AtBAG6, a novel calmodulin-binding protein, induces programmed cell death in yeast and plants

Calmodulin (CaM) influences many cellular processes by interacting with various proteins. Here, we isolated AtBAG6, an Arabidopsis CaM-binding protein that contains a central BCL-2-associated ethnogeny (BAG) domain. In yeast and plants, overexpression of AtBAG6 induced cell death phenotypes consistent with programmed cell death (PCD). Recombinant AtBAG6 had higher affinity for CaM in the absence of free Ca^2+ than in its presence. An IQ motif(IQXXXRGXXXR, where X denotes any amino-acid) was required for Ca^2+ -independent CaM complex formation and single amino-acid changes within this motif abrogated both AtBAG6-activated CaM-binding and cell death in yeast and plants. A 134-amino-acid stretch, encompassing both the IQ motif and BAG domain, was sufficient to induce cell death. Agents generating oxygen radicals, which are known to be involved in plant PCD, specifically induced the AtBAG6 transcript. Collectively, these results suggest that AtBAG6 is a stress-upregulated CaM-binding protein involved in plant PCD.

Recently, wave propagation imaging based on laser scanning-generated elastic waves has been intensively used for nondestructive inspection. However, the proficiency of the conventional software based system reduces when the scan area is large since the processing time increases significantly due to unavoidable processor multitasking, where computing resources are shared with multiple processes. Hence, the field programmable gate array (FPGA) was introduced for a wave propagation imaging method in order to obtain extreme processing time reduction. An FPGA board was used for the design, implementing post-processing ultrasonic wave propagation imaging (UWPI). The results were compared with the conventional system and considerable improvement was observed, with at least 78% (scanning of 100 × 100 ㎟ with 0.5 ㎜ interval) to 87.5% (scanning of 200 × 200 ㎟ with 0.5 ㎜ interval) less processing time, strengthening the claim for the research. This new concept to implement FPGA technology into the UPI system will act as a break-through technology for full-scale automatic inspection.

Protein O-phosphorylation often occurs reciprocally with O-GlcNAc modification and represents a regulatory principle for proteins. O-phosphorylation of serine by glycogen synthase kinase-3β on Snail1, a transcriptional repressor of E-cadherin and a key regulator of the epithelial-mesenchymal transition (EMT) programme, results in its proteasomal degradation. We show that by suppressing O-phosphorylation-mediated degradation, O-GlcNAc at serine112 stabilizes Snail1 and thus increases its repressor function, which in turn attenuates E-cadherin mRNA expression. Hyperglycaemic condition enhances O-GlcNAc modification and initiates EMT by transcriptional suppression of E-cadherin through Snail1. Thus, dynamic reciprocal O-phosphorylation and O-GlcNAc modification of Snail1 constitute a molecular link between cellular glucose metabolism and the control of EMT.

This might be the second report of a arrhenoblastoma in Korea. A 20 year old unmarried Korean woman was admitted to this hospital on Aug. 20, 1966, because of secondary amenrrhea, male type of trichosis(around the genital area and lower extremity), lower and deeper as male voice, many pimples, mamatrophy, prominent Adam`s apple etc. since 17 year old, and she also felt a mass in her lower abdomen recently. By physical examination, a hard mass was palpable as big as foetus head size in the right lower quadrant region, and the clitoris enlarged about 3.5 cm in length. A laparatomy was performed for removing the right ovaian tumor. General view of the tumor was yellowish-gray, glistening, uneven surface, and partially cystic oval form child head size. Micoscopically the tumor showed a mixed picture of tubular-adenomatous and sarcomatoid type (arrhenoblastoma). The post operative course was not complicated, her menses resumed regularly and pimples gradually disappeared 33 days and 2 months after operation respectively. The other refeminizing appearence were also seen steadily 5 months folling the surgical operation.

Redox-sensitive Transcriptional Regulators from Streptomyces coelicolor

We are investigating the regulation and the role of several oxidative defense enzyme systems in the growth and differentiation of the antibiotic-producing, Gram-positive bacterium Streptomyces coelicolor A3(2). Genes for catalase A(catA; monofunctional, vegetative catalase}, catalase B(catB; monofunctional, stationary phase specific), catalase C(catC; catalase-peroxidase, late exponential phase-specific), Fe-containing SOD(sodF), and Ni-containing SOD(sodN) have been examined for their regulation and the role in the growth and differentiation of S coelicolor. Transcriptional regulators governing the regulation of these genes in. response to thiol-oxidation(RsrA/SigR), nickel(SodR1), hydrogen peroxide(CatR, OxyR), osmolarity(RsbA/SigB), or others(FurA) were found and partially characterized. SigR(Φ^(R)) is a sigma factor responsible for inducing the thioredoxin system in response to oxidative stress in S coelicolor. A redox-sensitive, Φ^(R)-specific anti-sigma factor, RsrA, was identified. It binds <DR and inhibits Φ^(R)-directed transcription in vitro only under reducing conditions. Exposure to H₂O₂ or to the thiol-specific oxidant diamide caused the dissociation of the Φ^(R)-RsrA complex, thereby allowing Φ^(R)-dependent transcription. This correlated with intramolecular disulfide bond formation in RsrA. Thioredoxin was able to reduce oxidized RsrA, suggesting that Φ^(R), RsrA and the thioredoxin system comprise a novel feedback homeostasis loop that senses and responds to changes in the intracellular thiol-disulfide redox balance. Another redox-sensitive regulator CatR was identified as a repressor for catA gene expression encoding catalase A. The catR gene was isolated as a genetic locus whose mutation caused overproduction of catalase A. The catR gene encodes a 138 amino acids protein(15319 Da) showing homology to Fur proteins of other bacteria. Derepression of transcription from catRp1, catRp2, and catA promoters was observed within 10 min of H₂O₂ treatment, suggesting H₂O₂-mediated modification of CatR activity. Detailed mechanisms for the redox-dependent modulation of these transcriptional regulators are under investigation.
