        미세각막절개도 헤드의 선택을 통한 맞춤식각막절개방식의 라식

        Purpose: To compare the intraoperative and postoperative complication rates and visual outcomes with M2 130 head (thick flap group) and M2 110 head (thin flap group). Methods: One-hundred-ninety-five eyes of 104 patients who underwent LASIK with the Moria M2 microkeratome and Allegretto-wave laser were reviewed retrospectively. Selection of M2 heads was based on preoperative pachymetry and estimated ablation depth. Intraoperative and postoperative flap-related complications, mean postoperative uncorrected visual acuity (UCVA), and mean spherical equivalent (MSE) were evaluated and compared. Results: Mean follow-up was 8.1 months. The number of eyes, preoperative MSE and mean corneal thickness of M2 130 were 115, -4.04±1.63diopter (D) and 549.40±39.16??m, and 85, -6.61±3.43D and 525.16±24.53 ?m, respectively, in the 110 head group. Mean UCVA and MSE at postoperative 1 week, 3 months and 6 months were 1.00±0.18, -0.26±0.49D; 1.07±0.68, -0.40±0.51D; and 1.01±0.22, -0.51±0.50D, respectively, in the 130 head group, and 0.90±0.23, -0.46±1.02D; 0.91±0.23, -0.67±0.79D; 0.85±0.46, -0.75±0.88D, respectively, in the 110 head group. There were no significant differences in intraoperative and postoperative flap-related complication rates between the two groups (p=0.316). There were no statistically significant differences in postoperative mean UCVA or MSE between the two groups (p>0.05), except for MSE at the third and sixth postoperative months (p=0.005, 0.013). Conclusions: Proper selection of M2 heads by preoperative pachymetry allowed for an adequate residual stroma bed with good visual outcome. Utilizing one single microkeratome and switching between two heads was advantageous and cost-effective.

        Supplemented vaccination with tandem repeat M2e virus-like particles enhances protection against homologous and heterologous HPAI H5 viruses in chickens

        Highly pathogenic avian influenza (HPAI) H5 viruses derived from A/Goose/Guangdong/1/96 have been continuously circulating globally, severely affecting the public health and poultry industries. The matrix 2 protein ectodomain (M2e) is considered a promising candidate for a universal cross-protective influenza vaccine that provides more effective control over HPAI H5 viruses harboring variant hemagglutinin (HA)-antigens. Here, we evaluated the protective efficacy of a tandem repeat construct of heterologous M2e presented on virus-like particles (M2e5x VLPs) either alone or as a supplement against HPAI H5 viruses in a chicken model. Chickens immunized with M2e5x VLPs alone induced M2e-specific antibodies but were not protected against HPAI H5. The homo- and cross-protective efficacy of M2e5x VLP-supplemented vaccination of chickens was also examined. Importantly, supplementation with M2e5x VLPs induced significantly higher levels of antibodies specific for M2e and different viruses as well as provided improved protection against homologous and heterologous HPAI H5 viruses. Considering the limited efficacy of inactivated vaccines, supplement vaccination with M2e5x VLPs may be an effective measure for preventing outbreaks of HPAI viruses that have the ability to constantly change their antigenic properties in poultry.

        Identification of Aggregation-Sex Pheromone of the Korean Monochamus alternatus (Coleoptera: Cerambycidae) Population, the Main Vector of Pine Wood Nematode

        <P>We confirmed an aggregation-sex pheromone of the Korean Monochamus alternatus Hope (Coleoptera: Cerambycidae) population, which primarily transmitted the pine wood nematode in Korea. Only the M. alternatus male emitted monochamol which has been previously reported in the Chinese M. alternatus population. In field attraction testing, the monochamol+alpha-pinene+ethanol combination attracted more M. alternatus adult beetles than traps containing only monochamol, ethanol, or alpha-pinene. The bark beetle aggregation pheromone compounds of ipsenol, ipsdienol, and ipsenol+ipsdienol, did not show synergistic effects with the monochamol+alpha-pinene+ethanol combination. No synergistic effects were observed when each host volatile compound of 3-carene, beta-caryophyllene, limonene, myrcene, and beta-pinene was added to traps containing monochamol+alpha-pinene+ethanol. The ratio of female beetles was much higher in traps containing the monochamol+alpha-pinene+ethanol combination than that of the natural population. Our results indicated that pheromone and synergists could be useful for managing the M. alternatus population.</P>

      • The human AQP4 gene : Definition of the locus encoding two water channel polypeptides in brain

        The aquaporin family of membrane water transport proteins are expressed in diverse tissues, and in brain the predominant water channel protein is AQP4. Here we report the isolation and characterization of the human AQP4 cDNAs and genomic DNA. Two cDNAs were isolated corresponding to the two initiating methionines (M1 in a 323-aa polypeptide and M23 in a 301-aa polypeptide) previously indentified in rat [Jung, J. S., Bhat, R. V., Preston, G. M., Guggino, W. B. & Agre, P. (1994) Proc. Natl. Acad. Sci. USA 91, 13052-13056]. Similar to other aquaporins, the AQP4 gene is composed of four exons encoding 127, 55, 27, and 92 amino acids separated by introns of 0.8, 0.3, and 5.2 kb. Unlike other aquaporins, an alternative coding initiation sequence (designated exon 0) was located 2.7 kb upstream of exon 1. When spliced together, M1 and the subsequent 10 amino acids are encoded by exon 0; the next 11 amino acids and M23 are encoded by exon 1. Transcription initiation sites have been mapped in the proximal promoters of exons 0 and 1. RNase protection revealed distinct transcripts corresponding to M1 and M23 mRNAs, and AQP4 immunoblots of cerebellum demonstrated reactive polypeptides of 31 and 34 kDa. Using a P1 and a λEMBL subclone, the chromosomal site of the human AQP4 gene was mapped to chromosome 18 at the junction of q11.2 and q12.1 by fluorescence in situ hybridization. These studies may now permit molecular characterization of AQP4 during human development and in clinical disorders.

        신경섬유종증(렉클링하우젠병)에 의한 급사 1예 보고

        Neurofibromatosis type1(von Recklinghausen's disease) is characterized by multiple skin tumors and abnormal cutaneous pigmentation(cafe au lait spots). Unlikely the solitary neurofibroma, those encountered in von Recklinghausen's disease may cause significant morbidity. The vasculopathy, although poorly recognized manifestation of von Recklinghausen's neurofibromatosisis a major case of fatal cases. One of its most dramatic presentation is the spontaneous hemothorax. We report a case of Von Recklinghausen's disease with multiple neurofibromatosis accompanying intrathoracic hemorrhage due to vascular rupture, which is the first legal autopsy case in Korea.

        Quantitative Determination of Cyclosporine in Human Whole Blood by Ultra-Performance Liquid Chromatography with Triple Quadrupole Tandem Mass Spectrometry

        Cyclosporine is an immunosuppressant drug used in organ transplants or for the treatment of autoimmune diseases. We developed and validated a simple, sensitive, and specific method using UPLC-MS/MS to determine cyclosporine levels in human whole blood. MS/MS detection was performed in the positive electrospray ionization mode with multiple reaction monitoring. Cyclosporine was extracted from whole-blood samples using ascomycin as an internal standard. The mass transitions m/z 1203.49 -> 1185.53 and m/z 814.71 -> 796.67 were used to assay the analyte and IS. This method was validated with respect to linearity, specificity, accuracy, precision, recovery, and stability. The method exhibited a linear response from 10 to 1000 ng mL(-1) with correlation coefficient values >0.99. The precision and the accuracy values were within 15%, except at the lower limit of quatification (LLOQ). Cyclosporine was stable in whole blood with no evidence of degradation. This method was successfully applied to a pharmacokinetic study of cyclosporine in healthy volunteers following oral administration.

        성상세포교종에서의 BUdR, AgNOR's, PCNA에 대한 비교연구

        In 14 cases of astocytoma, we performed a comparative study of BUdR, AgNOR's, and PCNA with histopathology. Fourteen astrocytomas were classified histopathologically into low grade, anaplastic, and G.M.(glioblastoma multiforme) groups. These comprised 3 G.M., 5 anaplastic astrocytomas, and 6 low grade astrocytomas. The mean BUdR-LI(Labeling Index) was statistically different(P<0.005) between G.M.(10.4), anaplastic astrocytoma(3.6), and low grade astrocytoma(1.3). On the basis of correlation between BUdR-LI and histopathology which was reported in 1989 at PMC, we examined the relationship between BUdR-LI and AgNOR's count, and between BUdR-LI and PCNA-LI, Both AgNOR's count and PCNA-LI did not correlate with BUdR-LI, and also not with histopathology. This study indicates that, at present, for prognosis and differentiation between malignant and benign astrocytoma, histopathlogy and BUdR-LI are important.

      • Various shaped-ZnO nanocrystals via low temperature synthetic methods: Surfactant and pH dependence

        ZnO nanocrystals, rod-, carnation-, and flower-like structures, have been synthesized in a high yield through low-temperature synthetic methods. Well-aligned ZnO nanorods having hexagonal wurtzite structure were grown on the ZnO thin films assembled by a spin-coating method. The morphologies of ZnO seed films are affected by pHs of sol-gel solutions, resulting smaller sizes and homogeneous roughness at higher pHs and higher number of spin-coating times. The carnation-like structures, average size of about 2-3 μm, were assembled by tens of uniform ZnO nanosheet petals of ~50 nm in thickness when a different volume ratio of the precursory solution was used. ZnO nanocrystals on the facets of the compact ZnO nanorods have grown to linear nanorods having an average diameter of ~500 nm and length of ~2 μm. Furthermore, a noticeable difference in the growth of ZnO nanocrystals in the presence of various surfactants, polyvinylpyrrolidone, polyvinylsulphonic acid, and polyethyleneimine, has been observed and discussed.

        We report synthesis and quantitative analysis of calcium oxalate. Synthesis of calcium oxalate by treatment of oxalic acid with Ca(OH)_(2) and oxalic acid with CaCl_(2) was executed. quantitative analysis of Ca for bean sprouts, spinach, coffee, and brown seaweed was analyzed by ICP(Inductively Coupled plasma). We experienced reaction and formation of crystal for oxalic acid and CaCl_(2) with NaCl, KCl, FeCl_(3) or AlCl_(3).

        Quorum sensing activity of the plant growth-promoting rhizobacterium Serratia glossinae GS2 isolated from the sesame (Sesamum indicum L.) rhizosphere

        <P>Plant growth-promoting rhizobacteria (PGPR) affect plant growth through various mechanisms, such as indole-3-acetic acid (IAA) production, 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, and biofilm formation. The aim of the study reported here was to isolate and characterize rhizobacteria that produce quorum-sensing signal molecules and other PGPR-related molecules. A biofilm-forming bacterium, GS2, was isolated from the rhizosphere of a sesame plant and subsequently found to produce two quorum-sensing signal molecules that were identified as N-hexanoyl-L-homoserine lactone (m/z 200) and N-octanoyl-L-homoserine lactone (m/z 228) by liquid chromatography-tandem mass spectrometry analysis. The strain was also found to produce IAA (17.2 mu g mL(-1)), gibberellins (113.7 mu g mL(-1)), and ACC deaminase (9.7 mu M alpha-ketobutyrate mg(-1) protein h(-1)). The strain was identified as Serratia glossinae based on a comparison of 16S rRNA gene sequences. Inoculation of the strain promoted growth of a gibberellin-deficient rice dwarf mutant (Waito-C). Different growth attributes, including shoot and root elongation, chlorophyll content, and plant weight could be attributed to the PGPR characteristics of strain GS2. These results suggest that S. glossinae strain GS2 can serve as a microbial agent that improves plant growth.</P>
