        Development of an Adaptive-Feedrate Planning and Iterative Interpolator for Parametric Toolpath with Normal Jerk Constraint

        Compared to conventional linear and circular segments, the parametric curve has advantages in approximating and machining, guaranteeing higher machining precision and better machining quality. Therefore, in Computer Numerical Control (CNC) systems, parametric toolpath has become more and more popular for its high-performance in practical machining and the performance of the parametric interpolator has become a standard to illustrate the advanced CNC systems. In this paper, adaptive-feedrate planning and the iterative interpolator have been developed to generate sampling points with respect to error and higher order kinematic constraints. Firstly, a jerk-limited feedrate profile and look-ahead algorithm are utilized to provide reachable feedrate at critical points. Afterwards, normal jerk (i.e. the jerk in a normal direction) are limited to improve machining precision and quality. In theory, it is proved that the normal jerk limitations have relationship with curvature derivative and a novel iterative interpolator is proposed to ensure the normal jerk limitation by considering the corresponding constraints at curvature derivative extreme points. Finally, simulation and experiments are conducted to demonstrate the efficiency and contour performance of the proposed method compared to conventional method and time-optimal method.

        Two New Natural Products from the Fruits of Alpinia oxyphylla with Inhibitory Effects on Nitric Oxide Production in Lipopolysaccharide- Activated RAW264.7 Macrophage Cells

        Chemical investigation of the fruits of Alpinia oxyphylla led to an isolation of the two new natural products, 9-hydroxy epinootkatol (1) and (S)-2-pentanol-2-O-β-D-glucopyranoside (2), in addition to the nine known compounds, pinocembrin (3), tectochrysin (4), izalpinin (5), nookatone (6), yakuchinone A (7), protocatechuic acid (8), β-sitosterol (9), daucosterol (10) and β-sitosterol palmitate (11). Their structures were elucidated on the basis of physicochemical constants and NMR spectral data analysis. The effects of the isolated components on nitric oxide production in LPS-induced RAW 264.7 macrophages were examined. The two new natural compounds showed inhibitory activities with IC50 values of 21.8 and 32.8 μg/mL, respectively.

        A Local and Analytical Curvature-Smooth Method with Jerk-Continuous Feedrate Scheduling along Linear Toolpath

        Linear toolpath, which is always in forms of a large amount G01 codes, is most popular in practical Computer Numerical Control (CNC) systems. The linear toolpath discontinuity will bring the unavoidable fluctuations on velocity and acceleration, leading to unsatisfied machining efficiency and quality. To improve the G01 machining, corners are normally smoothened by various smooth curves and in this paper, a novel two-step method including path-smoothing and feedrate scheduling is proposed. In path-smoothing, a B-spline curve is adopted to blend sharp corners locally and the smooth curvature along transition toolpath is obtained. With constrained chord error, the analytical curvature extreme is proved to occur at the B-spline midpoint and optimal curvature extreme is searched to enhance machining efficiency. In feedrate scheduling, based on the extreme points, a look-ahead methodology is utilized to determine the reachable feedrate at these critical points and a jerk-continuous scheduling scheme is developed afterwards. To verify the proposed method, tracking and machining experiments are conducted to demonstrate the efficiency, contour performance as well as machining quality compared to point-to-point and curvature-continuous methods.

        Multi-Description Image Compression Coding Algorithm Based on Depth Learning

        Aiming at the poor compression quality of traditional image compression coding (ICC) algorithm, a multidescriptionICC algorithm based on depth learning is put forward in this study. In this study, first an imagecompression algorithm was designed based on multi-description coding theory. Image compression sampleswere collected, and the measurement matrix was calculated. Then, it processed the multi-description ICCsample set by using the convolutional self-coding neural system in depth learning. Compressing the waveletcoefficients after coding and synthesizing the multi-description image band sparse matrix obtained the multidescriptionICC sequence. Averaging the multi-description image coding data in accordance with the effectivesingle point’s position could finally realize the compression coding of multi-description images. According toexperimental results, the designed algorithm consumes less time for image compression, and exhibits betterimage compression quality and better image reconstruction effect.

      • Potential Therapeutic Targets for the Primary Gallbladder Carcinoma: Estrogen Receptors

        Gallbladder carcinoma, the most frequent malignant neoplasm of the biliary tract system, has always been considered to feature late clinical presentation and diagnosis, limited treatment options and an extremely poor prognosis. In recent years, while the incidence of gallbladder cancer has appeared to be on the increase, the available treatment methods have not greatly improved survival of the affected patients. Thus, exploring new therapeutic targets for this devastating disease is an urgent matter at present. Epidemical studies have demonstrated that the incidence of gallbladder carcinoma exhibits a distinct gender bias, affecting females two to three times more than males, pointing to crucial roles of estrogen. It is well known that estrogen acts on target tissues by binding to estrogen receptors (ERs), which are mainly divided into three subtypes, $ER{\alpha}$, $ER{\beta}$ and $ER{\gamma}$. $ER{\alpha}$ and $ER{\beta}$ appear to have overlapping but also unique even opposite biological effects. As important pathogenic mediators, ERs have been considered to relate to several kinds of tumors. In gallbladder carcinoma tissue, ERs have been shown to be positively expressed, and ERs expression levels are associated with differentiation and prognosis of this cancer. Nevertheless, the exact mechanisms of estrogen inducing growth of gallbladder carcinoma remain poorly understood. On the base of the current investigations, we deduce that estrogen participates in promotion of gallbladder carcinoma by influencing the formation of gallstones, stimulating angiogenesis, and promoting abnormal proliferation. Since ERs mediate the carcinogenic actions of estrogen in gallbladder, and therapy targeting ERs may provide new directions for gallbladder carcinoma. Therefore, it should be stressed that ERs are potential therapeutic targets for gallbladder carcinoma.

      • Antiproliferative Effects of Celecoxib in Hep-2 Cells through Telomerase Inhibition and Induction of Apoptosis

        Background: To investigate the effect of celecoxib on telomerase activity and apoptosis in a human laryngeal squamous carcinoma cell line (Hep-2 cells). Materials and Methods: The growth inhibition rate of Hep-2 cells in vitro was measured by MTT assay, and apoptosis by TUNEL assay and flow cytometry (FCM). The TRAP-ELISA method was used to determine telomerase activity in Hep-2 cells. The mRNA expression of human telomerase RNA component(hTR), human telomerase reverse transcriptase (hTERT) and human telomerase-associated protein(hTEP1) was determined by RT-PCR assay. Expression of Bax and Bcl-2 proteins was assessed by Western blotting. Results: Celecoxib can inhibit proliferation and induce apoptosis in a dose- and time-dependent manner, repress telomerase activity, decrease hTERT mRNA and Bcl-2 protein expression and increase Bax protein expression, PGE2 had no effect on telomerase. Conclusions: Celecoxib had the antiproliferative and pro-apoptotic effect in Hep-2 cells. Apoptosis was accompanied by a decrease in telomerase activity which was directly correlated with hTERT mRNA and up-regulation of Bax/Bcl-2. Bcl-2 may thus play an important role in telomerase activity as well as apoptosis.

        Transiting Exoplanet Monitoring Project (TEMP). II. Refined System Parameters and Transit Timing Analysis of HAT-P-33b

        <P>We present 10. R-band photometric observations of eight different transits of the hot Jupiter HAT-P-33b, which has been targeted by our Transiting Exoplanet Monitoring Project. The data were obtained by two telescopes at the Xinglong Station of National Astronomical Observatories of China (NAOC) from 2013 December through 2016 January, and exhibit photometric scatter of 1.6-3.0 mmag. After jointly analyzing the previously published photometric data, radial-velocity (RV) measurements, and our new light curves, we revisit the system parameters and orbital ephemeris for the HAT-P-33b system. Our results are consistent with the published values except for the planet to. star radius ratio (RP/R-*), the ingress/egress duration (tau) and the total duration (T-14), which together indicate a slightly shallower and shorter transit shape. Our results are based on more complete light curves, whereas the previously published work had only one complete transit light curve. No significant anomalies in Transit Timing Variations (TTVs) are found, and we place upper mass limits on potential perturbers, largely supplanting the loose constraints provided by the extant RV data. The TTV limits are stronger near mean-motion resonances, especially for the low-order commensurabilities. We can exclude the existence of a perturber with mass larger than 0.6, 0.3, 0.5, 0.5, and 0.3 M-circle plus near the 1: 3, 1: 2, 2: 3, 3: 2, and 2: 1 resonances, respectively.</P>

        Mitochondrial citrate accumulation drives alveolar epithelial cell necroptosis in lipopolysaccharide-induced acute lung injury

        Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.

      • Human Papillomavirus Infection and Prognostic Predictors in Patients with Oropharyngeal Squamous Cell Carcinoma

        This study focused on infection rates and subtypes of human papillomavirus (HPV) in patients with oropharyngeal squamous cell carcinoma (OSCC), and the relationship between HPV status and prognosis of the disease. We evaluated sixty-six OSCC patients who met the enrollment criteria during the period from January 1999 to December 2009. The presence or absence of oncogenic HPV types in tumors was determined using the SPF10 LiPA25 assay. Overall survival (OS) and disease specific survival (DSS) for HPV positive and HPV negative patients were estimated using Kaplan-Meier analysis. The Cox regression model was applied for multivariate analysis. HPV-DNA was detected in 11(16.7%) of all specimens. Among them, 7 were type HPV-16, while other types were HPV-16/11, HPV-35, HPV-58/52, and HPV-33/52/54. Patients with HPV positive tumors were more likely to be female, non-smokers and non-drinkers (p=0.002, 0.001 and 0.001, respectively). After a median follow-up of 24.5 months, patients with HPV positive tumors had significantly better overall survival (HR=0.106[95%CI=0.014-0.787], p=0.016,) and disease specific survival (HR=0.121[95%CI=0.016-0.906], p=0.030). Patients with HPV positive OSCC have significantly better prognosis than patients with HPV negative tumors. HPV infection is an independent prognostic factor.

        The Stent-Assisted Coil-Jailing Technique Facilitates Efficient Embolization of Tiny Cerebral Aneurysms

        Objective: Tiny cerebral aneurysms are difficult to embolize because the aneurysm’s sac is too small for a single small coil, and coils within the aneurysm may escape from the confinement of a stent. This study was performed to introduce the stent-assisted coil-jailing technique and to investigate its effect on the coil embolization of tiny intracranial aneurysms. Materials and Methods: Sixteen patients with tiny intracranial aneurysms treated with the stent-assisted coil-jailing technique between January 2011 and December 2013 were retrospectively reviewed and followed-up. Results: All aneurysms were successfully treated with the coil-jailing technique, and at the end of embolization, complete occlusion of the aneurysm was achieved in 9 cases (56.3%), incomplete occlusion in 6 (37.5%), and partial occlusion in 1 (6.3%). Intraprocedural complications included acute thrombosis in one case (6.3%) and re-rupture in another (6.3%). Both complications were managed appropriately with no sequela. Follow-up was performed in all patients for 3–24 months (mean, 7.7 months) after embolization. Complete occlusion was sustained in the 9 aneurysms with initial complete occlusion, progressive thrombosis to complete occlusion occurred in the 6 aneurysms with initial near-complete occlusion, and one aneurysm resulted in progressive thrombosis to complete occlusion after initial partial occlusion. No migration of stents or coils occurred at follow-up as compared with their positions immediately after embolization. At follow-up, all patients had recovered with no sequela. Conclusion: The stent-assisted coil-jailing technique can be an efficient approach for tiny intracranial aneurysms, even though no definite conclusion regarding its safety can be drawn from the current data.
