        Canonical Form and Mathematical Interpretation of Electrolyte Solution Systems

        We describe a canonical form of electrolyte solution systems for the mathematical interpretation of solid-liquid equilibrium. The canonical form is obtained from the analysis of the algebraic structure of electrolyte solutionsystems and the Karush-Kuhn-Tucker (KKT) conditions for the minimization of the total Gibs free energy. As a result,the mathematical role of solid species in the solid-liquid equilibrium problem is explained as a Lagrange multiplierof a sort of the linearly constrained optimization problem. This finding wil add to the development of an efficientnumerical algorithm for the simulation of electrolyte solution systems.

        Enrichment and verification of differentially expressed miRNAs in bursa of Fabricius in two breeds of duck

        Objective: The bursa of Fabricius (BF) is a central humoral immune organ belonging specifically to avians. Recent studies had suggested that miRNAs were active regulators involved in the immune processes. This study was to investigate the possible differences of the BF at miRNA level between two genetically disparate duck breeds. Methods: Using Illumina next-generation sequencing, the miRNAs libraries of ducks were established. Results: The results showed that there were 66 differentially expressed miRNAs and 28 novel miRNAs in bursa. A set of abundant miRNAs (i.e., let-7, miR-146a-5p, miR-21-5p, miR-17~92) which are involved in immunity and disease were detected and the predicted target genes of the novel miRNAs were associated with duck high anti-adversity ability. By gene ontology analysis and enriching KEGG pathway, the targets of differential expressed miRNAs were mainly involved in immunity and disease, supporting that there were differences in the BF immune functions between the two duck breeds. In addition, the metabolic pathway had the maximum enriched target genes and some enriched pathways that were related to cell cycle, protein synthesis, cell proliferation and apoptosis. It indicted that the difference of metabolism may be one of the reasons leading the immune difference between the BF of two duck breeds. Conclusion: This data lists the main differences in the BF at miRNAs level between two genetically disparate duck breeds and lays a foundation to carry out molecular assisted breeding of poultry in the future.

        Isolation and Proteomic Analysis of Extracellular Vesicles from Lactobacillus salivarius SNK-6

        The proteins carried by the extracellular vesicles of Lactobacillus salivarius SNK-6 (LsEVs) were identified to provide a foundation for further explorations of the probiotic activities of L. salivarius SNK-6. LsEVs were isolated from the culture media of L. salivarius SNK-6 and morphological analysis was conducted by scanning electron microscopy. Subsequent transmission electron microscopy and nanoparticle tracking analysis were performed to assess the morphology and particle size of the LsEVs. In addition, the protein composition of LsEVs was analyzed using silver staining and protein mass spectrometry. Finally, internalization of the identified LsEVs was confirmed using a confocal microscope, and enzyme-linked immunosorbent assay was employed to analyze the levels of inflammatory cytokines in LPS-challenged RAW264.7 cells. The results revealed that the membraneenclosed LsEVs were spherical, with diameters ranging from 100–250 nm. The LsEVs with diameters of 111–256 nm contained the greatest amount of cargo. In total, 320 proteins (10–38 kD) were identified in the LsEVs and included anti-inflammatory molecules, such as PrtP proteinase, cochaperones, and elongation factor Tu, as well as some proteins involved in glycolysis/gluconeogenesis, such as fructose-1,6-bisphosphate aldolase. Enrichment analysis showed these proteins to be related to the terms “metabolic pathway,” “ribosome,” “glycolysis/gluconeogenesis,” “carbohydrate metabolism,” and “amino acid metabolism.” Furthermore, the LsEVs were internalized by host liver cells and can regulate inflammation. These findings confirm that LsEVs contain various functional proteins that play important roles in energy metabolism, signal transduction, and biosynthesis.

        Molecular cloning of the duck MEF2C gene cDNA coding domain sequence and its expression during fetal muscle tissue development

        Myogenic enhancer transcription factor 2c (MEF2c), one of the members of the MEF2 family of transcription factors, plays an important role in mammalian muscle development. However, the role of MEF2c in avian muscle development still remains unclear. To understand the function of MEF2c in avian muscle development, we first cloned the duck MEF2c coding domain sequence (CDS) and analyzed MEF2c expression in duck muscle tissues of embryos from 10 days of incubation to 1 week after birth using real-time PCR technology. The results showed that the duck MEF2c CDS consists of 1,398nucleotides that encode 465 amino acids. The MEF2c duck protein contains a MADS domain, a MEF2 domain and a HJURP_C domain with high homology to related proteins in other organisms. Different expression levels of MEF2c were found in skeletal, smooth and cardiac muscle. Therefore, these results indicated that duck MEF2c has two conserved domains (a MADS and a MEF2 domain), is an indispensable regulator of muscle development, and plays an important role in the development of duck muscle.

        Transcriptome reveals genes involving in black skin color formation of ducks

        Background Skin color is colorful for birds, which has been reported to be associated with multi-biological functions, such as crypsis, camoufage, social signaling and mate choice, but little is known about its underlying molecular mechanism. Objective Studies on the major genes afecting the black skin color of ducks. Methods For this purpose, Silver ammonia staining and RNA-seq analysis were carried out to identify the diferences in tissue morphology and gene expressions between black and yellow skin ducks. Results The silver ammonia dyes slice results showed that in the development of black duck, the content of melanin in black skin gradually increased and then decreased, and the content of melanin in yellow and black skin was signifcantly diferent. Through transcriptome, a total of 102 and 84 diferentially expressed genes (DEGs) were identifed in beak skin and web skin, respectively. These DEGs were enriched in melanin biosynthesis and play a critical role in melanogenesis pathway. Co-expression analysis showed that EDNRB2 was the only gene associated with black skin color in DEGs, which was also consistent with qRT-PCR. Conclusions The melanin synthesis pathway dominated by EDNRB2 up-regulated the amount of melanin synthesis, leading to the formation of black skin in ducks.

        Identification of polymorphic loci in the deiodinase 2 gene and their associations with head dimensions in geese

        Objective: This study was conducted to clone and compare the molecular characteristics of the deiodinase 2 (DIO2) gene between Sichuan White geese and Landes geese, and to analyze the association between polymorphisms of the DIO2 gene and head dimensions in Tianfu meat geese. Methods: The coding sequence of the DIO2 gene was cloned by polymerase chain reaction and vector ligation and aligned by DNAMAN software. A total of 350 Tianfu meat geese were used to genotype the polymorphisms of the DIO2 gene and measure the head dimensions. Association analysis between the polymorphisms of the DIO2 gene and head dimensions was carried out. Results: An 840-bp coding sequence of the DIO2 gene was obtained and comparison analysis identified four polymorphic loci between Sichuan White geese and Landes geese. Further analysis showed that the dominant alleles for the four polymorphic loci were G, G, A, and T and the frequency of the heterozygous genotype was higher than that of the homozygous genotype in Tianfu meat geese. Compared to that in the population of non-knob geese of Tianfu meat geese, the head dimensions in the population of knob geese were significantly higher except for nostril height. However, in the non-knob geese, beak width 1, beak width 2, nostril length, cranial width 1, and maxillary length had significant differences among different genotypes or haplotypes/diplotypes. Conclusion: These results suggested that polymorphisms of the DIO2 gene could be considered molecular markers to select larger heads of geese in the population of non-knob geese.
