      • Curcumin Conjugates Induce Apoptosis Via a Mitochondrion Dependent Pathway in MCF-7 and MDA-MB-231 Cell Lines

Singh, Durg Vijay,Agarwal, Shikha,Singh, Preeti,Godbole, Madan Madhav,Misra, Krishna Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.10

In order to enhance the bioavailability of curcumin its conjugates with piperic acid and glycine were synthesized by esterifying the 4 and 4' phenolic hydroxyls, the sites of metabolic conjugation. Antiproliferative and apoptotic efficacy of synthesized conjugates was investigated in MCF-7 and MDA-MB-231 cell lines. $IC_{50}$ values of di-O-glycinoyl (CDG) and di-O-piperoyl (CDP) esters of curcumin were found to be comparable with that of curcumin. Both conjugates induced chromatin condensation fragmentation and apoptotic body formation. CDP exposure to MCF-7 cells induced apoptosis initiating loss of mitochondrial membrane potential (${\Delta}{\Psi}m$) followed by inhibition of translocation of transcription factor NF-${\kappa}B$ and release of Cytochrome-C. Reactive oxygen species (ROS) production was evaluated by fluorescent activated cell sorter. Change in ratio of Bcl2/Bclxl was observed, suggesting permeablization of mitochondrial membrane leading to the release of AIF, Smac and other apoptogenic molecules. DNA fragmentation as a hallmark for apoptosis was monitored by TUNEL as well as agrose gel electrophoresis. Thus, it was proven that conjugation does not affect the therapeutic potential of parent molecule in vitro, while these could work in vivo as prodrugs with enhanced pharmacokinetic profile. Pharmacokinetics of these molecules under in vivo conditions is a further scope of this study.

      • Intra-Operative Frozen Sections: Experience at A Tertiary Care Centre

Preeti, Agarwal,Sameer, Gupta,Kulranjan, Singh,Abhinav, Sonkar Arun,Preeti, Rani,Sunita, Yadav,Mati, Goel Madhu Asian Pacific Journal of Cancer Prevention 2016 Asian Pacific journal of cancer prevention Vol.17 No.12

The present study was conducted to assess error rates with diagnosis using intra-operative frozen sections, and to indicate ways to increase overall performance. Over a period of two years, 227 cases were biopsied intra-operatively. Errors were observed in 14 cases. Four of these were sampling errors, one by a pathologist and three by surgeons. In seven cases incorrect interpretations were made. Epithelial dysplasia was observed on definitive histology in two cases which was not reported intra-operatively. One case was of ectopic thyroid. In cases of oral cancer where sentinel lymph nodes were sampled, immunohistochemistry for cytokeratin was performed to facilitate identification of micrometastasis. Only single case displayed tumor deposits which was not evident morphologically. Resection margins were reported in seventy eight cases. Some 18% (14/50) benefited from revision of margins; overall sensitivity of intra-operative frozen sections for marginal status was 71.4%, with a specificity of 90.3%. Overall sensitivity was 75% and specificity was 97.5%. Careful observation, pathologist experience and knowledge of limitations help in improving the overall diagnostic outcome.

      • A Survey : Digital Image Watermarking Techniques

Preeti Parashar,Rajeev Kumar Singh 보안공학연구지원센터(IJSIP) 2014 International Journal of Signal Processing, Image Vol.7 No.6

Multimedia security is extremely significant concern for the internet technology because of the ease of the duplication, distribution and manipulation of the multimedia data. The digital watermarking is a field of information hiding which hide the crucial information in the original data for protection illegal duplication and distribution of multimedia data. This paper presents a survey on the existing digital image watermarking techniques. The results of various digital image watermarking techniques have been compared on the basis of outputs. In the digital watermarking the secret information are implanted into the original data for protecting the ownership rights of the multimedia data. The image watermarking techniques may divide on the basis of domain like spatial domain or transform domain or on the basis of wavelets. The spatial domain techniques directly work on the pixels and the frequency domain works on the transform coefficients of the image. This survey elaborates the most important methods of spatial domain and transform domain and focuses the merits and demerits of these techniques.

Quality of Chilled Ready-to-Bake Pizza Stored in Air and under Modified Atmospheres: Microbiological and Sensory Attributes

        Preeti Singh,Ali Abas Wani,G. K. Goyal 한국식품과학회 2011 Food Science and Biotechnology Vol.20 No.1

Modified atmosphere packaging (MAP) studies on microbiological and sensory analysis were conducted to extend the shelf life of ready-to-bake pizza stored at 7±1ºC. The gas combinations used were: atm1: air (control), atm2:CO_2 (100%), atm3: N_2 (100%), atm4: 50% CO_2/50% N2. Total plate count (TPC), yeasts/molds (Y/M), coliforms, lactic acid bacteria (LAB), psychrotrophs, and anaerobic spore formers were estimated at time intervals of 0, 5, 10,15, and 20 days. TPC and LAB of pizza samples (atm1)reached 7.10 and 8.14 log CFU/g after 10 days of storage, respectively. Coliforms, psychrotrophs, and Y/M were significantly higher (p<0.05) for pizza samples stored in atm1 than other storage conditions of MAP. Finally, counts of anaerobic spore formers were low (<3 log CFU/g)irrespective of the packaging conditions throughout the entire storage period. It was concluded that among the 4atmospheres examined, atm2 (100% CO_2) was the best, followed by atm4>atm3>atm1 respectively, in descending order. MAP conditions under this study may extend shelf life of pizza to considerable amount of time.

Cytokines Expression and Nitric Oxide Production under Induced Infection to Salmonella Typhimurium in Chicken Lines Divergently Selected for Cutaneous Hypersensitivity

        Singh, Rani,Jain, Preeti,Pandey, N.K.,Saxena, V.K.,Saxena, M.,Singh, K.B.,Ahmed, K.A.,Singh, R.P. Asian Australasian Association of Animal Productio 2012 Animal Bioscience Vol.25 No.7

In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.

In situ green synthesis of Au/Ag nanostructures on a metal-organic framework surface for photocatalytic reduction of p-nitrophenol

        Karanveer Singh,Deepak Kukkar,Ravinder Singh,Preeti Kukkar,Nardev Bajaj,Jagpreet Singh,Mohit Rawat,Akshay Kumar,Ki-Hyun Kim 한국공업화학회 2020 Journal of Industrial and Engineering Chemistry Vol.81 No.-

In this manuscript, we report the insitu growth of gold nanoparticles (GNPs) and silver nanoparticles(SNPs) over Ce/Tb-doped Y-benzene tricaroboxylate metal organic frameworks (MOF-76). Thephotocatalytic potential of these nanocomposites (NCs) was then assessed for the photocatalyticreduction of p-nitrophenol (p-NP) in aqueous medium. The NCs had large surface area to favorablyshowcase the efficient absorption of light energy. Correspondingly, it facilitated the electron transferbetween the valance and conduction bands to promote the reductive removal of p-NP. Among the testedcatalysts, GNPs@MOF-76 (Ce) (MOF-76(1a)) (dosage: 10 ml of 5 mg.ml 1) recorded the maximumcatalytic efficiency (96.3%) with an apparent rate constant (K0app: 0.33 min 1). Further, MOF-76 (1a) NCsexhibited good recyclability (e.g., a catalytic efficiency of 77.45% over 3 cycles). Moreover, the proposedNCs exhibited superior performance over commercial photocatalysts (e.g., p25 TiO2 nanopowder)towards p-NP reduction. As such, the results of this experimental study successfully demonstrated thesuperiority of the MOF immobilized nanoparticles (NPs) as an efficient platform for the removal ofindustrial grade pollutants.

Exceptionally stable green-synthesized gold nanoparticles for highly sensitive and selective colorimetric detection of trace metal ions and volatile aromatic compounds

Karanveer Singh,Deepak Kukkar,Ravinder Singh,Preeti Kukkar,김기현 한국공업화학회 2018 Journal of Industrial and Engineering Chemistry Vol.68 No.-

The manuscript reports synthesis of exceptionally stable gold nanoparticles (GNPs) using Momordica charantia fruit extract. The synthesis approach was optimized by refining three experimental variables including source of the fruit extract (peel, seed, and seed coat), pH of the solution, and temperature of the reaction medium. As synthesized GNPs showed excellent stability against various thiolated compounds (e.g., thioglycolic acid, thiourea, l-cystine, 1-dodecanethiol, and cysteamine hydrochloride). Moreover, these nanoparticles showed distinctive colorimetric responses against Cd2+ and thiophenol (TP) from their potential interferences. The limit of detection (LOD) values for Cd2+ and TP were determined as 0.186 and 0.154 μM, respectively.

<P><B>Abstract</B></P> <P>In this study, we investigated the optimal conditions for solvothermal-assisted synthesis of Eu-BDC-NH2 metal organic frameworks (MOFs) to improve their sensing capacity toward some selected target metal species. To this end, we focused on the interactive relationships between key synthesis variables (e.g., the relative molar ratios (and absolute amounts) of the reactants [EuCl3·6H2O vs. amino terephthalic acid (BDC-NH2)], temperature, and reaction duration). It was observed that different types of MOFs synthesized under diverse conditions consistently exhibited emission profiles of Eu3+. However, their luminescence properties were distinguished systematically by the synthesis conditions set for each sample group. The sensing potential of luminescent MOFs was explored for detection of toxic metal ions through two approaches, i.e., direct chemosensing and indirect biomolecule mediated sensing approach. In the former, the MOFs exhibited noticeable turn-off fluorescence behavior, especially with Hg2+ ions (i.e., a high Stern-Volmer quenching constant (Ksv) of 6.42 × 105 M−1 and limit of detection (LOD) of 0.007 M). In the latter, nanocomposites of MOFs-cyanocobalamin were characterized by a highly selective turn on fluorescence response towards Co2+ ions (Ksv = −6.25 × 103 M−1; LOD = 0.7 M). As such, the sensing capacity of these MOFs was distinguished efficiently by control of synthesis variables.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Optimized synthesis of Eu-BDC-NH2-MOFs by solvothermal approach. </LI> <LI> Effect of three variables i.e. molar ratio of reactants, temperature and time period reported has been investigated. </LI> <LI> Enhanced luminescence intensity, high reproducibility, and high stability of the MOFs upto 1 year. </LI> <LI> Significant turn off fluorescence in presence of 1 ppm Hg2+ metal ions. </LI> <LI> Distinct turn on fluorescence of MOF-CNCbl nanocomposites in presence of 1 ppm Co2+ ions. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Dielectric and Electro-Optical Properties of Ceramic Nanoparticles Doped Liquid Crystals

Preeti Porov,Vishal Singh Chandel,Rajiv Manohar 한국전기전자재료학회 2016 Transactions on Electrical and Electronic Material Vol.17 No.2

Liquid crystals are important materials because of their applications in display technology and many other scientific applications. Different mixtures of liquid crystals and their doped samples have gained interest because a single liquid crystal compound cannot fulfill all the required parameters for the display application. The doping can be accomplished with dyes, polymers, or composite nanoparticles among other substance. The addition of nanoparticles can modify the physical properties of the host liquid crystal and enhances the performance of electro-optical devices. The present study is focused on investigations of possible changes in dielectric and electro optical properties of liquid crystals caused by doping with ceramic nanoparticles. Including smaller nanoparticles were found to be better candidates for use in suppressing the unwanted ion effects in liquid crystal displays.
