        A NOVEL FUNCTIONAL MATERIAL OF Ag-S$i_3N_4$ NANOSTRUCTURED COMPOSITE

        $Ag-Si_3N_4$ nanostrutured composite has been investigated. The results show that it exhibits some novel properties such as strong piezpelectricity and high dielectricity. It can be constituted a battery using graphite and aluminum as the positive and the negative electrodes, respectively. The origins of these properties have been discussed.

        Coexpression of Kex2 Endoproteinase and Hac1 Transcription Factor to Improve the Secretory Expression of Bovine Lactoferrin in Pichia pastoris

        The large-scale production of functional recombinant lactoferrin has become a major goal because of its medicinal value and global demand. Secreting recombinant proteins into a culture medium offers a way to simplify protein purification and avoid toxicity from intracellularly accumulated materials. In this study, after 84 h of induction with methanol in a shaking flask, the recombinant bovine lactoferrin (rbLf) titer in the culture supernatant of the strain that integrated two copies of the rbLf gene was only 121.6 μg/L. A bottleneck might have existed in the folding and secretion pathways of rbLf. We then attempted to further improve the rbLf titer by overexpressing the transcription factor Hac1p and α-signal peptide-cutting protease Kex2p with different promoters. Results showed that the inducible coexpression of Hac1p and Kex2p linked with the 2A sequence improved the rbLf titer 5.0-fold (735.8 μg/L) after 84 h of induction with methanol. The maximal titer in a shaking flask was 1,150.5 μg/L after 120 h of induction. The rbLf titer achieved 35.6 mg/L in a 5 L fed-batch fermenter. Thus, Kex2 and Hac1 overexpression driven by methanol-induced promoter alleviated the bottleneck in the folding and secretion pathways and greatly improved the secretory expression of rbLf in Pichia pastoris.

        Effects of rehydration on physiological and transcriptional responses of a water-stressed rhizobium

        As a microsymbiont of soybean, Bradyrhizobium japonicum plays an important role in symbiotic nitrogen fixation and sustainable agriculture. However, the survival of B. japonicum cells under water-deplete (e.g., drought) and water-replete (e.g., flood) conditions is a major concern affecting their nitrogen-fixing ability by establishing the symbiotic relationship with the host. In this study, we isolated a water stress tolerant rhizobium from soybean root nodules and tested its survival under water-deplete conditions. The rhizobium was identified as Bradyrhizobium japonicum and named strain 5038. Interestingly, both plate counting and live/dead fluorescence staining assays indicate that a number of viable but non-culturable cells exist in the culture medium upon the rehydration process which could cause dilution stress. Bradyrhizobium japonicum 5038 cells increased production of exopolysaccharide (EPS) and trehalose when dehydrated, suggesting that protective responses were stimulated. As expected, cells reduced their production upon the subsequent rehydration. To examine differential gene expression of B. japonicum 5038 when exposed to water-deplete and subsequent waterreplete conditions, whole-genome transcriptional analysis was performed under 10% relative humidity (RH), and subsequent 100% RH, respectively. A total of 462 differentially expressed genes (DEGs, > 2.0-fold) were identified under the 10% RH condition, while 3,776 genes showed differential expression during the subsequent rehydration (100% RH) process. Genes involved in signal transduction, inorganic ion transport, energy production and metabolisms of carbohydrates, amino acids, and lipids were far more up-regulated than downregulated in the 10% RH condition. Notably, trehalose biosynthetic genes (otsAB, treS, and treYZ), genes ligD, oprB, and a sigma factor rpoH were significantly induced by 10% RH. Under the subsequent 100% RH condition, genes involved in transcription, translation, cell membrane regulation, replication and repair, and protein processing were highly up-regulated. Interestingly, most of 10%-RH inducible genes displayed rehydration-repressed, except three genes encoding heat shock (Hsp20) proteins. Therefore, this study provides molecular evidence for the switch of gene expression of B. japonicum cells when encountered the opposite water availability from water-deplete to water-replete conditions.

        Experimental Study On Solar Clothes Dryer using Evacuated Tube Collector and PV Panel

        A solar dryer for clothes and its testing results are presented in this paper. The dryer combines a solar thermal collector with a photo voltaic panel to supply hot dry air for clothes, but can also be used for food drying applications. Twin-glass evacuated tubes with heat pipes are used in the solar collector as the main heat source, while DC fans powered by a PV panel are used as the power source for hot air circulation in this unit. The performance of the dryer was tested under the UK real weather conditions. The air in the dryer is heated by more than 17℃ and the humidity is decreased more than 20% for a solar collector mea of 0.78㎡ and a PV panel mea of 0.68 ㎡. The average water removing rate is 0.464 ㎏/h for a solar irradiation of 646.5 W/㎡.

        An Optimal Random Carrier Pulse Width Modulation Technique Based on a Genetic Algorithm

        Since the carrier sequence is not reproducible in a period of the random carrier pulse width modulation (RCPWM) and a higher harmonic spectrum amplitude is likely to affect the quality of the power supply. In addition, electromagnetic interference (EMI) and mechanical vibration will appear. To solve these problems, this paper has proposed an optimal RCPWM based on a genetic algorithm (GA). In the optimal modulation, the range of the random carrier frequency is taken as a constraint and the reciprocal of the maximum harmonic spectrum amplitude is used as a fitness function to decrease the EMI and mechanical vibration caused by the harmonics concentrated at the carrier frequency and its multiples. Since the problems of the hardware make it difficult to use in practical engineering, this paper has presented a hardware system. Simulations and experiments show that the RCPWM is effective. Studies show that the harmonic spectrum is distributed more uniformly in the frequency domain and that there is no obvious peak in the wave spectra. The proposed method is of great value to research on RCPWM and integrated power systems (IPS).

        Mode Switching Smooth Control of Transient Process of Grid-Connected 400 Hz Solid-State Power Supply System

        The mode-switching control of transient process is important to grid-connected 400 Hz solid-state power supply systems. Therefore, this paper analyzes the principle of on-grid and islanding operation of the system with or without local loads in the grid-connected process and provides a theoretical study of the effect of different switching sequences on the mode-switching transient process. The conclusion is that the mode switch (MS) must be turned on before the solid-state switch (STS) in the on-grid process and that STS must be turned off before the MS in the off-grid process. A strategy of mode-switching smooth control for transient process of the system is proposed, including its concrete steps. The strategy utilizes the average distribution of peak currents and the smooth adjustment of peak currents and phases to achieve a no-shock grid connection. The simulation and experimental results show that the theoretical analysis is correct and that the method is effective.

        One-Carbon Metabolism Nutrients, Genetic Variation, and Diabetes Mellitus

        Diabetes mellitus (DM) affects about 9.3% of the population globally. Hyperhomocysteinemia (HHcy) has been implicated in the pathogenesis of DM, owing to its promotion of oxidative stress, β-cell dysfunction, and insulin resistance. HHcy can result from low status of one-carbon metabolism (OCM) nutrients (e.g., folate, choline, betaine, vitamin B6, B12), which work together to degrade homocysteine by methylation. The etiology of HHcy may also involve genetic variation encoding key enzymes in OCM. This review aimed to provide an overview of the existing literature assessing the link between OCM nutrients status, related genetic factors, and incident DM. We also discussed possible mechanisms underlying the role of OCM in DM development and provided recommendations for future research and practice. Even though the available evidence remains inconsistent, some studies support the potential beneficial effects of intakes or blood levels of OCM nutrients on DM development. Moreover, certain variants in OCM-related genes may influence metabolic handling of methyl-donors and presumably incidental DM. Future studies are warranted to establish the causal inference between OCM and DM and examine the interaction of OCM nutrients and genetic factors with DM development, which will inform the personalized recommendations for OCM nutrients intakes on DM prevention.

        Demographic potential of the pupal parasitoid Trichopria drosophilae (Hymenoptera: Diapriidae) reared on Drosophila suzukii (Diptera: Drosophilidae)

        Parasitoids represent a potential tool to control the invasive spotted wing drosophila, Drosophila suzukii, which has invaded Europe and America, recently. A good example is the pupal parasitoid, Trichopria drosophilae, would be an effective biocontrol agent. Populations of D. suzukii, and T. drosophilae were collected from a blueberry orchard in East China and reared in the laboratory at 25 °C. Life table and population growth data for T. drosophilae were obtained using D. suzukii pupae as a host. Mean adult longevity was 22.40 d and 26.45 d, for females and males respectively. The mean oviposition period was 18.20 d. The daily mean parasitization rate was 9.47% per female when 30 hosts were daily provided. The total number of T. drosophilae offspring throughout a lifetime was 63.45 per female, and the female offspring proportion was 65.06%. The intrinsic rate of natural increase and the finite rate of increase were 0.18 and 1.19, respectively. The generation time and the population doubling time were 21.29 d and 3.91 d, respectively. Our results suggest that T. drosophilae population differed among populations in population growth parameters, and those from China also has the parasitism potential to control D. suzukii.

        中国共产党党内法规制度建设的学理诠释

        The communist party of China has provided a model for the realization of the modernization of party governance structure by the Leninist party. During the transition process from the traditional Leninist party to the modern ruling party, the construction of inner-party law and regulation system plays an important role. To discuss the theoretical paradigm of governance structure of the communist party of China modernization from the national system level is not enough to explain the practice of the communist party of China, the communist party of China in the modernization of political party governance structure, follows the “revolution” paradigm, through inner germinal restricting power, to avoid external forces or mechanisms that threaten the party"s leadership; it can also maintain the party"s purity and sophistication and maintain the authority and unity of the party"s policy. In the process of self-revolution, the communist party of China realizes the “ruling of the party rules” through the construction of party rules and regulations to take the “ruling of the party rules” as the path of the modernization of party governance structure.
