Engineered bone tissue is thought to be the ideal alternative for bone grafts in the treatment of related bone diseases. BMP9 has been demonstrated as one of the most osteogenic factors, and enhancement of BMP9-induced osteogenesis will greatly accelerate the development of bone tissue engineering. Here, we investigated the effect of insulin-like growth factor 1 (IGF1) on BMP9-induced osteogenic differentiation, and unveiled a possible molecular mechanism underling this process. We found that IGF1 and BMP9 are both detectable in mesenchymal stem cells (MSCs). Exogenous expression of IGF1 potentiates BMP9-induced alkaline phosphatase (ALP), matrix mineralization, and ectopic bone formation. Similarly, IGF1 enhances BMP9-induced endochondral ossification. Mechanistically, we found that IGF1 increases BMP9-induced activation of BMP/Smad signaling in MSCs. Our findings demonstrate that IGF1 can enhance BMP9-induced osteogenic differentiation in MSCs, and that this effect may be mediated by the enhancement of the BMP/Smad signaling transduction triggered by BMP9. The esterase gene Est8 from the thermophilic bacterium Bacillus sp. K91 was cloned and expressed in Escherichia coli. The monomeric enzyme exhibited a theoretical molecular mass of 24.5 kDa and an optimal activity around 50°C at pH 9.0. A model of Est8 was constructed using a hypothetical YxiM precursor structure (2O14_A) from Bacillus subtilis as template. The structure showed an α/β-hydrolase fold and indicated the presence of a typical catalytic triad consisting of Ser-11, Asp-182, and His-185, which were investigated by site directed replacements coupled with kinetic characterization. Asp-182 and His-185 residues were more critical than the Ser-11 residue in the catalytic activity of Est8. A comparison of the amino acid sequence showed that Est8 could be grouped into the GDSL family and further classified as an SGNH hydrolase. Est8 is a new member of the SGNH hydrolase subfamily and may employ a different catalytic mechanism.
Breviscapine Ameliorates Fear Extinction and Anxiety in BALB/cJ Mice

Objective In this research, the influence of breviscapine on anxiety, fear elimination, and aggression and the potential mechanism was investigated. Methods Anxiety and locomotion were analyzed by elevated plus maze and open field test in mice. Bussey-Saksida Mouse Touch Screen Chambers were used to perform fear conditioning. Territorial aggression was assessed by resident intruder test. Protein levels were evaluated by Western blot. Breviscapine improved fear-extinction learning in BALB/cJ mice. Results Breviscapine at 20-100 mg/kg increased center cross number, total distance traveled, and velocity in a dose-dependent manner. On the other hand, breviscapine at 20-100 mg/kg decreased the immobility time in open field test. In addition, breviscapine at 20-100 mg/kg increased the ratio of time on the open arm, time on the distal parts of the open arm, and total distance traveled in elevated plus maze. Breviscapine at 100 mg/kg increased the average attack latency and decreased the number of attacks over the last 3 days of resident intruder test. In hippocampus, protein levels of postsynaptic density protein-95 and synaptophysin were elevated by breviscapine at these three doses. Conclusion The administration of breviscapine alleviates fear extinction, anxiety, and aggression, while increases locomotor in a dose-dependent manner, which might be associated with its influence on synaptic function.

Purpose: To investigate the efficacy and safety of selective radiotherapy after distant metastasis of nasopharyngeal carcinoma (NPC) treated with dose-dense cisplatin plus fluorouracil. Materials and Methods: Eligible patients were randomly assigned to a study group treated with dose-dense cisplatin plus fluorouracil following selective radiotherapy and a control group receiving traditional cisplatin plus fluorouracil following selective radiotherapy according to a 1:1 distribution using a digital random table method. The primary endpoint was overall survival (OS). Secondary endpoints were progression-free survival (PFS), objective response rate, relapse or progression rate in the radiation field and treatment toxicity. Results: Of 52 patients in the study group, 20 cases underwent radiotherapy., while in the control group of 51 patients, 16 underwent radiotherapy. The median PFS, median OS, survival rates in 1, 2 and 3 years in study and control group were 20.9 vs 12.7months, 28.3 vs 18.8months, 85.2%vs 65.9%, 62.2% vs 18.3%, and 36.6%vs 5.2% (p values of 0.00, 0.00, 0.04, 0.00 and 0.00, respectively). Subgroup analysis showed that the median OS and survival rates of 1, 2, 3 years for patients undergoing radiotherapy in the study group better than that in control group( 43.2vs24.1 months, 94.1% vs 86.7%, 82.4% vs 43.3%, 64.7% vs 17.3%, (p=0.00, 0.57, 0.04 and 0.01, respectively). The complete response rate, objective response rate after chemotherapy and three months after radiotherapy, relapse or progression rate in radiation field in study group and in control group were 19.2% vs 3.9%, 86.5% vs 56.9%, 85% vs 50%, 95% vs 81.3% and 41.3% vs 66.7% (p =0.03, 0.00, 0.03,0.30, 0.01 respectively). The grade 3-4 acute adverse reactions in the study group were significantly higher than in the control group (53.8% vs 9.8%, p=0.00). Conclusions: The survival of patients benefits from selective radiotherapy after distant metastasis of NPC treated with dose-dense cisplatin plus fluorouracil.

<P>A Sn-doped Ga<SUB>2</SUB>O<SUB>3</SUB> thin film was deposited on a n<SUP>+</SUP>-Si substrate by metal-organic chemical vapor deposition. The Ga<SUB>2</SUB>O<SUB>3</SUB> film was found to be amorphous-like and exhibited n-type conduction with Sn doping. Room-temperature electroluminescence was clearly observed from the Sn:Ga<SUB>2</SUB>O<SUB>3</SUB>/n<SUP>+</SUP> -Si diode, including an ultraviolet (UV) emission at ~ 370 nm, a yellow emission at ~ 580 nm, and a red emission at ~ 680 nm. The UV emission is assigned to the transition from Sn<SUB>Ga</SUB> donor to the <I>V</I><SUB>Ga</SUB> acceptor, whereas the visible emissions were assigned to be related to the dangling bond defects.</P>
Optimization of GEMV on Intel AVX Processor

To improve the performance of BLAS 2 GEMV subroutine under the latest instruction set, Intel AVX, this paper presents a new approach to analyze the new generation instruction set and enhance the efficiency of current data-oriented math subroutines. The whole optimizing process involves memory access optimization, SIMD optimization and parallel optimization. Also, this paper shows the comparison between the traditional SSE instruction set and the AVX instruction set. Experiments show that the optimized GEMV function has obtained considerable increase on performance. Compared with the Intel MKL, GotoBLAS, ATLAS, this optimized GEMV exceeds these BLAS implementations from 5% to 10%.
Thickness effects of aerosol deposited hygroscopic films on ultra-sensitive humidity sensors

<P><B>Abstract</B></P> <P>Hygroscopic film that has a super mesoporous structure exhibits high sensitivity and fast response/recovery in humidity sensing applications. Aerosol deposition (AD) via a shock-loading-solidification preparation mechanism, which is an unexplored potential hygroscopic film preparation technique, can produce a porous microstructure, and the AD hammering effect creates various interior and surface microstructures in different-thickness films. The objective of our study was to verify the feasibility of using AD as a humidity-sensing film preparation technique, and to investigate the thickness effects of the film on its microstructure and hygroscopic properties. Hygroscopic films with thicknesses ranging from 0.1–10.0 μm were aerosol deposited with BaTiO<SUB>3</SUB> powders before undergoing a 400 °C thermal treatment. The resulting surface morphology exhibited roughness increases for thicknesses from 0.1–6.0 μm, and a decrease at 10.0 μm. The cross-sectional structure was characterized by a transitional-density grain distribution in which higher density small-sized grains could be seen in the bottom layer. Films with thicknesses of 1.5 μm and 3.0 μm achieved excellent sensitivities of 178.6 ± 7.3 pF/%RH and 299.9 ± 5.4 pF/%RH, respectively. Further, the response and recovery time for the 1.5 μm film were both less than 5 s. The current study determined the feasibility of AD-based humidity-sensing film preparation and provided a reference for optimal thickness control. The surface hydrophilicity, pore volume, and open-pore ratio were analyzed as critical factors of the thickness related humidity sensing effects, and physical modeling indicated that AD-prepared films exhibited an expanded humidity detection range, enhanced water vapor adsorption and desorption, and improved sensitivity to humidity.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Aerosol deposition can be used to prepare ultra-sensitive ceramic humidity sensors. </LI> <LI> Film-based sensors were prepared using a shock-loading solidification mechanism of aerosol deposition. </LI> <LI> Their sensitivity depends on the hydrophilicity, pore volume, and open-pore ratio. </LI> <LI> The thickness of the film regularly affects the humidity-sensing properties. </LI> </UL> </P>
