        The quorum sensing regulator OpaR is a repressor of polar flagellum genes in Vibrio parahaemolyticus

        Vibrio parahaemolyticus possesses two types of flagella: asingle polar flagellum (Pof) for swimming and the peritrichouslateral flagella (Laf) for swarming. Expression of Lafgenes has previously been reported to be regulated by the quorumsensing (QS) regulators AphA and OpaR. In the presentstudy, we showed that OpaR, the QS regulator at high cell density(HCD), acted as a negative regulator of swimming motilityand the transcription of Pof genes in V. parahaemolyticus. OpaR bound to the promoter-proximal DNA regionsof flgAMN, flgMN, and flgBCDEFGHIJ within the Pof geneloci to repress their transcription, whereas it negatively regulatesthe transcription of flgKL-flaC in an indirect manner. Thus, this work investigated how QS regulated the swimmingmotility via direct action of its master regulator OpaR onthe transcription of Pof genes in V. parahaemolyticus.

        Experimental and numerical study on pre-peak cyclic shear mechanism of artificial rock joints

        The pre-peak cyclic shear mechanism of two-order asperity degradation of rock joints in the direct shear tests with static constant normal loads (CNL) are investigated using experimental and numerical methods. The laboratory testing rock specimens contains the idealized and regular two-order triangular-shaped asperities, which represent the specific geometrical conditions of natural and irregular waviness and unevenness of rock joint surfaces, in the pre-peak cyclic shear tests. Three different shear failure patterns of two-order triangular-shaped rock joints can be found in the experiments at constant horizontal shear velocity and various static constant normal loads in the direct and pre-peak cyclic shear tests. The discrete element method is adopted to simulate the pre-peak shear failure behaviors of rock joints with two-order triangular-shaped asperities. The rock joint interfaces are simulated using a modified smooth joint model, where microscopic scale slip surfaces are applied at contacts between discrete particles in the upper and lower rock blocks. Comparing the discrete numerical results with the experimental results, the microscopic bond particle model parameters are calibrated. Effects of cyclic shear loading amplitude, static constant normal loads and initial waviness asperity angles on the pre-peak cyclic shear failure behaviors of triangular-shaped rock joints are also numerically investigated.

        Emulsifying properties of different soy hull pectin polysaccharides and application in mayonnaise

        Soy hull pectic polysaccharide (SHPP) is a kindof biological macromolecule prepared from soy hull, whichhas certain thickening and gel properties. In present study,SHPP was extracted with citric acid and sodium citratefrom soybean hulls under the assistance of microwave,respectively. SHPPs were then compared with commercialpectin polysaccharide to test their emulsification ability. The emulsion prepared from SHPP extracted with sodiumcitrate has the best emulsifying effect, small particle sizeand uniform distribution. The rheological properties andparticle size distribution of mayonnaise did not changesignificantly after the addition of different SHPPs andcommercial pectin polysaccharides. However, microscopicobservations revealed that the droplets were more uniformlydistributed in the mayonnaise after the addition ofSHPP extracted with sodium citrate under the assistance ofmicrowave and commercial pectin APC141. SHPPextracted with sodium citrate may play an important role inmaintaining emulsion stability in the future.

        Development of Recombinase Polymerase Amplification Combined with Lateral Flow Strips for Rapid Detection of Cowpea Mild Mottle Virus

        Cowpea mild mottle virus (CPMMV) is a global plant virus that poses a threat to the production and quality of legume crops. Early and accurate diagnosis is essential for effective managing CPMMV outbreaks. With the advancement in isothermal recombinase polymerase amplification and lateral flow strips technologies, more rapid and sensitive methods have become available for detecting this pathogen. In this study, we have developed a reverse transcription recombinase polymerase amplification combined with lateral flow strips (RT-RPA-LFS) method for the detection of CPMMV, specifically targeting the CPMMV coat protein (CP) gene. The RT-RPA-LFS assay only requires 20 min at 40°C and demonstrates high specificity. Its detection limit was 10 copies/μl, which is approximately up to 100 times more sensitive than RT-PCR on agarose gel electrophoresis. The developed RT-RPA-LFS method offers a rapid, convenient, and sensitive approach for field detection of CPMMV, which contribute to controlling the spread of the virus.

        VDR promotes testosterone synthesis in mouse Leydig cells via regulation of cholesterol side chain cleavage cytochrome P450 (Cyp11a1) expression

        Background The vitamin D receptor (VDR) mediates the pleiotropic biological actions that include osteoporosis, immune responses and androgen synthesis.VDR is widely expressed in testis cells such as Leydig cells, Sertoli cells, and sperm. The levels of steroids are critical for sexual development. In the early stage of steroidogenesis, cholesterol is converted to pregnenolone (precursor of most steroid hormones) by cholesterol side-chain lyase (CYP11A1), which eventually synthesizes the male hormone testosterone. Objective This study aims to reveal how VDR regulates CYP11A1 expression and affects testosterone synthesis in murine Leydig cells. Methods The levels of VDR, CYP11A1 were determined by quantitative real-time polymerase chain reaction (RT-qPCR) or western blot. Targeted relationship between VDR and Cyp11a1 was evaluated by dual-luciferase reporter assay. The levels of testosterone concentrations in cell culture media serum by enzyme-linked immunosorbent assay (ELISA). Results Phylogenetic and motif analysis showed that the Cyp11a1 family had sequence loss, which may have special biological functions during evolution. The results of promoter prediction showed that vitamin D response element (VDRE) existed in the upstream promoter region of murine Cyp11a1. Dual-luciferase assay confirmed that VDR could bind candidate VDREs in upstream region of Cyp11a1, and enhance gene expression. Tissue distribution and localizatio analysis showed that Cyp11a1 was mainly expressed in testis, and dominantly existed in murine Leydig cells. Furthermore, over-expression VDR and CYP11A1 significantly increased testosterone synthesis in mice Leydig cells. Conclusions Active vitamin D3 (VD3) and Vdr interference treatment showed that VD3/VDR had a positive regulatory effect on Cyp11a1 expression and testosterone secretion. VDR promotes testosterone synthesis in male mice by up-regulating Cyp11a1 expression, which played an important role for male reproduction.
