        This paper describes experimental investigation on the forming limit for mash-seam welded sheets. The uniaxial tensile test was conducted to evaluate the mechanical properties of weld bead. Experimental forming limit diagrams were investigated for the different thicknesses and properties of welded sheets.

      • Baculovirus Vector System에 의해 발현된 재조합 Pseudorabies Virus Major Capsid Protein의 면역원성

        The recombinant pseudorabies virus major capsid protein (rMCP) was produced by expression of the MCP gene in Sf-9 cell using baculovirus transfer vector system. Following evaluation of the immunochemical properties of the rMCP, the immunogenicity of the recombinant subunit protiens were investigated in guinea pig and swine to obtain the preliminary guide line for the subunit vaccine using rMCP and gP50. It was proved that ultrasonication and 30% ammonium sulfate was most efficient to concentrate and purify the protein. The rMCP was safe in mice, guinea pigs and piglets. In guinea pigs, rMCP mixed with various adjuvants induced substantial degree of serum neutralizing antibody titers, but revealed incomplete protecivity against challenge. In swine, the combination of rMCP and gP50 showed the higher serum neutralizing antibody titers and cellular immune responses than rMCP alone. However, the protectivity was lower in comparison with the commercial gI-deleted inactivated vaccine. We expect these results to contribute to characterization of MCP gene of Korean isolate of PRV and to ultilize as preliminary information for prodution and evaluation of PRV recombinant subunit vaccines.

      • 수박 종자에서 Pseudomonas pseudoalcaligenes subsp. citrulli의 분리를 위한 선택배지

        A semiselctive agar medium(PSC) was developed for the isolation of Pseudomonas pseudoalcaligenes subsp. citrulli(Ps.c.) from watermelon seed. The medium contains following per liter : 20g proteose peptone #3, 5.0㎖ glycerol, 1.5g K2HPO4, 1.5g MgSO47H2O, 15g agar, 1㎎ ampicillin, 100㎎ cycloheximide 1㎖ 1.5% bromthymol blue, 1㎖ 1.0% methyl violet and 1㎎ vancomycin. Colonies of Ps.c. were 3-4㎜ in diameter, smooth, round, slight convext, dark green(lighter margin) and glistening after 5 days at 39C on the PSC medium. The recovery of 10 strains including ATTC type 29625, on PSC medium in comparison to KB ranged from 93% to 105%(mean 97.8%). Reduction in seed flora of 10 watermelon seed lots on PSC medium was 97.2%. Recovery of Ps.c. added to watermelon seed ranged from 91.3% to 103.4%(mean 96.7%).

      • Mirizzi 증후군의 변형된 분류와 치료

        Purpose : The Mirizzi syndrome is relatively rare and preoperative diagnosis of this disease is difficult. In 1978, Morelli suggested the subclassification of the Mirizzi syndrome into acute or chronic form. We experienced 5 cases of acute form. We analysed clinical features, preoperative radiologic findings and operative findings of 18 cases including acute forms which were diagnosed as Mirizzi syndrome and should suggest the modified classification of Mirizzi syndrome for choice of appropriate treatment. Method : From January 1995 to December 1998, 18 cases, of which 8 cases were diagnosed at Soonchunhyang University Chunan Hospital, and 10 cases were reported in the Korean Journal were retrospectively analysed with regard to clinical features, preoperative radiologic findings and operative findings. According to the clinical features, whole cases were divided into type Ⅰ(acute form) and type Ⅱ(chronic form) and then each type of cases were subclassified according to preoperative radiologic findings and operative findings. Results : Of 18 cases there were 5 cases in type Ⅰ(27.8%), 13 cases in type Ⅱ(72.2%). Type Ⅱb was most common. Type Ⅰa cases were treated only with cholecystectomy. We applied cholecystectomy, T-tube choledochostomy and patch technique in type Ⅰb and thpe Ⅰc cases. Cholectystectomies including removal of gallstones and internal drainage procedures were done in type Ⅱ chronic forms. Conclusion : The acute form(Type Ⅰ) of Mirizzi syndrome was suggested by Morelli might be subclassified into typeⅠa,Ⅰb and Ⅰc following the presence of the necrotic defect in common hepatic duct. Through the modified classification of Mirizzi syndrome based on clinical feature, preoperative radiologic findings and operative findings, we can choice appropriate treatment.

        흰쥐 해마박편에서 veratrine과 고농도 포타슘자극시 칼슘이온이 glutamic acid 유리에 미치는 영향에 관한 연구 : A role of calcium

        Present study was performed to clarify the effect of calcium on the release of glutamic acid employing hippocampal slices. Hippocampal slices(300∼400㎛ thick) were prepared by the method of Kim et al.(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. In case of veratrine induced glutamic acid release, pre-equilibrated slices were incubated in fresh KBM and then veratrine(25μM)-containing KBM for 10 min period in the presence or absence of 2.5mM Ca^++. In case of potassium-induced glutamic acid release, pre-equilibrated slices were incubated in fresh KBM and then potassium(50mM)-containing KBM for 5 min period in the presence or absence of 2.5mM Ca^++. Basal and veratrine and potassium-induced release of glutamic acid was determined from recovered medium by HPLC. The observed results were as follows: 1. The release of glutamic acid induced by the 10 min-exposure of 25μM veratrine and 5 min-exposure of 50mM potassium in the presence of 2.5mM Ca^++ was 330.9±25 nmol and 129.6±20.6 nmol, respectively. When compared with released amounts of glutamic acid during the corresponding spontaneous periods, these were 7.8 and 5.1-fold increase respectively. 2. the release of glutamic acid induced by the 10 min-exposure of 25μM veratrine and 5 min-exposure of 50mM potassium in the absence of Ca^++ was 361.8±31.9 nmol and 62.5±7.5 nmol, respectively. When compared with released amounts of glutamic acid during the corresponding spontaneous periods, these were 8.2 and 2.4-fold increase respectively.

        The purpose of this paper is to interpret social motives of Contemporary Korean architecture. Sociological interpretation of architecture have need to study coditions of politics, economics, society at that time. Theory of the SIM(Sociological interpretation Model) is found on 'Sociology of knowledge'. The SIM is consisted of three steps as follows; 1) To inquire into dominant ideology in the society. 2) To inquire into structure of society due to ideology. 3) To inquire into interrelation between architecture and structure of society. Contemporary Korean architecture is interpreted through the SIM as follows; 1) Historical style was influenced by Nationalism ideology. 2) Authoritarian style was influenced by Centralism ideology. 3) Modernistic style was influenced by Growthism ideology.

      • 실험 방광암 모델에 있어서 Proliferating Cell Nuclear Antigen이 발현되는 세포들의 색상-화상 분석

        Proliferating cell nuclear antigen(PCNA)에 대한 단클론성 항체에 의해 면역조직화학적으로 염색된 세포들의 평가에 있어서 색상-화상 분석의 역할을 살펴보고자 하였다. BBN 투여 기간에 따라 Fisher 344 백서 방광에 정상 상피로부터 단순 과증식, 결절성 혹은 유두상 과증식 및 표재성 방광암에 이르는 순차적인 병리세포학적 변화가 발생함을 관찰한 후, 이를 색상-화상 분석과 육안적 분석을 통해 PCNA에 대한 평균 labeling index를 구한 결과 두가지 방법에서 모두 각 병변 사이에 통계적으로 유의한 차이를 보였으나, 색상-화상 분석을 이용하여 구한 평균 labeling index의 변이계수가 육안적 분석을 이용하여 구한 평균 labeling index의 변이계수에 비해 전 병변에 대하여 낮게 나타나, 종양화과정이 진행함에 따라 PCNA에 대한 평균 labeling index가 증가하고 평균 labeling index가 높을 수록 증식도가 더 큼을 나타낸다는 결과를 얻었으며, 색상-화상 분석을 이용하여 얻은 결과가 육안적 분석을 이용하여 얻은 결과보다 신뢰도가 더 높은 것으로 나타났다. 이와같은 면역조직화학염색에 대한 색상-화상 분석을 통한 세포역동학적 연구는 정량화 및 재현성의 향상을 통해 면역조직화학염색의 평가에 있어 보다 신뢰도가 높은 정보를 제공하여줄 것이며, 이를 통해 방광종양의 증식도 및 악성도를 예측하는데 중요한 역할을 할 것으로 생각된다. A study was carried out to define the role of color-image analysis in evaluation of immunohistochemically labeled cells using anti-PCNA/cyclin monoclonal antibody. Sequential cellular changes(normal, simple hyperplasia, nodular or papillary hyperplasia, and transitional cell carcinoma-Ta, T1) were observed in Fisher 344 rat urinary bladder depending on the duration of BBN administration. The mean PCNA labeling index obtained by color-image analysis and visual analysis in normal, simple hyperplasia, nodular or papillary hyperplasia and bladder tumor showed statistically significant difference in each histological group. But coefficients of variation of mean PCNA labeling index obtained by color-image analysis were lower than those using visual analysis in all lesions. Higher PCNA labeling indices indicated greater biological malignant potential, and mean labeling index in each histological group obtained by both color-image and visual analysis progressively increased with tumorigenesis, but the coefficient of variation with color-image analysis was lower than that with visual analysis in general, therefore, the evaluation of labeling indices obtained using color-image analysis was more reliable than that obtained using visual analysis. The data suggest that cell kinetic evaluation using color-image analysis may provide more reliable information to evaluate immunohistochemical staining in terms of quantitation and reproducibility and play an important role in predicting proliferative activity and malignant potential for bladder tumor.
