      • 連翹敗毒散加味方이 알러지성 접촉피비염에 미치는 影響 : DNCB 재노출부위의 생쥐 피부손상완화와 비만세포 분포 변화를 중심으로 Based on the mitigation of skin damage and distributive change of mast cell on DNCB re-expasure region of Mice

        본 실험은 連翹敗毒散加味方이 알러지성 접촉피부염 유발시 발생되는 면역과민반응과 염증반응 저해에 미치는 영향을 조사하기위해 행해진 것으로 2,4-dinitrochlorobenzene(DNCB, 5% DNCB 25㎕로 감작, 7일경과 후 2.5% DNCB 5㎕로 유발) 처리로 알러지성 접촉피부염이 유발된 ICR계 암컷 생쥐에 連翹敗毒散加味方抽出物(0.51g/㎏/day)을 음용투여 한 다음 contact hypersensitivity assay, 피부의 일반적인 구조 그리고 비만세포의 분포 변화를 관찰하였다. Contact hypersensitivity assay의 결과, 連翹敗毒散加味方抽出物 투여군의 ear swelling이 DNCB군에 비해 유의성있게 감소한 것으로 나타났다. 알러지성 접촉피부염 유발 후 관찰되는 피부손상은 48시간에 제일 심한 것으로 관찰되었으나, 連翹敗毒散加味方抽出物 투여군에서는 DNCB군 비해 완화된 결과를 보였다. 連翹敗毒散加味方抽出物 투여군의 진피에서 림프구를 비롯한 염증관여세포의 이주와 혈관생성 그리고 표피내로의 림프구의 침윤 등이 감소되었고, 표피 기저층과 가시층세포에서는 세포질내 액포화와 세포사이 공간 확장도 줄어든 것으로 관찰되었다. 한편 알러지성 접촉피부염 유발 후 분포가 증가한 비만세포는 형태가 주로 세포내 함유물이 분비된 degranulated type로 나타났다. 이에 반해 連翹敗毒散加味方抽出物 투여군는 DNCB군에 비해 출현 비만세포'수가 일부 감소했고, 비만세포의 형태는 granulated t-ype였다. 또한 serotonin 면역조직화학적 염색결과로 連翹敗毒散加味方抽出物 투여군에서 se-rotonin 양성반응성이 높은 granulated type의 비만 세포가 주로 관찰되었다. 이상의 결과로 미루어 連翹敗毒散加味方抽出物은 알러지성 접촉피부염으로 유발된 피부 손상과 염증을 완화시키는 효과가 있는 것으로 사료된다. After allergic contact dermatitis elicitated by Dinitrochlorobenzene(DNCB) treatment, ICR female mice administered Yunkyopaedocksangamibang(YPGM) extract were observed to investigate the effect of YPGM on allergic contact dermatitis. This study investigated that contact hypersensitivity assay, abdominal skin morphologic changes including mast cells. At contact hypersensitivity assay, the right ear swelling in YPGM group were probability decrease than DNCB group. At obsevation of abdominal skin morphologic change, the infilteration of lymhocyte, lymphocyte insertion to epithelium, enlarged capillary, angiogenesis, and damages of epithelium as cytoplasmic vacuolation and enlarge of inter cellular space in YPGM were diminshed than DNCB group. The number of mast cell was increased both DNCB and YPGM group. The shape of mast cell in DNCB group was mainly appeared degranulated type, but granulated type in YPGM group. The number of serotonin positive cell was increased both DNCB and YPGM group. The shape of serotonin positive cell in DNCB group was mainly appeared degranuled type, but granulated type in YPGM group. As results indicated that the YPGM exrtact administration work on the mitigation of skin damages in mouse with allergic contact dermatitis.

        Intestinal malrotation with volvulus is generally presented as a bilious vomiting and acute intestinal obstruction in the newborn period. It could compromise vascular supply of the small bowel secondary to torsion of superior mesenteric artery (SMA) and without urgent surgical management, it could lead to detrimental outcomes such as transmural bowel infarction and sepsis. However, in chronic cases, it is rarely obstructs the vascular supply and propagates to an acute bowel infarction. Rarely, chronic malrotation with midgut volvulus may not reduce the mesenteric blood supply because of collateral vessels, and the chronically stagnated blood flow of the superior mesenteric vein (SMV) favors thrombus formation within the lumen. The recommended treatment is Ladd’s procedure and anticoagulation therapy. The authors present an unusual case of intestinal malrotation with chronic volvulus resulting in superior mesenteric vein and portal vein thrombosis in a 28-year-old patient.

        패류의 가공적성 2. 굴의 가공적성

        로우푸垂下式 양식굴의 加工適性에 관한 實驗을 하여 다음과 같은 結果를 얻었다. 1. 굴의 殼內부피에 대한 軟體部의 무게 또는 殼內부피에 대한 軟體部의 부피의 측정값으로서 肥灣度를 測定하는 指標로 利用할 수 있다. 2. 肉成分의 月別變化를 보면 水分과 脂肪은 대체로 逆相關關係가 있고, 단백질은 4月부터 약간 減少하기 시작하여 7月에 급격히 減少하였다가 8月에 다시 급격하게 增加하나 9月부터 다시 점차 減少하는 경향을 나타내었다. 글리코겐은 4月부터 급격하게 減少하기 시작하여, 6∼8月에 최저값을 나타내고, 9月부터 다시 增加하였다. pH는 6.0∼6.2로서 時期的으로 큰 變化를 찾아 볼수 없이 거의 一定하였다. 灰分은 6∼8月에 악간 減少하는 傾向이 있었다. 3. 肥灣度 및 肉成分 分析結果로써 加工適性을 判定한다면 加工原料 採取適期는 12月末에서 다음에 5月까지라고 보아진다. 4. 重金麗含量의 時期的變化範圍를보면 水銀은 0∼0.019 ppm, 카드뮴은 0.026∼0.053 ppm, 구리는 0.111∼0.594 ppm, 납은 0.061∼0.581 ppm로서 加工原料로서 安全하다고 볼 수 있다. 5. 생굴을 冷凍하기 전에 플리인산나토륨 10% 함유한 5% 食鹽水에 浸漬養處理한 것은 冷凍時에 drip流出防止效果가 있었다. 6. Na₂EDTA또는 BHA용액에 侵漬處理하는 前處理 操作만으로서는 굴 보일드통조림의 黃變을 防止할 수 없었다. 7. 2% 염화마그네슘 용액은 살아 있는 굴의 開殼活動을 촉진하는 效果가 있었다. The estimation of the pre-processing condition of oyster is of great importance for distributors and processors. This study was attempted to establish the basic data for evaluating the processing suitability of oyster, which is the most important shellfish for domestic use and export. The data were analysed by measuring the condition index, chemical composition and heavy metal content of oysters. In order to eliminate the manual work that has to be done on a tightly closed oyster shell and avoid shrinkage in the oyster meat which is attendant on the steaming process, chemical means to open oyster were examined. Finding the method of pretreatment of polyphosphate for frozen oysters were attempted to improve the product quality. The prevention of undesirable color change of the canned oyster meat is another problem to solve. The important results are as follows: 1. The ratio of meat volume and meat weight to the holding capacity by shells may be useful as an index to measure the condition index of oysters. 2. As a whole, monthly changes of moisture and fat content in oysters were reversely correlated. Protein content slightly decreased from April and rapidly decreased in July, and again rapidly increased in August but from September to November decreased slightly. In April, the content of glycogen was 4 percent. From this period to September, glycogen was rapidly decreased. From July to September, it was only 0. 7 to 1 percent but increased from October. There were little seasonal changes in pH value. The pH value of oyster meat was 6.0 to 6.2. The crude ash content was slightly decreased from June to August. 3. The range of monthly change of heavy metal content are as follows: Total mercury was 0 to 0.019 ppm, cadmium was 0.026 to 0.053 ppm, copper was 0.111 to 0.594 ppm, and lead was 0.061 to 0.581 ppm. 4. By the results of condition index, chemical composition and heavy metal content of oysters, the suitable harvest season as raw materials for processing was the end of December to the end of May of next year. 5. The pretreatment of 10 percent polyphosphate in 5 percent salt solution of oyster meat appeared effective to reduce thawing drip during cold storage. 6. The pretreatment of Na₂EDTA and BHA did not show the color prevention effect to the canned oyster meat during storage. 7. Magnesium chloride affected to open the valves of oysters.

      • 직렬과 병렬방식 이중효용 흡수식 냉동사이클의 성능 시뮬레이션

        Condensing the global environmental problems such as CFCs, energy losses in heat recovery systems as well as summer peak time power demands, the development of high efficiency absorption refrigeration systems is one of the most promising method in this problems. In this study, simulation models of both series and parallel type, both using double-effect absorption cycle refrigeration systems. The results summarized as follows : ⑴ In the parallel type, the solution maximum temperature and the pressure of high temperature generator were adjusted by varing the distribution ratio of the solution, and the optimum value was found to be about 0.4. ⑵ The efficiency of the cooling cycle in both parallel and series systems were increased by decreasing the solution circulation ratio in the absorbers, while the solution maximum temperature and high temperature generator pressure were slightly increased.

      • 5.7-diiodo-8-hydroxyquinoline-XAD침윤수지의 금속이온흡착 및 분리에 관한 연구

        5,7-diiodo-8-hydroxyquinoline(DIHQ)의 Amberilte XAD -4 및 XAD-7 수지에 대한 최적흡착 pH는 pH3.0-7.0 영역이다. 이 pH에서 XAD-4 및 XAD-7 수지에 대한 DIHQ의 침윤량은 각각 3.10×10?²mmol DIHQ/g, XAD-4 resin와 1.80× 10?²mmol DIHQ/g, XAD-7resin이었다. DIHQ-XAD-4 및 DIHQ-XAD-7 침윤수지는 pH 3.0-7.0 영역에서 안정하였으며, 특히 DIHQ-XAD-4수지의 경우에는 횹착된 금속이온의 회수에 사용되는 용리액인 HCI의 농도가 증가하여도 안정도가 감소하지 않았으며 DIHQ-XAD-7수지의 경우에는 HCI의 농도의 증가에 따라 안정도가 감소하였다. DIHQ-XAD침윤수지에 대한 금속이온의 흡착은 Cu(Ⅱ), Cd(Ⅱ) 및 Ni(Ⅱ)의 경우에는 pH 9.0에서 최대흡착을 보였으며 이때 각 금속이온의 흡착몰비(금속이온 : DIHQ)는 1:2이었으며, Fe(Ⅲ)의 경우에는 3.0-4.0 영역에서 최대흡착을 보였으며 이때 흡착몰비는 1 :3이었다. DIHQ-XAD-4 및 DIHQ-XAD-7 침윤수지에 흡착된 금속이온은 각각 5M HCl 및 1M HCI 용액으로 용리하였을 때 정량적으로 회수되었으며, DIHQ-XAD-4침윤수지의 경우엔는 5M HCl을 용리액으로 사용하였을때 침윤량의 감소없이 5회이상 재사용이 가능하였으나 DIHQ-XAD-7 침윤슈지의 경우에는 1M HCl을 용리액으로 사용하였을때 5회 재사용에서 침윤량이 1/2로 감소하였다. The adsorption behavior of 5,7-diiodo-8-hydroxyquinoline(DIHQ) on Amberlite XAD-4 and -7 resins was investigated by measuring its distribution coefficients under various experimental conditions, such as shaking time, pH and concentration of organic solvent in medium. The application of DIHQ-impregnated-XAD resins for adsorption and separation of metal ions was studied. The maximum adsorption of DIHQ on XAD resins was observed in 30% acetone solution having pH range from 3.0 to 7.0. The impregnation capacities of XAD resins for DIHQ were 3.10 × 10?²mmol DIHQ/g,XAD-4resin and 1.80 × 10?²mol DIHQ/g,XAD 7 resin respectively. The DIHQ-impregnated-XAD resin was stable in pH range from 3.0 to 7.0 and the amount of DIHQ leached from XAD-4resin by eluting with variation of hydrochloric acid concentration, 1 to 5M, was negligible. The optimum pH range for adsorption of metal ions on DIHQ-impregnated-XAD resin was 9.0 for Cu(Ⅱ), Cd(Ⅱ) and Ni(Ⅱ), and was 3.0 for Fe(Ⅲ), and the adsorption mole ratio of metal ion to DIHQ were 1:2 for Cu(Ⅱ), Cd(Ⅱ) and Ni(Ⅱ), and 1:3 for Fe(Ⅲ) at the above pH range. It was found that the adsorbed metal ions on DIHQ-impregnated-XAD-4 and -7 resin was recovered quantitatively with 5M HCl and 1M HCl respectively. The DIHQ-impregnated-XAD-4 resin could be reusable over 5times without decrease in its impregnation capacity, but DIHO-impregnated-XAD-7 resin was decreased in its impregnation capacity to 1/2 with 1M HCl.

        N-oleoyl-D-erythro-sphingosine-based Analysis of Ceramide by High Performance Liquid Chromatography and Its Application to Determination in Diverse Biological Samples

        Ceramide is involved in cell death as a lipid mediator of stress responses. In this study, we developed an improved method of ceramide quantification based on added synthetic ceramide and thin layer chromatography (TLC) separation, and applied to biological samples. Lipids were extracted from samples spiked with N-oleoyl-D-erythro-sphingosine ($C_{17}$ ceramide) as an internal standard. Ceramide was resolved by TLC, complexed with fatty-acidfree bovine serum albumin (BSA), and deacylated by ceramidase (CDase). The released sphingosine was derivatized with o-phthalaldehyde (OPA) and measured by high performance liquid chromatography (HPLC). The limit of detection for ceramide was about 1-2 pmol and the lower limit of quantification was 5 pmol. Ceramide recovery was approximately 86-93%. Ceramide concentrations were determined in biological samples including cultured cells, mouse tissues, and mouse and human plasma. TLC separation of ceramide provides HPLC chromatogram with a clean background without any interfering peaks and the enhanced solubility of ceramide by BSAceramide complex leads to the increased deacylation of ceramide. The use of an internal standard for the determination of ceramide concentration in these samples provides an accurate and reproducible analytical method, and this method can be applicable to diverse biological samples.

        Decursin induces apoptosis in glioblastoma cells, but not in glial cells via a mitochondria-related caspase pathway

        Decursin is a major biological active component of Angelica gigas Nakai and is known to induce apoptosis of metastatic prostatic cancer cells. Recently, other reports have been commissioned to examine the anticancer activities of this plant. In this study, we evaluated the inhibitory activity and related mechanism of action of decursin against glioblastoma cell line. Decursin demonstrated cytotoxic effects on U87 and C6 glioma cells in a dose-dependent manner but not in primary glial cells. Additionally, decursin increased apoptotic bodies and phosphorylated JNK and p38 in U87 cells. Decursin also down-regulated Bcl-2 as well as cell cycle dependent proteins, CDK-4 and cyclin D1. Furthermore, decursin-induced apoptosis was dependent on the caspase activation in U87 cells. Taken together, our data provide the evidence that decursin induces apoptosis in glioblastoma cells, making it a potential candidate as a chemotherapeutic drug against brain tumor.
