Background: Pruritis caused by atopic dermatitis (AD) is not always well controlled by topical corticosteroid therapy, but use of tacrolimus often helps to soothe such intractable pruritis in clinical settings. Objective: To determine the anti-pruritic efficacy of topical tacrolimus in treating AD in induction and maintenance therapy. Methods: Prior to the study, patients were randomly allocated into two groups, induction therapy followed by tacrolimus monotherapy maintenance, and induction therapy followed by emollient- only maintenance. In the induction therapy, the patients were allowed to use topical tacrolimus and emollients in addition to a low dose (<10 g/week) of topical steroids. Patients showing relief from pruritis were allowed to proceed to maintenance therapy. Recurrence of pruritis in maintenance therapy was examined as a major endpoint. Results: Two-thirds of patients (44/68; 64.7%) showed relief from pruritis after induction therapy. Pruritis recurred in 23.8% (5/21) of the tacrolimus monotherapy group and in 100% (21/21) of the emollient group during maintenance period, a difference that was statistically significant. Conclusion: Use of topical tacrolimus is effective in controlling pruritis of AD compared to emollient. (Ann Dermatol 24(2) 144∼150, 2012)

The present study examined whether light emitted by long-afterglow phosphorescent pigments (LumiNova) would stimulate gonadal development in fish during the nonbreeding season. Pellets containing LumiNova powder (treatment group) were prepared and placed on the calvaria of specimens of the sapphire devil Chrysiptera cyanea, a reef-associated damselfish that requires long days for gonadal recrudescence. A pellet without LumiNova powder was placed on the calvaria of the control fish (control group). Fish were reared at 26°C under a light.dark cycle (12 h photophase, 12 h scotophase; LD 12:12) for 4 weeks. No difference in the gonadosomatic index (GSI) or ovarian histology was observed among the control, sham-operation, and treatment groups 1 week after the start of the experiment. After 4 weeks, the GSI of the control and sham-operation groups remained at low levels, and ovaries contained immature oocytes at the perinucleolus stage. In contrast, the treatment group exhibited significantly higher values of GSI as well as developed ovaries with fully vitellogenic oocytes. These results demonstrated that long-day conditions were produced by light emitted from the LumiNova pellets, thus stimulating ovarian development in the damselfish. Therefore, long-afterglow phosphorescent pigments can be used as an alternative to standard light sources for purposes of artificial stimulation of gonadal development in fish.

<P><B>Abstract</B></P> <P>This study investigated how silver nanoparticles (AgNP) and ionic silver (AgNO<SUB>3</SUB>) undergo phase-transformations in soils under aerobic and anaerobic conditions using extended X-ray absorption fine structure (EXAFS) spectroscopy. After 30 days of aerobic incubation, 88% of AgNP added to the soil remained persistent, whereas AgNO<SUB>3</SUB> was completely transformed into Ag associated with humus and clay minerals. In the anaerobic soil, 83% of the spiked AgNP was transformed into Ag<SUB>2</SUB>S, accompanied by significant decrease in water- and acid-extractable Ag fractions. About 50% of AgNO<SUB>3</SUB> spiked to the anaerobic soil underwent transformations into metallic Ag and associations with clay minerals. Oxide (Ag<SUB>2</SUB>O) and carbonate (Ag<SUB>2</SUB>CO<SUB>3</SUB>) forms of Ag were not predominant in aerobic and anaerobic soils. Recent Development of Techniques and Devices in Colorectal Endoscopic Submucosal Dissection

Colorectal endoscopic submucosal dissection (ESD) is now a well-established endoscopic treatment for early-stage colorectal neoplasms, especially in Asian countries, including Japan. Despite the spread of colorectal ESD, there are still situations in which achieving successful submucosal dissection is difficult. Various novel techniques and devices have been developed to overcome these difficulties, and past reports have shown that some of these strategies can be applied to colorectal ESD. We review several recent developments in the field. The techniques reviewed include the pocket creation method and traction methods and the devices reviewed include the overtube with balloon and electrosurgical knives with water-jet function. These improved techniques and devices can facilitate safer, more reliable ESDs and expand its applicability and acceptability all over the world.

The antigen binding to the B cell receptor (BCR) induces a rise in cytoplasmic Ca2+ and initiates cell responses such as apoptosis, proliferation, and differentiation in B lymphocytes. We previously showed that the inhibition of mitochondrial Na+-Ca2+ exchange (NCXm) by 7-chloro-5- (2-chlorophenyl)-1,5-dihydro-4,1-benzothiazepin-2(3H)-one [CGP-37157] dose-dependently reduced BCR-mediated rise in cytoplasmic Ca2+ in DT40 B lymphocytes. However, the effect of CGP-37157 on BCR-mediated apoptosis is not yet fully understood. Here, we studied the effect of NCXm inhibition on BCR-mediated apoptosis in DT40 B lymphocytes. Apoptotic cells (Annexin V+ and PI−) were increased by the BCR-stimulation while the treatment of cells with 20-μM CGP-37157 attenuated the apoptosis induction. On the other hand, 24-hr treatments with 0.2 − 20 μM CGP- 37157 without the BCR stimulation did not affect the apoptotic-cell population. In the NCLX (a gene encoding NCXm) knockout cells, although the apoptotic cells were increased in the control, the appearance of late apoptotic cells (Annexin V+ and PI+) was decreased by BCR stimulation when compared to the wild-type cells. Our findings suggest that the inhibition of NCXm attenuates BCR-mediated apoptosis in DT40 B lymphocytes.
