<P><B>Abstract</B></P><P>The objective of this study is to demonstrate that a condition-monitoring system based on acoustic emission (AE) detection can provide timely detection of check valve degradation and service aging so that maintenance or replacement can be preformed prior to the loss of safety function. This research is focused on the investigation and understanding of the capability of the acoustic emission technique to provide diagnostic information on check valve failures.</P><P>AE testing for a check valve under controlled flow loop conditions was performed to detect and valve degradation such as wear and leakage due to foreign object interference. It is clearly demonstrated that the distinction of different types of failure were successful by systematically analyzing the characteristics of various AE parameters.</P>
Molecular Analysis of HLA - C Using Polymerase Chain Reaction - Sequence Specific Primers

Of all HLA class I molecules, HLA-C gene products are most poorly understood because they express at a low level on the cell surface compared to HLA-A and -B. In order to identify serologically detectable and undetectable HLA-C antigens, we have established a DNA-based tissue typing method for the HLA-C locus by PCR-SSP (polymerase chain reaction-sequence specific primers). Genomic DNA prepared from lymphoblastoid 21 B-cell lines and 120 Korean individuals by proteinase K digestion and phenol/chloroform extractions have been typed by PCR-SSP (23 primer mixes were used). The PCR-SSP results of control cell lines were discrepant from serology in 1 case among 21 cases: Cw6 which was negative by serology but positive by PCR-SSP (cell line: MANIKA). Twenty four HLA-Cw $quot;blank$quot; antigens among fifty Korean individuals were completely determined by PCR-SSP DNA typing. HLA-Cw*0101 (15.3%), Cw*1401 (12.3%) and Cw*0701 (11.7%) alleles were frequently found in 120 Korean individual samples. In conclusion. the high level of discrimination for HLA-C alleles may prove useful and informative in the study of transplant survival, and identify the importance of allelic differences, not readily detectable by serology, on host and donor compatibility.

<P>Oxalic acid has potent nematicidal activity against the root-knot nematode Meloidogyne incognita. In this study, fermentation parameters for oxalic acid production in submerged culture of Aspergillus niger F22 at 23, 25, and 30 A degrees C were optimized in 5-L jar fermenters. The viscosity of the culture broth increased with increasing temperature. There was a negative correlation between oxalic acid production and the apparent viscosity; high volumetric productivity of oxalic acid was obtained at low apparent viscosity (less than 1000 cP), with a productivity of more than 100 mg/L h. When the apparent viscosity was over 2500 cP, the volumetric productivity decreased below 50 mg/L h. In addition, the volumetric mass transfer coefficient, K (L) a, positively correlated with volumetric productivity. When the K (L) a value increased from 0.0 to 0.017 /s, the volumetric productivity proportionally increased up to 176 mg/L h. When the temperature decreased, K (L) a increased due to the decrease in viscosity, leading to increased volumetric productivity. The highest productivity of 7453.3 mg/L was obtained at the lowest temperature, i.e., 23 A degrees C. The nematicidal activity of culture filtrate was proportional to the content of oxalic acid. Based on a constant impeller tip speed, oxalic acid production was successfully scaled up to a 500-L pilot vessel, producing a final concentration comparable to that in the 5-L jar.</P>

The mechanization of domestic agriculture production and cultivation technology is ongoing by the lead of rice cropping. Among this, the possession rate of tractor is 46% of the major agricultural machinery such as tractor, rice transplanter and combine. Furthermore, the tractor possession keeps increasing in overseas market and the market size is also rapidly expanded. In order to strengthen competitiveness in overseas market, domestic tractor companies must put their efforts to reduce material costs and improve quality. To improve quality and reduce costs, need to analyze load characteristic according to the soil condition and work condition and appropriate design for power transport system based on the basic data on tractor design is required. While number of Utility and Row-Crop tractor is increasing domestically, load analysis study on agriculture operation is very weak. Also most of the studies are conducted on small tractors. Therefore, to increase competitive power for quality of domestic tractor and secure international competitiveness by cost reduction, load analysis study according to the increase in frequency of use of Utility and Row-Crop tractor agriculture operation is required. In this study, the operation condition according to the plowing operation has decided based on the survey of user in the Jeollabuk do. Finally load spectrum is written to use as a basic data for design and durability test.

팜 스테아린으로부터 분별온도(X₁=25, 30, 35℃)와 팜 스테아린에 대한 아세톤의 비율(X₂=1:3, 1:6, 1:9, w/v)을 요인변수로 하여 중심합성계획에 의한 반응표면분석을 이용하여 최적화한 POP purity(Y₁, %), POO purity(Y₂, %), POO+POP의 content(Y₃, g)의 조건을 살펴보았다. Y₁, Y₂, Y₃의 종속요인들을 모두 최대화하면서 순도를 가장 높일 수 있는 분별조건을 예측한 결과, 분별온도 29.3℃에서 팜 스테아린에 대한 사용 아세톤의 비율이 1:5.7일 경우 60.9%의 POP와 23.8%의 POO를 함유한 POO와 POP 분별유를 4 g의 팜 스테아린에서 3.0 g(75% 수율)의 획득을 예측할 수 있었다. POO와 POP의 순도를 높이면서 함량을 높이는 최적화 조건의 검증을 위하여 반복실험을 수행한 결과, POP purity와 POO purity, POO+POP의 content는 각각 60.18%, 24.10%, 74.38%(2.97 g)로, 반복 실험 간에 유의적인 차이가 없음을 확인하였다. 이렇게 얻어진 POO와 POP 함량 및 순도가 높은 유지는 앞으로 CBE의 주된 대칭형 TAG(POS와 SOS) 합성에 이용될 수 있을 것으로 사료된다. 1-Palmitoyl-2-oleoyl-3-oleoyl glycerol (POO) and 1-palmitoyl-2-oleoyl-3-palmitoyl glycerol (POP) were enriched from palm stearin using an acetone fractionation process. Response surface methodology was employed to optimize the purity of POO (Y₁, %) and POP (Y₂, %) along with POO+POP content (Y₃, g) based on independent variables such as fractionation temperature (X₁, 25, 30, and 35℃) and the ratio of palm stearin to acetone (X₂, 1:3, 1:6 and 1:9, w/v). Fractionation conditions were optimized to maximize Y₁, Y₂, and Y₃, in which fractionation temperature was 29.3℃ with a 1:5.7 acetone ratio. With such parameters, 60.9% of POP and 23.8% of POO purity were expected with a 75% yield (3.0 g) of POO and POP.

The contents of β-carotene and retinol in processed and restaurant foods, such as Korean noodles, mandus, rice cakes and bread products, were quantified by high-performance liquid chromatography (HPLC) with UV/visible and fluorescence detector, respectively. Samples were collected from different local areas (i.e. Gangwon-do, Gyeonggido, Gyeongsang-do, Seoul, Jeolla-do, and Chungcheong-do). After homogenization, samples were hydrolyzed by direct alkali saponification; thereafter, fat-soluble components were extracted by a mixture of n-hexane/ethylacetate (85:15, v/v), containing 0.01% butylated hydroxytoluene (BHT). β-carotene and retinol contents in infant formula used as an in-house material for the analytical quality control. Among 14 Korean noodles, high contents of β-carotene were found in Bibim-Guksu (average 442.43 μg/100 g) and Jjolmyeon (average 301.39 μg/100 g). In 4 Korean mandus, the highest contents of β-carotene was determined in Kimchi-mandu (average 197.64 μg/100 g), resulting in 33.3 RE of the converted vitamin A. Among 12 Korean rice cakes, Maeun-Tteokbokki and Modm-Chaltteok contained relatively high content of β-carotene with 205.11 and 41.33 μg/100 g, respectively, while retinol was detected only in Maeun- Tteokbokki (1.65~10.45 μg/100 g). In addition, among 8 bread products, 77.3 RE of pastry, 51.2 RE of buttercream- bread, and 41.4 RE of morning roll were found as the contents of the converted vitamin A.
Oxalic Acid Production in Submerged Culture of Aspegillus niger F22

Oxalic acid has a nematicidal activity against the root-knot nematode Meloidogyne incognita. High producer of oxalic acid was isolated, and then named as Aspergillus niger F22. Oxalic acid production was investigated under various temperatures from 20 – 33oC and rotational speeds in 5 L jar fermenters. Yield of oxalic acid increased with decreasing temperature. The highest yield was obtained at 23oC, showing the yield of oxalic acid of 8.7 g/L, whereas oxalic acid production was least at 33oC. At 20oC, the yield was lower than that of 23oC. At a rotational speed of 300 rpm, serious oxygen depletion was present from 48 - 72 h, resulting in low productivity of 26.2 mg /L·h. When a rotational speed was set at 600 rpm, dissolved oxygen tension was over 40% and oxalic acid production increased up to approximately 55%. Viscosity during the culture differed with temperatures. Viscosity increased with the increment of temperatures. When A. niger F22 was cultured at 23oC, viscosity was 810 cP, which was favorable for oxalic acid production.
