        Isolation and Characterization of Plant Growth-Promoting Rhizobacteria from Wheat Roots by Wheat Germ Agglutinin Labeled with Fluorescein Isothiocyanate

        Thirty-two isolates were obtained from wheat rhizosphere by wheat germ agglutinin (WGA) labeled with fluorescein isothiocyanate (FITC). Most isolates were able to produce indole acetic acid (65.6%) and siderophores (59.3%), as well as exhibited phosphate solubilization (96.8%). Fourteen isolates displayed three plant growth-promoting traits. Among these strains, two phosphate-dissolving ones, WS29 and WS31, were evaluated for their beneficial effects on the early growth of wheat (Triticum aestivum Wan33). Strain WS29and WS31 significantly promoted the development of lateral roots by 34.9% and 27.6%, as well as increased the root dry weight by 25.0% and 25.6%, respectively, compared to those of the control. Based on 16S rRNA gene sequence comparisons and phylogenetic positions, both isolates were determined to belong to the genus Bacillus. The proportion of isolates showing the properties of plant growth-promoting rhizobacteria (PGPR) was higher than in previous reports. The efficiency of the isolation of PGPR strains was also greatly increased by WGA labeled with FITC. The present study indicated that WGA could be used as an effective tool for isolating PGPR strains with high affinity to host plants from wheat roots. The proposed approach could facilitate research on biofertilizers or biocontrol agents.

        Synthesis of novel oleanolic acid and ursolic acid in C-28 position derivatives as potential anticancer agents

        A series of nitrogen-containing derivatives ofoleanolic acid and ursolic acid were prepared by a modificationat C-28 position via esterification with 2-hydroxyaceticacid followed by amidation with amines, such aspiperazine, N-methylpiperazine, and alkane-1, 2-diamines,alkane-1, 4-diamines, alkane-1, 6-diamines. In vitroantiproliferative activities of the compounds preparedtowards MCF-7, Hela and A549 cell lines were evaluatedby a MTT method to show that OA-5a, OA-5b, OA-5c andUA-5a showed somewhat improved antiproliferativeactivities against MCF-7, Hela and A549 cells comparingto that of the positive control, gefitinib.

        Prognostic Value of the Evolution of HER2-Low Expression after Neoadjuvant Chemotherapy

        Purpose Patients with human epidermal growth factor receptor 2 (HER2)–low advanced breast cancer can benefit from trastuzumab deruxtecan. Given the unclear prognostic characteristics of HER2-low breast cancer, we investigated the prognostic characteristics of HER2-low expression from primary tumor to residual disease after neoadjuvant chemotherapy (NACT). Materials and Methods The data of HER2-negative patients receiving NACT at our center were collected. Pathological complete response (pCR) rate were compared between HER2-0 and HER2-low patients. The evolution of HER2 expression from primary tumor to residual disease and its impact on disease-free survival (DFS) were examined. Results Of the 690 patients, 494 patients had HER2-low status, of which 72.3% were hormone receptor (HR)–positive (p < 0.001). The pCR rates of HER2-low and HER2-0 patients (14.2% vs. 23.0%) showed no difference in multivariate analysis regardless of HR status. No association was observed between DFS and HER2 status. Of the 564 non-pCR patients, 57 (10.1%) changed to HER2-positive, and 64 of the 150 patients (42.7%) with HER2-0 tumors changed to HER2-low. HER2-low (p=0.004) and HR-positive (p=0.010) tumors before NACT were prone to HER2 gain. HER2 gain patients had a better DFS compared with HER2-negative maintained patients (87.9% vs. 79.5%, p=0.048), and the DFS of targeted therapy group was better than that of no targeted therapy group (92.4% vs. 66.7%, p=0.016). Conclusion Although HER2-low did not affect the pCR rate and DFS, significant evolution of HER2-low expression after NACT creates opportunities for targeted therapy including trastuzumab.

      • A Novel Algorithm for Aggregating the Topological Nodes in Web GIS Network Management

        The problems of the icons covering each other and slow data loading are very challenging when the GIS technology is applied to load a large number of topological nodes. A novel algorithm for aggregating the topological nodes in GIS based on the hierarchical clustering algorithm is presented in this paper. Comparison with the hierarchical clustering algorithm and partitioning algorithm is explored. In addition, the implementation of the algorithm is derived. The experiment demonstrates that the algorithm solved the problems of the icons covering each other and slow data loading effectively, realized the aggregation of the topological nodes preferably, and presented the distributed trend of the topological nodes and the topology relation between each other in the topology. The experimental results verify the effectiveness of the method.

        Genetic variation and population structure of the oriental fruit moth Grapholita molesta in Shanxi, a major pome fruits growing region in North China

        Grapholitamolesta, commonly known as the oriental fruit moth, is one of the most destructive fruit pests both in China and in other regions of the world. We examined the genetic diversity and population structures of G. molesta populations from 11 locations in a major pome fruit production region (Shanxi province) in North China using seven microsatellite markers and sequence variations in two mitochondrial genes. We found that G.molesta in the studied region exhibited high levels of genetic diversity. Among 271 individual insects examined using microsatellite markers, mean number of alleles ranged from 5.86 to 9.00 with average 8.01. Population structure analysis revealed that populations from the eleven sampling sites formed two clusters without apparent distance isolation. Cluster 1 consisted of eight populations distributed in five regions of the sampling sites, whereas Cluster 2 consisted of three populations distributed in two distant locations of the sampling sites. Genetic exchange analysis revealed that about half of the haplotypes were shared among over 90% individual insects from different regions. Our results indicated that the population structure and genetic exchange of G. molesta in the studied region might have been affected by limited dispersal ability of the insect, environmental condition, colonization history and human-mediated dispersal. These findings should provide useful information for the implementation of area-wide pest management and for further characterization of genetic variation and population structure of this economically important pest.

        Construction of graphene/AuNPs based amperometric immunosensor for detecting bladder cancer biomarker apolipoprotein A1

        This paper presents the construction and characterization of an amperometric immunosensor based on the graphene/gold nanoparticles (AuNPs/GO) nanocomposite for the detection of the bladder cancer biomarker, apolipoprotein A1 (Apo-A1). The morphological analysis of the AuNPs/GO nanocomposite demonstrated an almost spherical shape of AuNPs and the successful coverage of their surface by graphene oxide. An increased G peak and decreased D peak after the association of AuNPs with GO, implied a reduction in graphene defects. The X-ray photoelectron spectroscopy (XPS) indicated a significant decrease in the quantity of oxygen-containing functional groups in the AuNPs/GO nanocomposite, as compared to the original GO. Furthermore, the developed sensor demonstrated commendable sensitivity and selectivity, with a wide linear range for Apo-A1 detection. Importantly, the immunosensor exhibited remarkable stability over a period of 14 days, signifying its potential for practical applications.
