The differentially virulent race T1 of common bunt (Tilletia tritici) was used to inoculate the wheat lines Neepawa (compatible) and its sib BW553 (incompatible) that are nearly isogenic for the Bt-10 resistance gene. Inoculated crown tissues were used to construct a suppression subtractive hybridization (SSH) cDNA library. Of the 1920 clones arrayed from the SSH cDNA library, approximately 10% were differentially regulated. A total of 168 differentially up-regulated and 25 down-regulated genes were identified and sequenced; 71% sequences had significant homology to genes of known function, of which 59% appeared to have roles in cellular metabolism and development, 24% in abiotic/biotic stress responses, 3% involved in transcription and signal transduction responses. Two putative resistance genes and a transcription factor were identified among the up regulated sequences. The expression of several candidate genes including a lipase, two non-specific lipid transfer proteins (ns-LTPs), and several wheat pathogenesis-related (PR)-proteins, was evaluated following 4 to 32 days post-inoculation in compatible and incompatible interactions. Results confirmed the higher overall expression of these genes in resistant BW553 compared to susceptible Neepawa, and the differential up-regulation of wheat lipase, chitinase and PR-1 proteins in the expression of the incompatible interaction.

<P>This paper presents an experimental study of the foaming behavior of polypropylene (PP)/(waste ground rubber tire powder) (WGRT) blends when using a chemical blowing agent in an extrusion foaming process. The effects of formulations (i.e., WGRT content, blowing agent content, compatibilizer) and the processing parameters (i.e., die temperature, screw speed) on the void fraction, average cell size, cell density, and cell morphology of the PP/WGRT foams were investigated. The blowing agent loading affected the cell structure of the foams and the average cell size, and the void fraction increased with increasing blowing agent loading. Both increasing the screw speed and decreasing the die temperature could establish a high pressure drop in the extruder die, and these were beneficial to the foaming extrusion. J. VINYL ADDIT. Construction and Expression of an Eukaryotic Expression Vector Containing the IER3 Gene

Background: More and more research indicate that the immediately early response gene 3 (IER3) is involved inmany biological provesses, such as apoptosis and immunoreaction, as well as viral infection, tumorigenesis and tumour progression. Methods: Here we describe the construction of an eukaryotic expression vector containing IER3 gene and its expression in A549 cells as assessed through fluorescence microscopyand Western-blotting. Results: Fluorescence detection displayed that GFP in cytoplasm was high during 48 and 72 hours post-transfection. In addition, Western blotting showed significant increase in IER3 gene expression in the transfected cells compared with controls. Conclusion: The recombinate plasmid expression vector was constructed successfully, which may provide a basis for further exploration of function of IER3 in lung cancer.
Molecular characterization and comparative analysis of two waxy alleles in barley

Two alleles of the barley waxy locus were characterized from non-waxy cultivar Bowman and waxy cultivar CDC Candle,respectively. Their nucleotide and protein sequences were compared with other known waxy genes. The comparison results indicated that there were 100 polymorphic sites, among which 69 were in the non-coding region and 31 were in the coding region. Out of 100 polymorphic sites, 45 were transversion,35 were transition and 20 were indels. A 397 bp deletion and a 193 bp insertion in the promoter region and a 15bp insertion in the coding region were found in CDC Candle,but not in Bowman. A deletion (11 bp) was detected in Bowman, which exhibited no effects on normal waxy expression. In summary, the 397 bp deletion was supposed to account for the reduction of GBSS I, resulting in the low amylose in CDC Candle; whereas other polymorphic sites might be not correlated with amylose synthesis.

State Estimation (SE) is the basis of a variety of advanced applications used in most modern power systems. An SE problem formed with enough phasor measurement units (PMUs) data is simply a linear weighted least squares problem requiring no iterations. Thus, designing a minimum-cost placement of PMUs that guarantees observability of a power system becomes a worthy challenge. This paper proposes an equivalent integer linear programming method for substation-oriented optimal PMU placement (SOOPP). The proposed method uses an exhaustive search to determine a globally optimal solution representing the best PMU placement for that particular power system. To obtain a more comprehensive model, contingencies and the limitation of the number of PMU measurement channels are considered and embodied in the model as changes to the original constraints and as additional constraints. The proposed method is examined for applicability using the IEEE 14-bus, 118-bus and 300-bus test systems. The comparison between SOOPP results and results obtained by other methods reveals the excellence of SOOPP. Furthermore, practical large-scale power systems are also successfully analyzed using SOOPP.

CCNG2 Suppressor Biological Effects on Thyroid Cancer Cell through Promotion of CDK2 Degradation

This study aimed to analyze the expression and clinical significance of cyclin G2 (CCNG2) in thyroid carcinoma and the biological effects of CCNG2 overexpression in a cell line. Immunohistochemistry and Western blotting were used to analyze CCNG2 protein expression in 63 cases of thyroid cancer and normal tissues to allow the relationship with clinical factors to be assessed. CCNG2 lentiviral and empty vectors were transfected into the thyroid cancer K1 cell line. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were applied to detect the mRNA and protein levels of CCNG2. MTT assay and cell cycle were also conducted to assess the influence of up-regulated expression of CCNG2 on K1 cell biology. The level of CCNG2 protein expression was found to be significantly lower in thyroid cancer tissue than normal tissues (P<0.05). Western blot: The relative amount of CCNG2 protein in thyroid cancer tissue was respectively found to be significantly lower than in normal tissues (P<0.05), correlating with lymph node metastasis, clinic stage and histological grade (P<0.05), but not gender, age or tumor size (P>0.05). Loss of CCNG2 expression correlated significantly with poor overall survival time on Kaplan-Meier analysis (P<0.05). The results for biological functions showed that K1 cell transfected CCNG2 had a lower survival fraction, a greater percentage in the G0/G1 phases, and lower cyclin-dependent kinase 2 (CDK2) protein expression compared with K1 cells non-transfected with CCNG2 (P<0.05). CCNG2 expression decreased in thyroid cancer and correlated significantly lymph node metastasis, clinic stage, histological grade and poor overall survival, suggesting that CCNG2 may play important roles as a negative regulator in thyroid cancer K1 cells by promoting degradation of CDK2.
Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.
