To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

The purpose of this study was to analyse brainstem auditory evoked potentials (BAEP) wave change data during microvascular decompression (MVD). The nerve function of Cranial Nerve VIII is at risk during MVD. Intraoperative monitoring of BAEP can be a useful tool to decrease the danger of hearing loss. Between January and December 2009, 242 patients had MVD for hemifacial spasm (HFS) and trigeminal neuralgia (TN). Among intraoperative BAEP changes, amplitude of V-V’ was the most frequently observed during cerebellar retraction and decompression step of the MVD procedure. 138 patients (57%) had no BAEP change while 104 patients (42.98%) had BAEP change. 69 patients (28.5%) had Type A-I, 16 patients (6.6%) had Type A-II, 5 patients (2.1%) had Type B, and 13 patients (5.37%) had Type C. MVD is a surgical procedure to relieve the symptoms (e.g. pain, muscle twitching) caused by compression of a nerve by an artery or vein. During BAEP intraoperative monitoring, the surgical step is important in interpreting the changes of wave V. Several potential mechanisms of injury may affect the cochlear nerve, and complete loss of BAEP is often associated with postoperative hearing loss. Intraoperative BAEP monitoring may provide an early warning of hearing disturbance after MVD.
Optimal Gas Detection System in Cargo Compressor Room of Gas Fueled LNG Carrier

본 연구는 가스추진 174K급 LNG 운반선의 가스 압축기실에서 발생하는 가스누출 모사를 통해 가스탐지기의 최적 위치를 분석 하였으며, 새로 개정된 IGC 코드에 명시된 안전규정을 만족하는 합리적인 방법도 함께 제안하였다. 가스압축기실에서의 LNG 가스누출 수치해석을 위해, 실제 ME-GI 엔진이 장착된 174K급 LNG 운반선의 압축기실 형상과 장비, 배관의 배치와 같은 치수로 3D 설계되었다. 가 스누설에 대한 시나리오는 305 bar의 높은 압력과 1 bar의 낮은 압력을 적용하여 진행하였다. 고압용 핀홀의 크기는 4.5, 5.0, 5.6 mm이고 저압용은 100, 140 mm이다. 해석 결과, 5.6 mm 핀홀(고압)과 100, 140 mm 핀홀(저압) 상태의 누출에 대한 환기평가에서 가연성 가스농도는 심 각한 위험이 없음을 확인하였다. 그러나 개정된 IGC 코드에 따라 설치된 압축기실의 가스 감지 센서의 실제 위치는 다른 지점으로 이동 해야 하고, 측정 지점이 현 규정에서 요구하는 것보다 더 추가되어야 함을 확인하였다. This study analyzes the optimal location of gas detectors through the gas dispersion in a cargo compressor room of a 174K LNG carrier equipped with high-pressure cargo handling equipment; in addition, we propose a reasonable method for determining the safety regulations specified in the new International Code of the Construction and Equipment of Ships Carrying Liquefied Gases in Bulk (IGC). To conduct an LNG gas dispersion simulation in the cargo compressor room-equipped with an ME-GI engine-of a 174 K LNG carrier, the geometry of the room as well as the equipment and piping, are designed using the same 3D size at a 1-to-1 scale. Scenarios for a gas leak were examined under high pressure of 305 bar and low pressure of 1 bar. The pinhole sizes for high pressure are 4.5, 5.0, and 5.6mm, and for low pressure are 100 and 140 mm. The results demonstrate that the cargo compressor room will not pose a serious risk with respect to the flammable gas concentration as verified by a ventilation assessment for a 5.6 mm pinhole for a high-pressure leak under gas rupture conditions, and a low-pressure leak of 100 and 140 mm with different pinhole sizes. However, it was confirmed that the actual location of the gas detection sensors in a cargo compressor room, according to the new IGC code, should be moved to other points, and an analysis of the virtual monitor points through a computational fluid dynamics (CFD) simulation.

A low endotoxic Outer membrane vesicles (OMVs) vaccine platform was established with an msbB genedeleted (ΔmsbB) Salmonella enteritidis mutant. The Salmonella ΔmsbB mutant displayed lipopolysaccharide phenotypic changes showing faster migration of the lipid A-core region in an SDS-PAGE analysis and the increased amount of penta-acyl lipid A due to the MsbB deficiency. In an endotoxicity assay, BALB/c mice injected intraperitoneally with the Salmonella ΔmsbB mutant survived for 10 days, whereas mice injected intraperitoneally with the wild type survived for 5 days. Also, all mice inoculated orally with the ΔmsbB mutant survived for 30 days but 70% of mice inoculated orally with wild type survived. Electron microscopically, the Salmonella ΔmsbB mutant produced a larger amount of OMVs than the wild type. In immunogenicity tests, all sera from mice groups immunized with the wild type, ΔmsbB mutant, and their OMVs, showed significantly higher IgG levels. This result was consistent with higher bactericidal activities against wild type S. enteritidis, compared to the negative control. However, there were no significant differences in serum IgG levels and the bactericidal activities of the immune sera between the four mice groups. The viable counts of S. enteritidis recovered from the spleen and liver of four preimmunized mice groups after 5 days of the bacterial challenge showed significant reductions of the live bacteria. Conclusively, the ΔmsbB mutant of S. enteritidis produced relatively low endotoxic OMVs, which was verified in this study for its potential to be a non-replicating Salmonella vaccine candidate.

The purpose of this study was to determine the influential factors on the early osteoblastic differentiation of MG63 osteoblast-like cells cultured on micro-grooved Ti substrata. Commercially pure Ti discs with surface microgrooves of various dimensions were fabricated using photolithography. Both ground and acid-etched Ti were used as controls. Surface widths of the Ti substrata were calculated and used as the surface area parameters. These surfaces were characterized by X-ray photoelectron spectroscopy (XPS) and water contact angle (WCA). Early osteoblastic differentiation of MG63 human osteoblast-like cells were analyzed using an alkaline phosphatase (ALP) activity test. Pearson`s correlation analysis and multiple regression analysis were used to analyze the correlations between experimental results and to determine the influential factors on osteoblastic differentiation, respectively. The results of experiments showed significant correlations between surface properties and osteoblastic differentiation. Surface width was determined to be the greatest influential factor on the ALP activity after 1 day of osteogenic culture. The water contact angles measured in perpendicular directions to the microgrooves showed greatest influence on the ALP activity on day 7. This study indicates that the factors influencing the early osteoblastic differentiation on micro-grooved Ti substrata show respective time-dependence.
