Aims: The current study aims to elucidate the treatment efficacy (defined as undetectable HCV RNA throughout 12 weeks of post-treatment follow-up, SVR12) and safety DCV/ASV plus ribavirin for 12 weeks in HCV-1b patients without NS5A RAS. Methods: This is a single-arm, open-label phase 2 study. Seventy directly acting antivirals (DAA)-naïve HCV-1b patients without L31/Y93 RAS are planned to receive daclatasvir (60 mg/ day) and asunaprevir (100 mg twice daily) plus weight-based ribavirin (1000-1200 mg/day) for 12 weeks. After treatment they were followed up for 12 weeks. Results: As of 31 Oct 2017, 58 eligible patients are allocated to treatment, with a mean age of 59.3 years and female predominance (67.2%, 39/58). The mean HCV RNA was 5.87+0.77 log10 IU/mL; 23 patients (39.7 %) had significant hepatic fibrosis (>F2). In the modified intention-to-treat analysis, the rate of undetectable HCV at week 1, week 2, week 4, week 8 and endof- treatment was 25 % (14/56), 84.8 % (39/46), 100 % (46/46), 100 % (38/38) and 100 % (27/27), respectively. Undetectable HCV RNA were observed in all of the patients with HCV RNA assessable 4 weeks (SVR4, 18/18) and 12 weeks (SVR12, 12/12) post treatment. None of the 18 patients who completed the 12-week treatment experienced relapse during post-treatment follow-up. The most common adverse event was fatigue (78.3 %), followed by pruritus (65.2 %) and dizziness (52.2 %), of which were considered as ribavirin related. None of the participating subjects withdrew treatment or follow-up throughout the trial peroid. Three serious adverse events were reported which included urosepsis, appendicitis and left ureteral stone. All were unrelated to the investigating drugs. Conclusions: 12 weeks of DCV/ASV plus ribavirin was highly effective and safe in HCV-1b patients without NS5A RAS in the interim analysis. The satisfactory results would be anticipated in the full patient set.

Shih-Chen Lee,Hwai-En Tseng,Chien-Cheng Chang,Yu-Ming Huang 한국정밀공학회 2020 International Journal of Precision Engineering and Vol.21 No.4
In addition to garbage sorting and resource recycling, green design should be a fundamental method for solving environmentalproblems, and design for disassembly is an important foundation of green design. This study focuses on providingquantitative assessment methods for designers’ reference. This study proposes interactive genetic algorithms to solve theproblem of disassembly sequence planning. First, the disassembly factor is measured by the fuzzy scoring procedure method, and then the genetic algorithm is used to select the optimal sequence. With the penalty value provided from the process, areference is provided for the revised design. Finally, examples are discussed to demonstrate that the proposed approach isa feasible method.
Development of Novel Tool Center Point Velocity Planning Algorithm for Five Axis Machine Tool
Shih-Kai Wu,Meng-Shiun Tsai,Ming-Tzong Lin,Hong-Wei Huang 한국정밀공학회 2018 International Journal of Precision Engineering and Vol.19 No.8
In this paper, a tool center point (TCP) feedrate scheduling algorithm for 5-axis machine tool is developed to generate the axes interpolation commands. The proposed algorithm considers not only the constraints of TCP velocity, acceleration and jerk, but also the velocity differences of each axes at the junction of each block. First, the proposed method determines the maximum speed for each block segment at the TCP coordinate based on the computed length. Then the kinematics of the five axis machine tool is employed to derive the five-axis corner velocity difference (FCVD) formulation. The FCVD utilizes the axis velocity difference at the junction of blocks as the designed variable. As the starting and end velocities of each block are determined, the S-shape acceleration/ deceleration (Acc/Dec) method is adopted to generate both smooth TCP and rotary axis profile based on the given interpolation parameters. The servo dynamics of the five axis machine tool are utilized to evaluate the performances of the FCVD. Simulation results demonstrate that the FCVD can achieve better contour accuracy with less machining time as compared to the five-axis feedrate regulation formulation (FFRF) algorithm. Furthermore, the FCVD are compared with Heidenhain CNC controller and the results show that the FCVD has similar behaviors as the Heidenhain controller, but it can achieve less machining time.

Toxic air pollutants are investigated in a petrochemical industrial park in Taiwan by using a movable open path FTIR. The results show the qualitative and quantitative analysis of emission gases from plants, and also provide the emission rate of different compounds. More than twenty compounds under usual operation are found from this industrial park. The concentration variation with time can be correlated exactly with wind direction. It means that by changing the measuring points, the source of emission can be unambiguously identified. An EPA proved PAL model is applied to estimate the emission rate of either a point or an area source. Local atmospheric stability is determined by releasing the $SF_6$ tracer. The origins of errors come mainly from the uncertainty of source's configuration and the variation of meteorological condition. Through the continuous measurement (half an hour base in this study) of OP-FTIR sensor, the maximum value of emission rate and the annual amount of emission can be derived. The emission rate of the measured toxic gases are derived by the model technique and the results show that the emission amount are in the order of ten to hundred tons per year.
Design of a hydraulic anti-lock braking system (ABS) for a motorcycle
Chun-Kuei Huang,Ming-Chang Shih 대한기계학회 2010 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.24 No.5
This work presents a hydraulic anti-lock braking system (ABS) for a motorcycle. The ABS has a hydraulic modulator and an intelligent controller. The hydraulic modulator is analyzed, and then equipped on a scooter for road tests. The intelligent controller controls the hydraulic modulator by estimated vehicle velocity to calculate the slip ratio of the wheels in real time. The performance of the hydraulic modulator and intelligent controller are assessed by the hardware-in-the-loop (HIL) simulations and road tests. In HIL simulation, the ABS is tested for different initial braking velocities on roads with different adhesive coefficients. Furthermore, both HIL simulations and road tests are conducted on a one-phase pavement road and three-phase pavement road.

Purpose: Cancer cells develop acquired resistance induced by chemotherapeutic drugs. In this study, we investigated the effects of brief treatment with cytotoxic drugs on the phenotype of breast cancer cells. Methods: Breast cancer cells MCF7 and BT-474 were briefly treated with paclitaxel or doxorubicin. Clonogenic, migration, and invasion assays were performed on the treated cells. Western blot analysis and RhoA activity assay were also performed. Results: Breast cancer cells when briefly treated with paclitaxel or doxorubicin showed reduced clonogenic ability. Doxorubicin, but not paclitaxel, augmented cell migration and invasion. The invasion-promoting effects of doxorubicin were lost when the two drugs were sequentially used in combination. Myosin light chain (MLC) 2 phosphorylation and RhoA activity were upregulated by doxorubicin and downregulated by paclitaxel. Pretreatment with RhoA inhibitors abolished the migration- and invasion-promoting effects of doxorubicin. Conclusion: Doxorubicin activates the RhoA/MLC pathway and enhances breast cancer cell migration and invasion. Therefore, this pathway might be explored as a therapeutic target to suppress anthracycline-enhanced tumor progression.

Background and ObjectivesAge is a traditional risk factor for open-heart surgery. The efficacy and safety of transcatheter edge-to-edge mitral valve repair, using MitraClip (Abbott Vascular), has been demonstrated in patients with severe mitral regurgitation (MR). Since octogenarians or older patients are usually deferred to receive open-heart surgery, the main interest of this study is to elucidate the procedural safety and long-term clinical impact of MitraClip in elderly patients. MethodsPatients with symptomatic severe MR were evaluated by the heart team. For those with high or prohibitive surgical risks, transcatheter mitral valve repair was performed in hybrid operation room. Transthoracic echocardiography (TTE), blood tests, and six-minute walk test (6MWT) were performed before, 1-month, 6-months, and 1 year after index procedure. ResultsA total of 46 consecutive patients receiving MitraClip procedure were enrolled. Nineteen patients (84.2±4.0 years) were over 80-year-old and 27 (73.4±11.1 years) were younger than 80. Compare to baseline, the significant reduction in MR severity was achieved after the procedure and sustained. All the patients benefited from significant improvement in New York Heart Association functional class. The 6-minute walk test (6MWT) increased from 259±114 to 319±92 meters (p=0.03) at 1 year. The overall 1-year survival rate was 80% in the elderly and 88% in those <80 years, p=0.590. Baseline 6MWT was a predictor for all-cause mortality (odds ratio, 0.99; 95% confidence interval, 0.982–0.999; p=0.026) after the MitraClip procedure. ConclusionsTrans-catheter edge-to-edge mitral valve repairs are safe and have positive clinical impact in subjects with severe MR, even in advanced age.

Transient luminous events (TLEs; sprites, elves, jets and etc.) are lightning-related optical flashes occurring above thunderstorms. Since the first discovery of sprites in 1989, scientists have learned a great deal about the morphological, spectroscopic and electromagnetic characteristics of TLEs through ground and spacecraft campaigns. However, most of the TLE studies were based on events recorded over US High Plains. To elucidate the possible biasing effects, space-borne observations are needed and have their merits. Imager of sprites and Upper Atmospheric Lightning (ISUAL) on the FORMOSAT-2 satellite is the first instrument to carry out a true global measurement of TLEs from a low- earth orbit. In this short paper, we apply a common astronomical data analysis technique, two-color diagram, on the ISUAL spectrophotometer (SP) data. By choosing appropriated bandpasses and converting the measured flux of TLEs into the unit of magnitude, two-color diagrams of TLEs can be constructed. We demonstrate that two-color diagrams, which were constructed from the narrow-band spectrophotometer data, can be used to classify different types of TLEs and trace their temporal evolution. The amount of reddening due to Earth's atmosphere can also be estimated from two-color diagrams assembled from the broad-band spectrophotometer data.
