Background The present study investigates the protective effect of myricitrin (MCT) on the retinal tissues in diabetic retinopathy of diabetic rats. Objective The diabetic rat model was created by treating the rats with streptozotocin along with high-energy diet. The blood glucose was evaluated by glucose estimation kit, and rats measuring glucose ≥ 12.00 mmol/L were considered diabetic and were selected for the study. The diabetes-induced rats were divided randomly as diabetic control rats (vehicle treated) (DB), diabetic rats MCT (100 mg/kg) (DB + MCT 100 mg/kg) and diabetic rats MCT 200 mg/kg (DB + MCT 200 mg/kg). Rats received MCT for 12 weeks via oral route daily. The rats after 12 weeks were sacrificed, and the eyes were removed. The retina tissue was evaluated for thickness, oxidative stress markers, levels of VEGF and Bax and Bcl-2. In silico docking analysis was done by Autodock Vina tools for target confirmation of MCT. Results MCT decreased the serum glucose levels and also improved the body weight in diabetic rats. MCT improved thickness and apoptosis of cells. It was also found that MCT decreased oxidative stress, and improved levels of VEGF in retinal tissues of diabetic rats. MCT also exerted anti-apoptotic activity as shown by over-expression of Bcl-2 and suppression of Bax in retinal tissues of rats. Docking study confirmed potential binding affinity of MCT with VEGF. Conclusions MCT could be a potential therapeutic molecule in treating diabetic retinopathy which could modulate glucose levels, oxidative stress and levels of VEGF in diabetic rats.
Effects of Lactobacillus plantarum SCS2 on blood glucose level in hyperglycemia mice model

In this study, the hyperglycemia mice model was established with 1-week high sugar and fat diet plus with 70 mg/kg body weight of streptozotocin injection for 3 days. Sixty male Kunming mice of 3 weeks old in a specific-pathogen-free grade were divided into six groups randomly, which includes normal group (NG), prevention group (PG), treatment group for low dose (TGL), middle dose (TGM), high dose (TGH), and model group (MG). NG and MG mice were fed with sterile physiological saline (10 mL/kg body weight). PG mice were fed with the concentration of 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions from the second to third week. TGL, TGM, and TGH mice were fed with the concentration of 2.0 × 109, 4.0 × 109, and 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions (10 mL/kg body weight), respectively from fourth to tenth week. The results showed that the fasting and postprandial 2 h blood glucose levels of TGH mice were reduced by L. plantarum SCS2 significantly (p < 0.05) as compared with MG. The body weight of TGH mice came to normal level at tenth week. Content of K+ in plasma of TGH mice was increased and contents of Na+ and Cl− in the plasma of TGH mice were decreased as compared with MG. Meanwhile, content of glycogen in TGH mice was reduced. However, the effect of L. plantarum SCS2 on the prevention of hyperglycemia in PG mice was not significant as compared with NG mice during the experiment. These results suggested that L. plantarum SCS2 showed a hypoglycemic effect on hyperglycemic mice model.
Laser Joining of Ti3Al-Based Alloy to Ni-Based Superalloy using a Titanium Interlayer

Joining Ti3Al-based alloy to Ni-based superalloy is of great interest for applications in the aerospace fields. Direct welding of these two materials was very difficult and the joint usually fractured during laser welding which cannot form an effective welding joint. In this work, a pure titanium interlayer with a thickness of 0.4 mm was used between the base metals. The results indicated that the addition of titanium interlayer has a great effect on the joint performance. No macrocracks were found through the whole joint. The average room-temperature tensile strength of the joint with titanium was 177 MPa which still much lower than the two base metals. The weak link of the dissimilar joint was the Ti3Al/weld interface. The presence of Ti2Ni, α2-Ti3Al and AlNi2Ti brittle intermetallic compounds in the Ti3Al/weld interface deteriorated the joint properties.

Seven polyketides, including four new onesnamed as varilactones A–B (1–2) and wortmannilactonesM–N (3–4), as well as three biogenetically related knownwortmannilactones E, F and H (5–7), were isolated fromthe fungus Penicillium variabile HXQ-H-1, cultivated inpotato-based medium with the histone deacetylase inhibitorsuberoylanilide hydroxamic acid (SAHA). Their structures(1–7) were established by analysis of NMR and MS data,and the absolute configuration of oxabicyclo[2.2.1]heptaneunit was deduced from 1H-NMR analyses on MPA estersafter chemical reduction reaction. Among them, varilactonesA and B (1 and 2), features a novel skeleton with atriene unit linking an oxabicyclo[2.2.1]heptane and anoxabicyclo[3.3.0]octane rings.

To evaluate the behavior of the advanced unbonded pre-stressed concrete containment vessel (UPCCV) for one typical China nuclear power plant under Japan’s March 11 earthquake, five nonlinear time history analysis and a nonlinear static analysis of a 1:10 scale UPCCV structure have been carried out with MSC.MARC finite element program. Comparisons between the analytical and experimental results demonstrated that the developed finite element model can predict the earthquake behavior of the UPCCV with fair accuracy. The responses of the 1:10 scale UPCCV subjected to the 11 March 2011 Japan earthquakes recorded at the MYG003 station with the peak ground acceleration (PGA) of 781 gal and at the MYG013 station with the PGA of 982 gal were predicted by the dynamic analysis. Finally, a static analysis was performed to seek the ultimate load carrying capacity for the 1:10 scale UPCCV.

Three new diketopiperazine derivatives (DKPs), saroclazines A–C (1–3) along with three known DKPs (4– 6) were isolated from mangrove-derived fungi Sarocladium kiliense HDN11-84. Saroclazines A–B (1 and 2) possessed a free amide structure, which was first found in sulfurcontaining aromatic DKPs. Their structures were elucidated by NMR, HRESIMS and X-ray. The cytotoxic activity of new compounds (1–3) was tested against HeLa cell lines, among which compound 2 showed an IC50 value of 4.2 lM.

Improving health and delaying aging is the focus of medical research. Previous studies have shown that mesenchymal stem cell (MSC) senescence is closely related to organic aging and the development of aging-related diseases such as osteoarthritis (OA). m6A is a common RNA modification that plays an important role in regulating cell biological functions, and ALKBH5 is one of the key m6A demethylases. However, the role of m6A and ALKBH5 in MSC senescence is still unclear. Here, we found that the m6A level was enhanced and ALKBH5 expression was decreased in aging MSCs induced by multiple replications, H2O2 stimulation or UV irradiation. Downregulation of ALKBH5 expression facilitated MSC senescence by enhancing the stability of CYP1B1 mRNA and inducing mitochondrial dysfunction. In addition, IGF2BP1 was identified as the m6A reader restraining the degradation of m6A-modified CYP1B1 mRNA. Furthermore, Alkbh5 knockout in MSCs aggravated spontaneous OA in mice, and overexpression of Alkbh5 improved the efficacy of MSCs in OA. Overall, this study revealed a novel mechanism of m6A in MSC senescence and identified promising targets to protect against aging and OA.
