        Separation of arenols from a low-temperature coal tar by liquid-liquid extraction

        Low-temperature coal tar (LTCT) is a sticky liquid mixture produced mainly from coal pyrolysis, which contains various value-added chemicals (VACs). Liquid-liquid extraction is considered as one of the green and effective ways to explore the organic composition and separate the VACs from LTCT. Herein, petroleum ether, methanol, and carbon disulfide were used to extract arenols from a LTCT. As a result, the relative content and absolute content of arenols extracted from the LTCT are ca. 96.3% and 85.9%, respectively. Among them, p-cresol is predominant, accounting for 22.2%. The isolated contents of arenols are up to 84.6%. Moreover, a kilogram-scale operation was carried out under the same conditions, which offers a potential application in industrial production.

        Dynamic and Damage Characteristics of Mortar Composite under Impact Load

        In the process of underground construction, it is common to spray concrete on the working surface, which will form a concrete-rock composite structure, to achieve the expected reinforcement effect. Compared with pure concrete or rock structures, composite structures have different mechanical properties, energy evolution and damage characteristics. Understanding the dynamic mechanics and damage characteristics of composite structures under impact loads is important to the safety of underground engineering construction. First, dynamic compression tests on different grades of mortar-sandstone composite specimens were carried out by using a Hopkinson pressure bar system to discuss the effect of impact velocities on the mechanical properties and failure modes of composite specimens. The results show that the dynamic peak stress, strain energy density and fragmentation block of the mortar-sandstone composite have an obvious strain rate effect. With an increase in impact velocity, the internal cracks of the sample do not have sufficient time to expand, and the energy absorbed by the sample leads to the continuous initiation of new cracks in the sample, resulting in an increasing damage degree of composite specimens. The increase in mortar strength can significantly increase the peak stress, improve the load-bearing capacity of the sample, and reduce the degree of sample damage. At the same impact velocity, the fragmentation degree of the mortar sample is more severe than that of sandstone, and the mortar sample is broken into powder debris, while the sandstone is broken into fragments. In addition, according to the failure properties of the composite sample, the mortar and sandstone samples are described by different damage models, and the dynamic damage constitutive model of the mortar-sandstone composite is established based on the overlay principle. The dynamic test results were fitted. The research shows that the fitting curves of the model are well fitted with the test curves and that the model accurately reflects the changes in the mechanical parameters of the composite specimens.

      • Radiation Induces Phosphorylation of STAT3 in a Dose- and Time-dependent Manner

        Background: We have reported the radiation could activate STAT3, which subsequently promotes the invasion of A549 cells. We here explored the dose- and time-response of STAT3 to radiation and the effect of radiation on upstream signaling molecules. Materials and Methods: A549 cells were irradiated with different doses of ${\gamma}$-rays. The expression of and nucleus translocation of p-STAT3 in A549 cells were detected by immunoblotting and immunofluorescence, respectively. The level of phosphorylated EGFR was also assessed by immunoblotting, and IL-6 expression was detected by real time PCR and ELISA. Results: Radiation promoted the phosphorylation of STAT3 at Y705 in a dose- and time-dependent manner and nuclear translocation. The level of phosphorylated EGFR in A549 cells increased after radiation. In additional, the mRNA and protein levels of IL-6 in A549 cells were also up regulated by radiation. Conclusions: STAT3 is activated by radiation in a dose-and time-dependent manner, probably due to radiation-induced activation of EGFR or secretion of IL-6 in A549 cells.

        Identification of Polymorphisms in Thymic Stromal Lymphopoietin (TSLP) Gene and Their Association with Allergic Rhinitis

        Dendritic cell (DC) activator, thymic stromal lymphopoietin (TSLP) induces DCs to produce Th2-attracting chemokines, causing differentiation of CD4+ and CD8+ T cells into effector cells. The differentiated cells are characterized by a typical pro-allergic phenotype. In this study, we identified four single nucleotide polymorphisms (SNPs) and one variation site (g.-305delT) from the results of scanned human TSLP gene. The SNPs are as following: g.-1914A>G and g.-847C>T in promoter, g.-82C>T in 5` UTR and g.1117C>T in intron 2. We also evaluated the association of genotype and allele frequencies of these SNPs between non-allergic rhinitis controls and allergic rhinitis patients. We further investigated possible correlations between each genotype with serum total IgE levels and peripheral blood eosinophil counts in allergic rhinitis patients. The frequencies of haplotype constructed by these SNPs between these two groups were also compared. Our results demonstrate the two SNPs (g.-1914A>G and g.-847C>T) were associated with susceptibility of allergic rhinitis (P=0.006 and P=0.015 respectively), however, serum total IgE levels were not significantly related with the SNPs.

        The MALATANG Survey: The <i>L</i><sub>GAS</sub>-<i>L</i><sub>IR</sub> Correlation on Sub-kiloparsec Scale in Six Nearby Star-forming Galaxies as Traced by HCN <i>J</i> = 4 → 3 and HCO<sup>+</sup><i>J</i> = 4 → 3

        <P>We present HCN J = 4 -> 3 and HCO+ J = 4 -> 3 maps of six nearby star-forming galaxies, NGC 253, NGC 1068, IC 342, M82, M83, and NGC 6946, obtained with the James Clerk Maxwell Telescope as part of the MALATANG survey. All galaxies were mapped in the central 2' x 2' region at 14 '' (FWHM) resolution (corresponding to linear scales of similar to 0.2-1.0 kpc). The L-IR-L'(dense) relation, where the dense gas is traced by the HCN J = 4 -> 3 and the HCO+ J = 4 -> 3 emission, measured in our sample of spatially resolved galaxies is found to follow the linear correlation established globally in galaxies within the scatter. We find that the luminosity ratio, L-IR/L'(dense), shows systematic variations with L-IR within individual spatially resolved galaxies, whereas the galaxy-integrated ratios vary little. A rising trend is also found between L-IR/L'(dense) ratio and the warm-dust temperature gauged by the 70 mu m/100 mu m flux ratio. We find that the luminosity ratios of IR/HCN (4-3) and IR/HCO+ (4-3), which can be taken as a proxy for the star formation efficiency (SFE) in the dense molecular gas (SFEdense), appear to be nearly independent of the dense gas fraction (f(dense)) for our sample of galaxies. The SFE of the total molecular gas (SFEmol) is found to increase substantially with f(dense) when combining our data with those on local (ultra) luminous infrared galaxies and high-z quasars. The mean L'(HCN(4-3))/L'(HCO+(4-3)) line ratio measured for the six targeted galaxies is 0.9 +/- 0.6. No significant correlation is found for the L'(HCN(4-3)) L'(HCO+(4-3)) ratio with the star formation rate as traced by L-IR, nor with the warm-dust temperature, for the different populations of galaxies.</P>

        Ram semen preserved at 0℃ with soybean lecithin Tris-based extender substituted for egg yolk

        Objective: The present study evaluated the preservation of ram semen at 0℃ using soybean lecithin with a Tris-fructose extender. Methods: Semen was collected by artificial vagina ejaculation from six rams with proven fertility. High quality ejaculates were diluted by soybean lecithin (0.25%, 0.5%, 0.75%, 1.0%, 1.25%) using Tris-fructose extender and control (Tris-fructose egg yolk extender), respectively. The ejaculates were diluted to a concentration of 5×108 sperm/mL, followed by cooling to 0℃ in 90 min and maintaining the temperature for 12 days. The diluted semen samples were examined and recorded for sperm progressive motility, acrosome integrity at 0, 24, 72, 144, 216, 288 h, respectively. Two hundred and twenty-three ewes were inseminated for 216 h with optimal soybean lecithin concentrated semen or control via trans-cervical insemination. Results: The results showed that there were no differences in sperm progressive motility at 0, 24, 72, and 144 h (p>0.05). After 216 h, the sperm progressive motility in the control group and 0.5% concentration groups was significantly higher when compared to 0.25% concentration (p<0.05). The 0.5% concentration group demonstrated the highest survival rate and had no difference with the control group (p>0.05). At 216 h, the sperm progressive motility of all groups was still above 50%. The acrosome integrity of all groups was decreased with prolongation of storage time, but there was no difference at each time point (p>0.05). There was no significant difference in the lambing rate and pregnancy rate between the 0.5% concentration group and the control group (p>0.05). Conclusion: These results suggest that ram sperm is capable of fertilization after preservation at 0℃ with 0.5% of soybean lecithin in Tris-based extender substituted for egg yolk and produce normal offspring after insemination.

      • Low Expression of Tyrosine-protein Phosphatase Nonreceptor Type 12 is Associated with Lymph Node Metastasis and Poor Prognosis in Operable Triple-negative Breast Cancer

        Background: Low tyrosine-protein phosphatase nonreceptor type 12 (PTPN12) expression may be associated with breast cancer growth, proliferation, and metastasis. However, the prognostic value of PTPN12 in breast cancer has not been clearly identified. Patients and Methods: 51 triple-negative breast cancer (TNBC) patients and 83 non-TNBC patients with a histopathology diagnosis from October 2001 to September 2006 were included in this study. Immunohistochemical staining for PTPN12 on tissue microarrays was conducted. Results: High PTPN12 expression was seen in 39.2% of TNBC and 60.2 % of non-TNBC cases. Low PTPN12 expression was associated with lymph node status (p = 0.002) and distant metastatic relapse (p = 0.002) in TNBC patients. Similarly, low PTPN12 expression in non-TNBC patients was significantly correlated with lymph node status (p = 0.002), stage (p = 0.002) and distant metastatic relapse (p = 0.039). The high PTPN12 expression group was associated with longer DFS and OS compared with low PTPN12 expression group only in TNBC cases (p = 0.005, p = 0.015), according to univariate Cox regression analysis. Conclusion: These findings provide evidence that low expression of PTPN12 is associated with worse prognosis and may be used as a potential prognostic biomarker in TNBC patients.

        QTL mapping and identification of candidate genes for cold tolerance at the germination stage in wild rice

        Background Cold damage stress significantly affects rice growth (germination and seedling) and causes serious losses in yield in temperate and high-altitude areas around the globe. Objective This study aimed to explore the cold tolerance (CT) locus of rice and create new cold-tolerant germplasm. We constructed a chromosome segment substitution line (CSSL) with strong CT and fine mapped quantitative trait loci (QTLs) associated with CT by performing the whole-genome resequencing of CSSL with phenotypes under cold treatment. Methods A chromosome CSSL, including 271 lines from a cross between the cold-tolerant wild rice Y11 (Oryza rufipogon Griff.) and the cold-sensitive rice variety GH998, was developed to map QTLs conferring CT at the germination stage. The whole-genome resequencing was performed on CSSL for mapping QTLs of associated with CT at the germination stage. Results A high-density linkage map of the CSSLs was developed using the whole-genome resequencing of 1484 bins. The QTL analysis using 615,466 single-nucleotide polymorphisms (SNPs) led to the identification of 2 QTLs related to germination rate at low-temperature on chromosome 8 (qCTG-8) and chromosome 11 (qCTG-11). The qCTG-8 and qCTG-11 explained 14.55% and 14.31% of the total phenotypic variation, respectively. We narrowed down qCTG-8 and qCTG-11 to 195.5 and 78.83-kb regions, respectively. The expression patterns of important candidate genes in different tissues, and of RNA-sequencing (RNA-seq) in CSSLs, were identified based on gene sequences in qCTG-8 and qCTG-11 cold-induced expression analysis. LOC_Os08g01120 and LOC_Os08g01390 were identified as candidate genes in qCTG-8, and LOC_Os11g32880 was identified as a candidate gene in qCTG-11. Conclusions This study demonstrated a general method that could be used to identify useful loci and genes in wild rice and aid in the future cloning of candidate genes of qCTG-8 and qCTG-11. The CSSLs with strong CT were supported for breeding cold-tolerant rice varieties.

        3,7-Dihydroxy-2,4,6-trimethoxyphenanthrene, A New Phenanthrene from Bulbophyllum Odoratissimum

        Efficient stereoselective synthesis of α,β-Unsaturated acids, α-Cyanoacrylonitriles and α-Cyanoacrylates has been carried out in the presence of NaHSO4·SiO2 under solvent?free conditions with an E-geometry. A new phenanthrene derivative 3,7-dihydroxy-2,4,6-trimethoxyphenanthrene was isolated from the all plant of Bulbophyllum odoratissimum, and its structure was elucidated by extensive spectral studies and chemical transformation. The compound displayed cytotoxicity against the growth of human leukemia cell lines K562 and HL-60, human lung adenocarcinoma A549, human hepatoma BEL-7402 and human stomach cancer cell lines SGC-7901 with IC50 values of 14.23, 10.02, 3.42, 15.36 and 1.13 mg/ml respectively.
