        Er doped SiO_x films have been synthesized by ion beam assisted deposition (IBAD). The morphology and microstructure of films and their annealing behaviors have been examined by using scanning electron microscopy and x-ray diffraction. The composition and properties of films have been systematically investigated.

      • Neutrophil Count and the Inflammation-based Glasgow Prognostic Score Predict Survival in Patients with Advanced Gastric Cancer Receiving First-line Chemotherapy

        Purpose: To explore the value of systemic inflammatory markers as independent prognostic factors and the extent these markers improve prognostic classification for patients with inoperable advanced or metastatic gastric cancer (GC) receiving palliative chemotherapy. Methods: We studied the prognostic value of systemic inflammatory factors such as circulating white blood cell count and its components as well as that combined to form inflammation-based prognostic scores (Glasgow Prognostic Score (GPS), Neutrophil-Lymphocyte Ratio (NLR), Platelet Lymphocyte Ratio (PLR), Prognostic Index (PI) and Prognostic Nutritional Index (PNI)) in 384 patients with inoperable advanced or metastatic gastric cancer (GC) receiving first-line chemotherapy. Univariate and multivariate analyses were performed to examine the impact of inflammatory markers on overall survival (OS). Results: Univariate analysis revealed that an elevated white blood cell, neutrophil and/or platelet count, a decreased lymphocyte count, a low serum albumin concentration, and high CRP concentration, as well as elevated NLR/PLR, GPS, PI, PNI were significant predictors of shorter OS. Multivariate analysis demonstrated that only elevated neutrophil count (HR 3.696, p=0.003) and higher GPS (HR 1.621, p=0.01) were independent predictors of poor OS. Conclusion: This study demonstrated elevated pretreatment neutrophil count and high GPS to be independent predictors of shorter OS in inoperable advanced or metastatic GC patients treated with first-line chemotherapy. Upon validation of these data in independent studies, stratification of patients using these markers in future clinical trials is recommended.

        Ram semen preserved at 0℃ with soybean lecithin Tris-based extender substituted for egg yolk

        Objective: The present study evaluated the preservation of ram semen at 0℃ using soybean lecithin with a Tris-fructose extender. Methods: Semen was collected by artificial vagina ejaculation from six rams with proven fertility. High quality ejaculates were diluted by soybean lecithin (0.25%, 0.5%, 0.75%, 1.0%, 1.25%) using Tris-fructose extender and control (Tris-fructose egg yolk extender), respectively. The ejaculates were diluted to a concentration of 5×108 sperm/mL, followed by cooling to 0℃ in 90 min and maintaining the temperature for 12 days. The diluted semen samples were examined and recorded for sperm progressive motility, acrosome integrity at 0, 24, 72, 144, 216, 288 h, respectively. Two hundred and twenty-three ewes were inseminated for 216 h with optimal soybean lecithin concentrated semen or control via trans-cervical insemination. Results: The results showed that there were no differences in sperm progressive motility at 0, 24, 72, and 144 h (p>0.05). After 216 h, the sperm progressive motility in the control group and 0.5% concentration groups was significantly higher when compared to 0.25% concentration (p<0.05). The 0.5% concentration group demonstrated the highest survival rate and had no difference with the control group (p>0.05). At 216 h, the sperm progressive motility of all groups was still above 50%. The acrosome integrity of all groups was decreased with prolongation of storage time, but there was no difference at each time point (p>0.05). There was no significant difference in the lambing rate and pregnancy rate between the 0.5% concentration group and the control group (p>0.05). Conclusion: These results suggest that ram sperm is capable of fertilization after preservation at 0℃ with 0.5% of soybean lecithin in Tris-based extender substituted for egg yolk and produce normal offspring after insemination.

        Sputasol (Dithiothreitol 0.54%) Improves the Detection of Human Papillomaviruses Using the Cobas 4800 System

        Dear Editor, Persistent infection with human papillomavirus (HPV) is the main cause of cervical cancer. Screening of cervical specimens for high-risk human papillomaviruses (HR-HPV) has become a routine part of patient care in most countries. The Roche cobas 4800 system (Roche Molecular Systems Inc., Pleasanton, CA, USA), a fully automated system that uses in vitro qualitative real-time PCR technology, displays high sensitivity and specificity for HR-HPV detection [1]. However, a small proportion of invalid results may occur in specimens showing mucopurulent discharge because of pipetting errors, clot formation, or failed amplification, which could cause a delay in the results and lead to additional waiting time for patients and extra work hours for staff, although most cases are resolved by vortexing the specimens and retesting. To the best of our knowledge, no researcher has tried to overcome this problem by pretreating specimens with an appropriate concentration of Sputasol (Oxoid Ltd., Basingstoke, United Kingdom); Sputasol (dithiothreitol 1.4%) has been used to aid culturing and DNA extraction from sputum specimens in some laboratories [2]. Here, we propose a standard method to improve HR-HPV detection without affecting the performance of the cobas 4800 HPV system.

        Corrosion Resistance and Compressive Strength of Cemented Soil Mixed with Nano-Silica in Simulated Seawater Environment

        Cemented soil structures are frequently exposed to the corrosive environment of seawater for an extended period, severely affecting their interior structure and mechanical qualities. This study presents laboratory-based approach to investigate the effect of nano-SiO2 on the structural and mechanical properties of cemented soil in a simulated seawater environment. The unconfined compressive strength (UCS), elastic modulus and other mechanical properties of cemented soil mixed with nano-SiO2 and ordinary silicate cemented soil were evaluated. X-ray diffraction (XRD) and scanning electron microscopy (SEM) were used to deduce the crystalline composition and microstructure characteristics of the modified cemented soil. The results show that the addition of nano-SiO2 greatly increased the compressive strength and corrosion resistance of the cemented soil. In a 3C (105% salinity) simulated saltwater curing environment, the compressive strength of the cemented soil containing 2% nano-SiO2increased by 86% and 158% at 30days and 60days, respectively. XRD and SEM showed that nano-SiO2 enhanced the interface structure of cemented soil and increased the compactness of the cement-soil system. The current study demonstrates that nano-SiO2 could considerably improve the mechanical characteristics and corrosion resistance of cemented soil when exposed to simulated seawater.

      • Radiation Induces Phosphorylation of STAT3 in a Dose- and Time-dependent Manner

        Background: We have reported the radiation could activate STAT3, which subsequently promotes the invasion of A549 cells. We here explored the dose- and time-response of STAT3 to radiation and the effect of radiation on upstream signaling molecules. Materials and Methods: A549 cells were irradiated with different doses of ${\gamma}$-rays. The expression of and nucleus translocation of p-STAT3 in A549 cells were detected by immunoblotting and immunofluorescence, respectively. The level of phosphorylated EGFR was also assessed by immunoblotting, and IL-6 expression was detected by real time PCR and ELISA. Results: Radiation promoted the phosphorylation of STAT3 at Y705 in a dose- and time-dependent manner and nuclear translocation. The level of phosphorylated EGFR in A549 cells increased after radiation. In additional, the mRNA and protein levels of IL-6 in A549 cells were also up regulated by radiation. Conclusions: STAT3 is activated by radiation in a dose-and time-dependent manner, probably due to radiation-induced activation of EGFR or secretion of IL-6 in A549 cells.

        Embryological and genetic evidence of amphimixis and apomixis in Boehmeria tricuspis

        Apomixis is a widespread alternative mode of sexual reproduction resulting in offspring that are genetically identical to the maternal plant. Boehmeria tricuspis (Hance) Makino is a perennial, wind-pollinated, herbaceous plant in the nettle family Urticaceae. The diploid B. tricuspis is monoecious but the triploid B. tricuspis is gynoecious, bearing female inflorescences only. Apomixis in B. tricuspis was first reported 50 years ago, but the mode of apomixis in the species has not been described yet. Here, we provide embryological observations of the embryo sac formation proving that triploid B. tricuspis reproduced apomictically following the Antennaria type of diplospory, and that the diploid individuals were the sexual genotype with the classical Polygonum-type maturation pattern of embryo sac development. A subsequent flow cytometry seed screen (FCSS) showed that the triploids were obligate apomicts with autonomous endosperm development, and the diploids reproduced sexually. In addition, a progeny test by molecular marker assays further demonstrated the above results.

        Dynamic and Damage Characteristics of Mortar Composite under Impact Load

        In the process of underground construction, it is common to spray concrete on the working surface, which will form a concrete-rock composite structure, to achieve the expected reinforcement effect. Compared with pure concrete or rock structures, composite structures have different mechanical properties, energy evolution and damage characteristics. Understanding the dynamic mechanics and damage characteristics of composite structures under impact loads is important to the safety of underground engineering construction. First, dynamic compression tests on different grades of mortar-sandstone composite specimens were carried out by using a Hopkinson pressure bar system to discuss the effect of impact velocities on the mechanical properties and failure modes of composite specimens. The results show that the dynamic peak stress, strain energy density and fragmentation block of the mortar-sandstone composite have an obvious strain rate effect. With an increase in impact velocity, the internal cracks of the sample do not have sufficient time to expand, and the energy absorbed by the sample leads to the continuous initiation of new cracks in the sample, resulting in an increasing damage degree of composite specimens. The increase in mortar strength can significantly increase the peak stress, improve the load-bearing capacity of the sample, and reduce the degree of sample damage. At the same impact velocity, the fragmentation degree of the mortar sample is more severe than that of sandstone, and the mortar sample is broken into powder debris, while the sandstone is broken into fragments. In addition, according to the failure properties of the composite sample, the mortar and sandstone samples are described by different damage models, and the dynamic damage constitutive model of the mortar-sandstone composite is established based on the overlay principle. The dynamic test results were fitted. The research shows that the fitting curves of the model are well fitted with the test curves and that the model accurately reflects the changes in the mechanical parameters of the composite specimens.

        Synthesis, Antibacterial Activity, and Structure–Activity Relationship of Fusaric Acid Analogs

        Forty-one fusaric acid analogs possessing a pyridine carboxylic acid scaffold have been synthesized. The antibacterial activity results demonstrated that compounds 5b, 7b, 8c, and 8d displayed strong antibacterial activities against Staphylococcus aureus ATCC25923 with minimum inhibitory concentrations (MICs) of 4–16 μg/mL. Molecular docking study indicated that these compounds have strong hydrogen-bonding interactions with TyrRS. Meanwhile, 8c and 8d showed promising antibacterial activities against Pseudomonas aeruginosa ATCC9027. Compound 4 exhibited pronounced antibacterial activities against a clinically isolated multidrug-resistant strain of Escherichia coli (MIC: 64 μg/mL as compared 64 μg/mL of levofloxacin and 1024 μg/mL of ceftriaxone sodium). Moreover, compound 17e displayed strong synergistic antibacterial effect with levofloxacin against the multidrug-resistant strain, decreasing the MIC value of levofloxacin to 1/16 of its original MIC. No obvious cytotoxic activities against LO2 was observed for compounds 4, 5b, 8c, 8d, 17d, and 17e at 50 μM. The preliminary structure–activity relationship of fusaric acid analogs was also discussed.
