It has been found that a number of diseases are associated with mutations in the mitochondrial DNA. Therapeutic gene delivery to mitochondria has been suggested as a clinical option for these diseases. In this study, we developed a gene carrier to mitochondria by the conjugation of mitochondrial leader peptide (LP) to polyethylenimine (PEI). Mitochondrial LP conjugated PEI (PEI-LP) was synthesized with low molecular weight PEI (2,000 Da, PEI2K). Gel retardation assay showed that PEI2K-LP formed complexes at a 1.0/1 weight ratio. In addition, PEI2K-LP protected DNA from the enzymatic degradation for at least 60 min, while naked DNA was completely degraded within 20 min. PEI2K-LP was compared with LP conjugated high molecular weight PEI (25,000 Da, PEI25K) in terms of toxicity and delivery efficiency. MTT assay showed that PEI2K-LP had much lower cytotoxicity than PEI25K-LP to 293 cells. In addition, cell-free DNA delivery assay showed that PEI2K-LP delivered more DNA to mitochondria at a 1.8/1 weight ratio than naked DNA or PEI. This result suggests that PEI2K-LP may be useful for the development of mitochondrial gene therapy system with lower cytotoxicity.

The purpose of this study is to examine the toxic effects caused by Glucose Oxidase(GO) and the effects of herbal extracts such as Acori Rhizoma(AR) on the treatment of the toxic effects. For this purpose, experiments with the cultured cell from the cerebrums of new born mice were done. The results of these experiments were as follows. 1. GO, a oxygen radical, decreased the survival rate of the cultured cells on NR assay and MTT assay. 2. GO, a oxygen radical, decreased the amount of neurofilaments and total protein. 3. AR have efficacy of increasing the amount of neurofilament. 4. AR have efficacy of increasing the amount of total protein. From the above results, It is concluded that AR has marked efficacy as a treatment for the damages caused in the GO-mediated oxidative process. And AR is thought to have certain pharmacological effects on controlling over aging. Further clinical study of this pharmacological effects of AR should be complemented.

Foot-and-mouth disease (FMD) is a very deadly and highly contagious disease that occurs in various cloven-hoofed animals. Korea imports vaccines including non-vaccinated serotype for the vaccine bank. The two company vaccines imported from 2019 to 2021 were evaluated through multiple tests based on national standard assay. SN titers for the vaccine of company A were more than 1.5 log10, those of company B vaccine in the geometric mean were at least 1.72. All imported vaccines exceeded the evaluation criteria, and the potency of each vaccine was above its own company standard. Stockpiled vaccines for emergency may help to control non-vaccinated serotypes of FMD outbreak nationally.

Recently, research on various metabolic diseases such as obesity and diabetes has been in the spotlight. A representative example of this is the study of metabolic function analysis using Metabolic cage. It is believed that these studies will greatly contribute to the prevention and treatment of various metabolic diseases in humans. This study focuses on the study of obesity in mice. Mouse metabolic phenotyping has been widely used for detecting metabolic disorder in genetically engineered mouse. Korea Mouse Phenotyping Center (KMPC), nation-wide program for mouse production and phenotyping in Korea has been establishing several pipelines for disease-specific mouse phenotyping to support the mouse research. KMPC has been providing mouse metabolic phenotyping including high fat diet, exercise and cold challenges. Multiple parameter including energy expenditure (EE), O2/CO2 consumption (RER), heat generation and activity has been provided with histology service and body composition. Also temperature can be measured during metabolic chamber with telemetric system. Brown adipogenesis is one of key mechanism for developing new candidate drug target for curing obesity and type 2 diabetes. Several factors including cold, adrenergic b3 agonist and exercise are well known factor leading to brown adipogenesis. Here we introduce mouse metabolic phenotyping services with environmental control chamber at KMPC. Temperature-controlled metabolic cage, the climate chambers can control temperatures from 4 °C to 35 °C, while measuring energy consumption using oxygen consumption, carbon dioxide production, and respiratory exchange ratio and food intake. Using this, exposing the mouse to a low temperature environment can activate BAT by browning WAT, which causes various diseases such as heart disease, diabetes, and obesity. The changes of metabolic parameter was finely measured during cold exposure with temperature-controlled metabolic cage.
The improving effects of Saengmaeksan on ulcerative colitis

Saengmaeksan (SMS) is a well known Korean traditional herbal prescription, which consists of 3 different herbs, Radix Ophiopogonis, Radix Ginseng, and Fructus Schisandrae. SMS has been commonly used in various disease including respiratory system and cardiovascular System in Korea. The objective of this study is to find out the improving effects of SMS against Dextran Sulfate Sodium (DSS)-induced ulcerative colitis. SMS reduced clinical signs of DSS-induced colitis, including body weight loss, shorten colon length, and increased disease activity index. The results showed that SMS significantly inhibited the activation of nuclear factor-${\kappa}B$ p65 in the colon tissues of DSS-treated mice. In addition, we observed that result showed that the levels of IL-6 in plasma were increased in DSS treated group compared to those of the normal group, but these increased levels were reduced by administration with SMS. Taken together, these findings suggest that SMS has improving effects on DSS-induced ulcerative colitis, which may explain its beneficial effect in the regulation of chronic intestinal inflammation.
Risk of osteoporosis and fracture in long-term breast cancer survivors

Purpose: High incidence of osteoporosis has been reported in breast cancer patients due to early menopause triggered by adjuvant treatment and temporary ovarian function suppression. In this study, we sought to determine whether long-term breast cancer survivors had an elevated risk of low bone density compared to the general population. Methods: Long-term breast cancer survivors who had been treated for more than 5 years were selected for this study. Data were obtained from medical records and using a questionnaire from the Korea National Health and Nutrition Examination Survey (KNHANES). An agematched non-cancer control group was selected from the KNHANES records. Incidence of fracture and bone mineral density (BMD) were compared between the two groups. Results: In total, 74 long-term breast cancer survivors and 296 non-cancer controls were evaluated. The incidence of fracture did not differ between the two groups (P=0.130). No differences were detected in lumbar BMD (P=0.051) following adjustment for body mass index, while hip BMD was significantly lower in breast cancer survivors (P=0.028). Chemotherapy and endocrine treatment were not related to low BMD in breast cancer survivors. In more than half of the survivors, the 10-year risk of osteoporotic fracture was less than 1%. Conclusion: Long-term breast cancer survivors had low bone density but a comparable risk of fracture compared to non-cancer agematched controls. Further studies on the factors related to low bone density in long-term breast cancer survivors are required

This paper presents high a PSR low drop-out (LDO) voltage regulator without external capacitor. To satisfy stability, we use nested miller compensation using small internal capacitors. Also, we connect PSR enhancer to the gate of pass transistor to get high PSR at DC area. It regulates output at 1.8V from 2V-3.3V supply voltages and get -102dB PSR at 10uA load current using 5.3pF internal capacitors.
