From rabbit antibody repertoires to rabbit monoclonal antibodies

In this review, we explain why and how rabbit monoclonal antibodies have become outstanding reagents for laboratory research and increasingly for diagnostic and therapeutic applications. Starting with the unique ontogeny of rabbit B cells that affords highly distinctive antibody repertoires rich in in vivo pruned binders of high diversity, affinity and specificity, we describe the generation of rabbit monoclonal antibodies by hybridoma technology, phage display and alternative methods, along with an account of successful humanization strategies.

Lemierre’s syndrome is characterized by anaerobic septicemia, internal jugular vein thrombosis, and septic emboli associated with infections of the head and neck. We describe an unusual and clinically confusing case of a young woman with an acute paresis of the abducens nerve and partial paresis of the right oculomotor nerve. After an extensive imaging diagnostic procedure, we also documented a peritonsillar abscess and various types of thromboses in intracranial and extracranial veins. Furthermore, we found brain and lung abscesses, which led us to establish the diagnosis of Lemierre's syndrome. Despite intensive anti-coagulation and antibiotic therapy,the patient developed a mycotic aneurysm in the right internal carotid artery directly adjacent to the previously thrombosed cavernous sinus. In summary, we were able to confirm that Lemierre's syndrome may occur in conjunction with uncharacteristic symptoms. Due to the sometimes confusing clinical symptoms as well as clinical and radiological specialties, we had to work on an interdisciplinary basis to minimize the delay prior to establishing the diagnosis and therapy.
Quantitative Review of Oral Cancer Research Output from Pakistan

Background: Oral cancer is the most common cancer among men and second only to breast cancer among women in Pakistan. For the effective control and prevention of oral cancer, Pakistan needs to recognize the importance of research and generation of the evidence-base which can inform policy making and planning and implementation of intervention programs. The objective of this review was to quantify oral cancer research output in Pakistan. Materials and Methods: A systematic electronic search in "Medline", "ISI-Web of Science" and "Pakmedinet", supplemented by a Google search, was carried out in January and February, 2014, to identify literature from Pakistan relevant to oral cancer. The selection of publications for the review was carried out according to preset criteria. Data were recorded and analyzed using Microsoft Excel. Results: A total of 166 publications comprising 62 case series, 36 cross sectional, 31 case control, 10 basic laboratory research, eleven reviews and two trials, were included in this review. Some 35 % of the publications focused on risk factors for oral cancer. COMSATS Institute of Information Technology was the institution with the highest contribution. Conclusions: There is a lack of research in the field of oral cancer research in Pakistan. Focused efforts should be put in place to improve both quality and quantity of oral cancer research in the country.
Modeling, Simulation and Fault Diagnosis of IPFC using PEMFC for High Power Applications

An Interline Power Flow Controller (IPFC) is a converter based controller which compensates and balance the power flow among multi-lines within the same corridor of the multi-line subsystem. The Interline Power Flow Controller consists of a voltage source converter based Flexible AC Transmission System (FACTS) controller for series compensation. The reactive voltage injected by individual Voltage Source Converter (VSC) can be controlled to regulate active power flow in the respective line in which one VSC regulates the DC voltage, the other one controls the reactive power flows in the lines by injecting series active voltage. In this paper, a circuit model for IPFC is developed and simulation of interline power flow controller is done using the proposed circuit model. Simulation is done using MATLAB Simulink and PSPICE. The results obtained by MATLAB are compared with the results obtained by PSPICE and compared with theoretical values.

Ruspolia differens (Serville) (Orthoptera: Tettigoniidae) is a highly valued edible grasshopper species in Africa. However, the effects of plant diets on lipid content and fatty acid composition of R. differens are not well understood. We tested the effects of four diets on the total lipid content and fatty acid composition of R. differens. Sixth instar nymphs of R. differens were reared on one, and mixtures of two, three, and six natural plant inflorescences. Individuals collected from the field constituted a control treatment. We extracted lipids and analyzed the fatty acid methyl esters using gas chromatography–mass spectrometry. We analyzed if the total lipid content, body weight, and fatty acid composition differed among diets and between the sexes using two‐way ANOVAs and a PERMANOVA model, respectively. The total lipid content and weight of R. differens did not differ among the diets. The nine common fatty acids were palmitic (mean across treatments, 26%), oleic (22%), palmitoleic (18%), linoleic (13%), stearic (7%), myristic (6%), myristoleic (4%), α‐linolenic (2%) and arachidic acid (1%). The composition of fatty acids and the proportion of essential fatty acids significantly differed among the diets. The proportion of essential fatty acids was highest in the control treatment (21%) but low in less diversified (one to three feed) diets (12–13%). This study demonstrates that the fatty acid composition in R. differens can be influenced through diet. Thus, with dietary manipulations, using local plants in Africa, it is possible to produce R. differens with preferred high quality essential fatty acids for human consumption.
Properties and Applications of Magnetic Tunnel Junctions

The discoveries of antiferromagnetic coupling in Fe/Cr multilayers by Grünberg, the Giant Magneto Resistance by Fert and Grünberg and a large tunneling magnetoresistance at room temperature by Moodera have triggered enormous research on magnetic thin films and magnetoelectronic devices. Large opportunities are especially opened by the spin dependent tunneling resistance, where a strong dependence of the tunneling current on an external magnetic field can be found. We will briefly address important basic properties of these junctions like thermal, magnetic and dielectric stability and discuss scaling issues down to junction sizes below 0.01 ㎛² with respect to single domain behavior, switching properties and edge coupling effects. The second part will give an overview on applications beyond the use of the tunneling elements as storage cells in MRAMs. This concerns mainly field programmable logic circuits, where we demonstrate the clocked operation of a programmed AND gate. The second ‘unconventional’ feature is the use as sensing elements in DNA or protein biochips, where molecules marked magnetically with commercial beads can be detected via the dipole stray field in a highly sensitive and relatively simple way.
