      • Weighted Gene Co-expression Network Analysis in Identification of Endometrial Cancer Prognosis Markers

        Objective: Endometrial cancer (EC) is the most common gynecologic malignancy. Identification of potential biomarkers of EC would be helpful for the detection and monitoring of malignancy, improving clinical outcomes. Methods: The Weighted Gene Co-expression Network Analysis method was used to identify prognostic markers for EC in this study. Moreover, underlying molecular mechanisms were characterized by KEGG pathway enrichment and transcriptional regulation analyses. Results: Seven gene co-expression modules were obtained, but only the turquoise module was positively related with EC stage. Among the genes in the turquoise module, COL5A2 (collagen, type V, alpha 2) could be regulated by PBX (pre-B-cell leukemia homeobox 1)1/2 and HOXB1(homeobox B1) transcription factors to be involved in the focal adhesion pathway; CENP-E (centromere protein E, 312kDa) by E2F4 (E2F transcription factor 4, p107/p130-binding); MYCN (v-myc myelocytomatosis viral related oncogene, neuroblastoma derived [avian]) by PAX5 (paired box 5); and BCL-2 (B-cell CLL/lymphoma 2) and IGFBP-6 (insulin-like growth factor binding protein 6) by GLI1. They were predicted to be associated with EC progression via Hedgehog signaling and other cancer related-pathways. Conclusions: These data on transcriptional regulation may provide a better understanding of molecular mechanisms and clues to potential therapeutic targets in the treatment of EC.

        Microcrystal Structure and C/O Element Occurrence State of Diesel PM by Non-Thermal Plasma Oxidation at Different Reaction Temperatures

        To reveal the effect of reaction temperature on the reduction of diesel particulate matter (PM) by non-thermal plasma (NTP) using oxygen as a gas source. The changes in the microcrystalline structure and the elemental state of PM before and after NTP oxidation at different temperatures were explored by Raman and X-ray photoelectron spectroscopy. After NTP oxidation, the disorder in the PM microcrystal structure and the amorphous carbon structure was reduced. The full width at half maximum (FWHM) of the D1 and D3 peaks decreased, and the FWHM of the G peak increased slightly. During the oxidation of PM, the carbon microcrystals grew and became restructured, and the graphitization of PM increased. After NTP oxidation, the content of O in PM increased as the reaction temperature increased, resulting in a gradual change in the binding form of O with C from C-O to C=O. The ability of temperature rise to promote the oxidation activity of NTP was gradually weakened for the thermal decomposition of NTP active substances. The microcrystalline structure and the occurrence state of C and O of PM changed with reaction temperature, indicating that the oxidizability of NTP on PM differed at different reaction temperatures.

        Post-meiotic Deficient Anther1 (PDA1) Encodes an ABC Transporter Required for the Development of Anther Cuticle and Pollen Exine in Rice

        The tapetum of the anther locule encloses the malereproductive cells and plays a supportive role for normalpollen development. However, the underlying mechanismremains less understood. Previously, we identified a completerecessive male sterile mutant, post-meiotic deficient anther1(pda1), with abnormal postmeiotic tapetal development. Inthis study we comprehensively characterized pda1. Chemicalanalysis uncovered that pda1 anther had significant lowerlevels of cutin monomers and cuticular waxes. PDA1 geneencodes an ATP-binding cassette (ABC) half-transporter,namely OsABCG15, which is conserved from algae tohigher plants. In situ RNA hybridization assay showed thatPDA1 is strongly expressed in tapetal cells, and weakly inmicrospores during the anther development. Additionally,the expression of two pollen exine biosynthetic genesCYP704B2 and CYP703A3 was dramatically reduced inpda1 mutant anthers. Altogether, these observations suggestthat the tapetum-expressed ABC transporter PDA1 plays acrucial role in secreting lipidic precursors from the tapetumto developing microspores and the anther epidermis.

      • Business-Driven Process Fragment Selections in RESTful Business Processes

        Past work showed that runtime adaptability of business processes can be improved by applying Representational State Transfer (REST) to design and implementation of business processes. However, the existing solutions for RESTful business processes (RESTfulBP) were focused on manual selection of process fragments to be composed at runtime. In this paper, we propose solutions that enable semi-automatic selection of process fragment at each decision-making point of RESTfulBP. The new built-in middleware MiniZnMASC can provide user based process fragment advice to knowledge workers in ways that achieve better overall business value while satisfying all existing constraints. In addition, we redesign the architecture of RESTfulBP in order to allow the business-driven decision-making solutions. The solutions are evaluated for feasibility, functional correctness, business benefits, and performance.

        Analysis of the Genome Sequence of Strain GiC-126 of Gloeostereum incarnatum with Genetic Linkage Map

        Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum.

        Genetic linkage map construction and quantitative trait loci mapping of agronomic traits in Gloeostereum incarnatum

        Gloeostereum incarnatum is an edible medicinal mushroom widely grown in China. Using the whole genome of G. incarnatum, simple sequence repeat (SSR) markers were developed and synthetic primers were designed to construct its first genetic linkage map. The 1,048.6 cm map is composed of 10 linkage groups and contains 183 SSR markers. In total, 112 genome assembly sequences were anchored, representing 16.43 Mb and covering 46.41% of the genome. Selfing populations were used for quantitative trait loci (QTL) targeting, and the composite interval mapping method was used to co-localize the mycelium growth rate (potato dextrose agar and sawdust), growth period, yield and fruiting body length, and width and thickness. The 14 QTLs of agronomic traits had LOD values of 3.20–6.51 and contribution rates of 2.22– 13.18%. No linkage relationship was found between the mycelium growth rate and the growth period, but a linkage relationship was observed among the length, width and thickness of the fruiting bodies. Using NCBI’s BLAST alignment, the genomic sequences corresponding to the QTL regions were compared, and a TPR-like protein candidate gene was selected. Using whole-genome data, 138 candidate genes were found in four sequence fragments of two SSR markers located in the same scaffold. The genetic map and QTLs established in this study will aid in developing selective markers for agronomic traits and identifying corresponding genes, thereby providing a scientific basis for the further gene mapping of quantitative traits and the marker-assisted selection of functional genes in G. incarnatum breeding programs.

      • Cecropin Suppresses Human Hepatocellular Carcinoma BEL-7402 Cell Growth and Survival in vivo without Side-Toxicity

        Conventional chemotherapy against hepatocellular carcinoma typically causes various side effects. Our previous study showed that cecropin of Musca domestica can induce apoptosis in human hepatocellular carcinoma BEL-7402 cells in vitro. However, whether cecropin inhibits BEL-7402 cell in vivo and the question of possible side effects remained undentified. The present study confirmed tumor-inhibitory effects of cecropin in vivo, and furthermore strongly suggested that cecropin cytotoxicity in BEL-7402 cells in vivo may be mainly derived from its pro-apoptotic action. Specifically, we found that cecropin exerted no obvious side effects in tumor-bearing mice as it had no significant hematoxicity as well as visceral toxicity. Therefore, cecropin may be a potential candidate for further investigation as an antitumor agent against hepatocellular carcinoma.

        Tension–Shear Experimental Analysis and Fracture Models Calibration on Q235 Steel

        Tension–shear loading is a common loading condition in steel structures during the earthquake shaking. To study ductile fracture in structural steel under multiple stress states, experimental investigations on the diff erent fracture mechanisms in Chinese Q235 steel were conducted. Diff erent tension–shear loading conditions achieved by using six groups of inclined notch butterfl y confi gurations covering pure shear, tension–shear and pure tension cases. Numerical simulations were carried out for all the specimens to determine the stress and strain fi elds within the critical sections. Two tension–shear fracture models were calibrated based on the hybrid experimental–numerical procedure. The equivalent fracture strain obtained from the round bar under tensile loading was used for evaluating these two models. The results indicated that the tension–shear criterion as a function of the shear fracture parameter had better performance in predicting the fracture initiation of structural steel under diff erent loading conditions.

        In vitro differentiation of spermatogonial stem cells using testicular cells from Guangxi Bama mini-pig

        In this study, we attempted to establish a culture system for in vitro spermatogenesis from spermatogonial stem cells (SSCs) of Bama mini-pig. Dissociated testicular cells from 1-month-old pigs were co-cultured to mimic in vivo spermatogenesis. The testicular cells were seeded in minimum essential medium alpha (a-MEM) supplemented with Knockout serum replacement (KSR). Three-dimensional colonies formed after 10 days of culture. The colonies showed positive staining for SSC-associated markers such as UCHL1, PLZF, THY1, OCT4, Dolichos biflorus agglutinin, and alkaline phosphatase. Induction of SSCs was performed in a-MEM + KSR supplemented with retinoic acid, bone morphogenetic protein 4, activin A, follicle-stimulating hormone, or testosterone. The results showed that STRA8, DMC1, PRM1, and TNP1 were upregulated significantly in the colonies after induction compared to that in testis from 1-month-old pigs, while expression levels of those genes were significantly low compared to those in 2-month-old testis. However, upregulation of ACROSIN was not significant. Replacement of a-MEM and KSR with Iscove’s modified Dulbecco’s medium and fetal bovine serum did not upregulate expression of these genes significantly. These results indicate that SSCs of Bama mini-pig could undergo differentiation and develop to a post-meiotic stage in α-MEM supplemented with KSR and induction factors.
