        A Prediction Model of the Sum of Container Based on Combined BP Neural Network and SVM

        The prediction of the sum of container is very important in the field of container transport. Many influencing factors can affect the prediction results. These factors are usually composed of many variables, whose composition is often very complex. In this paper, we use gray relational analysis to set up a proper forecast index system for the prediction of the sum of containers in foreign trade. To address the issue of the low accuracy of the traditional prediction models and the problem of the difficulty of fully considering all the factors and other issues, this paper puts forward a prediction model which is combined with a back-propagation (BP) neural networks and the support vector machine (SVM). First, it gives the prediction with the data normalized by the BP neural network and generates a preliminary forecast data. Second, it employs SVM for the residual correction calculation for the results based on the preliminary data. The results of practical examples show that the overall relative error of the combined prediction model is no more than 1.5%, which is less than the relative error of the single prediction models. It is hoped that the research can provide a useful reference for the prediction of the sum of container and related studies.

        Background: The association between altered expression of miR-34a and pathophysiological features of nonalcoholic fatty liver disease (NAFLD) and whether there is a connection between susceptibility to NAFLD has not been completely clarifi ed. Methods: The vitro model was established by culturing L02 cells with a high concentration of free fatty acid (FFA) and the vivo model was established by feeding C57BL/6 mice with HFD. To determine the effects of miR-34a, cultured L02 cells transfected with miR-34a inhibitor and C57BL/6 mice injected with miR-34a inhibitor through vein tail were analysed for the level of PPARaand the metabolic sensor AMPK. In functional experiments, TG content and steatosis degree were measured by TG assay kit, HE and Oil Red O staining. Results: miR-34a expression is signifi cantly upregulated in steatosis-induced hepatocytes and in liver tissues of HFD fed mice. The upregulation of miR-34a resulted in the downregulation of hepatic peroxisome proliferator-activated receptor-a(PPARa), the direct target of miR-34a. Moreover, the action of miRNA-34a on PPAR-a depends on the presence of a single miRNA-34a binding site. Silencing miR-34a led to an initial increase the level of PPARaand the targets of PPARa, including CPT1, CPT2, SLC27A4, SLC27A1 and ACBD3. Activation of the central metabolic sensor AMPK was also increased. In functional experiments, miR-34a inihibitor suppressed cell and mice liver TG content and improve steatosis degree. Furthermore, inhibition of PPARa expression aggravated hepatocellular steatosis in vitro models. Conclusions: NAFLD is associated with altered hepatic miR-34a expression. Decreased expression of miR-34a potentially contributes to altered lipid metabolism implicated in the pathogenesis of NAFLD. These results suggest that regulating it`s target PPARa by down-regulation of miR-34a levels may be a therapeutic strategy against NAFLD.

        Background: The mechanisms of pathogenesis underlying nonalcoholic fatty liver disease (NAFLD) remain unclear at present and the depth study of HADHA in the development of NAFLD has never been investigated. Methods: The NAFLD cell model was established by treating L02 cells with free fatty acid (FFA) overload. Small interfering RNA (siRNA) was used to knock down HADHA level. The expression of HADHA and key enzymes associated with fatty acid beta- oxidation in L02 cells were determined by q-PCR. Key protein associated in energy metabolism, endoplasmic reticulum(ER) stress and infl ammatory were determined by western blotting. ATP, hydroperoxide (H2O2), catalase (CAT) and mitochondrial membrane potential (MMP) were measured. The prediction of HADHA upstream regulation of miRNA was carried out and luciferase reporter assays were implemented to validate the prediction. Results: After culturing L02 cells by FFA for 48h, we detect the increased protein level of HADHA. HADHA knockdown in L02 cells resulted in an increased of lipid accumulation and downregulation of gene expression involved in fatty acid beta-oxidation, including PPARa, ACOX1, CPT2, EHHADH, ECHS1, HADHB and HADH. Additionally, administering HADHA siRNA exhibited improvement of oxidative stress, embodied in decreased level of H2O2 and MDA, meanwhile, increased levels of ATP, CAT and MMP. Furthermore, HADHA knockdown demonstrated weakened AMPK pathway, activation of MAPK and MKK3 pathway, and improve ER stress by downregulation of C/EBPa and C/EBPß. Moreover, HADHA was regulated directly by upstream gene of miR-124. Conclusions: Our results show that HADHA may plays a role of double-edged sword in hepatic steatosis and cell injury in NAFLD, and provide a new insight into the pathogenic mechanisms of NAFLD, may becoming a potential new therapeutic target for NAFLD.

        Removal efficiency and mechanism of bio-treated coking wastewater by catalytic ozonation using MnO₂ modified with anionic precursors

        In this paper, MnO₂ catalyst were firstly prepared and modified by four kinds of anionic precursors (i.e., NO₃⁻, AC⁻, SO₄<SUP>2-</SUP> and Cl-) through redox precipitation method. After that, bio-treated coking wastewater (BTCW) was prepared and employed as targeted pollutants to investigate the catalytic ozonation performance of prepared-MnO₂ catalyst was investigated and characterized by the removal efficiencies and mechanism of the prepared bio-treated coking wastewater (BTCW), which was employed as the targeted pollutants. Specifically, the effects of specific surface area, crystal structure, valence state of Mn element and lattice oxygen content on catalytic activity of MnO₂ materials were characterized by BET, XRD and XPS, respectively. Results showed that COD of BTCW could be removed 47.39% under MnO₂-NO₃⁻ catalyst with 2 h reaction time, which was much higher than that of MnO₂-AC⁻ (3.94%), MnO₂-SO₄<SUP>2-</SUP> (12.42%), MnO₂-Cl⁻ (12.94%) and pure O₃ without catalyst (21.51%), respectively. So, MnO₂-NO₃⁻ presented the highest catalytic performance among these catalysts. The reason may be attributed to a series of better physiochemical properties including the smaller average grain, the larger specific surface area and active groups, more crystal defect and oxygen vacancy, higher relative content of Mn<SUP>3+</SUP> and adsorbed oxygen (Oads) than that of another three catalysts.

        Excessive mechanical strain accelerates intervertebral disc degeneration by disrupting intrinsic circadian rhythm

        Night shift workers with disordered rhythmic mechanical loading are more prone to intervertebral disc degeneration (IDD). Our results showed that circadian rhythm (CR) was dampened in degenerated and aged NP cells. Long-term environmental CR disruption promoted IDD in rats. Excessive mechanical strain disrupted the CR and inhibited the expression of core clock proteins. The inhibitory effect of mechanical loading on the expression of extracellular matrix genes could be reversed by BMAL1 overexpression in NP cells. The Rho/ROCK pathway was demonstrated to mediate the effect of mechanical stimulation on CR. Prolonged mechanical loading for 12 months affected intrinsic CR genes and induced IDD in a model of upright posture in a normal environment. Unexpectedly, mechanical loading further accelerated the IDD in an Light-Dark (LD) cycle-disrupted environment. These results indicated that intrinsic CR disruption might be a mechanism involved in overloading-induced IDD and a potential drug target for night shift workers.

      • Overexpression of Platelet-derived Growth Factor-D as a Poor Prognosticator in Endometrial Cancer

        Background: Emerging evidence implicates the platelet-derived growth factor-D (PDGF-D) in many types of human solid tumors. We investigated whether PDGF-D plays an important role in endometrial cancer (EC) in relation to clinicopathologic phenotype, angiogenesis, and patient prognosis. Materials and Methods: We analyzed PDGF-D protein expression by Western blotting in twenty-seven human endometrial cancer tissues, and matched normal endometrial controls collected at the third Affiliated hospital of Sun Yat-sen University during 2012-2013 (n=27). Immunohistochemical staining was performed using a human PDGF-D antibody on the endometrial cancer patients collected in the same facility during January 2001 and October 2013 (n=152). Patients were followed from the time of primary surgery in 2001-2013 until death or last follow-up. We correlated the PDGF-D expression levels with clinicopathologic parameters and prognosis in human endometrial cancer patients. Results: Compared with matched normal endometrial cases, PDGF-D was up-regulated in endometrial cancer. Expression of PDGF-D protein, found in 78% of the cases, was associated with nonendometrioid histologic type (p=0.028), FIGO stage III/IV (p=0.039), >50% solid tumor growth (p=0.048), pelvic LN metastasis (p=0.035) and ER and PR negativity (p=0.04 and 0.002). PDGF-D expression was also significantly associated with expression of VEGF-A (p=0.021). In multivariate analysis, PDGF-D expression proved to be an independent prognostic factor in addition to histologic grade and FIGO stage. Patients with high expression levels of PDGF-D had a significantly poorer overall survival rate compared with patients with no expression. Conclusions: PDGF-D expression is frequently up-regulated in endometrial cancer, and is associated with aggressive features and poor prognosis.

        間斷-均衡理論與中國政策終結的生成機理

        On December 28, 2013, the Lao Jiao system was finally terminated through legal procedures. People wonder why it could run more than 50 years, though it violated the rule of law, human rights and other values obviously. From the view of Punctuated-Equilibrium theory, the policy image and policy institutional venues interacted, which made the policy equilibrium or policy punctuation. The research shows trajectory on the termination of Lao Jiao policy is consistent with the theory of punctuated-equilibrium, taking on the form of equilibrium-punctuation-equilibrium-punctuation. At the same time, the termination of Lao Jiao policy can also be attributed the interaction between the policy attributes、policy environment and constraints. In these factors, the political ideology play an important role in the policy termination.
