      • Designation of fingerprint glycopeptides for targeted glycoproteomic analysis of serum haptoglobin : insights into gastric cancer biomarker discovery

        Gastric cancer (GC) is one of the leading causes of cancer-related death worldwide, largely because of difficulties in early diagnosis. In spite of accumulating evidence linking aberrant glycosylation and GC, site-specific localization of the glycosylation for improving clinical specificity and sensitivity is still in analytical challenge. Here, we constructed an analytical platform with targeted glycoproteomic approach for GC biomarker discovery. Unlike conventional glycomic approach characterized by untargeted MS profiling of released glycan, developed platform can be characterized by three key features: target protein-specific, glycosylation site-specific, and structure-specific platform with one-shot non-specific enzyme digestion. Serum haptoglobin (Hp) was enriched by immunoaffinity purification and subjected to pronase to generate site-specific glycopeptides. Glycopeptides were identified and quantified by nano LC/MS and -LC/MS/MS, resulting in full characterization of glycan macro- and micro-heterogeneity. A total of 96 glycopeptides, each corresponding to a unique glycan/glycosite pairing, were tracked across all cancer and control samples. Significant alterations in site-specific glycosylation have been observed in Hp in GC patients.

        기업휴게시설 명상공간의 디자인 특성 연구

        (연구배경 및 목적) 산업사회에서의 휴식은 근무 시간과의 명확한 구분을 통해 만들어지는 개념이었고 공간의 구분 역시 이에 적합한 형태였다면, 현대 기업 환경에서의 휴식은 보다 복합적이고 자유로운 형태로 나타난다. 또한 업무 중 가지게 되는 휴식의 형태 역시 단순한 육체적 재충전을 벗어나 정신적인 측면, 즉 스트레스 등에 대한 대응을 고려하는 형태로 전환되고 있다. 그리고 이러한 정신적 대응의 구체적 형태의 하나로서 명상이라는 행위를 기업 휴게시설 측면에서 주목할 수 있다. 이러한 관점에서 본 연구는 업무 환경 변화에 따라 변화하는 휴게공간의 형태에 주목하고, 기업의 휴게시설에서 새롭게 요구되어지고 있는 명상공간의 디자인 개념과 특성을 고찰하는데 목적이 있다. (연구방법) 첫째, 다양한 선행 연구에 대한 조사를 통해 기업 환경의 시대적 변화과정을 파악하고, 이를 바탕으로 기업의 휴게시설이 가지는 중요한 특징들을 고찰한다. 둘째, 현대 기업의 휴게공간에서 등장하고 있는 명상공간의 개념을 명확히 하고, 이러한 새로운 현상의 시대적 흐름을 파악하고 명상공간의 개념과 특징을 이해한다. 셋째, 기업 휴게공간의 명상공간에 대한 주요 사례들을 수집하여 명상공간에서 나타나는 디자인 특성을 분석하여 주요한 경향을 파악한다. 그리고 이를 통해 기업 휴게시설의 명상공간의 디자인이 가지는 개념과 의미를 제시하고자 한다. (결과) 주요기업 10곳의 평균값을 분석한 결과, 공간에서 주된 요소만 구성되어 운영이 용이한 점으로 명료성이 가장 높았으며, 불필요한 기능과 불편한 요소가 배제된 배치계획으로 접근성과 편의성이 비교적 높았다. 하지만 공간이 서로 다른 요구를 충족시키지 못하고 공간이 제공하는 활동과 기회들이 다양하지 못해 다양성 요소 점수가 가장 낮았으며, 명상공간에 대해 이질적인 거부감은 없으나 시설환경에서 친근감이 느껴지기 어려웠다. 또한 유연한 환경으로 자유로운 커뮤니케이션과 상호작용을 통한 공감대 형성이 어렵고, 유지관리의 측면, 다양한 요소의 통일감 부족으로 편안하지 않다는 점에서 유대감과 효율성이 낮게 나타났다. (결론) 새롭게 요구되는 명상공간에 대한 적절한 계획이 현재의 업무환경변화에 따른 휴게시설의 디자인 방향이며, 디자인 측면에서 명상공간이 가져할 조건은 효율성, 접근성, 명료성, 편의성, 다양성 측면의 물리적인 환경요소와 편안함, 균형감, 친근감, 쾌적함, 유대감의 심리적 환경요소를 반영해야 한다. 또한 지속적인 프로그램 개발 및 공간의 선택가능성과 시설환경의 색채, 공간배치, 위생적인 환경조성 및 유지관리로 지속가능한 공간이 되도록 계획해야 한다. 본 연구를 통해 많은 기업의 근무자들이 업무와 대인관계로 인한 스트레스에서 벗어나 심적, 신체적으로 편안하고 행복감을 가져 일의 능률을 향상시키고, 이것이 기업의 생산성도 높여 그것이 다시 더 높은 수준의 복지향상이라는 결과를 가져오기를 기대해본다. (Background and Purpose) Recently, the concept of rest has been clarified and the division of spaces has traditionally been distinct. In the modern working environment, the concept of rest is more complex and focused on stress responses. Meditation, a form of self-awareness of one`s own thoughts and movements, has become popular within corporate facilities and areas for meditation can be seen in corporate lounges or break rooms. This study focuses on the form of resting spaces as they change the work environment, and aims to examine the design concept and characteristics of the typical meditation space. (Method) First, this study investigates changes in design of corporate environments through a literature review before examining common features used in lounge facilities. Second, this study hopes to clarify the concept of a meditation space, determine typical characteristics of these spaces, and understand trends within this relatively new phenomenon. Third, this study collects specific examples of meditation spaces included in corporate lounges and other rest areas, analyzing the design characteristics used in these spaces. Finally, the author discusses findings and presents the importance of both concept and meaning in the designing of meditation spaces for corporate break rooms. (Result) Results of an internal and external analysis of ten different corporate meditation spaces shows that clarity is rated highest, due to ease of operation. Accessibility and convenience are also high, thanks to a successful layout that excluded unnecessary functions and inconveniences. However, diversity was the lowest rated, since the space was appropriate for a limited number of activities and opportunities, and thus failed to meet the needs of multiple groups. The meditation spaces studied also lacked any sense of heterogeneity, making it difficult to feel familiarity in these facility environments. Finally, fellowship and efficiency were low because it is often difficult to form a consensus through free communication and interaction, causing discomfort with the lack of unity. (Conclusions) Design choices made have the potential to change the work environment if meditation spaces are thoughtfully planned. Characteristics of meditation spaces should reflect its physical elements (efficiency, accessibility, clarity, convenience, diversity), psychological considerations (comfort, balance, affection, agreement, fellowship), and environmental factors. These spaces should also be sustainable and considerate of various types of users. This study suggests that providing meditation spaces in corporate lounges decreases employee reports of stress, both in regards to work stressors and interpersonal relationships, and improves work efficiency by providing an employee area that encourages mental and physically comfort. Further, increased efficiency is often linked to an increase in productivity, which can have positive effects on the local economy and lead to improvements in the welfare of neighboring communities.

      • 청년 혜택 알리미 어플리케이션 설계와 구현

        청년들을 위해 국가에서 시행하는 다양한 혜택들이 많다. 자신에게 해당하는 혜택임에도 인지도가 낮아 신청하지 않는 청년들이 많다. 이에 본 논문에서는 청년들에게 정보를 제공하고 신청까지의 과정이 쉽고 빠르게 이루어지는 편리한 어플리케이션을 설계하였다. 본 논문의 어플리케이션 시스템과 UI는 시장조사와 설문조사, 벤치마킹 등을 통하여 설계하였다. 본 논문의 어플리케이션은 많은 청년들에게 도움이 될 것이라 기대한다. There are many various benefits implemented by country for young people. However, there are also many young people who do not apply due to low awareness, even though it is a benefit that corresponds to them. Therefore, in this paper, an application was designed. It is convenient to provide information to young people and to make applications easily and quickly. We designed the application system and UI through market research, surveys, and benchmarking. The application of this paper is expected to help many young people.

      • Developmental profile of human frontal cortex using glycomic approach

        Glycan plays numerous roles in the nervous system during development, regeneration and synaptic plasticity, forming of a complex meshwork at the cell surface. We newly established an analytical platform for exploring two molecules- glycan attached to proteins and glycan attached to lipids- from human brain using mass spectrometry. 69 brain samples were obtained from the University of Cambridge. Glycan extraction method from previous study, and new ganglioside extraction method were paired with MS. Briefly, each brain tissues were grinded by sonication. Then for glycans, the membrane was pelleted by ultracentrifugation. N-glycan release and associated processing steps were performed by previously optimized procedures. For sphingolipids, homogenized brain is mixed with chloroform/methanol/water and centrifuged. Total lipids extracts were partitioned again with chloroform/methanol to collect gangliosides. After further enrichment by SPE, N-glycans and gangliosides were profiled by positive and negative ion mode nano-LC/MS respectively. Stucture information was obtained by nano-LC/MS/MS. Brain samples were divided into the 7 groups: neonates, infants, toddlers, school-ages, teenagers, young adults. We successfully profiled brain glycome and sphingolipids, identifying approximately 100 N-glycan compositions and 120 ganglioside compounds with several unique “brain type” characters. This study promise insights into involvement of glycans to massive flow of information that underpins brain, with providing meaningful understanding to neuropathology related with aging.

        제왕절개술을 위한 Atracurium 의 점적주입속도와 근이완에서의 자연회복

        Background: Atracurium, a nondepolarizing muscle relaxant, is eliminated by Hofmann elimination and not affected by pseudocholinesterase(pChe). Many reports show fall in pChe activity during pregnancy and our measurement failed to prove it. However, the authors found previously the infusion rate of mivacurium, which is metabolized by pChe, to achieve surgical relaxation is decreased. So, we compared the difference in the infusion rate of atracurium between full-term pregnant and nonpregnant women. Methods: Muscle relaxation of full-term pregnant women(C group, n=10) and nonpregnant women (Non-C group, n=10) was maintained by continuous infusion of atracurium to keep 1st response of TOF at 5 1%. After discontinuance of infusion, recovery index was measured without reversals using the accelerograph. Results: There was no significant difference in infusion rate of atracurium to maintain 1st twitch response of TOF at 5 1% and recovery index between two groups. There was little correlation between the total infusion time and infusion rate, or between the total infusion time and recovery index. Conclusion: The atracurium infusion rate in parturients to maintain muscle relaxation was not different from that in non-parturients. For Cesarean section, the usual infusion rate of atracurium is recommended to achieve the adequate surgical relaxation. (Korean J Anesthesiol 1997; 32: 561∼566)

        MS-Based Technologies for the Study of Site-Specific Glycosylation

        Glycosylation, which is one of the most common post-translation modification (PTMs) of proteins, plays a variety of crucial roles in many cellular events and biotherapeutics. Recent advances have led to the development of various analytical methods employing a mass spectrometry for glycomic and glycoproteomic study. However, site-specific glycosylation analysis is still a relatively new area with high potential for technologies and method development. This review will cover current MS-based workflows and technologies for site-specific mapping of glycosylation ranging from glycopeptide preparation to MS analysis. Bioinformatic tools for comprehensive analysis of glycoprotein with high-throughput manner will be also included.

        Effects of carbon and nitrogen additions on microstructure and mechanical properties of Ti-48.5Al-1.5Mo (at.%) alloys were investigated. The alloys added with 0.3%, 1.0% and 2.0%C or N were prepared by vacuum arc melting, and heat-treated at 1390℃ for lamellar microstructure and then aged at 900℃. Tensile tests were performed in air with the strain rate of 2×10-4S-1 at room temperature and 800℃. It was found that C (or N) addition decreased the grain size of lamellar structure. Especially, 1.0%C(or N) and 2.0%C(or N) addition refined the grain size to 1/10 of that of the mother alloy. Only fine Ti3AlN precipitates were formed at the lamellar boundaries for the alloy containing 0.3% N, but Ti2AlN precipitates of a few micrometers in size were also formed in the matrix in addition to Ti3AlN precipitates for the alloys with 1.0 and 2.0% N. Tensile elongation at room temperature decreased markedly for the alloys with 0.3%C or N but recovered for the alloys with 1.0% and 2.0% addition compared to the value of the mother alloy. Yield strength at room temperature for the alloys containing 1.0% and 2.0% C or N increased up to two times of the value of the mother alloy, and the high strength was maintained up to 800℃. The aged alloys showed much higher tensile elongation at room temperature than the alloys only heat-treated to obtain the lamellar microstructure.
