Backgroud : Gagamtongsun-san(加減通順算) is a herbal medicine which has been used for the traditional therapeutic agent of removal of Phlegm and tumor by Phlegm in the oriental medicine. Objective : This study was performed to investigate the effect of Gagamtongsun-san on the tumor and immune response in the mouse B16 melanoma tumor model. Materials and Methods : The tumor was induced by subcutaneous inoculation of B16BL6 melanoma cells in the shaved dorsal region of mice. Mice were orally administered with Gagamtongsun-san extract (28mg/mouse) for 14days after inoculation. For making examination of antitumor effect, the Increase of life span, Tumor growth inhibition rate, change of body weight were measured and evaluated. For the immune response increasing effect, the percentage of T lymphocyte and B Lymphocyte in the peripheral blood, the percentage of CD4+ T-cell, CD8+ T-cell CD4+/CD8+ ratio in the peripheral blood and spleen, interleukin-2 productivity were measured and evaluated. Result : Gagamtongsun-san showed 18.55% increase of life span, 35.23% tumor growth inhibition rate and increase of body weight. Gagamtongsun-san increased the percentage of T lymphocyte in the peripheral blood, CD4+ T cell percentage of peripheral blood and spleen, and Interleukin-2 productivity as compared with the control group. Whereas Gagamtongsun-san had no effect on the percentage of B lymphocyte in the peripheral blood, the percentage of CD8+ T cell. CD4+/CD8+ T-cell ratio in both of peripheral blood and spleen as compared with the control group. Conclusion : This study shows that Gagamtongsun-san has anti-tumor effects and immunoregulatory effects on the B16 melanoma tumor model. It is suggested that Gagamtongsun-san could be a useful immunomodulator and anti-tumor agent.
Modeling and Parameter Extraction for the Series Resistance in Thin-Film Transistors

<P>A new parameter extraction method is proposed for the series resistance of thin-film transistors (TFTs). By analyzing the gate-source overlap region of staggered structure TFTs, the model for the series resistance is derived and utilized for the parameter extraction. To verify the extraction method, the characteristics of amorphous silicon TFTs obtained from TCAD simulation are used. For the devices with different overlap lengths, the extracted parameters are identical to each other, although the series resistances are different due to the narrow overlap length. When the actual channel length is different from the mask-specified length, the offset length can be effectively corrected by the new method, so that accurate parameters can be obtained. Because the new method has several advantages such as the accuracy and generality over the conventional method, it can be used for further analysis of TFT characteristics.</P>

Cystic parathyroid adenoma is one of the rare causes of hyperparathyroidism, and is usually located in neck and mediastinum. This type of adenoma tends to cause increased serum level of parathyroid hormone and alkaline phosphatase, similar serum calcium level when compared to those of solid adenoma. Thanks to current radioimmunoassay and easy accessibility to serum autoanalyzer, serum calcium level and parathyroid hormone level are more easily measured, which in turn lead to more easier diagnosis of hyperparathyroidism. Also improvement in imaging and nuclear diagnostic method of parathyroid lesion are suggested to enable easy diagnosis of cystic parathyroid adenoma. A 35-year-old male presented with easy fatigability for 12 months. The serum calcium, phosphate, alkaline phosphotase were 11.5mg/dL, 1.4mg/dL, 194IU/L respectively and his parathyroid hormone level in serum was 126.42pg/mL. Neck CT showed enlargement of right lobe of thyroid gland with well defined inhomogenously enhanced density inside the right thyroid gland. The patient was diagnosed of hyperparathyroidim due to parathyroid adenoma and was surgically removed. The surgical biopsy showed cystic parathyroid adenoma. After operation his general condition was improved and serum calcium, phosphate, parathyroid hormone level were normalizd. We report a case of hyperparathyroidism caused by cystic parathyroid adenoma with brief review of literature. (J Kor Soc Endocrinol 12:75-80, 1997)

<P><B>Background</B></P><P>Nanoparticles fabricated from the biodegradable and biocompatible polymer, polylactic-co-glycolic acid (PLGA), are the most intensively investigated polymers for drug delivery systems. The objective of this study was to explore fully the development of a PLGA nanoparticle drug delivery system for alternative preparation of a commercial formulation. In our nanoparticle fabrication, our purpose was to compare various preparation parameters.</P><P><B>Methods</B></P><P>Docetaxel-loaded PLGA nanoparticles were prepared by a single emulsion technique and solvent evaporation. The nanoparticles were characterized by various techniques, including scanning electron microscopy for surface morphology, dynamic light scattering for size and zeta potential, x-ray photoelectron spectroscopy for surface chemistry, and high-performance liquid chromatography for in vitro drug release kinetics. To obtain a smaller particle, 0.2% polyvinyl alcohol, 0.03% D-α-tocopheryl polyethylene glycol 1000 succinate (TPGS), 2% Poloxamer 188, a five-minute sonication time, 130 W sonication power, evaporation with magnetic stirring, and centrifugation at 8000 rpm were selected. To increase encapsulation efficiency in the nanoparticles, certain factors were varied, ie, 2–5 minutes of sonication time, 70–130 W sonication power, and 5–25 mg drug loading.</P><P><B>Results</B></P><P>A five-minute sonication time, 130 W sonication power, and a 10 mg drug loading amount were selected. Under these conditions, the nanoparticles reached over 90% encapsulation efficiency. Release kinetics showed that 20.83%, 40.07%, and 51.5% of the docetaxel was released in 28 days from nanoparticles containing Poloxamer 188, TPGS, or polyvinyl alcohol, respectively. TPGS and Poloxamer 188 had slower release kinetics than polyvinyl alcohol. It was predicted that there was residual drug remaining on the surface from x-ray photoelectron spectroscopy.</P><P><B>Conclusion</B></P><P>Our research shows that the choice of surfactant is important for controlled release of docetaxel.</P>
Quantitative Analysis and Preformulation of Extracts from Alnus Japonica

Alnus japonica has been known to exert antioxidative, anti-inflammatory, anti-cancer and immune response inhibitory effects. The aim of this study was to figure out the characteristics of extracts obtained with different extraction solvent such as water, 100% ethanol, 70% ethanol or 70% methanol because characteristic components such as oregonin and hirsutanone extracted from Alnus japonica might be essential for the biological activity. For this purpose, oregonin and hirsutanon of four extracts, index ingredient of Alnus japonica, were analyzed with HPLC and physicochemical studies such as SEM, particle size and zeta potential were conducted. In cell cytotoxicity study, extract of water showed the highest cytotoxicity among four extracts. In case of oregonin, 70% MeOH and water extracts showed high contents and in case of hirsutanone, all extracts showed similar contents except 70% EtOH extracts. The extract of 70% MeOH from Alnus japonica for both oregonin and hirsutanone appeared to have the highest content. Both oregonin and hirsutanone extracted from Alnus japonica using 70% methanol showed stability in pH 1.2.

Since July 2005, survey of chestnut ink disease was carried out in chestnut stands located at southern parts of Korea. Dead chestnut trees showing inky ooze on necrotic trunks were found in two different locations. In order to isolate and identify the causal fungus, infected tissues and soil samples around dead or dying trees were collected and placed on Phytophthora-selective medium. Rhododendron and chestnut tree leaves were used as a bait to isolate the fungus from soil samples by attracting zoospores in soil suspensions. On V-8 culture medium, the isolates produced homothallic oogonia with protuberances ($34.0-46.2{\times}21.9-26.7{\mu}m$) abundantly, but did not produced sporangia. Mass production of sporangia was possible by immersing agar plugs with actively growing mycelium in the creek water at $18^{\circ}C$ for 3 days. Sporangia were papillate, and ovoid to obpyriform ($17.0-38.9{\times}14.6-29.2{\mu}m$) in shape. Comparison of the ITS sequences revealed that the isolates had 100% identity to the P. katsurae isolates from Japan and New Zealand and 99.6% identity to other P. katsurae isolates. All of the examined isolates from Korea were completely identical to each other in ITS sequence. Numerous sporangia were formed in filtered as well as unfiltered creek water, but no sporangia formed in sterilized distilled water. Light induced sporangia formation, but has no influence on oospore formation. Amendments of ${\beta}$-sitosterol in culture media have no significant effect on mycelial growth but significantly stimulate oospore and sporangia formation.

저압 축소형 연소 시험을 위해 제작되는 연소기 분사기면의 구조 안정성 해석을 수행하였으며, 시험압력의 1.2배의 압력에서 분사기면에서 큰 변형이 발생하여 항복강도 이상의 응력이 발생하는 것으로 나타났다. 더미 분사기를 추가하여 개선된 설계를 사용하였을 때 구조적으로 안전여유 132% 이상의 안정된 분사기면을 가진 축소형 연소기로 해석 결과가 나타났다. Structural analysis of injector face plate for low-pressure subscale thrust chamber has been performed, and the results of structural analysis showed that the stresses over yield strength was observed because of the severe deformation of the injector face plate at 1.2 times its operating pressure. After design modification with additive dummy injector, the injector face plate of the subscale thrust chamber was found to be structurally stable with more than 132% safety margin.
