Effect of ER signal peptides (KDEL) on expression and function of monoclonal antibody in plant

Plant genetic engineering led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current animal expression systems. In this study, the expression level of mAbP SO57 between with and without Lys-Asp-Glu-Leu (KDEL) was compared in transgenic plant. PCR and PT-PCR analyse showed stable gene insertion transcription of heavy and light chain genes of mAbP SO57 with or without KDEL in plant, respectively. We validated expression of mAbP SO57 by western blot. Western blot showed the significantly higher expression level of mAbP SO57 with KDEL compared to without KDEL. Flow cytometry (FACS) analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb (mAbM) evidenced similar binding activity to the FcγRI receptor (CD64). High performance liquid chromatography (HPLC) analysed, glycosylation patterns of mAbP SO57 with or without KDEL. The mAbP SO57 with KDEL had glycan profile with both oligomannose type (47.6%) and golgi type (52.4%), while the mAbP SO57 without KDEL had only golgi type (100%) glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest the potential of mAbP SO57 for rabies immunotherapy, regardless of plant specific glycan structures.

PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. Pierre Robin syndrome 환아에서 기관내 삽관 경험 : 증례보고

Pierre Robin syndrome is characterized by micrognathia, glossoptosis, and cleft palate. Neonates with Pierre Robin are at risk of upper airway obstruction and may require surgical fixation of the tongue to the mandible. Such neonates are at high risk of hypoxia and difficult to intubation during induction of anesthesia. We report a case of an 11-day old neonate with Pierre Robin syndrome was intubated in operative room and emergency room. While under general anaesthesia, awake intubation was attempted and facilitated oxygenation. After a 4 day discharge, he revisited to emergency room due to airway obstruction symptom. Repeated attempts at intubation (again with spontaneous breathing) failed. Finally, laryngeal mask airway (LMA) was introduced, and as a result of this ventilation was achieved. The next day, we awake fiberoptic intubation through the laryngeal mask and were easily achieved.
Social and stylistic variation in vowel raising in Seoul Korean

The purpose of this study is to examine extralinguistic factors that influence vowel raising of /o/ in constituent-final -ko and -to in Seoul Korean, focusing mainly on the influence of social variations and stylistic variations on this phenomenon. The Sociolinguistic interview data showed that older speakers used the most raised variant of /o/ in the AP-medial position, which is evidence for age-grading for the following reasons: (i) the linguistic marketplace, which is an important concept in age-grading, supports this idea because older people use the non-standard form of /o/, which is the raised variant, after they leave the workforce, and (ii) the unraised variant does not push out the raised variant, but rather, they co-exist. In addition, stylistic variations related to the formality of the speech setting and the solidarity between interactants affect the vowel, leading to vowel raising (i) in casual speech situation and (ii) in interaction with an addressee with whom the speaker is intimate; these trends are especially salient for younger speakers.

We analyzed the transcriptional profile of the Xoo infected bbr1 mutant using a commercial rice gene chip containing 51,279 transcripts. Microarray revealed 92 genes with increased levels of expression and 22 genes with decreased levels of expression in bbr1. Some of the differentially expressed genes were validated by qRT-PCR. Higher expression of defense-related genes and AP2 domain containing transcription factors along with lower expression of reactive oxygen scavenging enzymes may be responsible for defense signaling in the bbr1 upon Xoo infection. The putative target genes of AP2 domain containing transcription factors also showed differential gene expression during Xoo infection, some of which encoded bacterial pathogen resistance-related protein. Induction of AP2 domain containing transcription factors along with up-regulation of their putative target genes during Xoo infection may inhibit pathogen spread in the bbr1. This observation supports the hypothesis that AP2 domain containing transcription factors is involved in the regulation of differentially expressed genes in bbr1.
