        치환 Terpyridine 리간드를 가진 옥소루테늄 착물에 의한 알켄산화반응의 반응속도론적 연구

        The 4'-substituted terpyridines(Xtpy; Phtpy, EtOtpy, Cltpy, Me_(2)Ntpy) were synthesized. Their spectral properties have been investigated by means of ^(1)H-NMR and mass spectrometer. Also, a series of new aqua- of oxo-ruthenium complexes containing the tridentate meridonal ligand Xtpy ligand substituted in the 4' positions with electron-donating and electron withdrawing groups of general formula [Ru(Y)(Xtpy)(bpy)]^(2+)(X = ph, EtO, Cl, Me_(2)N ; Y = H_(2)O, O) have been prepared and characterized. Their specral properties have been investigated by means of UV-Vis, ^(1)H-NMR and FT-IR spectrometer. The kinetics of styrene oxidation by [RuO(Xtpy)(bpy)]^(2+) complexes were studied spectrophotometrically by monitoring the increasing of the 400-500nm of oxo ruthenium complexes in acetonitrile. The results of the oxidation reaction clearly demonstrate that the rate of oxidation is dependent on the electronic properties of the oxoruthenium complexes. The oxidation reactions were studied at three different temperatures(15, 25, 35℃). The experimental data support the operation of an associative interchange mechanism in which cleavage of the Ru=O bond and formation of an O-styrene bond have equal importance.

      • 12-O-Tetradecanoylphorbol-13-Acetate에 의한 HL-60 세포 분화유도중 Vimentin 유전자 전사조절에 대한 AP-1의 역할

        Purpose: To gain insight on the role of AP-1 in transcriptional regulation of vimentin gene during differentiation of HL-60 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA), the levels of vimentin mRNA and AP-1 have been investigated with Northern blot hybridization and DNA mobility shift assay. Materials and Methods: HL-60 cells were grown in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum and antibiotics in a humidified 5% CO_(2) at 37°C. Total RNA was prepared by a modification of the method of Karlinsey et al. Northern blot hybridization was performed by the method of Virca et al. EcoRI fragment of pVIM-GEM was used as probe for vimentin mRNA. DNA mobility shift assay was performed by the method of Lim et al. End labeled DNA probe(Upper strand, 5'-CGCTTGATGAGTCAGCCG- 3') for AP-1 binding activity was mixed with nuclear extracts in a 20 μl reaction volume containing 300 mM KC1, 60 mM HEPES, pH 7.9, 25 mM MgCl_(2), 1 mM EDTA, 1 mM DTT, 60% glycerol, and 2μg of poly[dI-dC]. Results: TPA increased vimentin mRNA levels, with maximal stimulation reached at 24 hr. The level of vimentin mRNA was induced in proportion to the concentration of TPA. TPA-induced vimentin mRNA was almost reduced by actinomycin-D pretreatment. TPA-induced stimulation of vimentin gene was completely reduced by staurosporin pretreatment. In DNA mobility shift assay, AP-1 newly appeared at 24 hr during TPA-induced differentiation and was almost not detected after the pretreatment of staurosporin. Conclusions: These results suggest that the induction of vimentin mRNA during TPA-dependent differentiation in HL-60 cells may be mediated by protein kinases C signal transduction and AP-1 is important to transcriptional regulation.

      • RAW264.7 세포에서 interferon-r 및 LPS에 의해 유도되는 NO생성에 미치는 TALT-35의 영향

        TALP-35 purified from human term placenta is known to increase microtubule polymerization and stabilize the polymerized microtubule. To examine the effect of TALP-35 on immune system this study was performed. MTT assay was performed to investigate the effect of TALP-35 on the proliferation of RAW264.7 cells. TALP-35 dose dependently suppress the proliferation of RAW264.7 cells at high concentration (above 1 μM) in unstimulated cells, in case of 10 μM TALP-35 treated cells the suppression was 25% but in stimulated cells it was only 15%. Cosedimentation assay was carried out to investigate whether TALP-35 can bind to tubulin of RAW264.7, monocyte/macrophage lineage of mouse, and polymerize it. TALP-35 polymerize the tubulin of RAW264.7 cells and sedimented in dose-dependent manner. To investigate the effect of TALP-35 on the expression of iNOS protein western blotting was performed. The expression level of iNOS was decreased dose dependently in high concentration of TALP-35 treatment. To examine the activity of iNOS, secreted NO was determined by method based on Griess reaction. Interferon-γ and LPS-stimulated production of NO from RAW264.7 cells was decreased dose dependently above 0.1 μM concentration of TALP-35 and 50% is decreased at 10μM of it. This study shows TALP-35 can control cytokine induced-iNOS expression therefore it might control inflammatory diseases.

      • Erwinia sp. 에 의한 고추 마디 썩음병(무름병)에 관한 연구

        A bacterial disease of peppers (Capsicum annuum L.) that rotted the stem nodes to blacken was newly found in peppers that cultivated in plastic house by hydroponic culture at Chungdo, Kyung pook, Korea in March 1998. Bacterial isolates derived from the diseased peppers was also pathogenic to potato, eggplant and Chinese cabbage but was not be pathogenic to chrysanthemum by artificial inoculation. Growth of the pathogenic bacteria was inhibited by 5r/ml Streptomycin H₂SO₄ and Friadiomycin H₂SO₄. On the basis of bacteriological characteristics and pathogenicity on the host plants of the organism, the causal bacteria to the pepper bacterial disease is identified as Envinia carotovora subsp. carotovora and this disease is proposed to name "Bacterial node black rot of pepper"

      • 高眞空下에서 브롬화벤질류와 이소퀴놀린과의 親核性 置換反應에 관한 速度論的 硏究

        전기전도도법에 의해 아세토니트릴 용매내에서 브롬화벤질류(p-CH₃, p-H, p-NO₂)와 isoquinoline과의 반응을 고진공하에서 온도(15, 20, 25℃)변화에 따라 속도론적으로 연구하였다. 이때 반응속도상수는 온도가 증가함에 따라 증가하였으며, 또한 기질에 전자 주게 치환체(p-CH₃)가 치환된 경우 증가하였다. 이들 반응속도상수로 부터 활성화에너지(Ea), 활성화엔탈피(???), 활성화엔트로피(???), 활성화자유에너지(???) 및 Hammett 반응상수 ρ를 구하였으며, 이러한 속도론적 연구로 부터 전반적인 반응메카니즘은 dissociative ??? 반응으로 진행됨을 알 수 있었다. Rate of reactions of p-substituted benzyl bromides(p-CH₃, p-H, p-NO₂) with isoquinoline have been measured by conductivity method in acetonitrile, which was determined under vapor pressure of the solution obtained by using high vacuum apparatus, by kinetic studies at several temperatures. The rate constants are increased as a function of temperatures and are further increased by introducing the electron donating substituents in substrates. The activation energies (Ea), activation parameters(???,???,???)and Hammett ρ values are calculated from the rate constants and, generally, the results of these studies show a greater degree of bond breaking than bond formation in the transition state.

      • 고진공내의 데칸용매하에서의 브롬화메틸이 니트로벤젠 및 톨루엔과의 착물형성에 관한 연구

        고진공내의 데칸용매하에서 브롬화메틸의 용해도를 8, 15, 25℃에서 니트로벤젠과 톨루엔이 존재할 때와 존재하지 않을 때의 두 경우에 대하여 각각 측정하여 보았다. 데칸용매에 니트로벤젠과 톨루엔이 존재할 때 브롬화메틸의 용해도가 존재하지 않을 때 보다 더 크고, 톨루엔과 브롬화메틸의 상호작용이 니트로벤젠과의 상호작용보다 더 강하게 작용한다는 것을 알 수 있었다. 그리고 니트로벤젠과 톨루엔이 데칸용매내에서 존재할 때 브롬화메틸과 니트로벤젠 및 톨루엔이 각각 불안정한 complex를 형성한다고 생각되어 이 complex를 여러 경우의 비율로 가정하여 계산해 본 결과 1 : 1 complex를 형성한다고 생각되어 이 complex를 여러 경우의 비율로 가정하여 계산해 본 결과 1 : 1 complex를 형성한다고 보는 경우의 평형상수 K값이 비교적 일정한 값을 나타내었다. 그러므로 1 : 1 complex 즉 ??과 ??이 형성됨을 알았다. 또한 이 complex에 대한 표준엔탈피, 자유에너지 및 엔트로피 변화도 산출하였다. The solubilities of methyl bromide in decane on high vacuum were measured at 8℃, 15℃ and 25℃ in the presence and absence of nitrobenzene and toluene. When nitrobenzene and toluene exist in system individually, the solubilities of methyl bromide in the presence of nitrobenzene and toluene are greater than in the absence, and the interaction of methyl bromide with toluene is stronger than with nitrobenzene. In the presence of nitrobenzene and toluene, it could be thought that methyl bromide forms unstable complex with nitrobenzene and toluene. The complex was assumed in various ways and it was evaluated that the equilibrium constant K value was relatively constant under the assumption of 1 : 1 complex. Thus, 1 : 1 complexes, ?? and ??, are formed in the solution. Also, the change of standard enthalpy, free energy and entropy for the formation of the complex were calculated.

      • 고진공내의 데칸용매하에서 요오드화메틸이 피리딘 및 α-피콜린과의 착물형성에 관한 연구

        고진공내의 데칸용매하에서 요오드화메틸의 용해도를 5, 15 25℃에서 피리딘과 α-피콜린이 존재할때와 존재하지 않을 때의 두 경우에 대하여 각각 측정하였다. 데칸용매내에 피리딘과 α-피콜린이 존재할 때 요오드화메틸의 용해도가 존재하지 않을 때 보다 더 크고 α-피콜린과 요오드화메틸과의 상호작용이 피리딘과의 상호작용보다 더 강하게 작용한다는 것을 알 수 있었다. 그리고 피리딘과 α-피콜린이 데칸용매내에 존재할 때 요오드화메틸과 피리딘 및 α-피콜린이 각각 불안정한 Complex를 형성한다고 생각되어 이 Complex를 여러 경우의 비율로 가정하여 게산해 본 결과 1 : 1Complex를 형성한다고 보는 경우의 평형상수 K 값이 비교적 일저한 값을 나타내었다. 그러므로 1 : 1Complex 즉 CH₃I·??과 CH₃I·α-??이 영액속에 형성됨을 알았다. 그리고 피리딘과 α-피콜린의 요오드화메틸과의 Complex의 안정도를 이와 대응하는 요오드화에틸과 요오드화 n-프로필과 비교하여 보았다. 또한 이 Complex의 생성에 대한 엔탈피, 자유에너지 및 엔트로피의 변화도 산출하였다. And the stability of the complex of methyl iodide with pyridine and α-picoline are compared with similar complexes of pyridine and α-picoline with ethyl iodide and n-propyliodide. Also, the changes of enthalpy, free energy and entropy for the formation of the complex were calculated.

        The Kinetics of tris (malonato) Cobaltate(Ⅲ) anion in aquation reaction was carried by spectrophotometric method. Themperature range was 20˚ to 30℃ and pressure was varied up to 1500 bars. The rate constants were increased with increasing temperature, but were decreased with increasing pressure. Activation volume, energy, other activation parameters were reported.
