        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • Genetic Diversity of Korean Rice Breeding Parents as Measured by DNA Fingerprinting with Simple Sequence Repeat (SSR) Markers

        Molecular markers are useful tools for evaluating genetic diversity and determining cultivar identity. Present study was conducted to evaluate the genetic diversity within a diverse collection of rice accessions used for Korean breeding programs. Two hundred eighty-seven rice cultivars, composed of temperate japonica, tropical japonica, indica, and Tongil-type of Korean crossing parents were evaluated by means of 15 simple sequence repeat (SSR) markers. A total of 99 alleles were detected, and the number of alleles per marker ranged from 4 to 11, with an average of 6.6 per locus. Polymorphism information content (PIC) for each of the SSR markers ranged from 0.2924 to 0.8102 with an average of 0.5785. These results, with the result that use of only 15 SSR markers made all rice cultivars examined could be uniquely distinguished, imply the efficiency of SSR markers for analysis of genetic diversity in rice. Cluster analysis was performed on similar coefficient matrics calculated from SSR markers to generate a dendogram in which two major groups corresponding to japonica (Group I) and indica and Tongil type rice (group II) with additional subclasses within both major groups. The narrowness of the Korean breeding germplasm was revealed by the fact that most of the Korean-bred and Japan-bred temperate japonica cultivars were concentrated into only 2 of the sub-group I-1 (143 cultivars) and I-2 (58 cultivars) among six sub-groups in major group of japonica. This is because of the japonica accessions used in this study was a very closely related ones because of frequent sharing of the crossing parents with similar genetic background with synergy effect of the inherited genetic difference between indica and japonica. A rice breeding strategy with the use of molecular markers was discussed for overcoming of genetic vulnerability owing to this genetic narrowness.

        1992년 첫 삽을 뜨면서 시작한 경부고속철도건설사업이 2004년 4월, 1단계 개통을 한데 이어 지난 2010년 11월 완전 개통하였다. 경부고속철도 개통으로 경부축 지역의 2시간대 서비스 제공을 통한 국토의 균형발전 촉진 및 여가·관광사업 활성화, 기술발전, 환경비용 및 에너지 절감 등 긍정적인 효과를 이룬 반면, 잦은 기본계획 변경으로 인한 사업비 및 사업기간의 증가, 현실과 맞지 않은 설치기준으로 인한 과잉설계 문제 등 부정적 효과도 초래했다. 본 연구를 통해 사업추진 단계별 주요 이슈에 대해 객관적인 시각으로 문제점을 분석하고 시사점을 도출함으로써, 현재 추진하고 있는 호남고속철도와 수도권고속철도 사업에서는 시행착오를 줄이고 사업을 효율적으로 추진하는데 활용하고자 한다. Launched in 1992, the Gyeung-Bu High Speed Railway (HSR) was fully opened in November 2010 following the partial opening of the first phase in April 2004. Since the opening it has made tremendous achievements, including but not limited to, technical innovation, balanced regional development, growth of leisure and tourism industries, and savings of environmental costs and energy. On the other hand, it has brought negative effects such as increase in project time and cost by frequent changes of the basic plan and over-design based on unrealistic criteria. In this study we will analyze major issues that occurred at each stage of the Gyeong-Bu HSR project and draw implications. Thus, this study will enhance to efficiently implement the Ho-Nam and Seoul Metropolitan HSR project through minimizing possible trials and errors.

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

        Protective Effect of Heme Oxygenase-1 on High Glucose-Induced Pancreatic β-Cell Injury

        <P><B>Background</B></P><P>Glucose toxicity that is caused by chronic exposure to a high glucose concentration leads to islet dysfunction and induces apoptosis in pancreatic β-cells. Heme oxygenase-1 (HO-1) has been identified as an anti-apoptotic and cytoprotective gene. The purpose of this study is to investigate whether HO-1 up-regulation when using metalloprotophyrin (cobalt protoporphyrin, CoPP) could protect pancreatic β-cells from high glucose-induced apoptosis.</P><P><B>Methods</B></P><P>Reverse transcription-polymerase chain reaction was performed to analyze the CoPP-induced mRNA expression of HO-1. Cell viability of INS-1 cells cultured in the presence of CoPP was examined by acridine orange/propidium iodide staining. The generation of intracellular reactive oxygen species (ROS) was measured using flow cytometry. Glucose stimulated insulin secretion (GSIS) was determined following incubation with CoPP in different glucose concentrations.</P><P><B>Results</B></P><P>CoPP increased HO-1 mRNA expression in both a dose- and time-dependent manner. Overexpression of HO-1 inhibited caspase-3, and the number of dead cells in the presence of CoPP was significantly decreased when exposed to high glucose conditions (HG). CoPP also decreased the generation of intracellular ROS by 50% during 72 hours of culture with HG. However, decreased GSIS was not recovered even in the presence of CoPP.</P><P><B>Conclusion</B></P><P>Our data suggest that CoPP-induced HO-1 up-regulation results in protection from high glucose-induced apoptosis in INS-1 cells; however, glucose stimulated insulin secretion is not restored.</P>

        Dynamic job analysis effectively links vertical job analysis with its horizontal component. Dynamic job analysis can be used to obtain the systematic information that is necessary for the performance of future jobs, as well as current ones, and to design future organizations and personnel management systems. Dynamic job analysis is composed of three stages. In the first stage, the structure of current jobs can be systematically clarified through vertical analysis. In the second stage, the horizontal linkage of jobs can be obtained through horizontal job analysis, and desirable future job process can be designed by analyzing current problems. In the third stage, vertical and the horizontal analyses are combined together. At this stage, the effect of the new job process designed by horizontal analysis upon the current vertical structure of job is measured, and systematic information is obtained on job characteristics and the qualifications of workers for future jobs These three stages provide a foundation on which new processes and organizational structures for more competitive enterprises can be designed. The concrete procedure of combining vertical and the horizontal analyses is as follows. First, impact assessment is made to analyses the effect of new processes on existing organizations or the performance of workers involved in the concerned jobs. Second. Modifications in task are described by checking what changes the workers in charge of a job undergo after the introduction of the new process. In the final stage, the necessary information for future jobs is drawn up by describing performance methods and workers qualifications for the jobs.

        동기적으로 적응적인 컴퓨터 보조수업 : 개념과 미래 방향 Concepts and Future Directions

        학습자의 동기는 학습도중에 변화한다. 그러나 학습자의 동기변화에 적응적인 CAI를 설계하고 개발하려는 실제 노력이 시작된 것은 비교적 최근의 일이다. 즉, 개별화 수업을 제공하려는 노력과 좀더 동기유발적인 CAI를 제공하려는 노력은 이제 동기적으로 적응적인 CAI를 설계하려는 노력으로 발전하고 있다. 하지만 이러한 노력이 성공하기 위해서는 4가지 이슈에 대한 연구와 논의가 필요하다:1) 학습동기를 실제적으로 정의하기, 2) 학습동기를 측정하기, 3) 동기유발전략을 체계화하기, 4)이론과 경험적으로 검증된 통합적 모델을 구축하기, 본 논문에서는 이러한 이슈들을 고려하여 최근의 노력들을 살펴보면서 동기적으로 적응적인 CAI의 발전방향을 탐색한다. Learners motivation changes during their learning with computer - assisted instruction. However, it is quite recent that some efforts have been made to design and develop CAI which is adaptive to learners motivational change. The efforts to provide more individualized instruction and the efforts to design more motivating CAI are now heading in the same direction toward designing a motivationally adaptive CAI. The purposes of this article are to introduce the idea of motivationally adaptive computer-assisted instruction( MACAl) and to present the issues to be solved in order to develop a functioning protytype. The following four issues are pointed out. 1. Need for practical definition of motivation to learn. 2. Measuring motivation to learn. 3. Comprehensive list of motivational strategies. 4. An integrated model validated in theory and practice.

        한국 및 러시아 한국계 청소년 우울증의 사회심리적 위험요인에 대한 횡문화 비교 연구

        연구목적 : 본 연구는 사회문화적 배경이 다른 한국과 러시아, 두 나라의 한국인 청소년을 대상으로 우울증의 유병율을 비교 조사하고, 우울증과 관련된 사회심리적 위험요인들을 알아보고자 하였다. 방 법 : 연구도구의 한국어판을 제작하고 두 나라의 청소년 우울증 환자 40명씩을 대상으로 우울척도의 cut-off 점수를 산정하기 위한 예비연구를 본 연구에 앞서 시행하였다. 한국의 청소년 대상은 서울 소재의 중고등학생 838명이었고, 러시아의 청소년은 우즈베키스탄 공화국의 중고등학생 중 양부모가 모두 한국인인 652명이었다. 연구도구로는 우울증의 사회심리적 요인들과 횡문화적 요인들을 조사하기 위하여 기초자료 설문지 및 Beck 우울척도를 적용하였다. 결 과 : 우울증 환자를 대상으로 시행한 예비연구에서 얻은 Beck 우울척도의 cut-off 점수 19점 이상을 기준으로 하였을 때, 한국 청소년 집단에서 우울증의 유병률은 26.5%인데 반하여 러시아 청소년에서는 7.4%의 유병률을 보였다. 한국인 청소년들은 한국인-러시아 청소년에 비하여 염세주의(pessimism), 실패감(failure), 자살사고, 죄책감 등, 절망감(helplessness)과 관련된 증상들이 유의하게 많이 나타났다. 한국인 청소년과 한국인-러시아 청소년 간에 우울증의 유의한 사회심리적 위험요인들로서 여성, 낮은 학업성적, 부모와의 갈등이 공통적으로 나타났으며, 그외에 친구교제 요인, 만성 신체질환, 형제간이나 친구들과의 갈등이 한국인 청소년에서는 또 다른 위험요인으로 존재하였다. 결 론 : 본 연구를 통하여 한국인 청소년의 우울증 유병율이 한국인-러시아 청소년의 유병률에 비하여 유의하게 높았으며, 한국과 러시아의 한국인 청소년들 간에 우울증의 유의한 사회심리적 위험요인들이 차이가 있음을 시사한다. Objectives : In order to study a cross-cultural epidemiology of the adolescent depressives with Korean ethnicity in Korea and Russia, we compared psychosocial characteristics and risk factors of depressive disorder between two school-based populations in different sociocultural environments. Methods : We measured psychosocial variables of 838 subjects in Korea, and 652 in Russia(Uzbekitan) using Basic Questionnaires and Beck Depression Inventory(BDI). Results : The cut-off point of BDI for adolescent depression in Koreans was 19 points. Higher prevalence of depression was noted in Korean adolescents(26.5%) compared to Korean-Russian adolescents(7.4%). We found signicant risk factors of adolescent depression in two study samples : female, academic failure, meeting friends less frequently, presence of chronic physical illness, interpersonal problems with parents, and same-sex and opposite-sex friends. Conclusions : Our data will provide information on cross-cultural understanding of psychosocial characteristics and risk factors in adolescent depression compared between Koreans and Korean-Russians.

        뮤코다당증(Mucopolysaccharidosis)환아의 치은 증식

        뮤코다당증(Mucopolysaccharidosis, MPS)은 gIycosaminoglycans (GAGs)의 분해에 필요한 라이소좀 효소(lysomal enzymes)의 결함으로 GAGs의 대사 산물이 세포의 라이소좀내에 축적되어 점차 세포와 조직, 기관의 기능 이상을 초래하고 신체 질환과 정신 발달 지연을 가져오는 질환이다. 본 증례는 심각한 치은 증식을 가진 뮤코다낭증 환아를 전신 마취 하에 치은 절제술을 시행한 치험례이다. 소아치과 의사는 뮤코다당증 환아의 구강내 증상과 치과 치료시 주의 사항에 대해 숙지해야 하고, 내과의와 연계하여 환아의 전신 상태 변화에 민감하게 대처하여 치과 치료를 시행해야 한다. Mucopolysaccharidosis (MPS) is a disorder of storage in which there is excessive accumulation of glycosaminoglycans(GAGs) from lysosomal enzyme defect. Lysosomal accumulation of GAGs eventually results in cell, tissue and organ dysfunction. This patient may manifest mental retardation and physical disorders. This clinical report presents a girl with MPS having severe gingival hyperplasia. Gingivectomy was performed under general anesthesia. The pediatric dentist must be aware of oral manifestations present in the MPS. The approach to dental management will require teamwork between the dentist and the patient' s physician.
