      • MicroRNA-146a Enhances Helicobacter pylori Induced Cell Apoptosis in Human Gastric Cancer Epithelial Cells

        Helicobacter pylori (H. pylori) infection induces apoptosis in gastric epithelial cells, and this occurrence may link to gastric carcinogenesis. However, the regulatory mechanism of H. pylori-induced apoptosis is not clear. MicroRNA-146a has been implicated as a key regulator of the immune system. This report describes our discovery of molecular mechanisms of microRNA-146a regulation of apoptosis in human gastric cancer cells. We found that overexpression of microRNA-146a by transfecting microRNA-146a mimics could significantly enhance apoptosis, and this upregulation was triggered by COX-2 inhibition. Furthermore, we found that microRNA-146a density was positively correlated with apoptosis rates in H. pylori-positive gastric cancer tissues and intratumoral microRNA-146a density was negatively correlated with lymph node metastasis among H. pylori-positive gastric cancer patients. Understanding the important roles of microRNA-146a in regulating cell apoptosis in H. pylori infected human gastric cancer cells will contribute to the development of microRNA targeted therapy in the future.

      • Effects of MicroRNA-106 on Proliferation of Gastric Cancer Cell through Regulating p21 and E2F5

        Objective: To investigate the effects of miR-106b on malignant characteristics of gastric cancer cells, and explore possible mechanisms. Methods: Expression of miR-106b, p21 and E2F was determined by real-time PCR. Transfection with miR-106b mimics was conducted, and gastric cancer cells with miR-106b overexpression were obtained. Cells transfected with mimic mutants and those without transfection served as negative and blank controls, respectively. Flow cytometry and transwell assays were adopted to detect the effects of miR-106b overexpression on cell cycle, migration and invasion of gastric cancer cells. Results:. The expression of miR- 106b in gastric cancer cells was significantly higher than that in normal gastric mucosa cells. Furthermore, the expression level of miR-106b rose according to the degree of malignacy among the three GC cell strains (MKN- 45 > SGC-7901 > MKN-28). Overexpression of miR-106b shortened the G0/G1 phase and accelerated cell cycle progression, while reducing p21 and E2F5, without any significant effects on the capacity for migration and invasion of gastric cancer cells. Conclusions: miR-106b may promote cell cycling of gastric cancer cells through regulation of p21 and E2F5 target gene expression.

        Ultrasonic Vibration Assisted Cutting of Nomex Honeycomb Core Materials

        Nomex honeycomb core materials have been widely used in the aviation industry due to their special structure and performance. Conventional high-speed machining have resulted in the poor machinability of the honeycomb core so that the ultrasonic machining technology was applied. The kinematic characteristics in the ultrasonic vibration assisted cutting process were analyzed according to the movement of the sharp tool. Based on slide effect, a cutting force model was proposed to study the relationship between cutting parameters and cutting force. Ultrasonic vibration assisted cutting and ordinary cutting tests of Nomex honeycomb core material were conducted by considering feed rate, the inclined angle and the deflected angle. Besides, the effects of cutting parameters on machined surface quality of honeycomb core wall were studied. The test results show that slide effect caused by ultrasonic vibrations can reduce cutting resistance compared with ordinary cutting. The developed cutting force model can be applied to evaluated the cutting force in the ultrasonic vibration assisted cutting of Nomex honeycomb core material. The inclined angle has a great influence on the cutting force during ultrasonic vibration assisted cutting. High-speed reciprocating sliding action can effectively cut aramid fibers so that burrs and tearing defects of the incision have been greatly improved under condition of ultrasonic vibration assisted cutting.

        Development and verification of a Monte Carlo two-step method for lead-based fast reactor neutronics analysis

        With the rise of economic and safety standards for nuclear reactors, new concepts of Gen-IV reactors and modular reactors showed more complex designs that challenge current tools for reactor physics analysis. A Monte Carlo (MC) two-step method was proposed in this work. This calculation scheme uses the continuous-energy MC method to generate multi-group cross-sections from heterogeneous models. The multi-group MC method, which can adapt locally-heterogeneous models, is used in the core calculation step. This calculation scheme is verified using a Gen-IV modular lead-based fast reactor (LFR) benchmark case. The influence of homogenized patterns, scatter approximations, flux separable approximation, and local heterogeneity in core calculation on simulation results are investigated. Results showed that the cross-sections generated using the 3D assembly model with a locally heterogeneous representation of control rods lead to an accurate estimation with less than 270 pcm bias in core reactivity, 0.5% bias in control rod worth, and 1.5% bias on power distribution. The study verified the applicability of multi-group cross-sections generated with the MC method for LFR analysis. The study also proved the feasibility of multi-group MC in core calculation with local heterogeneity, which saves 85% time compared to the continuous-energy MC.

        A phase I dose-finding trial of hyperthermic intraperitoneal docetaxel combined with cisplatin in patients with advanced-stage ovarian cancer

        Objective: To identif y the maximum tolerated dose (MTD) of docetaxel combined with afixed dose of cisplatin (75 mg/m2) delivered as hyperthermic intraperitoneal chemotherapy(HIPEC) in patients with ovarian cancer. Methods: In this phase I trial, a time-to-event Bayesian optimal inter val design was used. Docetaxel was given at a starting dose of 60 mg/m2 and was increased in 5 mg/m2 incrementsuntil the MTD was determined or the maximum dose level of 75 mg/m2 was reached. The dose-limiting toxicity (DLT) rate was set at 25%, with a total sample size of 30 patients. HIPEC wasdelivered immediately following debulking surger y at a target temperature of 43°C for 90 minutes. Results: From August 2022 to November 2022, 30 patients were enrolled. Among thepatients who received a dose of docetaxel ≤65 mg/m2, no DLT was reported. DLTs wereobser ved in one patient who received 70 mg/m2 docetaxel (grade 3 anaemia) and in threepatients who received 75 mg/m2 docetaxel (one case of grade 3 anaemia, one case of grade3 hepatic impairment and one case of grade 4 thrombocytopenia). Patients treated withdocetaxel 75 mg/m2 in combination with cisplatin 75 mg/m2 had an estimated DLT rate of25%, which was the closest to the target DLT rate and was therefore chosen as the MTD. Conclusion: Docetaxel, in combination with a fixed dose of cisplatin (75 mg/m2), can be usedsafely at intraperitoneal doses of 75 mg/m2 in ovarian cancer patients who received HIPEC(43°C, 90 minutes) following debulking surger y.         Feasibility, accuracy and acceptability of self-sampled human papillomavirus testing using careHPV in Cambodia: a cross-sectional study

        Objective: Self-sampled human papillomavirus (HPV) testing is a potential option forcer vical cancer screening, but research is scarce in Cambodia. We evaluated the feasibility,accuracy, and acceptability of self-sampled HPV testing using careHPV. Methods: A cross-sectional study including women aged 20–49 years attending 2 nationalhospitals in the capital city was conducted. Women under went both self-sampling andclinician-sampling of specimens, and were then asked to complete an acceptabilityquestionnaire. The paired samples were analyzed for high-risk HPV by careHPV andgenotyped by polymerase chain reaction (PCR). Results: A total of 375 women were eligible for inclusion. Based on PCR, 78.9% were negativefor HPV in both self and clinician-samples, 9.9% had a complete HPV type match, and 6.1%had all HPV types in clinician-samples also detected in self-samples. In 5.1%, one or more HPVtypes identified in the clinician-samples were missed in self-samples. When using careHPV,the overall agreement between the 2 sampling methods was 95.7% (95% confidence inter val[CI]=95.8–95.6) with good concordance (κ=0.66, 95% CI=0.56–0.76). Nearly 90% of the womenpreferred clinician-sampling over self-sampling, citing greater comfort, ease, and speed. Conclusion: Self-sampled HPV testing using careHPV could be an option for cer vical cancerscreening in Cambodia; however, it requires periodic quality control of handling procedures. In addition, women’s health education regarding the accuracy of self-sampled HPV testingand the importance of follow-up in cases of positive results is needed.

        Transport with minimum time cost and distance remains to be an important research area in intelligent transport systems. Shortest path algorithms are primary methods to address simplified problems, which could not be well applied in high-dimensional real situations. We realized the minimum cost and maximum flow result via classical iterative algorithm based on graph theory, adjacency matrix is well applied to express the relationship between transport nodes, a topological sorting transport map is adopted to verify these approaches.

      • Hypothesis on the Role of Cytoplasmic "Short Base Sequences" in Carcinogenesis

        Cancer is a highly complex medical problem with ramifications for public health throughout the world. Most studies have mainly focused on change in the nuclei as being aetiologically responsible. Few have examined the relationship between the cytoplasm and cancer, despite the fact that research has indicated that the cytoplasmic environment is an important factor for cellular differentiation and that the genetic information provided by the nucleus is entirely dependent on this environment for its expression. Gene mutations may be the result, rather than the cause of carcinogenesis. We submit a new concept - "short base sequences" (50-500 bps, including DNA or RNA sequences) in the cytoplasm which could play an important role in carcinogenesis. This is a new theory to explain the origin of the cancer.

        A Virtual Optical Encryption System Based on Polarization Optics

        In this paper, an encryption system is proposed based on polarization optics. A modulated image at reference plane was obtained by using a virtual magneto-optical modulator, and the field from the reference plane interfered with that from the information plane having the original image to form an encrypted image. This system not only has the advantages of multiple freedom degrees, high security strength and data manipulation in high-dimension as the traditional optical information processing system but also possesses the advantages of data processing flexibility in the computer information process. Numerical experiments prove that images encrypted by using this system have a high level of security, so it is hard for attackers to analyze the original images.
