처하는 위기상황 극복 노력이 거의 기존의 지원체계에 의존하고 있는 실정이며, 일부 심리상담 자원봉사가 시도되기는 했으나 전문가와 전문프로그램의 부재 등으로 인해 그 실효성이 대우 미미한 실정 이다. 본 연구 결과는 위기 유가족들의 스트레스 대처능력을 강조하며, 적응적 대처전략의 습득에 대한 구체적인 지침을 제공해 줄 것이다. 위기 유가족들은 스트레스 상황에 대한 부정적 지각을 통해 자신의 우울, 불안, 분노감을 더욱 증대시키는 경향이 있다. 따라서, 위기 유가족들이 효과적이고 적응적인 대처전략을 습득할 수 있다면, 이는 사회적 지원 체계와 결합되어 그들의 역기능적인 증상 유발을 감소시키는 데 크게 기여할 것이다. This study aims to develop a brief group intervention program for the families which have lost their member(s) through a calamity. In particular, it is through various comparisons with the former intervention programs that we have developed the program in terms of cognitive-behavioral therapy, which can help the families get more sufficient control of their own lives. Considering the papers concerning crisis intervention programs that have been published so far, the focus of major programs lies on the medical, material, and financial supports without mention of psychological and emotional ones. For example, in case of recent subway conflagration in Daegu, every effort was made to support those involved in the calamity only in terms of the former support system. Any professional psychological and emotional intervention was not provided for the families who suffered from the accident. This study may contribute to increasing the abilities for such families to overcome the critical experience and it may help them to learn sufficient coping strategies with various stress situations. The negative effect of critical events leads to the paychological symptoms such as depression, anxiety, and rage and so on. Therefore, the efficient psychological intervention program will be very helpful and necessary to overcome the crisis experiences that those exposed to various calamities face.

      • 유기 능동 소자 제작을 위한 유기 박막의 제작 및 전기적 특성

        There are currently considerable interest in the applications of conjugated polymers, oligomers, and small molecules for thin-film electronic devices. Organic materials have potential advantages to be utilized as semiconductors in field-effect transistors and light-emitting diodes. In this study, pentacene thin-film transistors (TFTs) were fabricated on glass substrate. Aluminums were used for gate electrodes. silicon dioxide was deposited as a gate insulator by PECVD and patterned by reactive ion etching (R.I.E). Gold was used for the electrodes of source and drain. The active semiconductor pentacene layer was thermally evaporated in vacuum at a pressure of about 10?? Torr and a deposition rate 0.3Å/s. The fabricated devices exhibited the field-effect mobility as large as 0.07㎠/V.s and on/off current ratio as larger than 10??.

      • 유자종실의 Sterol 조성

        남해산 柚子種實油을 分析試料로 하여 日本, 基準油脂分析試驗法에 따라서 不檢化物量을 定量하고 T. L. C法으로 不檢化物을 分劃하였으며 G. C法에 依해 sterol組成을 分析하였다. 유자종실기름의 불검화물량은 기름 1g당 19.2mg으로서 1.9%이였고 불검화물의 분획 pattern은 less polar compounds가 20.3%, 4,4-dimethylsterol이 9.6%, 4-desmethylsterol이 47.1% 및 미전개물질이 22.9%이였다. sterol조성은 cholesterol이 0.3%, campesterol이 11.9%, stigmasterol이 9.3%, β-sitosterol이 72.2% 및 未知 sterol이 6.3% 이였다. 불검화물량, 불검화물의 분획 pattern 및 sterol조성은 他植物種實油와 거의 같은 경향을 나타냈다. This experiment was performed to the unsaponifiable matters content of the citrus seed oil. Also, the fractionated sterol pattern and the sterol composition of the citrus seed oil were examined. The citrus seed oil were contained 19.2mg unsaponifiable matters per gram. The unsaponifiable matters in seed oil were fractionated into less polar compounds(20.3%), 4, 4-dimethylsterol(9.6%), 4-monomethylsterol(0%), 4-desmethylsterol(47.1%) and undeveloped matters(22.9%). The sterol composition in citrus seed oil were cholesterol 0.3%, campesterol 11.9%, stigmasterol 9.3% and beta-sitosterol 72.2%. But brassicasterol and avenasterol were not detected.

      • CRP* 의존성 maltose 대사 촉진 유전자 sfs4의 클로닝 및 염기배열 결정

        CRP (cAMP receptor protein)은 cAMP와 결합하여 cAMP-CRP 복합체를 형성하여 전사조절의 조절인자로서 작용한다. crp 유전자에 변이를 도입하여 cAMP의 비존재 상태에서 cAMP-CRP와 비슷한 기능을 가진 crp* 유전자가 도입된 대장균 MK2001 (crp*¹, cya::km)을 숙주로 사용하여 cAMP 혹은 cGMP의 비존재하에서도 mal 유전자의 발현을 촉진시키는 유전자 sfs (sugar fermentation stimulation) 수 종을 클로닝하였다. 본 실험에서는 이미 밝혀진 nlp (Ner like protein) 유전자와 같이, sfs의 새로운 유전자를 탐색하여, 그 중 sfs4의 2126bp 전 염기배열을 결정하고, 잠정적인 sfs4의 promoter 영역에는 CRP 단백질과의 결합 DNA 공통 염기배열(5' AAT TGTGA ACACCA TCACC CGT 3')이 존재함을 확인했다. lacZ 융합 유전자를 작성하여 TP2010R1과 MK2001의 균주에서 cAMP를 첨가할 경우 각각 2.3배, 1.8배의 β-galactosidase 활성이 증가 하는 것으로 보아 sfs4는 cAMP-CRP에 의해 발현 조절을 받는 것으로 나타났다 In Escherichia coli, CRP forms a complex with cAMP and acts as a transcriptional regulator of many genes, including sugar metabolism operons. The E. coli MK2001, which is introduced the altered crp*¹, is functional in the expression of lac, ara and man, in the absence of cAMP. However, the expression of mal gene is fully activated by the addition of cAMP or cGMP. The object of the study is cloning of the sfs (sugar fermentation stimulation) genes, which was involved in regulation of mal gene expression with the altered crp*¹gene, and structural analysis and characterization of the genes at the molecular level. We have cloned 5 different E. coli genes which stimulate the maltose metabolism in a crp*¹, cya::km (MK2001) background. Newly identified genes were designated as sfs. One of the sfs genes (pPC1), located at the 53.2 min map position in the E. coli chromosome, was further analyzed. Expression of the genes, which is involved in maltose metabolism, malQ (amylomaltase), was increased to 5.8-fold in the presence of a plasmid, pAP5, containing the subcloned sfs4 gene. The nucleotide sequence of a common 2,126bp segment of the pPCM1 was determined and two open reading frames (ORF1 and ORF2) were detected. The ORF1 encodes the sfs4 gene and ORF2 encodes a truncated protein. Potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. Expression of the cloned sfs4 gene was positively regulated by the cAMP-CRP complex.
