Probiotics have been suggested as a prophylactic measure in colon cancer. The aim of this study was to investigate the impact of Lactobacillus salivarius Ren (Ren) in modulating colonic microbiota structure and colon cancer incidence in a rat model after injection with 1,2-dimethyl hydrazine (DMH). The results indicated that oral administration of Ren could effectively suppress DMH-induced colonic carcinogenesis. A significant decrease in cancer incidence (87.5% to 25%) was detected in rats fed with a dose of 5 × 1010 CFU/kg bodyweight per day. Using denaturing gradient gel electrophoresis and Real-time PCR combined with multivariate statistical methods, we demonstrated that injection with DMH significantly altered the rat gut microbiota, while Ren counteracted these DMH-induced adverse effects and promoted reversion of the gut microbiota close to the healthy state. Tvalue biplots followed by band sequencing identified 21 bacterial strains as critical variables affected by DMH and Ren. Injection of DMH significantly increased the amount of Ruminococcus species (sp.) and Clostridiales bacteria, as well as decreasing the Prevotella sp. Administration of Ren reduced the amount of Ruminococcus sp., Clostridiales bacteria, and Bacteroides dorei, and increased the amount of Prevotella. Real-time PCR results were consistent with the results derived by t-value biplots. These findings suggested that Ren is a potential agent for colon cancer prevention. In conclusion, the results in the present study suggest a potential therapeutic approach based on the modulation of intestinal microflora by probiotics may be beneficial in the prevention of colorectal carcinogenesis.

AlN ¯lms and nc-AlN/a-SiNx nanocomposite ¯lms are deposited on (111) silicon and glass sub-strates by using medium-frequency reactive magnetron sputtering under N2/Ar atmosphere, and then the ¯lms are analyzed in detail by using transmission electron microscopy (TEM) and X-ray diraction (XRD). Hardness of the nc-AlN/a-SiNx ¯lms is measured. The microstructures of AlN ¯lms under dierent nitrogen concentrations are compared. The XRD patterns indicate that the ¯lm shows (002) orientation, strongly independent of N2 partial °ow rate. The TEM images show that the uniform AlN crystal, the clear polycrystalline diraction circles and the dierence of the grain size under dierent conditions are signi¯cant. Columnar grains of AlN ¯lm on (111) silicon substrate under 70 % N2 partial °ow rate are observed. The results indicate that the preferred orientation of AlN ¯lm is aected by N2 partial °ow rate. The hardness of nc-AlN/a-SiNx ¯lms increases with increasing a-SiNx content and the hardness decreases when a-SiNx is excessive. The maximal hardness of nc-AlN/a-SiNx nanocomposite ¯lms can reach 30 GPa by adjusting a-SiNx content. The transmission of the nanocomposite ¯lms is over 85 %. The results suggest that it has potential application for optical protective coatings.

Objective: To identify potential risk factors of unsatisfactory screw position during robot-assisted pedicle screw fixation. Methods: A retrospective analysis of robot-assisted pedicle screw fixation performed in Beijing Jishuitan Hospital from March 2018 to March 2019 was conducted. Research data was collected from the medical record and imaging systems. Univariate tests were performed on the potential risk factors (patient’s characteristics and surgical factors) of unsatisfactory screw position during robot-assisted pedicle screw fixation. For statistically significant variables in univariate tests, a logistic regression test was used to identify independent risk factors for unsatisfactory screw position. Results: A total of 780 pedicle screws placed in 163 robot-assisted surgeries were analyzed. The rate of perfect screw positions was 93.08%, and the unsatisfactory rate was 6.92%. In patients with severe obesity (body mass index≥30 kg/m2) (odds ratio [OR], 2.459; 95% confidence interval [CI], 1.199–5.044; p=0.014), osteoporosis (T≤-2.5) (OR, 1.857; 95% CI, 1.046–3.295; p=0.034), and the segments 3 levels away from the tracker (OR, 2.216; 95% CI, 1.119–4.387; p=0.022), robot-assisted pedicle screw placement has a higher risk of screw malposition. Conclusion: During robot-assisted pedicle screw placement for patients with severe obesity, osteoporosis, and segments 3 levels away from the tracker, vigilance should be maintained during surgery to avoid postoperative complications due to unsatisfactory screw position.

Background/Aims: The recognition of a correlation between patatin-like phospholipase domain containing-protein 3 (PNPLA3) rs738409 (C>G) and the severity of liver steatosis or fibrosis in chronic hepatitis C (CHC) has not reached a consensus. This meta-analysis sought to investigate with accuracy the association between the PNPLA3 rs738409 (C>G) polymorphism and liver steatosis and advanced fibrosis in CHC patients. Methods: We performed a comprehensive literature search from the PubMed, Embase, Web of Science, and Google Scholar databases up to December 31, 2014. Crude odds ratios (ORs) with 95% confidence intervals (CIs) were calculated. Statistical analyses were performed using Stata 12.0 software. Results: The meta-analysis revealed the severity of liver fibrosis was significantly higher in CHC patients with PNPLA3 rs738409 GG in Caucasians (versus CC+CG: OR, 2.29; 95% CI, 1.57 to 3.35; p<0.05) but not Asian populations. In Caucasians, liver steatosis was also more severe in CHC patients with rs738409 GG (versus CC+CG; OR, 4.33; 95% CI, 2.59 to 7.22; p<0.05). The sensitivity analysis indicated the results of this meta-analysis were stable and no publication bias was detected. Conclusions: PNPLA3 rs738409 (C>G) was associated with the risk of both advanced liver fibrosis and steatosis in patients with CHC, especially among Caucasian populations. (Gut Liver 2016;10:456-463)

This paper deals with free vibration analysis of non-uniform column resting on elastic foundations and subjected to follower force at its free end. The internal pores and graphene platelets (GPLs) are distributed in the matrix according to different patterns. The model is proposed with material parameters varying in the thickness of column to achieve graded distributions in both porosity and nanofillers. The elastic modulus of the nanocomposite is obtained by using Halpin-Tsai micromechanics model. The differential quadrature method as an efficient and accurate numerical approach is used to discretize the governing equations and to implement the boundary conditions. It is observed that the maximum vibration frequency obtained in the case of symmetric porosity and GPL distribution, while the minimum vibration frequency is obtained using uniform porosity distribution. Results show that for better understanding of mechanical behavior of nanocomposite column, it is crucial to consider porosities inside the material structure.

To understand better the mechanisms that regulate the water stress response in wheat, we conducted a comparative analysis of transcript profiles in roots from two wheat genotypes -- drought-tolerant ‘Luohan No. 2’ (LH)and drought-susceptible ‘Chinese Spring’ (CS). In LH roots,3831 transcripts displayed changes in expression of at least two-fold over the well-watered control when drought treatment was applied. Of these, 1593 were induced while 2238 were repressed. Relatively fewer transcripts were drought-responsive in CS; i.e., 1404 transcripts were induced and 1493 were repressed. In common between LH and CS,569 transcripts were induced and 424 transcripts were repressed. In all, 689 transcripts (757 probe sets) identified from LH and 537 transcripts (575 probe sets) from CS were annotated and classified into 10 functional categories. Among those annotated transcripts from LH and CS that had fold-change ratios of at least 4, 92 induced transcripts were common to both, while 23 transcripts were specifically induced in LH. Gene ontology analysis of these induced genes showed highly significant enrichment for multiple terms related to abiotic stimuli, organic acid biosynthesis, and lipid metabolism. This suggests that these gene groups play important roles during the stress response in LH and CS, and might also be responsible for differences in drought tolerance between those genotypes.

In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition. [BMB reports 2010; 43(3): 193-198]
