      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

      • General Introduction of American Ginseng Indigenous in USA and Canada

        American ginseng (Panax quinquefolium) is herbaceous perennial plants indigenous to North American forests. This is highly valued as medicinal herbs with a long history of collection from wild populations since 1716. Wild American ginseng distributed from Quebec in Canada to northern Florida in USA. A heavy concentration is found in the Appalachian mountains, although wild American ginseng is considered endangered. The price paid for field cultivated ginseng has dropped dramatically in the past 10 years, while the price for wild or woods cultivated ginseng has rised significantly. The price curve for ginseng resembles a roller coaster, reflecting not only supply and demand but many other factors. This information will be useful to understand American ginseng compared to Korean ginseng.

        DJ-1 deficiency impairs synaptic vesicle endocytosis and reavailability at nerve terminals

        <P>Mutations in DJ-1 (PARK7) are a known cause of early-onset autosomal recessive Parkinson's disease (PD). Accumulating evidence indicates that abnormalities of synaptic vesicle trafficking underlie the pathophysiological mechanism of PD. In the present study, we explored whether DJ-1 is involved in CNS synaptic function. DJ-1 deficiency impaired synaptic vesicle endocytosis and reavailability without inducing structural alterations in synapses. Familial mutants of DJ-1 (M26I, E64D, and L166P) were unable to rescue defective endocytosis of synaptic vesicles, whereas WT DJ-1 expression completely restored endocytic function in DJ-1 KO neurons. The defective synaptic endocytosis shown in DJ-1 KO neurons may be attributable to alterations in membrane cholesterol level. Thus, DJ-1 appears essential for synaptic vesicle endocytosis and reavailability, and impairment of this function by familial mutants of DJ-1 may be related to the pathogenesis of PD.</P>

        콜롬비아 소가모스에서 미세먼지 감축 시나리오 분석

        콜롬비아 소가모스 지역에서 미세먼지 감축 시나리오를 TAPM 모델을 활용하여 도출하고 평가하였다. 시나리오 1의 제철소 및 금속 등 중공업 오염원을 대상으로 미세먼지 방지 기술을 이용하여 배출량을 80% 감축하는 방안에서는 도시 중심 평균 농도는 6.5 μg m-3로 1.5 % 감소하였고, 모델영역에서 최고 평균농도는 387 μg m-3 로 변동 없었다. 시나리오 2의 벽돌 및 석회공장 등을 대상으로 연료를 석탄에서 코크스로 교체하여 90 % 감축하는 방안에서는 도시 중심 평균 농도는 4.5 μg m-3로 31.8%, 모델영역에서 최고 평균농도는 111 μg m-3 로 71.6 % 감소하였다. 시나리오 1과 시나리오 2를 결합한 시나리오 3에서는 도시 중심에서 평균 농도는 4.3 μg m-3로 34.8 %, 모델영역에서 최고 평균농도는 110 μg m-3 로 71.7 % 감소하였다. 소가모스 지역에서 대기환경을 개선하는 로드맵 수립에는 수공업에서 주로 사용되는 석탄 연료를 청정연료인 코크스로 교체하는 방안이 최우선으로 고려되어야 하겠다. 이후 미세먼지 방지 시설을 굴뚝에 설치하는 방안도 차선책으로 고려하여 환경과 경제가 선 순환하는 녹색성장관점에서 장기적인 미세먼지 개선 마스터 플랜이 수립되어야 하겠다. Reduction scenarios for PM10 concentration in Sogamoso, Colombia were analyzed using the TAPM( The Air Pollution Model). The average concentration of PM10 in the downtown area was 6.5 μg m-3, a reduction of 1.5 %. The maximum average concentration in the model domain was 387 μg m-3 without any reduction in scenarios 1 which the PM10 emissions from heavy industries such as iron and metal was reduced by 80 % using the prevention technology. In scenario 2 which the PM10 emissions from the brick and lime industries was reduced by 90 % substituting coke (a cleaner fuel) for coal, the average concentration of PM10 in the downtown area was 4.5 μg m-3, a reduction of 31.8 % , the maximum average concentration in the model domain was 111 μg m-3 , a reduction of 71.6 %. In scenario 3 which the scenarios 1 and 2 were combined, the average concentration of PM10 in the downtown area was 4.3 μg m-3, a reduction of 34.8 % and the maximum average concentration was 110 μg m-3, a reduction of 71.7 %. To put forward a road map for the reduction of PM10 in the Sogamos region, substituting coke for coal in manual industries should have priority over reduction policies. Also, it is highly recommended that a longterm master plan for reducing PM10 concentrations should be established with a focus on environmentally-friendly development and future green growth policy with a win-win relationship between economy and environment.

        한방 닭죽 개발 및 최적 배합비율 확립

        죽의 기능성 향상을 위하여 2:1:1:1의 배합으로 혼합한 한약재료(가시오가피, 숙지황, 산약, 백복령)와 닭가슴살 등의 재료를 사용해 한방 영양죽을 제조하였다. 한방 닭죽의 DPPH radical 소거능, SOD 유사활성, 맛의 선호도, cost는 수학적인 canonical model과 trace plot을 이용하여 분석한 결과, SOD 유사활성 이외에는 각 성분들이 독립적으로 작용하여 상호작용이 없는 선형 모델을 보여주었다. DPPH radical 소거능, SOD 유사활성, 맛의 선호도를 최대로, cost를 최소로 목적 범위를 설정하고, 각각의 canonical 계수를 이용하여 수적 최적화를 통하여 최적 배합비를 구한 결과, 한약 혼합물 101g, 닭가슴살 38.66g, 찹쌀 비율 41.34g의 최적 배합비를 구하였으며, 이 배합비에 따른 맛의 선호도는 6.39, DPPH radical 소거능은 17.25㎎VCE/ℓ, SOD 유사활성은 2.25%, cost는 1,382.39 won으로 나타났으며, 여러 결과들 사이에 최적화된 한 값에 수렴하는 desirability는 0.714로 나타났다. 이는 한약혼합물이 많을수록 기능성 면에서는 높게 나타나지만 맛의 선호도면에서는 낮게 나타냈고, 닭가슴살의 경우에도 그 함량이 많을수록 맛의 선호도는 올라갔지만 그에 반해 cost가 높게 올라간 것을 반영한 결과로 사료된다. The objectives of this study were to develop porridge with medicinal herbs and chicken breast meat and to find out the optimum mixing ratio. Several herb extracts such as Acanthopanacis senticosus, Rehmannia glutinosa, Disocorea japonica, and Poria cocos Wolf were used in the ratio of 2, 1, 1, and 1, respectively. The optimum processing condition for herbal chicken porridge was determined by a design expert program. Seventeen experimental points were selected, and herb extracts (82∼101g), chicken breast meat (30∼50g), and glutinous rice (40∼60g) were chosen as the independent variables. The measured responses were preference of taste, DPPH radical scavenging, SOD-like activity, and cost. The optimum formulation of herbal chicken porridge using the numerical analysis was set at herbal extracts (101g), chicken breast meat (38.66g), and glutinous rice (41.34g) with a 0.714 desirability value. DPPH radical scavenging effect, preference of taste and the cost showed a linear model, whereas SOD-like activity showed a quadratic model indicating a higher interaction among the mixture. As a result of proximate composition of optimized herbal chicken porridge, the contents of moisture, carbohydrate, crude protein, crude lipid, and ash contents were 76.4, 9.0, 1.4, 0.5, and 1.6%, the calories of the porridge was 46.1kcal/100g.

        제왕절개 분만율의 지역간 변이와 관련요인에 대한 연구

        The purpose of this study is to estimate cesarean section rate in Korea and analyze the socioeconomic variables and health resources which affect regional variation in the rate. Samples were drawn from the record of vaginal and cesarean section deliveries based upon insurance claim bills which have been submitted to the National Federation of Medical Insurance for the first three months, January through March, 1991. The results are obtained as follows : It was found that. cesarean section rate was increasing rapidly up to 23.1% in 1991. Cesarean section per 10 thousand insured people was 4.8 and the number of cesarean section per 10 thousand insured eligible($15{\sim}49$ years old) female was 7.6. The fee for normal delivery was 109,489 won and that for cesarean section was 390,024 won. The average days of hospitalization in normal delivery was 2.3 days, and those in cesarean section was 7.6 days. On the average cesarean section has a longer of stay as much as by 4.3 days and cost 3.6 times more than normal deliveries. Cesarean section rates vary among medical facilities 19.8% at clinics 37.6% in small-scale hospitals, and 29.1% in general hospitals. The regional variation of cesarean section rates was also fairly prominent. The South Cheju Gun has the highest rate of cesarean section, 56.2%. Meanwhile no cesarean section cases has been reported in Sunchang Gun during the period of this study. The variation is noted among provinces. The rate for Cheju province has been 3.4 times higher than that for Chunnam. The number of cesarean section per 10 thousand insured people vary greatly among regions, too. This study has found that there exists significant regional variations among various geographic units in terms of average length of stay, average cost, number of obsretricians and number of beds. Multiple regression analysis was done to identify factors explaining the regional variance of various cesarean section rates : In the urban areas, no significant explaining variables were noted except the number of beds for the dependent variable of cesarean section cases per 10 thousand insured eligible females. The smaller the number of bed, the more cases of cesarean section was noted for an urban area. The is mostly because the rate of cesarean section is higher in medium-size hospitals than in large general hospitals. In the rural areas, the factor of education has been found significant for all three deplendent variables. The higher the educational level, the rate of cesarean section is most likely to rise. An income variable measured by the amount of monthly insurance contribution has been identified a powerful predictor in explaining the valiance of cesarean section rates. The same has been noted for the number of obstetricians. Similar findings are observed for the country as a whole. The income level has veen found as the most powerful explaining factor in the regional variance of cesarean section rates. In general the rate is higher in the urban areas, and lower in the area with more small hospitals. As this is the initial attempt to identify the factors relevant to the regional difference in the rates of cesarean section, more elaborated study is urgently required.

        Background: We report a rare case of CAPD peritonitis caused by Ochrobacterium anthropi. Introductions: Among the continuous ambulatory peritoneal dialysis(CAPD) patients, peritonitis is one of the most commonly taken complications, and also the general cause of dialytic modality exchange to hemodialysis. Usual pathogens of CAPD peritonitis may be bacteria, fungi, mycobacteria. Coagulase negative staphylococci, S. aureus, campylobacter, pseudomonas are common reported pathogens among the bacteria, of CAPD peritonitis cases, meanwhile candida is among fungi. Ochrobacterium anthropi is one species of Brucellaceae, which is rare pathogen of human disease. Case (Methods and results): 73 years old female, who was on CAPD due to diabetic end stage renal disease visited Kyung-Hee University hospital with intermittent abdominal pain. Body fi uid analysis showed increased white blood cell(WBC) count of 26,750/mm3 with her peritoneal fi uid. Culture study with peritoneal fi uid suggested O. anthropi, and DNA sequencing with PCR was consistent with O. anthropi. Intraperitoneal ceftazidime and cefazolin were administrated as empirical antibiotics. Ceftazidime resistance was noted with the result of antibiotics sensitivity test at 7th day of hospitalization, and antibiotics were changed into intraperitoneal gentamicin, which showed sensitivity to the pathogen. CAPD catheter removal and antibiotics re-exchange into imipenem and cefazolin, which were other sensitive antibiotics by the sensitivity test, were done since clinical manifestation and peritoneal fi uid WBC count was repeatedly improved and aggravated. The patient discharged with improved lab test results and resolving clinical symptoms afterward. Conclusions: We presentators report rare case of CAPD peritonitis with pathogen of O. anthropi. The pathogen of the case confi rmed by classical microbiologic, and molecular biologic Methods: The patient was unable to treat only with antibiotics, thus CAPD catheter, which might be act as colonizing source, was removed, and the disease resolved.

      • 소아 알레르기비염의 경제적 부담 평가를 위한 다기관 조사

        목적: 알레르기비염의 유병률은 전 세계적으로 급격히 증가하고 있으며, 그에 따른 경제적 부담도 증가하고 있다. 그러나, 소아 알레르기 질환이 미치는 경제적 부담에 관한 국내 연구는 제한적이어서 소아 알레르기비염이 환자와 그 가족에게 미치는 경제적 부담을 산출하고자 본 연구를 진행하였다. 방법: 2008년 7월 1일부터 9월 31일까지 서울시 6개 2-3차 의료기관을 방문한 18세 이하 소아 알레르기비염 환자 및 보호자를 대상으로 설문 조사를 진행하였다. 설문을 통해 직접 의료비 (병·의원 진료비, 및 약제비 한방 진료비 및 약제비, 보완/대체요법비), 직접비의료비 (교통비, 환경개선비), 그리고 간접비용 (월 평균 노동 손실)을 조사하였고 그 결과를 질환의 중증도 및 유병 기간에 따라 분석하여 그 차이를 비교하였다. 결과: 모집된 262명의 대상 중 174명(66.4%)이 남자였고, 평균 연령은 6.54세였다. 대상군의 연간 평균 직접 의료비는 177만 원이었고 직접비 의료비는 57만 원이었다. 비록 통계학적 유의성은 없었지만, 알레르기비염의 중증도가 증가할수록 직접 의료비가 증가하는 경향을 보였고, (P=0.053) 유병 기간 또한 직접 의료비의 증가와 유의한 양의 상관 관계를 보였다. (R=0.195, P=0.002) 대상 환자 보호자의 약 17%가 아이의 알레르기비염으로 인해 직장에 결근 또는 조퇴를 경험한 것으로 조사되어 사회적 간접비용을 미루어 생각할 수 있었다. 결론: 알레르기비염이 미치는 경제적 부담은 질환의 중증도가 심할수록, 그리고 유병 기간이 길수록 증가하며, 이는 특히 한방 진료 및 치료비와 보완/대체요법비의 증가에 기인한 것으로 조사되었다. 따라서 알레르기비염으로 인한 경제적 부담을 줄이기 위해서는 환자와 보호자를 대상으로 근거 중심의 치료를 할 수 있도록 지속적이고 체계적인 교육이 필요할 것으로 사료된다. Purpose: The prevalence of allergic rhinitis is rapidly increasing and results in relatively high socio-economic burden on their family and community. However, studies on the economic burden of pediatric allergic rhinitis in Korea are limited. Therefore, we conducted this study to investigate the impact of pediatric allergic rhinitis on economic burden. Methods: Two hundred sixty two children with allergic rhinitis were enrolled in 6 secondary or tertiary medical centers in Seoul from July to September, 2008. We collected data of the economic burden of allergic rhinitis (direct medical costs, direct nonmedical costs, and indirect costs) by face to face questionnaire survey. We compared the economic burden according to the severity and the duration of allergic rhinitis. Results: The mean age of subjects was 6.54 years, and male were 174 (66.4%). Direct medical costs (10,000 Korean Won/yr) were 177.75, and direct nonmedical costs were 57.92. Although, there was no statistical significance, direct medical costs showed increasing trends in severe allergic rhinitis. (P=0.053) In addition, direct medical costs were positively correlated with duration of allergic rhinitis.( R=0.195, P=0.002). About 17% of the parents who care the allergic rhinitis children experienced the work absence due to their child s illness. Conclusion: The economic burdens of allergic rhinitis were positively correlated with the severity and duration of illness. Particularly costs for alternative medicine including oriental medicine` were related with severity and duration allergic rhinitis. Therefore, special efforts for education with evidence based treatment strategy are necessary to decrease the economic burden of allergic rhinitis.

        Notes on bird species newly recorded in Korea

        A total of 527 species of birds have been recorded in Korea. We summarize the records of three genera and six species that have been newly reported in Korea, discovered through the avian monitoring programs carried out by The National Institute of Biological Resources from 2013 to 2016. These six newly recorded species are; Ring-necked Duck (Aythya collaris), Lesser Scaup (Aythya affinis), Bufflehead (Bucephala albeola), Black-winged Kite (Elanus caeruleus), Mongolian Lark (Melanocorypha mongolica) and Crested Myna (Acridotheres cristatellus). One Ring-necked Duck was observed on March 23, 2014 in Jungrangcheon Stream. One male Lesser Scaup was recorded on February 24, 2014 in Gyeongpoho Lagoon. A male bufflehead was first seen in Yeongrangho Lagoon on January 26, 2013. The Black-winged Kite was first observed on January 23, 2013 at the Gangseo Wetland Ecological Park. One Mongolian Lark was also recorded on May 11, 2016 and May 12, 2017 in Marado Island. The Crested Myna was first observed on April 20, 2016 in Seomando, Island.
