        이온화방사선에 의한 느타리버섯(Rleurotus ostreatus)의 ITS2 염기서열 변화

        rDNA의 ITS2(internal transcribed spacer 2)부위에 대한 이온화방사선(gamma-ray)의 영향을 알아보기 위해서 감마선을 조사한 느타리버섯(Pleurotus ostreatus)의 5종의 변이중에서 ITS2의 염기서열을 결정하였다. ITS2의 염기서열은 1 kGy 조사군(PO-5, PO-6, PO-15와 PO-16)에서 변화되지 않았으나, 2kGy 조사군(PO-14)에서는 1개의 염기가 치환되었다. RAPD(ramdomly amplified polymorphic DNA) 양상에 의한 유전유사도의 변화가 다양하였고 섬유소성 고체검정 배지에서도 다양한 생장특성을 나타내었다. 본 연구결과는 ITS염기서열이 이온화 방사선에 의해 선량의존적으로 변화될 수 있음을 시사해 주었다.

        연안부근 복잡지형의 대기유동장 수치실험 1 : 선형이론을 이용한 국지순환모형의 타당성 검토 A Verification of Propriety of Local Circulation Model Using the Linear Theory

        A sea/land breeze circulation system and a regional scale circulation system are formed at a region which has complex terrain around coastal area and affect to the dispersion and advection of air pollutants. Therefore, it is important that atmospheric circulation model should be well designed for the simulation of regional dispersion of air pollutants. For this, Local Circulation Model, LCM which has an ability of high resolution is used. To verify the propriety of a LCM, we compared the simulation result of LCM with an exact solution of a linear theory over a simple topography. Since they presented almost the same value and pattern of a vertical velocity at the level of 1 ㎞, we had a reliance of a LCM. For the prediction of dispersion and advection of air pollutants, the wind field should be calculated with high accuracy. A numerical simulation using LCM will provide more accurate results over a complex terrain around coastal area.

        Openings through beams are required for the passage of utility ducts and pipes, and they are frequently encountered in the design of modern buildings. They are particularly important in multistory buildings where the use of openings in beams can mean lower story heights, leading in turn to a multiplicity of savings. Therefore, the designers are often faced with the necessity of providing adequate openings in the buildings. However, provision of openings may cause problems of excessive cracking and deflection, and may severely affect the strength of a beam. An experimental investigation is carried out on ten reinforced concrete beams with rectangular openings. The size of opening and it's location are considered as major variables. The specimens were tested under two point loading. Loads were usually applied in steps of 0.5 ton up to failure to observe the crack initiation and propagation, initial diagonal cracking midspan deflection, concrete strain and reinforcement strain.

      • 항산화제 복합비타민 섭취기간에 따른 운동강도별 SOD·CAT 활성도의 변화

        The purpose of this study was to investigate the shift of antioxidant enzymes SOD, CAT for the period of vitamin mixture supplementation during continued exercise. The subjects were consisted of male 14 Korea National Bowlers who have been trained at training center of Korea Sports Council. To know the shift of variables was expreimented at point of before supplement-action, after 4 weeks & 8 weeks at 80%HRmax & 85%HRmax used treadmill by balke protocol. In a day, the 2 capsules were supplemented for 8 weeks with the vitamin mixture containing 500㎎ ascorbic acid of vitamin C, 400I.U a-Tocopherol of vitamin E, 50㎎ β-Carotene, 92.6㎎ selenium. The results of the study can be summerized in the followings ; SOD(superoxide dismutase) activity increased in exercise intensity of 85%HRmax than 60%HRmax before supplementation of vitamin mixture. But, the tendancey of SOD activity was decreased according to the period of supplement. In particular, 85%HRmax after 4 weeks was lower than the exercise intensity of 60%HRmax. It was a result to remove for SOD in high exercise intensity induced much oxygen radical. CAT(catalase) level increased in exercise intensity of 85%HRmax than 60%HRmax before supplementation of vitamin mixture were significantly different between exercise intensities. So, higher CAT activity in high exercise intensity was controled free radical in high exercise intensity. It was expected as a antioxidants when vitamin mixture was supplemented over 4 weeks in exercise intensity of 60%HRmax and 85%HRmax.

        느타리버섯(Pleurotus ostreatus) 재배균상에 오염되는 Rhizoctonia solani 유사균의 특성

        느타리버섯(Pleurotus ostreatus) 재배균상은 흔히 토양, 공기 중 곰팡이에 의해 오염되어 자실체 생산량이 감소되곤 한다. 느타리버섯 재배균상에서 3종류(Rs-1, RS-2와 RS-3)의 오염균주를 분리하여 internal transcribed spacer(ITS)의 염기서열을 분석하였다. 이들의 ITS염기서열은 식물 병원성 사상균의 일종인 Rhizoctonia solani와 매우 높은 상동성을 보였다. 이들 오염균주들의 영양배지와 lignin 배지에서 균사체 생장율은 느타리버섯보다 각각 수배 및 수십배나 높았다. 세포외분비효소의 활성도는 오염균주들의 경우 느타리버섯과 비교하여 매우 낮은 활성도를 보였다. 느타리버섯과의 혼합배양 결과 오염균주들은 느타리버섯의 균사체 생장 및 자실체 형성을 심하게 저해하였다. The contamination of air- and soil-borne microbes could occur and result in the reduction of fruiting body formation during cultivation of Pleurotus ostreatus. Three contaminants(RS-1, RS-2 and RS-3) were isolated and showed higher homology to the plant pathogenic fungus, Rhizoctonia solani by internal transcribed spacer(ITS) analysis. The rates of their mycelial growth on the lignin specific medium and complete medium were several times higher than those of P. ostreatus. The activities of the extracellular enzymes of three contaminants were lower than those of P. ostreatus. During oyster mushroom cultivation(P. ostreatus) together with these contaminants, the mycelial growth and fruiting body formation of P. ostreatus was seriously repressed.

        The accuracy and applicability of numerical simulation model for land-sea breezes are studied by the computative study between analytical model and numerical simulation results. In the model, Defant's equation which has analytical solution was selected. Both numerical and analytical solutions represented well land and sea breezes under the condition of various diffusion coefficients and hydrostatic and nonhydrostatic conditions. By the results, the numerical simulation method was proved as an appliable technique to simulate accurate land-sea breezes.

        Bacillus lentimorbus WJ5의 감마선유도 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자의 microarray 분석

        본 연구에서는 감마선으로 유도된 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자들의 발현을 연구하기 위하여, B. lentimorbus WJ5의 방사선 유도 돌연변이체에서 발현되는 유전자를 DNA microarray로 동시에 탐색하였다. DNA microarray는 B. lentimorbus WJ5 genome을 무작위로 절단하여 2,000단편으로 구성하였으며, 감마선 (^(60)Co)으로 유도된 7돌연변이체의 발현을 정량적으로 관찰하였다. 클러스터 분석결과 발현된 408 유전자 중 27개가 감마선 유도 돌연변이체 모두에서 유의하게 발현이 증가되었다. 특히, 복구(mutL, mutM)에너지 대사 (acsA, sdhB, p하, yhjB, citB), protease(npr), 산화자극에 대한 환원 (HMM)관련 유전자들이 동시에 증가되었다. 이는 감마선 유도 돌연변이체들에서 자발적인 직/간접 복구 관련 유전자의 발현 증가는 방사선 노출 직후 보이는 stress response와는 다른 현상임을 나타내는 것으로 생각된다. To study the radiation related gene expression in mutants of Bacillus lentimorbus WJ5 induced by gamm radiation, the simultaneous gene expression was analyzed by DNA microarray. We constructed DNA chips including two thousand randomly digested genome spots of B. lentimorbus WJ5 and compared its quantitative aspect with seven mutants induced by gamma radiation (^(60)Co). From the cluster analysis of gene expression pattern, totally 408 genes were expressed and 27 genes were significantly upregulated by the gamma radiation in all mutants. Especially, genes involved in repair (mutL, mutM), energy metabolism (acsA, sdhB, pgk, yhjB, citB), protease (npr), and reduction response to oxidative stress (HMM) were simultaneously upregulated. It seems that the induction of the direct and/or indirect repair related genes in mutants induced by gamma radiation could be remarkably different from the adaptive responses against acute exposure to radiation.

        Bacillus sp. MS202에 의한 Dinitroaniline계 제초제인 Pendimethalin의 부분환원

        토양과 지하수에서 pendimethalin의 지속성은 환경에 해로운 영향을 미친다. 경남 마산에서 분리한 pendimethalin분해 균주는 API CHB50 kit 시험, FAME 분석, 그리고 165 rDNA 염기서열분석 결과로 Bacillus sp. MS202로 잠정적으로 동정하였다. TLC, GC, 그리고 GC-MS 분석에 의해 Bacillus sp. MS202가 pendimethalin의 -NO₂를 -NH₂로 환원시킨다는 것을 알 수 있었다. 이는 일반적으로 알려진 호기성 미생물에 의한 pendimethalin 분해가 탈알킬화가 우선한다는 보고와 상반되는 새로운 결과이다. The persistence of pendimethalin in soil and ground water has an injurious effect on ecosystem. Pendimethalin-degrading bacterium was isolated from Masan, Gyeongnam province and temporarily identified as Bacillus sp. MS202 by the analysis of API CHB50, kit, FAME, and 16S rDNA sequence. From the analysis of pendimethalin metabolite using TLC, GC, and GC-MS, we found that the degradation of pendimethalin by Bacillus sp. MS202 did not result in the dealkylated form, but the formation of the reduced compound, 6-amino-2-nitro-N(1-ethylpropyl)-3,4-xylidine or 2-amino-6-nitro-N(1-ethylpropyl)-3,4-xylidine.

        김수영 시의 관념성

        In Kim su-young's poetry, there are many non-poetic words and conceptional expressions ; abstract ideas, slang expressions, naturalized words and etc. Some researchers regard it as various one, others disordered. But just making a list of vocabulary means nothing. We could recognize that Kim su-young used words and phrases to achieve the new and vivid meaning, especially in the aspect of using ideological words represented in his poetry. So we must have a thorough grasp the effect and direction of using poetic words. Everyone says that the blooming of the flower is beautiful and miraculous. However, when Kim, su-young had witnessed the moment of blooming, he discovered a popular fallacy in the event and broke the usual diction in his poem "꽃(flower)". In his poem, a flower comes to have whole different meaning from the usual diction. It becames 'a firm flower blooming in disorder'. Kim, su-young frequently raise a question of the meaning and importance of writing in our daily life. In a work titled "눈(snow)", he looked at the snow falling and reflected a poem writing at the same time. Ultimately, Kim, su-young represents that not the ruins but the snow-falling ruins imply the reality. Also in "절망(Despair)", lots of ideological words were repeated and juxtaposed on the surface in the poems. However, the repetition and justaposition of ideological words let us obstinately resist from the reality. In a work titled "구라중화(gladiolus)", Kim, su-young introduced a method of the infinitive mood. By using the infinitive mood, he has denied an ideality of ideological words. He repeat the ideological dictions to identify the substance of a flower but he failed. But at the point of failure, he came up to see the shadow of a flower and surroundings of it. Through this process, he was confronted with death and he secure the power to overcome his time. Kim, su-young used many ideological words, but he did not ignore details of the reality. He gives a shape to ideological words in his poems, and poetic diction achieves a concrete meaning. He did not take shelter behind transcendental rhetorics and did not hide himself behind a poetic diction. He found out a distortion in daily lives and discolsed a suppression in our society.
