        Realgar transforming solution suppresses angiogenesis and tumor growth by inhibiting VEGF receptor 2 signaling in vein endothelial cells

        Realgar (As4S4), as an arsenic sulfide mineraldrug, has a good therapeutic reputation for anticancer inTraditional Chinese Medicine, and has recently beenreported to inhibit angiogenesis in tumor growth. However,considering the poor solubility and low bioavailability ofrealgar, large dose of realgar and long period of treatmentare necessary for achieving the effective blood medicineconcentration. In present study, we resolved the crucialproblem of poor solubility of realgar by using intrinsicbiotransformation in microorganism, and investigatedunderlying mechanisms of realgar transforming solution(RTS) for antiangiogenesis. Our results demonstrated thatRTS had a strong activity to inhibit HUVECs proliferation,migration, invasion, and tube formation. Moreover, RTSinhibited VEGF/bFGF-induced phosphorylation ofVEGFR2 and the downstream protein kinases includingERK, FAK, and Src. In vivo zebrafish and chickenchorioallantoic membrane model experiments showed thatRTS remarkably blocked angiogenesis. Finally, comparedwith the control, administration of 2.50 mg/kg RTSreached more than 50% inhibition against H22 tumorallografts in KM mice, but caused few toxic effects in thehost. The antiangiogenic effect was indicated by CD31immunohistochemical staining and alginate-encapsulatedtumor cell assay. In summary, our findings suggest thatRTS inhibits angiogenesis and may be a potential drugcandidate in anticancer therapy.

        Precision Manufacturing of a Linear Fiber Assembly with Axially Varying Compositions and Structures by Using Computer Numerically Controlled Ring Spinning

        Ring spinning is an effective technology for forming multi-component spun yarns. There is an increasing need formultifunctional textiles with characteristics, such as antibacterial properties, UV protection, easy cleaning, and coloralteration, which can be achieved via various physical and mechanical processing approaches, such as electrospinning,friction spinning, hollow spindle spinning, and ring spinning. In particular, ring spinning is based on the mechanicalprocessing of fibers, and it is suitable for mass customized production systems. However, conventional ring spinning involvesa fixed fiber blending ratio and yarn linear density, which limits its usefulness for fabricating multifunctional textiles due tothe uncontrollable composite yarn structure. In the present work, we propose a new method for manufacturing a threecomponentspun yarn by combining a computer numerical control system with a ring spinning frame, which increases theflexibility of yarn production. The as-prepared composite yarns possess axially varying compositions and structures, whichcan be readily tuned by changing the feeding speeds of the back rollers, and these aspects can facilitate the fabrication ofmultifunctional textiles for mass production at a low cost. To the best of our knowledge, this is the first work to demonstratethe feasibility of fabricating a three-component spun yarn with tunable compositions and structures by employing a ringspinning frame.

        <P>We report a new reducing agent system: hydriodic acid with trifluoroacetic acid, which can chemically convert graphene oxide into reduced graphene oxide at temperatures below 0 °C in solution.         cAMP induction by ouabain promotes endothelin-1 secretion via MAPK/ERK signaling in beating rabbit atria

        Adenosine 3',5'-cyclic monophosphate (cAMP) participates in the regulation of numerous cellular functions, including the $Na^+-K^+$-ATPase (sodium pump). Ouabain, used in the treatment of several heart diseases, is known to increase cAMP levels but its effects on the atrium are not understood. The aim of the present study was to examine the effect of ouabain on the regulation of atrial cAMP production and its roles in atrial endothelin-1 (ET-1) secretion in isolated perfused beating rabbit atria. Our results showed that ouabain ($3.0{\mu}mol/L$) significantly increased atrial dynamics and cAMP levels during recovery period. The ouabain-increased atrial dynamics was blocked by KB-R7943 ($3.0{\mu}mol/L$), an inhibitor for reverse mode of $Na^+-Ca^{2+}$ exchangers (NCX), but did not by L-type $Ca^{2+}$ channel blocker nifedipine ($1.0{\mu}mol/L$) or protein kinase A (PKA) selective inhibitor H-89 ($3.0{\mu}mol/L$). Ouabain also enhanced atrial intracellular cAMP production in response to forskolin and theophyline ($100.0{\mu}mol/L$), an inhibitor of phosphodiesterase, potentiated the ouabain-induced increase in cAMP. Ouabain and 8-Bromo-cAMP ($0.5{\mu}mol/L$) markedly increased atrial ET-1 secretion, which was blocked by H-89 and by PD98059 ($30{\mu}mol/L$), an inhibitor of extracellular-signal-regulated kinase (ERK) without changing ouabain-induced atrial dynamics. Our results demonstrated that ouabain increases atrial cAMP levels and promotes atrial ET-1 secretion via the mitogen-activated protein kinase (MAPK)/ERK signaling pathway. These findings may explain the development of cardiac hypertrophy in response to digitalis-like compounds.

        Filtering for Discrete-time Linear Noise Delay Systems

        Optimal state estimation problem for discrete-time systems with delays in noise sequence is investigated. Predictor similar to the traditional Kalman ones is derived based on projection formula in Hilbert space. Filter and fixed-lag smoother are obtained at the same time. The estimators are presented by solving two coupled Riccati-type difference equations. One example shows the effectiveness of the proposed approach.

        A Model for Machine Fault Diagnosis based on Mutual Exclusion Theory and Out-of-Distribution Detection

        The primary task of machine fault diagnosis is to judge whether the current state is normal or damaged, so it is a typical binary classification problem with mutual exclusion. Mutually exclusive events and out-of-domain detection have one thing in common: there are two types of data and no intersection. We proposed a fusion model method to improve the accuracy of machine fault diagnosis, which is based on the mutual exclusivity of events and the commonality of out-of-distribution detection, and finally generalized to all binary classification problems. It is reported that the performance of a convolutional neural network (CNN) will decrease as the recognition type increases, so the variational auto-encoder (VAE) is used as the primary model. Two VAE models are used to train the machine's normal and fault sound data. Two reconstruction probabilities will be obtained during the test. The smaller value is transformed into a correction value of another value according to the mutually exclusive characteristics. Finally, the classification result is obtained according to the fusion algorithm. Filtering normal data features from fault data features is proposed, which shields the interference and makes the fault features more prominent. We confirm that good performance improvements have been achieved in the machine fault detection data set, and the results are better than most mainstream models.

      • Nonvolatile Memory Device Using Gold Nanoparticles Covalently Bound to Reduced Graphene Oxide

        <P>Nonvolatile memory devices using gold nanoparticles (AuNPs) and reduced graphene oxide (rGO) sheets were fabricated in both horizontal and vertical structures. The horizontal memory device, in which a singly and doubly overlayered semiconducting rGO channel was formed by simply using a spin-casting technique to connect two gold electrodes, was designed for understanding the origin of charging effects. AuNPs were chemically bound to the rGO channel through a π-conjugated molecular linker. The π-conjugated bifunctional molecular linker, 4-mercapto-benzenediazonium tetrafluoroborate (MBDT) salt, was newly synthesized and used as a molecular bridge to connect the AuNPs and rGOs. By using a self-assembly technique, the diazonium functional group of the MBDT molecular linker was spontaneously immobilized on the rGOs. Then, the monolayered AuNPs working as capacitors were covalently connected to the thiol groups of the MBDT molecules, which were attached to rGOs (AuNP-frGO). These covalent bonds were confirmed by XPS analyses. The current–voltage characteristics of both the horizontal and vertical AuNP-frGO memory devices showed noticeable nonlinear hysteresis, stable write–multiple read–erase–multiple read cycles over 1000 s, and a long retention time over 700 s.         Tailoring the Impact Toughness of Sintered NdFeB Magnets via Surface Coating

        Metallic coating by electroplating is commonly attractive for improving the corrosion resistance of sintered NdFeB magnets. However, its tailoring of mechanical characteristics for sintered NdFeB magnets has been seldom concerned. Herein, the impact toughnesses of sintered NdFeB magnets with various metallic coatings (Ni or Ni/Sn) were comparatively investigated. The results indicate that the impact toughnesses of sintered NdFeB magnets are both improved by Ni coating and Ni/Sn bilayer coating. And Ni/Sn bilayer coating exhibits more enhancement of the impact toughness, increased by 41.6% compared with the original magnet. Moreover, the microstructural observations of the metallic coatings and the fracture were conducted, and the enhanced mechanism of impact toughness for the magnet is analyzed. These findings may provide a reference for toughening the brittle materials.
