        Signaling and Function of Caspase and c-Jun N-terminal Kinase in Cisplatin-induced Apoptosis

        Caspases and c-Jun N-terminal kinase (JNK) are activated in tumor cells during induction of apoptosis. We investigated the signaling cascade and function of these enzymes in cisplatin-induced apoptosis. Treatment of Jurkat T-cells with cisplatin induced cell death with DNA fragmentation and activation of caspase and JNK. Bcl-2 overexpression suppressed activation of both enzymes, whereas p35 and CrmA inhibited only the DEVDase (caspase-3-like) activity, indicating that the activation of these enzymes may be differentially regulated. Cisplatin induced apoptosis with the cytochrome c release and caspase-3 activation in both wild-type and caspase-8-deficient JB-6 cells, while the Fas antibody induced these apoptotic events only in wild-type cells. This indicates that caspase-8 activation is required for Fas-mediated apoptosis, but not cisplatin-induced cell death. On the other hand, cisplatin induced the JNK activation in both the wildtype and JB-6 cells, and the caspase-3 inhibitor ZDEVD- fmk did not inhibit this activation. The JNK overexpression resulted in a higher JNK activity, AP-1 DNA binding activity, and metallothionein expression than the empty vector-transfected cells following cisplatin treatment. It also partially protected the cells from cisplatin-induced apoptosis by decreasing DEVDase activity. These data suggest that the cisplatin- induced apoptotic signal is initiated by the caspase- 8-independent cytochrome c release, and the JNK activation protects cells from cisplatin-induced apoptosis via the metallothionein expression.

        소타액선에 발생한 기저세포선암의 치험례

        Basal cell adenocarcinoma is an epithelial neoplasm which is cytologically and histomorphologically similar to basal cell adenoma but is different because of the infilitrative growth. This tumor, a rare salivary gland tumor newly classified as basal cell adenocarcinoma by the WHO in 1991, is infiltrative, locally destructive and tends to recur but metastasis is less common. The differential diagnosis includes basal cell adenoma, adenoid cystic carcinoma, and basaloid squamous carcinoma. Nearly 90 percent of these tumors occurr in the parotid gland and can be classified into low grade carcinomas with a relative good prognosis. Basal cell adenocarcinoma of minor salivary gland is very rare and has a less favorable clinical course compared with that of the major salivary glands. This is a case of basal cell adenocarcinoma occurring at the minor salivary gland of the soft palate. We treated this patient with block excision and adjunctive radiation therapy.

        김승희 시에 나타난 여성 의식

        This thesis examines Kim Seung-hee's attiude toward women reflected in some of her poems. Her attitude can be summarized as follows: First, Kim Seung-hee, a famous korean woman poetress, throws a keen glance at the order and logic of male-oriented society, where women are oppressed and ill-treated. By showing women's oppression, Kim indicates that women's problems should be solved not only by men but also women themselves. Second, Kim asserts that women should stive to find out the door to the world of freedom and liberation and be born again as human beings. She encourages women to fly high in order to gain absolute purity and time of absolute freedom. Kim shows the ways and the amount of liberation women acquire after being thrown out to the world, breaking the wall with the strenuous effort of hatching. She finds the power in the wounds women experience under painful oppression. She adds that women are also responsible for liberation they have achieved. She asserts that pains can prove to be powerful. If women would be courageous enough to embrace wounds, they could be able to soar high above the wounds and meet the golden stars. Thus, women should not succumb to pains; rather, they need courage and power to embrace and surpass them. Lastly, Kim Seung-hee encourages women to overcome the culture of sexual discrimination and probe the path to liberation and self-realization. Inquiry of women's problems does not simply mean women's liberation, but it aims at the world where every person can live as a human being, free from oppressions. Women have to acknowledge 'the tiger spirit' (masculinity) in them and turn around from the accustomed attitude of 'taking it for granted.' Kim sings for the women who make efforts to fly high and challenge the status quo. In sum, Kim Seung--hee's poems reveal strong women's spirits of overcoming the routinous oppressions, of giving birth to a new life, and of challenging the possibilities the future bringes.

      • 최근 3년간 혈액에서 분리된 세균의 항생제 감수성 양상

        Total 1,661 strains were isolated from blood culture at Seoul National University Hospital during the period of 1984 through 1986. The most frequent isolates were E. coli(16.7%) followed by coagulase-negative, Staphylococci(16.0%), S. aureus(11.2%), Pseudomonas spp(10.4%), S. typhi(7.2%), Klebsiella spp(6.6%). Antibiotic high susceptibility test of S. aureus and coagulase-negative Staphylococci revealed relatively high susceptibility to CEP, but very and coagulase-negative Staphylococci revealed relatively high susceptibility to CEP, but very low susceptibility to PCN, AMP and TC, Enterococei showed 40~70% and 40~94% of susceptibility to PCN and AMP, respectively. S. pneumoniae showed increasing tendency of resistance to PCN. Most of the gram-negative bacilli revealed relatively high susceptibility to GM, AMK and TOB, while, A. calcoaceticus and S. marecscens disclosed susceptibility to AMK as 30~63% and 44~71%, respectively. The proportion of multiresistant bacteria was 2.8% of total number of isolates, A. colcoaceticus var, anilralus(13.8%), Enterccocci(11.8%), S. marcescens(8.6%), S.aureus(5.4%) showed the relatively high proportion of multiresistant strain.

      • A Study on Evaluation of Exfoliation Efficacy and Erythema Relief by Carnitine Salicylate Ion Pair System

        In this study, a carnitine salicylate ion pair (called IP-BHA) was prepared through hydrogen bonding and dipole-dipole interaction, which are known to possess exfoliating and anti-inflammation properties. In an in vitro study, the efficacy of carnitine salicylate on keratin exfoliation at weakly acidic pH and cytotoxicity and alleviation of the pro-inflammatory factor nitric oxide was superior to that of salicylic acid. As a result of analyzing the degree of keratin exfoliation using a colorimeter in the clinical evaluation of keratin peeling performed on the forearms of 12 participants aged 20 ∼ 30 years, the IP-BHA (0.5% salicylic acid) applied experimental group improved by 76.37 ± 9.16% after 14 days compared to the vehicle applied group. Erythema mitigation effect was evaluated by inducing erythema using applying 1% sodium dodecyl sulfate solution to the forearms of 9 participants aged 20 ∼ 30 years for 24 hours. As a result, the IP-BHA applied g roup improved by 74.03 ± 14.82% a f ter 12 days compared to the vehicle applied g roup. Through sensory evaluation, it was verified that the skin texture was improved and skin soothing was felt by IP-BHA. This study indicates that IP-BHA can be used as a cosmetic material with excellent effects on exfoliation and erythema relief.

      • Salmonella typhimurium에 의한 고관절염 1예

        Salmonella septic arthritis is unusual and rare manifestation noted in fewer than 1% of salmonellosis. S. typhimurium is the most prevalent among the salmonella species from clinical specimens including bone and joint in United States and other countries, on the other hand, S. typhi is the most frequent organism in Korea. There has been no case report of septic arthritis caused by S. typhimurium in Korea. We report the first case of septic arthritis due to S. typhimurium in a patient with diabetes mellitus. The patient was a 43-year-old male. He had undergone sequestrectomy and iliac bone graft of the hip joint because of tuberculosis 16 years age, thereafter he took oral anti-tuberculous medication for a year. Six months before admission high fever and copious pus discharge were noted. And he was admitted to the our hospital via local clinic in Cheju Island, where new onset of diabetes mellitus was detected, for further proper managements. There was no history of Salmonella septicemia, dysenteric infection or pulmonary tuberculosis. Physical examination revealed warm, swollen and tender areas with two active draining sinuses on the anterolateral aspect of right hip joint, which was in fused state. And there were also an old operation scar and five healded draining sinuses. Cultures of pus obtained from right hip at reoperation grew group B Salomonella which was positive for H₂S, motility, citrate, fermentation of mannitol and sorbitol. Salmonella was finally identified as S. typhimurium having 1, 4, 5 and 12 of somatic (O) antigen, i on phase 1, 1 and 2 on phase 2 of flagella (H) antigens by Serological tests performing at National Institute of Health. In the other laboratory findings were as follows; WBC 12,000/㎕, Hb/Hct 15.2/46, platelets 510,000/㎕ on CBC, FBS/pp2h:330/482, glucosuria (3+), CRP(3+), Cryglobulin(-), anti-HIV Ab(-), 1:40 in both O and H titer of Widal tests, and HLA B27(-). AFB was not isolated on stained smears and cultures. He was treated with cefradine 1.0g and kanamycin 1.0g initially and chloramphenicol 1.0g daily for 6 weeks. And 8 U RI and 16 U human insulin(Novolyn N) were administrated for DM control. He responded to the treatment and he was retransferred to Cheju local clinic for continuous DM control.

      • Corynebacterium Group JK 분리 1예

        Antimicrobial resistant Corynebacterium group JK (Johnson-Kay strain) has increasingly been reported as causes of serious sepsis in immunosuppressive patients with long period of hospitalization and receiving intensive antibiotic therapy. One characteristic strain Corynebacterium group JK was isolated from surveillance urine culture in 62 year old patient with cervix carcinoma, stage IIIb. The JK strain isolated was aerobic, slow growing catalase-positive, oxidase-negative, fermentative, Gram-positive cocco-bacillus, which was sensitive only to vancomycin and resistant to the other antibiotics tested.
