Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p< 0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism. Downregulation of olfactomedin-4 (OLFM4) is associated with tumor progression, lymph node invasion and metastases. However, whether or not downregulation of OLFM4 is associated with epigenetic silencing remains unknown. In this study, we investigate the role of OLFM4 in gastric cancer cell invasion. We confirm the previous result that OLFM4 expression is increased in gastric cancer tissues and decreases with an increasing number of metastatic lymph nodes, which are associated with OLFM4 promoter hypermethylation. Overexpression of OLFM4 in gastric cancer cells had an inhibitory effect on cell invasion. Furthermore, we found that focal adhesion kinase (FAK) was negatively correlated with OLFM4 in regards to lymph node metastasis in gastric cancer tissues. Also, inhibition of FAK induced by OLFM4 knockdown resulted in a decrease in cell invasion. Thus, our study demonstrates that epigenetic silencing of OLFM4 enhances gastric cancer cell invasion via activation of FAK signaling. [BMB Reports 2015; 48(11): 630-635]

The geological conditions surrounding the Jijiapo Tunnel of the Three Gorges Fanba Highway project in Hubei Province are very complex. In this paper, a 3-D physical model was carried out to study the evolution process of filling-type fracture water inrush and mud gush based on the conditions of the section located between 16.040 km and 16.042 km of the Jijiapo Tunnel. The 3-D physical model was conducted to clarify the effect of the self-weight of the groundwater level and tunnel excavation during water inrush and mud gush. The results of the displacement, stress and seepage pressure of fracture and surrounding rock in the physical model were analyzed. In the physical model the results of the model test show that the rock displacement suddenly jumped after sustainable growth, rock stress and rock seepage suddenly decreased after continuous growth before water inrushing. Once water inrush occured, internal displacement of filler increased successively from bottom up, stress and seepage pressure of filler droped successively from bottom up, which presented as water inrush and mud gush of filling-type fracture was a evolving process from bottom up. The numerical study was compared with the model test to demonstrate the effectiveness and accuracy of the results of the model test.

Triaxial compression tests had been performed to determine the properties of the surrounding rock. The displacement and stress of surrounding rock had been analyzed according to Silin 2 at Yungui railway under three kinds of conditions: the initial state, excavation without supporting , and supporting after excavation. Plane strain FEM was established to analyze the stability of the surrounding rock. Contrasted the monitor measuring data, the results showed that the most unfavorable position in the tunnel was found at the ridge side of the arch, and the implementation of support afforded to ensure the stability of surrounding rock, and the design of the early support approach had the ability to meet the safety requirements. The results had certain significance for understanding the mechanical behavior of shallow tunnel in unsymmetrical strata. tunnel, mechanical behaviour, numerical simulation, deformation, stability.
Fluid Solid Coupling Analysis of Large Underground Oil Storage Caverns in Containment of Groundwater

Triaxial compression tests had been performed to determine the properties of the rock mass around unlining underground crude oil a storage cavern which was the first one in China. The execution situation of the tunnel project and the seepage law of groundwater were taken into account. The stress and the seepage field around the tunnel in different working states were simulated by applying Comsol around a underground crude oil storage caverns. According to the test results, it was found that the excavation process may arise the local damage. The extension of the loose zone induced by excavation ranged from 0 to 15.6 m, depending on the buried depth of the caverns. According to numerical simulation results, the crown settlement and stress concentration was depended on the buried depth and the water pressure distribution after excavation of the main cavity. This research results can provide the reference for analysis on the stability of the underground cavities under low stress level and on the water sealed underground petroleum storage rock caverns.

본 논문은 주로 중국어의 ‘형용사+명사’구조의 예를 통하여 의미문법의 이론적 문제와 양방향문법의 방법론적인 문제를 논의하였다. 우선, 기존의 중국어 문법연구에서 보이는 양방향의 개념을 역사적인 관점에서 검토하였다. 둘째, 방법적인 면에서 양방향문법의 개념과 방법론적 의의를 분석하였고, ‘형용사+명사’구조를 예로 그 과학성과 타당성을 밝혔다. 또한, 의미적 관점에서 의미문법이론의 본질적인 특징을 주로 고찰하였으며, 방법론으로서의 양방향문법과 의미문법이론이 상보적인 관계에 있음을 주장하였다. 즉, 양방향문법은 의미문법이론에 기초할 때 그 방법론적 가치가 드러나며, 반대로 의미문법이론은 양방향문법을 통해 조합규율을 증명할 때에 비로소 의미적 특징을 잘 제시할 수 있다.

Mo2 FeB2 cermet coating and stainless steel substrate were integrally fabricated by one-step forming technology of powder metallurgy. The phase and microstructure of the coating and interface were analysed through X-ray diffraction and scanning electron microscopy. Microhardness, bending strength, and wear resistance were also measured by Vickers tester, the electronic universal testing machine and wear tester. The microstructure of Mo2 FeB2 cermet coating showed Mo2 FeB2 hard phase and Fe binder phase, which was uniform and dense. A compacted interface without pores or defects was formed between the cermet coating and stainless steel substrate. The formation of eutectic liquid phase and applied pressure in sintering process promoted densification of the coating. Mo2 FeB2 cermet coating presented a microhardness of 1275 HV200 , which was obviously higher than that of 316L stainless steel substrate. The results of three-point bending test and wear test showed that Mo2 FeB2 cermet coating on steel substrate maintained good metallurgical interface bonding with the substrate, and the coating exhibited high bending strength and excellent wear resistance.

Quantitative detection of protein, fat, starch,and amino acids in foxtail millet using Fourier transformnear-infrared spectroscopy (NIRS) was investigated. Foxtail millet samples (n=259) were analyzed using NIRS. Spectral data were linearized with data from chemicalanalyses. Calibration models were established using apartial least-squares (PLS) algorithm with cross-validation. Optimized models were tested using external validation setsamples with coefficients of determination in the externalvalidation (R2val) of >0.90. Residual predictive deviation(RPD) values were nearly equal to or >2.5 for crudeprotein, alanine, aspartic acid, glutamic acid, isoleucine,leucine, and serine. However, for glycine, histidine,phenylalanine, proline, threonine, tyrosine, and valine, theR2val values were >0.83 and RPD values were nearly equalto or >2.0. For crude fat, total starch, arginine, and lysine,the R2val values were >0.70 and RPD values were >1.5. NIRS is a rapid determination tool for foxtail milletbreeding, and for quality control.
