고삼 메탄올 추출물로부터 분리한 flavonoids를 IR, NMR등의 분광학적인 방법으로 Leachianone A(LA)과 Sophoraflavanone G(SFG)로 동정하였고, 카드뮴의 세포독성효과를 고삼 및 고삼추출물이 방어할 수 있는 지를 검색하기 위하여 NIG 3T3 세포에 고삼 및 LA와 SFG, LA+SFG를 처리하고 MTT assay 및 광학현미경으로 세포의 생존률을 검색하여 다음과 같은 결론은 얻었다. MIT의 흡광도는 카드뮴의 농도에 의존하여 감소하였으며, IC_50인 MTT50은 12.5??이었으며 카드뮴을 IC_50농도로 처리하고 고삼 및 LA, SFG, LA+SFG를 각 화합물의 자체 세포독성을 갖지 않는 농도로 처리한 후 MTT assay로 세포 생존율을 측정한 결과 LA, SFG, LA+SFG 처리군들은 카드뮴 처리군(MTT_50)에 비해 세포생존률이 증가되었으며 이들은 고삼 및 고삼추출물의 농도에 의존적으로 증가되었고 각각의 단일 화합물보다 두 화합물 LA+SFG 병용처리군에서 수복 효과가 더 좋은 것으로 나타났으며 광학현미경적 소견에서도 세포재생이 뚜렷하게 보였다. 이상과 같이 고삼에서 분리한 flavonoids가 카드뮴 독성에 의하여 손상된 NIH 3T3 섬유아세포의 재생효과에 영향이 있는 것으로 판단된다. The aim of this study was to assess the antitoxic compounds, flavanones (Leachinanone A=LA and Sophora flavanone G=SFG), from Sophora flavescents (S. Flavescens). We investigated the possibility of antitoxicity of LA and SFG against NIH 3T3 fibroblasts cell lines using colorimetric MTT[3-(4-5-dimethylthiazol-2-yl)-2-5diphenyl-2H-tetra-azolium bromide] assay. The results were as follows : After cadmium was treated against NIH 3T3 cell lines, we determined IC_50. Accordingly we have examined the detoxification effects of S. flavescens, LA and SFG under cadmium IC_50=12.5?? and was carried out to observe morphological changes by the light microscopic study. In NIH 3T3 cells, Sophora flavescens, LA, SFG and LA+ SFG showed inhibitory effects on the cytotoxicity of cadmium and these detoxication effects increased in proportion to the concentration of these drugs. These results suggest that LA and SFG from S. flavescens retain a potential antitoxic activity.
Distribution of Heavy Metal Content in Plants and Soil from a Korean Shooting Site

In this research we determined the levels of heavy metals in soil and metal-accumulating plants from a D military shooting site in the Kyungkido district of Korea. The data obtained may be useful in the development of methods for the efficient phytoremediation of contaminated soil. The total Cd, Cu, Pb, and Zn concentrations in the soil were found to be 1.67-5.04 mg/kg, 52.51-106.26 mg/kg, 37.24-90.32mg/kg, and 111.45-188.19mg/kg, respectively. These results show that the soil is contaminated with Cd and Cu, and this contamination is particularly severe in the case of Cd because of its high bioavailability (25-57% of the total metal in the soil is exchangeable). The high concentrations of heavy metals in the shoots of Persicaria thunbergii and Artemisia princeps var. orientalis indicate that these plants (all perennial herbs) accumulate heavy metal efficiently. Further, these plants were found to contain more Cd in its shoots (>60% of the total metal found in the plant) than any other plant; these results indicate that these native species are particularly suited to use in Cd phytoextraction.

Androgen receptor (AR) is ligand-inducible nuclear hormone receptor which has been focused on key molecular target in growth and progression of prostate cancer. We synthesized a series of 4-aryl 2-substituted aniline-thiazole analogs and evaluated their anti-cancer activity in AR-dependent human prostate cancer LNCap cells. Among them, the compound 6 inhibited the tumor growth in LNCap-inoculated xenograft model.
Detection of Nocardia sp. Hl7-1 by PCR during Bioremediation of Crude Oil-Contaminated Soil

For the detection of the oil-degrading bacterium, Nocardia sp. Hl7-1, inoculated during the bioremediation of oil-contaminated soil, a species-specific primer was constructed based on the 16S rDNA sequence of this strain. Two forward primers and two reverse primers were designed and tested against both closely and distantly related bacterial strains. All the primers designed were specific to the Nocardia sp. H17-1. Particularly, primer sets NH169F-NH972R and NH575F-NH972R could be used to detect 50 fg of template DNA and $1.2${\times}$10^4$ CFU/g of sandy soil. These two PCR primer sets successfully detected the H 17-1 strain in the oil-con-laminated soil samples containing heterogeneous DNA. We also conformed the primer specificity by restriction-enzyme cleavage of the PCR products and denaturing gradient gel electrophoresis. 원유로 오염된 토양의 생물학적 복원과정 동안 접종된 Nocardia sp. Hl7-1 균주를 확인하기 위해 165 rDNA sequence에 기초하여 균주에 특이적인 primer를 제작하였다 14균주의 16S rDNA sequence비교를 통해 제작된 4개의 primer set는 Hl7-1 균주를 특이적으로 검출할 수 있었다. 특히 NH169F-NH972R과 NH575F-NH972R의 primer set는 50 fg의 DNA와 $1.2${\times}$10^4$ cfu/g-soil의 균체농도까지 민감하게 검출할 수 있었다. 이 두 primer set는 원유로 오염된 토양의 bioremediation과정 동안 접종된 Hl7-1 균주의 특이적 검출을 가능케 하였으며, 이는 사용된 primer set에 의해 증폭된 PCR산물을 제한효소(EcoRI)로 절단한 결과와 DGGE를 통한 Hl7-1 균주의 확인을 통해 본 연구에서 제작된 primer set의 특이성을 검증하였다.

We investigated the effects of high calorie and low caloriediets on skeletal integrity, and whether β adrenergic blockade(BB) attenuates bone loss induced by dietary calorie alteration. Male 6-week old C57BL/6 mice were assigned to either anad-lib fed control diet (CON), a high calorie diet (HIGH), or alow calorie diet (LOW) group. In each diet group, mice weretreated with either vehicle (VEH) or propranolol, a β-adrenergicantagonist. Over 12-weeks, β-blockade mitigated bodyweight and fat mass increases induced by the high calorie diet. Femoral trabecular bone mineral density and the expressionlevels of osteogenic marker genes in bone marrow cells were reduced in HIGHVEH and LOWVEH mice, and BB significantly attenuated this decline only in HIGH mice. In summary, the magnitude of bone loss induced by low calorie diet was greater than that caused by high calorie diet in growing mice, and β-blockade mitigated high calorie diet-induced bone loss.
