      • Prognostic Role of Nucleophosmin in Colorectal Carcinomas

        Aim: Recent research suggests that nucleophosmin (NPM) may be a prognostic marker in colorectal carcinomas (CRC). We here tested its use to predict the survival of CRC patients. Methods: We investigated NPM expression by immunohistochemistry in histologically normal to malignant colorectal tissues and evaluated its association with clinicopathological variables. Overall and disease-free survival after tumor removal were calculated by the Kaplan-Meier method, and differences in survival curves were analyzed by the log-rank test. The Cox proportional hazards model was used for multivariate analysis of prognostic factors. Results: NPM expression was found significantly upregulated in CRC compared to adjacent colorectal tissue, villous adenoma, tubular adenoma and normal colorectal mucosa (p<0.05 for all). NPM expression was statistically linked to cancer embolus, lymph node metastasis, differentiation grade, and recurrence of CRC. Overall and disease-free survival of NPM-negative CRC patients tended to be better than those for patients with NPM-positive lesions (log-rank statistic, p<0.05 for all). Multivariate analysis indicated NPM expression as an independent prognostic indicator for CRC patients (p<0.05 ). Conclusion: Our results suggest that NPM expression can predict the survival of CRC patients. Prognosis of CRC is determined by not only many known prognostic factors but also by NPM expression.

      • hARIP2 is a Putative Growth-promoting Factor Involved in Human Colon Tumorigenesis

        Activin is a multifunctional growth and differentiation factor of the growth factor-beta (TGF-${\beta}$) superfamily, which inhibits the proliferation of colon cancer cells. It induces phosphorylation of intracellular signaling molecules (Smads) by interacting with its type I and type II receptors. Previous studies showed that human activin receptor-interacting protein 2 (hARIP2) can reduce activin signaling by interacting with activin type II receptors; however, the activity of hARIP2 in colon cancer has yet to be detailed. In vitro, overexpression of hARIP2 reduced activin-induced transcriptional activity and enhanced cell proliferation and colony formation in human colon cancer HCT8 cells and SW620 cells. Also, hARIP2 promoted colon cancer cell apoptosis, suggesting that a vital role in the initial stage of colon carcinogenesis. In vivo, immunohistochemistry revealed that hARIP2 was expressed more frequently and much more intensely in malignant colon tissues than in controls. These results indicate that hARIP2 is involved in human colon tumorigenesis and could be a predictive maker for colon carcinoma aggressiveness.

      • Ten Year Literature on Psychological and Behavioral Interventions Against Cancer: a Terms Analysis

        We here performed a systematic review of PBIC literature using terms analysis in a hope of both identifying potential trends and patterns and exploring methods leveraging traditional literature reviews in this specific area. Articles meeting inclusion criteria were retrieved from PUBMED and translated into dichotomized article records representing presence or non-presence of MeSH terms and a metric consisting of numbers of times of co-occurrence between all pairs of terms identified using a self-designed program. The occurrence of and relations among the terms were calculated and visualized using Excel2007 and UCINET respectively. A total of 1,742 terms were identified from 997 articles retrieved. Put in a descending order, the lines representing the times of term occurrence formed a typical hyperbolic curve; when plotted along the x-axis of whole MESH terms, the lines clustered within four specific regions. Comparison of term occurrence between 2002 and 2011 revealed priority changes in population and subjects (from general groups to priority groups), intervention approaches (from medicine to exercise and psychotherapy), methodology and techniques (from cohort studies to randomized controlled trials) and outcomes (from health and mental health to quality of life, depression etc.). Networks of the terms featured a number of closely linked groups of topics including method and questionnaires, therapy and outcomes, survival management, psychological assessment and intervention, behavioral intervention (individual and community oriented). Terms analysis revealed interesting trends and patterns about PBIC publications and both the analysis methods and findings have implications for future research and literature reviews.

      • Laser-processing of transparent solid materials for advanced photonic device applications

        We demonstrate the laser processing for fabrication of advanced photonic devices including fiber gratings and high transmittance biomimetic windows. The fiber gratings, including fiber Bragg gratings (FBGs) and long period fiber gratings (LPFGs), are fabricated in non-sensitized standard telecom single-mode fiber by femtosecond (fs) laser, making them hightemperature (1000 ℃) stable. The FBG written by fs laser combined with phase mask method is used for monitoring the thermal effect in fs laser interaction with silica glass. The peak temperature of the thermal impulse is estimated about 4800 ℃, which decays to around 500 ℃ in the pulse interval for irradiation of 1.1 mJ at repetition rate of 1 kHz. When the pulse energy and repetition rate increase in the fabrication process of FBGs, the fs-laser-induced grating regions extend from the fiber core to the cladding?air boundary due to the laser self-focusing. With the help of selective chemical etching of the laser modified grating regions, a microhole array structured FBG is formed, which is demonstrated in the application of simultaneous sensing for refractive index (RI) and temperature. For the fs laser direct writing of LPFGs, the period is 100 μm, making the total length of the LPFG only 5 mm. The LPFGs can couple the guided mode to high-order cladding modes of second-order diffraction, resulting in a high RI sensitivity of 5265 nm/RI unit in the RI range of 1.433 ∼ 1.443 and a low-temperature sensitivity of ?15.52 pm/℃ from 20 ℃ to 500 ℃. In addition, the high transmittance biomimetic windows are produced by direct laser interference ablation. The interfered laser beams are introduced into the surface of a bare optical substrate, where structured surfaces consisting of a micropillar array are produced by the multiple exposure of the two-beam interference ablation. As an example of the application, zinc sulfide substrates are processed with this technology.

      • Post-diagnosis Soy Food Intake and Breast Cancer Survival: A Meta-analysis of Cohort Studies

        Background and Objectives: Data on associations between soy food intake after cancer diagnosis with breast cancer survival are conflicting, so we conducted this meta-analysis for more accurate evaluation. Methods: Comprehensive searches were conducted to find cohort studies of the relationship between soy food intake after cancer diagnosis and breast cancer survival. Data were analyzed with comprehensive meta-analysis software. Results: Five cohort studies (11,206 patients) were included. Pooling all comparisons, soy food intake after diagnosis was associated with reduced mortality (HR 0.85, 95%CI 0.77 0.93) and recurrence (HR 0.79, 95%CI 0.72 0.87). Pooling the comparisons of highest vs. lowest dose, soy food intake after diagnosis was again associated with reduced mortality (HR 0.84, 95%CI 0.71 0.99) and recurrence (HR 0.74, 95%CI 0.64 0.85). Subgroup analysis of ER status showed that soy food intake was associated with reduced mortality in both ER negative (highest vs. lowest: HR 0.75, 95%CI 0.64 0.88) and ER positive patients (highest vs. lowest: HR 0.72, 95%CI 0.61 0.84), and both premenopausal (highest vs. lowest: HR 0.78, 95%CI 0.69 0.88) and postmenopausal patients (highest vs. lowest: HR 0.81, 95%CI 0.73 0.91). In additioin, soy food intake was associated with reduced recurrence in ER negative (highest vs. lowest: HR 0.64, 95%CI 0.44 0.94) and ER+/PR+ (highest vs. lowest: HR 0.65, 95%CI 0.49 0.86), and postmenopausal patients (highest vs. lowest: HR 0.67, 95%CI 0.56 0.80). Conclusion: Our meta-analysis showed that soy food intake might be associated with better survival, especially for ER negative, ER+/PR+, and postmenopausal patients.

        Processing, microstructure and mechanical properties of porous silicon nitride by a slip casting method using petroleum coke as pore former

        In this study, porous Si3N4 ceramics were obtained by a slip casting method which would be easier applied in industry. The most stable and dispersing aqueous suspension was obtained from a solid slurry content of 56.2%, a carboxymethyl cellulose concentration of 0.3% as the adhesive and 0.05% glycerol as the defoamer. Petroleum coke was used as pore-former, and at the same time, it was helpful for the stability of the suspension. The burning-out of the petroleum coke seemed to be an economical way for increasing the porosity of porous Si3N4. After sintering at 1,800℃ for 2 h, samples with a pore size about 10 μm, porosities of 45-60%, and flexural strength of 52.0-112.1MPa were obtained.         Bioactivity of essential oil of Litsea cubeba from China and its main compounds against two stored product insects

        During our screening program for agrochemicals from Chinese medicinal herbs and wild plants, the essential oilof Litsea cubeba fruits was found to possess strong contact toxicity against the cigarette beetle Lasiodermaserricorne adults and the booklouse Liposcelis bostrychophila, with LD50 values of 27.33 μg/adult and71.56 μg/cm2, respectively, and also showed strong fumigant toxicity against the two stored product insectswith LC50 values of 22.97 and 0.73 mg/L, respectively. The essential oil obtained by hydrodistillation wasinvestigated by GC MS. The main components of the essential oil were identified to be E-citral (geranial)(27.49%), Z-citral (neral) (23.57%) and D-limonene (18.82%) followed by β-thujene (3.34%), β-pinene (2.85%), α-pinene (2.57%), 6-methyl-5-hepten-2-one (2.40%) and linalool (2.36%). Citral (Z/E-citral), D-limonene, β-pinene,α-pinene and linalool were separated and purified by silica gel column chromatography and preparative thinlayer chromatography, and further identified by means of physicochemical and spectrometric analysis. Citral andlinalool showed strong contact toxicity against L. serricorne and L. bostrychophila (LD50 = 11.76, 12.74 μg/adultand 20.15, 99.97 μg/cm2, respectively) and fumigant toxicity against L. serricorne and L. bostrychophila (16.54,18.04 mg/L air and 0.14, 0.71 mg/L air, respectively). Otherwise, citral, D-limonene and linalool were strongly repellentagainst the cigarette beetle L. serricorne as the essential oil whereas β-pinene and α-pinene exhibited weakerrepellency against the cigarette beetle compared with the positive control, DEET. Moreover, except α-pinene andlinalool, the other three compounds as well as the essential oil exhibited comparable repellency against thebooklouse relative to DEET.

        Molecular characterization and functional analysis of a protease-related protein in Chang-Liver cells

        In this study, the cDNA library of Chang-liver cells was immunoscreened using common ADAMs antibody to obtain ADAM related genes. We found one positive clone that was confirmed as a new gene by Blast, which is an uncharacterized helical and coil protein and processes protease activity, and named protease-related protein 1 (ARP1). The submitted GenBank accession number is AY078070. Molecular characterizations of ARP1 were analyzed with appropriate bioinformatics software. To analyse its expression and function, ARP1 was subcloned into glutathione S-transferase fusion plasmid pGEX-2T and expressed by E. coli system. The in vitro expression product of ARP1 was recognized by common ADAMs antibody with western blot. Interestingly, ARP1 cleaves gelatine at pH9.5, which suggests it is an alkaline protease. Semi-quantitative RT-PCR result indicates that ARP1 mRNA is strongly transcribed in the liver and the treated Chang-liver cells. [BMB reports 2010; 43(5): 375-381]
