      • ZNF509S1 downregulates PUMA by inhibiting p53K382 acetylation and p53-DNA binding

        Expression of the POK family protein ZNF509L, and -its S1 isoform, is induced by p53 upon exposure to genotoxic stress. Due to alternative splicing of the ZNF509 primary transcript, ZNF509S1 lacks the 6 zinc-fingers and C-terminus of ZNF509L, resulting in only one zinc-finger. ZNF509L and -S1 inhibit cell proliferation by activating p21/CDKN1A and RB transcription, respectively. When cells are exposed to severe DNA damage, p53 activates PUMA (p53-upregulated modulator of apoptosis) transcription. Interestingly, apoptosis due to transcriptional activation of PUMA by p53 is attenuated by ZNF509S1. Thus we investigated the molecular mechanism(s) underlying the transcriptional attenuation and anti-apoptotic effects of ZNF509S1. We show that ZNF509S1 modulation of p53 activity is important in PUMA gene transcription by modulating post-translational modification of p53 by p300. ZNF509S1 directly interacts with p53 and inhibits p300-mediated acetylation of p53 lysine K382, with deacetylation of p53 K382 leading to decreased DNA binding at the p53 response element 1 of the PUMA promoter. ZNF509S1 may play a role not only in cell cycle arrest, by activating RB expression, but also in rescuing cells from apoptotic death by repressing PUMA expression in cells exposed to severe DNA damage.

        S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

        S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

        1H NMR Measurements of the Phase Transition of (NH₄)₃H(SO₄)₂ Single Crystals

        $^1$H nuclear magnetic resonance (NMR) experiments have been performed in the temperature range of 30 -- 300 K at 7 T to investigate the phase-dependent nature of the dynamic network of hydrogen bonds in a ((NH₄)₃H(SO₄)₂ single crystal. The crystal has six phases, which are ferroelectric, antiferroelectric, incommensurate, antiferroelectric, ferroelastic, and superionic with the respective transition temperatures of 63, 133, 139, 256 and 413 K. The spin-lattice relaxation time, T₁, of ¹H NMR is similar for the ammonium protons and the hydrogen-bond protons over the entire range of experimental temperatures. The T₁, of ¹H NMR gradually decreases down to 120 K and starts to steeply increase below 100 K. Then, the T₁ shows an abrupt decrease below 68 K with a sharp minimum at 63 K, where the ferroelectric transition occurs. The ¹H NMR spectrum shifts to the high-frequency side at temperatures below 63 K due to the ferroelectric phase transition. The spectrum at 65 K shows a distinctive change in line shape toward the ferroelectric transition at 63 K. The measured values of T1 for ammonium and hydrogen-bond protons are similar in the whole range of temperature. T1 of 1H NMR shows a gradual decrease down to 120 K and starts to steeply increase below 100 K. Then T1 shows abrupt decrease below 70 K with a sharp minimum at 63 K, where the ferroelectric transition occurs. This temperature dependence of spectrum and T1 clearly prove that the large change in the dynamics of hydrogen bond network is associated with the ferroelectric phase transition at 63 K.

All rights reserved</P><P>Background</P><P>The presence of latent tuberculosis (TB) infection (LTBI) should be evaluated before kidney transplantation. Although a new T cell-based assay for diagnosing LTBI gave promising results, this assay has not yet been compared with the tuberculin skin test (TST) for diagnosing LTBI in renal transplant candidates before transplantation.</P><P>Patients and methods</P><P>All adult patients admitted to a single institute for renal transplantation over a 1-year period were prospectively enrolled. A clinically predictive risk of LTBI was defined as: (i) recent close contact with a person with pulmonary TB; (ii) abnormal chest radiography; (iii) a history of untreated or inadequately treated TB; or (iv) a new infection (i.e., a recent conversion of TST).</P><P>Results</P><P>Of 209 renal recipients, 47 (22%) had a positive TST≥5 mm, 21 (10%) had a positive TST≥10 mm, 65 (30%) had a positive T-SPOT.<I>TB</I> test, and 25 (12%) had an indeterminate T-SPOT.<I>TB</I> test. The induration size of TST was significantly associated with a high positivity rate on T-SPOT.<I>TB</I> (<I>P</I><0.001). Agreement between T-SPOT.<I>TB</I> test and TST≥10 mm was fair (<I>k</I>=0.24, 95% confidence interval 0.11–0.36). However, neither univariate nor multivariate analysis showed any association between the clinical risk for LTBI and positivity on T-SPOT.<I>TB</I> or TST.</P><P>Conclusion</P><P>T-SPOT.<I>TB</I> test was more frequently positive than TST in renal transplant candidates. However, further longitudinal studies are awaited to determine whether the ability of T-SPOT.<I>TB</I> assay to detect LTBI in renal transplant recipients can better predict the development of TB than can TST after transplantation.</P>

© 2010 John Wiley & Sons A/S.</P><P><B>Background – </B> It has not been clarified whether the disparity in ischemic stroke outcome between populations is caused by ethnic and geographic differences or by variations in case mix. Propensity score matching (PSM) analysis can overcome some analytical problems but is rarely used in stroke outcome research. This study was to compare the ischemic stroke case‐fatality between two PSM cohorts of Sweden and Korea.</P><P><B>Methods – </B> Prognostic variables related to baseline characteristics and stroke care were included in our PSM model. Then, we selected 7675 Swedish and 1220 Korean patients with ischemic stroke from each stroke registers and performed one‐to‐one matching based on propensity scores of each patient.</P><P><B>Results – </B> After PSM, all measured variables were well balanced in 1163 matched subjects, and the 90‐day case‐fatality was identical 6.2% (HR 0.997, 95%CI 0.905–1.099) in Sweden and Korea.</P><P><B>Conclusions – </B> No difference is found in the 90‐day case‐fatality in propensity score‐matched Swedish and Korean patients with ischemic stroke.</P>

        한우개량을 위한 유단백질의 유전적 다형현상에 관한 연구

        Genetic polymorphisms of milk proteins, αs₁-casein (αs₁-CN) β-casein (β-CN), k-casein (k- CN) and β-lactoglobulin (β- LG), in milk samples from 175 Korean native cattle were analyzed by using starch gel elec-trophoresis. Their genotype and gene frequencies were estimated and their genetic relationships with other breeds were compared by using genetic distance and dendogram. The results obtained were summarized as fallows ; 1. Three genetic variants each were identified at αs₁-CN (BB, BC, and CC), k-CN (AA. AB and BB) and β-LG (AA, BB and BB) and nine genetic variants (A¹A¹, A²A², A¹A², A¹A⁴, A²A⁴, BA¹, BA², BA⁴ and BB) existed at β-CN. 2. Genotype distributions of the milk protein loci were as fallows: αs₁-CN BB 76% BC 21.1%, CC 2.9% β-CN A²A²40%, A¹A²38.3%, BA²11.4%, A¹A¹3.4%,, A²A⁴2.9%, BA¹1.7%, A¹A⁴1.1%, BA⁴0.6%, BB 0.6%, k-CN AB 52%, AA 37.1%, BB 10.9%, β-LG BB 76%, AB 23.4%, AA 0.6%. The distributions of genotypes in each milk protein locus were corresponded closely to expectations of Hardy-Weinberg equilibrium. 3. Gene frequencies of the milk protein loci were: αs₁-CN^B 0.866, αs₁-CN^C 0.134; β-CN^A2 0.660, β-CN^A1 0.235, β-CN^B 0.080, β-CN^A4 0.025: k-CN^A 0.631. k-CN^B 0.369; β-LG^A 0.877, β-LG^B 0.123. 4. From the analysis of` the genetic distance values and the dendogram drawn from the genetic distances matrix, the results indicate that there was a close relationship between Korean native cattle and Japanese native cattle, especially Japanese Brown. Consequently, it is supposed that Japanese native cattle originated from Korean native cattle.
