      • 두경부 종양세포주에 대한 Mithramycin A의 세포자멸사 유도성 및Cisplatin 항암작용 증가효과

        Background and Objectives : Cisplatin (CP) is a major chemotherapeutic agent for head and neck cancer. However, cancer cells can get drug resistance through recovering the DNA damage by CP. Mithramycin A (MA) can inhibit gene expression of multidrug resistance gene 1, c-myc or h-ras. The goal of this study is to evaluate the therapeutic effect of MA alone and combination of MA and CP on head and neck cancer cell lines. Materials and Methods : Two head and neck cancer cell lines (AMC-HN4, AMC-HN7) were treated with MA. The cellular proliferation, apoptosis and protein expression were measured with XTT assay, FACS analysis, and western blotting, respectively after MA treatment in cell lines. In order to know the combination effect of MA with CP, the cancer cell lines were treated with MA, CP, and MA/CP combination, and cellular proliferation, apoptosis and expression of apoptosis-associated proteins were measured. Results : MA inhibited the proliferation of head and neck cancer cells and induce dapoptosis. Increase of caspase activation, PLC-?1 fragmentation and percent apoptosis by MA treatment was blocked by z-VAD-fms, but not blocked by N-Acetyl Cystein. Combination of MA and CP significantly inhibited cancer cell growth and increased apoptosis compared to MA and CP alone, and it also suppressed the expression of anti-apoptotic proteins such as XIAP and Mcl-1. Conclusions : The present study suggests that MA alone andMA/CP combination chemotherapy could be a potential regiment for the control of the head and neck cancer.

        초임계 이산화탄소를 이용하는 친환경 공정에 의한 다공성 고분자의 제조

        An experimental study on the preparation of monolithic porous polymers by environmentally friend process in supercritical carbon dioxide has been carried out. Polymerization mixture composed of a cross-linking monomer, initiator and functional co-polymer was charged in the reactor with sapphire window. After the system was purged with a flow of CO₂ for 15 min, the reactor was pressurized with liquid CO₂ up to 100 bars. The reactor was isolated from and placed back to the system via quick connector for shaking until the mixture had become fully homogeneous. The reactor was then heated and pressurized to the required reaction conditions and left overnight. After cooling and CO₂ evacuation, the polymer was removed from the reactor as dry, white, continuous monoliths. The effect of experimental conditions on the physical properties of porous polymer was systematically examined, and it was found that monomer content had a major effect on the physical properties of the polymers.

      • 급성 담낭염에 대한 내과적 치료의 역할

        In 58 patients who were admitted to department of internal medicine, ChungNam national university hospital from January 1989 to March, 1993. We evaluated for medical treatments and the following results were obtained. 1) The ratio of male and female was 1 : 1 and mean age of the patients was 57.4 years. 2) At arrival on hospital, the most common clinical symptom was pain in right upper quadrant and epigastrium(93.1%), the remainders were fever and chill(79.3%), nausea(51.7%) and vomiting(36.2 %) in order of frequency. 3) The most common physical findings was tenderness on the right upper quadrant and epaigastrium(98%), the remainders were jaundice(50%) and palpable gall bladder(13.8%) in order of freguency. 4) In laboratory examination, elevated levels of ESR(82.8%), leukocytosis(67.2%), elevated levels of AST(62.0%), ALT(50.0%), serum total bilirubin(51.7%) and alkaline phosphatase (43.1%) were noted. 5) The mean period of NPO was 3.9 days and the mean hospital period was 13.5 days. 6) The rates of combined complications in patients with acute cholecystitis were 17.2%; these are gall bladder empyema(5.2%), hydrops(5.2%), perforation(3.4%), sepsis(3.4%) and intestinal obstruction (1.7%). 7) In 96.6 percent of 58 cases, the symptoms of acute cholecystitis were improved by conservative management. 8) The mean follow-up period was 25.5 months and the recurrence rate was 10% during this period.

      • 넙치, Paralichthys olivaceus 치어의 생존, 성장 및 산소소비에 미치는 수은의 만성적 독성

        산업활동의 증가로 인해 해양에 유입되는 수은 (Hg)이 저서성 연안정착 어류인 넙치, Paralichthys olivaceus 치어의 생존, 성장, 사료효율 및 산소소비에 미치는 영향을 조사하였다. 6주 동안 0~0.13mg/L의 아치사 농도에서 넙치를 사육하며 Hg 독성에 대한 영향을 조사하였다. Hg 농도와 노출기간이 증가할수록 생존육은 감소했으며 6주 후, 0.05mg/L 이상의 농도 구에서 생존율이 급격히 감소했다. 성장률과 사료효율 또한, 각각 0.028mg/L (P<0.01)와 0.05mg/L (P<0.01) 이상의 농도 구에서 현저하게 감소하였다. 산소소비율은 0.05mg/L와 0.13mg/L의 농도구에서 각각 대조 구에 비해 25(P<0.01)와 32% (P<0.01) 감소해, 유의적인 차이를 나타내었다. 이상의 결과로부터 Hg 독성은 넙치의 성장, 사료효율 및 산소소비와 같은 생리적 기능을 저해해 생존율을 감소시키는 것으로 나타났다. Effect of mercury (Hg) toxicity on survival, growth, feed efficiency and oxygen consumption were examined in the juvenile olive flounder. Paralichthys olivaceus. Fishes were exposed to sublethal concentrations of Hg ranging from 0 to 0.13mg/L for 6 weeks. Hg reduced survival rate in a concentration and exposure period-dependent way and suddenly reduction occurred at Hg concentrations greater than 0.05mg/L after 6 weeks. Growth rate and feed efficiency also significantly decreased at greater than 0.028 and 0.05 mg/L respectively. Oxygen consumption rate was significantly decreased to 25 and 32% than that of the control at the Hg concentration of 0.05 and 0.13 mg/L respectively. These results suggest that Hg toxicity inhibit physiological function including growth, feed efficiency and oxygen consumption in the juvenile olive flounder, resulting in survival failure at high concentration.

        한국판 아동용 예일-브라운 강박 척도:신뢰도와 타당도 연구

        목적 : 본 연구는 소아, 청소년의 강박 증상을 평가하기 위한 도구인 아동용 예일-브라운 강박척도(Childen's Yale-Brown Obsessive Compulsive Scale : Y-BOCS)의 한국판을 제작하여 그 신뢰도와 타당도를 검증하고자 시행되었다. 방법 : 강박장애로 진단된 42명의 아동과 청소년들을 대상으로 CY-BOCS를 시행하였다. 평가자간 신뢰도를 측정하기 위하여 20명의 환자를 2명의 평가자가 동시에 면담하였다. 수렴타당도와 변별타당도의 검증을 위해 총괄적인 임상 인상척도-강박장애(Clinical Global Impression-Obsessive Com-pulsive Disorder : CGI-OCD). 아동용 Leyton 강박 척도(Lryton Obsessional Inventory-Child Version : LOI-CV), 소아우울척도(Child Depression Inventory : CDI), 소아 상태-특성 불안척도(State-Trait Anxiety Inventory for Children : STAI-C)를 시행한다. 결과 : Cronbach's α값으로 평가한 내적 일치도는 0.86으로 높게 나타났다. CY-BOCS 전체 점수 강박사고, 강박행동 소척도 점수에 대한 군내 내적일치도는 각각 0.94. 0.94. 0.84로 우수한 평가자간 일치도를 보였다. CY-BOCS 전체 점수와 CGI-OCD 점수간의 상관관계는 매우 높게 나타났고(r-0.88), LOI-CY 총점과도 통계적으로 유의한 상관관계(r=0.51)가 관찰되었다. CY-BOCS 전체 점수와 STA-I-C의 상태불안 점수간에는 상관관계가 나타나지 않았고(r=0.25), 특성불안 점수(r=0.43) 및 CDI 점수(r=0.49)와는 유의한 상관관계를 나타냈다. 결론 : 본 연구결과, 한국판 CY-BOCS의 높은 타당도 및 신뢰도가 입증되었으며, 향후 강박 증상의 객관적인 평가 및 정량화를 위해 유용하게 사용될 수 있을 것이다. Objective : This study was carried out to evaluate the reliability and validity of the Korean form of the Children's Yale-Brown Obsessive Compulsive Scale(CY-BOCS). Method : Forty-two children with obsessive-compulsive disorder(OCD)(31 boys and 11 girls, aged 7 to 18 years) were assessed with CY-BOCS. Interrater agreement was assessed by two raters in subsample(n=20). discriminant and convergent validity were assessed by comparing CY-BOCS scores to Clinical Global Impression-OCD(CGI-OCD), Leyton Obsessional Inven-tory-Child Version(LOI-CV), State-Trait Anxiety Inventory for Children(STAI-C), and Child Depression Inventory( CDI ). Results : Internal consistency was high, measuring 0.86 for the 10 items and total score. The intraclass correlations for the CY-BOCS total, obsession, and compulsion scores were 0.94, 0.94, 0.84, suggesting excellent interrater agreement for subscale and total scores. The CY-BOCS total score showed highest correlation with CGI-OCD(r=0.88), and significant correlation with LOI-CV(r=0.51), Trait Anxiety score(r=0.43), and CDI scores(r=0.49), but it was not correlated with State-Anxiety score(r=0.25). Conclusion : The results of this study indicate that the Korean form of CY-BOCS is a reliable and valid scale for rating obsessive-compulsive symptom severity.         한국어판 예일 틱 증상 평가척도 : 신뢰도 및 타당도 연구

        목 적 : 틱 증상의 심한 정도를 평가하기 위한 도구인 예일 틱 증상 평가척도(Yale Global Tic Severity Scale)를 한국판 가정 평가용 설문지 및 임상 평가용 척도로 개발하기 위하여 본 연구를 시행하였다. 방 법 : 예일 틱 증상 평가척도는 뚜렛 장애 및 기타 틱 장애에서 나타나는 운동틱과 음성틱 각각의 증상의 수, 빈도, 심한 정도, 복합성, 방해 정도 및 장해도를 포괄적으로 평가하도록 고안되어져 있다. 틱 증상을 주소로 정신과외래를 방문한 만 4.3세에서 19세까지의 100명의 환아 및 부모들을 대상으로 한국어로 번역한 예일 틱 증상 평가척도의 가정평가용 설문지를 평가전에 완료하도록 한 후, 면담을 통한 임상평가용 척도를 실시하였으며 수렴타당도 및 변별타당도의 검증을 위해 총괄적인 임상 인상척도-뚜렛 증후군, 강박장애, 주의력결립/과잉운동장애(Clinical Global Impression-TS, OCD, ADHD)를 시행하였다. 결 과 : 자료분석결과 내적일치도, 수렴타당도, 변별타당도 및 검사자간 신뢰도 모두 매우 높게 나타났으며 요인분석상 전 항목척도와 장해도는 운동틱과 음성틱에 해당하는 2개의 요인으로 묶여졌다. 결 론 : 본 연구결과 한국판 예일 틱 증상 평가척도의 높은 타당도 및 신뢰도가 입증되었으며 이는 향후 틱 증상의 객관적인 평가 및 정량화를 위해 유용하게 사용되어질 수 있을 것이다. Objectives : This study was carried out to develop the Korean form of Yale Global Tic Severity Scale(YGTSS)-family & clinical rating version. The severity of motor and phonic tics was rated according to five separate dimensions : number, frequency, intensity, complexity, and interference. Methods : The Korean form of YGTSS was applied to 100 children who visited psychiatric outpatient clinic with chief complaints of tic symptom. Together with YGTSS, Clinical Global Impression for Tourette's syndrome(CGI-TS), Obsessive-Compulsive disorder(CGI-OCD), Attention-Deficit/Hyperactivity Disorder(CGI-ADHD) were administered to all subjects for examining convergent and discriminant validities. Results : We could confirm high internal consistency, convergent and discriminant validities and interrater reliability of YGTSS by analysing data from 100 children with tic disorder. In factor analysis, items were clustered to 2 factors which were identical to motor and phonic tic subscales. Conclusion : The results of this study indicate the Korean form of YGSS is a reliable and valid rating scale for rating tic symptom severity. It can be used to evaluate tic symptom objectively and to quantify the tic severity in the studies for tic disorder.

        청각장애학교에서의 진로교육 현황 및 개선방안

        The purpose of this study is to examine and analysis the realities of career education and to propose the its improvements in school for the deaf. We gathered opinions of 135 teachers in school for the deaf from questionnaire.The results are shown as following. First, Teachers have a correct understanding about the concept, the instruction steps, and instruction contents of career education. Second,Teachers know the necessity and importance of career education in classroom,but they didn't give the career guidance and information in classroom. Third, Teachers didn't give the career guidance and information in extra activity and counseling. Fourth,The materials and programs are not enough to provide the career education. Finally, We suggest improvements of career education based on the realities and teachers' recommendation 본 연구는 청각장애학교의 진로교육 현황을 분석하기 위해 전국에 소재하고 있는 11개 청각장애학교에 근무하고 있는 교사 135명을 대상으로 진로교육에 대한 인식과 진로교육 현황을 교과지도, 특별활동 및 진로행사, 상담활동, 진로정보자료 및 프로그램의 활용으로 나누어 조사하였다. 조사결과, 진로교육은 초등부부터 시작되어야 하며 그 필요성도 느끼고 있었을 뿐 아니라 진로교육의 의미나 학교급별 진로교육의 단계와 그에 적절한 교육 내용에 대해서도 바르게 인식하고 있었다. 그러나 교과활동을 통한 진로교육의 제공, 특별활동과 상담활동을 통한 진로교육 현황은 필요성 인식에 미치지 못하고 있는 실정이다. 이러한 현황과 교사들이 제안하는 개선방안을 바탕으로 진로교육 개선방안을 제시하였다.

      • 임의 형상의 개수로에서의 복잡한 흐름에 대한 고정도 수치해석

        본 연구의 목적은 일반곡선좌표계에서의 St. Venant 방정식을 MacCormack 차분법으로 처리한 2차원 부정류 모델을 개발함과 동시에 수리실험결과를 통해 흐름의 메카니즘을 파악하고 개발한 모델의 적합성 및 적용성을 검증한다. 본 수치모델이 상·살가 혼재하는 2차원 흐름을 잘 재현하고 있다고 생각되므로 본 수치모델은 낙차공동의 설치에 의한 급변 흐름의 유동을 표현하는데 유용할 것으로 생각된다. The purpose of this is to develop a numerical model of two-dimensional unsteady flow, which is composed of St. Venant equation on the general curve-linear coordinates by MacComack's scheme. First, this study comprehends the flow mechanism through the results of hydraulic experiments. Second, it verifies and feasibility of the present numerical model. The numerical model developed by this study regenerated well a coexisting sub-and super-critical two dimensional flow. Therefore, this model will be useful to express suddenly changed flow by hydraulic structures.

      • Pb의 노출에 따른 넙치, Paralichthys olivaceus, 치어의 생존, 성장 및 산소소비율의 변화

        Experiments were carried out to investigate the effects of lead exposure on survival, growth and oxygen consumption of juvenile oliver flounder, Paralichthys olivaceus. Survival rate of the oliver flounder was significantly affected above 150㎍/L. Growth of the olive flounder exposed to ≥ 150 ㎍/L of lead concentration was significantly lower than that of fish reared in normal condition. Metabolic rate of fish exposed to lead ≥ 80 ㎍/L concentration was significantly reduced with increasing lead concentration than that in normal condition. This study revealed that high lead concentration (≥ 80 ㎍/L) reduced growth and metabolic rates of juvenile olive flounder suggesting potential influence of lead on the natural mortality of olive flounder in the coastal areas.

        치주염의 원인균 및 정상상주균간의 길항기전 규명 및 치주치료에의 응용

        It has been reported that microbial distribution and colonization in oral cavity have a relation to antagonism, synergism or symbiosis among microorganisms and bacterial antagonism have a relation to bacteriocin, hydrogen peroxide and bacterial metabolic end-products. The purpose of this study is to investigate on the in vivo and in vitro ecological interrelation and to detect the growth inhibition subatance and to clarify the antagonistic mechanism of the periodontopathic bacteria. In vivo ecological interrelation was studied by bacterial isolation & identification and in vitro ecological was studied by stab culture method. Bacteriocin-like substance was examined, purified, electrophoresed, and stained with silver stain. After the producer microorganism was cultured in various conditions, the pH & bacterial end-products of it was examined and the susceptibility of indicator bacteria by pH was measured. The presence of plasmid DNA was investigated by hot-boiling method. In in vivo ecologic interrelationship, Actinobacillus actinomycetemcomitans(Aa) was not isolated, black-pig-mented Bacteroides was less in the presence of Actinomyces species than in the absence of it in refractory periodontitis. In in vitro ecologic interlationship, S. mutans showed strong growth inhibitory activity against Porphyromonas gingivalis and Aa strains and more strong growth inhibitory activity against Aa Korean isolates than S. sanguis. After bacteriocin-like substance was examined, purified, electrophoresed, and stained with silver stain, a band was appeared whose molecular weight was 36 kd, pH and the production rate of lactic acid was varied according to the presence and absence of oxygen and glucose. These results suggested that there exists in vivo and in vitro growth inhibition among oral microorganisms and growth inhibition was affected by bacteriocin-like substance, pH and bacterial end-products of producer strains and sensitivity of indicator strains. Animal study is needed by using strains of growth inhibition and growth inhibitory substance.
