        축양 멸치의 월동시험

        1971년 11월 초에 어획하여 22일간 蓄養한 활멸치를 12월 1일부터 익년 3월 31일까지 4개월간 慶南 忠武市 仁坪洞 地先 海上에서 越冬試驗한 結果, (1) 試驗期間中 水溫은 最低 7.2℃까지 下降했으나, 이 水溫의 下降으로 인한 폐사는 일어나지 않았다. 그러나 시험의 初期, 蓄養槽의 移動 직후부터 9일간에 약 1.3%, 또 15일 후부터 20일간에 약 1.7%의 폐사가 일어났다. 前者의 폐사 原因은 蓄養槽의 移動에 의한 것이며, 後者는 蓄養槽의 綱地交替로 인한 것이다. (2) 越冬可能水城은 水溫, 潮流, 水深 등을 고려할 때 巨齊島 南 및 南西部, 忠武市 南 및 南西部부터 南海島 東, 南, 西部, 突山島 以東의 海域이 適地로 생각되며, 鎭海灣內는 水溫下降이 심하여 좋지 않다고 추정된다. (3) 肥滿度는 試驗 初期 약 두 달간은 낮아지는 경향이 있었으나 그 후 試驗初에 비하여 20%정도까지 증가했다. 이것은 멸치가 蓄養槽 안에서의 생활과 低水溫에 적응되어 攝餌가 왕성해진 것에 기인한다고 보아진다. Since no medium-sized anchovies for skipjack baits are caught during the winter season in the Korean waters, the demand for the live baits in the winter season is met with crawl-stocked anchovies. Authors conducted a test on the wintering of the anchovies which were caught in the early November anb acclimated for 22days in a crawl. The wintering crawl was kept in the inlet near Chungmu City, which is situated in the southern coast of Korea, for 4 months through December 1, 1971 to March 31, 1972. 1. The lowest water temperature of 7.2℃ recorded on March 1, caused no significant mortality of the wintering anchovies. However, two operations, moving the crawl and repacling the crawl webbing, brought about a considerable mortality. The mortality of the anchovies, due to the crawl moving to the wintering place two kilometers away from the transient acclimating place, amounted to 1.5 percent in 9 days following the crawl moving. The replacement of the fouled crawl webbing with a new one also caused a mortality amounting to 1.7 percent in 5 days. 2. It is judged that the favorable wintering waters lie in the south and southeast coasts of Keoje Island, the south and southwest coast of Chungmu City, coasts around Namhae Island, and the east coast of Dolsan Island. Jinhae Bay is not suited for tha wintering because of its low water temperature in winter. 3. The winterng anchovies were fed, once a day through the test period, with 4 ㎏ of finely minced fresh anchovy for 57days in the early period, and with 2㎏ of compound pellet fish food for the rest of the period. Fatness coefficient of the wintering anchovies showed a slight increase in the latter half of the period.

        한우 및 유우에서의 indocyanine green 배설시험 및 혈장효소 활성치의 변화

        This experiment was carried out to establish a proper method of indocyanine green(ICG) excrection test for a applicable liver function test in three Korean native cattle average weighing about 450kg and dairy cattle parity of 3~5. The results obtained the half life($T^1/_2$), fractional clearance rate(KICG), retention rate and plasma enzyme activities before or after injection of ICG were as follows. 1. The maximum absorbance of ICG in plasma was at 805nm. 2. Average half life and fractional clearance rate following the injection of ICG 0.25mg/kg body weight were $5.53{\pm}1.27$ minute and $0.131{\pm}0.031$/minute in Korean native cattle, $4.55{\pm}0.68$ minute and $0.156{\pm}0.031$/minute in dairy cattle, respectively. The ICG removal rate was exponentially liner for the first 15 minutes after injection both of Korean native cattle and dairy cattle. 3. Average plasma retention rate when 10, 15, 30 minutes after injection was $35.7{\pm}13.9$, $23.2{\pm}7.1$, $10.8{\pm}3.5%$ in Korean native cattle, $26.8{\pm}3.3$, $14.2{\pm}1.2$, $5.5{\pm}2.2%$ in dairy cattle, respectively. 4. Plasma enzyme activities(AST, ALT, r-GTP) were no variation among the before, during and after injection of ICG. From these results, ICG excretion test to cattle is applicable to evaluation of liver funtion in both clinical and research, and adopted the 15 minutes plasma sample as the sample taken at the ideal time for comparative purposes.

        Progesterone 측정을 위한 액상(液相) 효소면역측정법(酵素免疫測定法)의 최적조건에 관한 연구

        This study was carried out to develop an effective liquid-phase double antibody enzyme immunoassay for determining of progesterone. The optimum conditions of assay system, 1st and 2nd antibodies, enzyme conjugate, and time reaction were invested. The bovine plasma progesterone level in dairy cattle and korean native bulls were also analyzed. The results obtained were as follows; 1. The reproducibility of petroleum ether was superior to that of ethyl ether as extract solvent of progesterone in plasma. 2. The optimum dilution rate of 1st and 2nd antibody was 30,000 and 10 times, respectively. Affer the reaction of enzyme conjugate to progesterone 1st antibody, and then 2nd antibody competition reaction was enough for over 1hr. 3. Average plasma progesterone level in 4 pregnant and 9 nonpregnant Holstein was $2.5{\pm}0.5$ and $0.7{\pm}0.2ng/m{\ell}$, respectively. Average plasma progesterone level of 10 Korean native bulls was $0.1{\pm}0.001ng/m{\ell}$ From these results, by using liquid phase double antibody enzyme immunoassay for progesterone is applicable to detect of early pregnancy diagnosis, factorial analysis of reproductive disorder, and also reproductive physiological function such as monitoring of cyclicity during the post-partum period.

        개에서의 indocyanine green 배설시험 및 혈장효소 활성치의 변화

        This experiment was carried out to establish a proper method of indocyanine green(ICG) excretion test for a applicable liver function test in dogs. The half life(T1/2), fractional clearance rate(KICG) and retention rate after injection of ICG with or without administred carbon tetrachloride($CCl_4$) were also invested. The results obtained were as follows ; 1. The maximum absorbance of ICG in plasma was at 810nm. 2. Half life and fractional clearance rate when administered 0.25 and 0.50mg of ICG per Kilogram body weight were $6.33{\pm}0.58$ minutes and $0.11{\pm}0.99$/minute in the former, $10.01{\pm}1.0$ minutes and $0.07{\pm}0.007$/minute in the latter, respectively. The ICG removal rate was exponentially linear for the first 15 minutes after injection both, of 0.25 and 0.50mg of ICG. 3. One day following the administration of $0.0042m{\ell}\;CCl_4$ kilogram body weight which injected 0.50mg of ICG, half life was more longer and fractional clearance rate was significantly reduced than that of ICG single injection. 4. Plasma retention rate when 15, 30, 45 minutes after injection dose of 0.25 and 0.50mg ICG per Kilogram body weight, $14.7{\pm}4.8$, $5.1{\pm}3.1$, $2.6{\pm}1.6%$ in the former, $26.9{\pm}1.8$, $11.1{\pm}2.4$, $4.8{\pm}1.3%$ in the latter, respectively. However, after administration of $CCl_4$, plasma retention rate of ICG at a dose of 0.50mg, it was $39.3{\pm}0.9$, $16{\pm}2.9$, $10.7{\pm}0.1%$, respectively. 5. Plasma enzyme(AST, ALT, r-GTP) activities administered with $CCl_4$ were increased, but there was no change which injected any dose of single ICG injection. From these results, ICG excretion test to dog is applicable to evaluation of liver function in both clinical and research.

        멸치 축양의 예비시험

        定置綱과 들망에 의해 어획된 멸치를 蓄養하여 魚體 크기, 漁獲方法, 移送方法등에 따른 生殘率의 차이를 조사하였다. 1. 中形 멸치 (全長 7.8∼9.6㎝, 體重 3.6∼4.8g)를 20∼25일간 축양한 生殘率이 70∼92%였음에 비하여, 小形 멸치 (全長 5.2㎝, 體重 1.0g)는 어획후 12시간 동안의 生殘率이 16%였다. 2. 어획된 멸치를 運搬槽에 수용하여 蓄養場까지 移送할 때, 運搬槽를 曳引한 距離가 멀수록, 또 務送速力이 빠를수록 蓄養중의 生殘率이 낮다. 3. 定置網과 들망으로 어획된 멸치가 다 같이 상당히 좋은 생잔율을 보였으나, 이 두 어법 사이의 생잔율의 차이는 이번 試驗의 결과로는 단정할 수 없다. Preliminary creel-holding test of live anchovies which are to be used as bait for the skipjack fishery, was conducted on the fish caught by set net and lift net. They were held in the creels constructed with bamboo frame and minnow netting, during the period of Oct. 24-Nov. 28 in 1972. It was found that the survival rate of the creel-held anchovies varied by the size of fish, the towed distance and speed of fish-carrying creel in which the fish were accomodated after catchi. 1. The survival rate of the medium size anchovies, 7.8-9.6㎝ in length and 4.6-4.8 g in weight, was 70-92% during the holding period of 20-35 days. Whereas, only 16% of juvenile anchovies, 5.2㎝ in length and 1.0 g in weight, were survived for 12 hours after catch. 2. During the time of transport, the farther and faster the creel containing fish were towed, the less fish were survived in the subsequent holding period. 3. Though both the set net and lift net could be fairly used to catch live anchovy, no decision between these two gears could be made in this experiment to determine the superiorness for the subsequent survival rate.

        기선기현강의 연구 1, 재래식 어구의 유구저황과 그물꼴에 관하여

        자루 입구의 둘레가 력자망지 150골(75m)되는 권현망어구의 $\frac{1}{10}$ 크기의 모형을 제작하여, 어구의 유분저항과 린망중의 그물꼴에 관하여 실험한 결과 다음과 같은 결론을 얻었다. 1. 모형 어구로부터 환산된 실물어구의 유분저항은 (R은 kg, v는 m/sec) 전저항 $$R_1=30,000\;v^{1.2}\;(0.2{\leqq}v{\leqq}1.0)$$ 자루 저항 $$R_2=16,000\;v^2\;(0.2{\leqq}v{\leqq}0.6)$$ 2. 예망중의 그물꼴은 (1) 오비기(extension wing) 발줄의 전 길이 중, 앞 끝부터 약 $70\%$ 까지는 직선상으로 깊어지며, 해저에 나란한 부분은 안쪽 약 $30\%$ 정도이다. (2) 수비(inside-wing)의 마깥 언저리는 예망속도가 0.2 m/sec이상 되면, 자루와 수비의 연결부 보다 더 뒤로 쏠려서, 어군을 자루로 유도하는 데 합리적인 형상이 아니다. 이것을 시정하기 위해서는, 수비의 깊이 방향의 코스를 $\frac{1}{2}\~\frac{2}{3}$ 정도로 줄이거나, 또는 그물코의 크기를 그런 정로로 줄임과 동시에, 자루 입구에서 오비기 발줄로 가는 힘줄을 넣어서 수비에는 예망장력이 적게 미치도록 하는 것이 좋다고 생각된다. (3) 앞치마(lower bosom)는 예망속도가 0.2m/sec이상 되면, 뒤끝치 들어 올려져서 수심이 깊은 어장에서는 수비에까지 유도된 어군도 이리로 해서 도피할 우려가 있다. (4) 문턱(upper bosom)의 계관 상의 경사 각도는 $35\~40^{\circ}$이다. 이것은 어군을 자루그물로 유도하게 유도하기에는 너무 크다고 보아진다. 따라서, 문턱은 좀 더길게 해서 경사 각도를 작게 하는 것이 좋다고 생각된다. (5) 자루(bag net)의 모양은 예망속도가 0.2m/sec 이상 되면, 뒤쪽이 앞쪽보다 좁아져서, 구성상 앞쪽보다 뒤쪽을 넓게한 의도가 나타나지 않는다. 따라서 자루 그물은 전체 모양이 길다란 원통형이 되게 구성하는 것이 합리적이라고 생각된다. A boat seine has been used as a major fishing gear for catching anchovy (Engraulis japonica) in the southern coastal waters of Korea since 1920s. The original seine was operated by two rowing boats as the haul seine. The rowing boats were, in recent, replaced by powered boats. The net size was enlarged by more than three times the original nit as they began to by operated in the deeper waters of approximately sixty meters. However, there are many problems in the efficiency of the fishing gear to be improved. The authors studied on the hydrodynamic resistance and performance of the boat seine net of the 1/10 scale model in tow. The results are summaried as follows. 1. The hydrodynamic resistance converted from model experiment into the full scale is: $$R_1=30,000\;v^{1.2}\;(0.2{\leqq}v{\leqq}1.0)$$ $$R_2=16,000\;v^2\;(0.2{\leqq}v{\leqq}0.6)$$ where $R_1$ and $R_2$ denote the resistance of whole gear and of bag net in kg respectively, and v the speed of flow in m/sec. 2. In the extension wing, approximately seventy percent of the length of the ground rope from the towing end to the inside-wing slopes down from sea level toward the sea bottom, while the thirty percent of the inside of it remains parallel with th: 5:a level. The performance is regarded to b: inefficient for driving fish shoal into the inside-wing, especially for the shoal diving suddenly. 3. At the towing speed higher than 0.2 m/sec, the trailing edge of the inside-wing is blown backward beyond the seaming line connecting the inside-wing and the mouth of the bag net. It is regarded as an unreasonable performance to drive the fish shoal smoothly into the bag net. 4. At the towing speed higher than 0.2 m/sec, the posterior end of the lower bosom is lifted up above the level of the ground rope of the inside-wing. It is considered that the fish shoal diving suddenly can escape through the discrepancy between the lower bosom and the sea bottom, even if the ground rope of the inside-wing sweeps the sea bottom. 5. The angle of inclination of the upper bosom is estimated as $35\~40^{\circ}$. It seems that the inclination is too steep to drive smoothly the fish shoal diving toward the sea bottom into the bag net. 6. In structure, circumference of the posterior section of the bag net is wider by 1.3 times that of the anterior section. Actually in towing at a speed higher than 0.2m/sec, however, the circumference of the posterior section becomes smaller than that of the anterior section. It is recommended to be designed in a long cylindrical form.
