        Development of Multifunctional Active Film and Its Application in Modified Atmosphere Packaging of Shiitake Mushrooms

        <B>ABSTRACT</B><P>Agar-based films with multiple functions (CO2 absorption, water vapor absorption, and antimicrobial activity) were developed, tested for their properties, and then applied to the packaging of fresh shiitake mushrooms as an insert label. The films were cast from an agar-based aqueous solution containing a dissolving plasticizer (glycerol), a CO2 absorbent (sodium carbonate [SC] alone or a combination of SC and sodium glycinate [SC-SG]), and a volatile antimicrobial agent (carvacrol [CRV]). The agar of the film matrix is designed to serve as a water vapor absorbent. The multifunctional films tended to have poor mechanical properties, with a hard texture and an opaque and yellowish color. The CO2 absorbent, either SC alone or SC-SG, affected CRV retention and release along with the CO2 and water vapor absorption behavior. Both films (SC-CRV and SC-SG-CRV films) showed good inhibitory effects against Pseudomonas fluorescens and Saccharomyces cerevisiae. SC-CRV film had a higher and faster CO2 absorption property, higher retention and extended release of CRV, and lower and slower water vapor absorption and was assessed to be better suited for use in shiitake mushroom packaging. The packaging in which the SC-CRV film with an appropriate amount of CRV was used as an insert label was able to generate the desired atmosphere and less moisture condensation inside the package, producing the best preservation of quality in terms of mushroom color, firmness, flavor score, and microbial counts after 6 days of storage at 10°C. A tailored modified atmosphere packaging system using multifunctional film would be useful in the preservation of CO2-sensitive fresh commodities.</P>

      • Eco approaching at an isolated signalized intersection under partially connected and automated vehicles environment

        <P><B>Abstract</B></P> <P>This research proposed an eco-driving system for an isolated signalized intersection under partially Connected and Automated Vehicles (CAV) environment. This system prioritizes mobility before improving fuel efficiency and optimizes the entire traffic flow by optimizing speed profiles of the connected and automated vehicles. The optimal control problem was solved using Pontryagin’s Minimum Principle. Simulation-based before and after evaluation of the proposed design was conducted. Fuel consumption benefits range from 2.02% to 58.01%. The CO<SUB>2</SUB> emissions benefits range from 1.97% to 33.26%. Throughput benefits are up to 10.80%. The variations are caused by the market penetration rate of connected and automated vehicles and v/c ratio. No adverse effect is observed. Detailed investigation reveals that benefits are significant as long as there is CAV and they grow with CAV’s market penetration rate (MPR) until they level off at about 40% MPR. This indicates that the proposed eco-driving system can be implemented with a low market penetration rate of connected and automated vehicles and could be implemented in a near future. The investigation also reveals that the proposed eco-driving system is able to smooth out the shock wave caused by signal controls and is robust over the impedance from conventional vehicles and randomness of traffic. The proposed system is fast in computation and has great potential for real-time implementation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Is beneficial as long as there is a connected and automated vehicle in the traffic system. </LI> <LI> Is able to smooth out the shock wave caused by signal controls. </LI> <LI> Is robust over the impedance from conventional vehicles and randomness of traffic. </LI> <LI> Is less affected by stochastic human-driven vehicles. </LI> <LI> Can potentially be used in real-time. </LI> </UL> </P>

      • 3D Modeling Of GNSS Vector Tracking Receiver in Urban Area

        3D maps may be treated as a new data source for urban navigation and used to improve positioning accuracy in urban canyons However, 3D digital maps are not available in most cities. This paper proposed a new approach to build a 3D model by using a GNSS VT receiver. The system architecture was described first, and then the method of 3D modeling by analyzing received data was presented. The premise for building the 3D model is signal classification. Analysis of data collected by the VT receiver was described in detail. Using a GNSS signal recorded in a dense urban environment showed the effectiveness of the proposed approach.

        Fast 3D reconstruction method based on UAV photography

        3D reconstruction of urban architecture, land, and roads is an important part of building a “digital city.” Unmanned aerial vehicles (UAVs) are gradually replacing other platforms, such as satellites and aircraft, in geographical image collection; the reason for this is not only lower cost and higher efficiency, but also higher data accuracy and a larger amount of obtained information. Recent 3D reconstruction algorithms have a high degree of automation, but their computation time is long and the reconstruction models may have many voids. This paper decomposes the object into multiple regional parallel reconstructions using the clustering principle, to reduce the computation time and improve the model quality. It is proposed to detect the planar area under low resolution, and then reduce the number of point clouds in the complex area.

        Fe-curcumin Nanozyme-Mediated Immunosuppression and Anti-inflammation in Experimental Autoimmune Uveitis

        Background EAU is an inflammatory disease usually characterized by autoinflammation and autoimmunity and is aggravated by excessive generation of ROS. Conventional hormone therapy often has more adverse effects. It is urgent to find a therapeutic drug with higher efficiency and fewer adverse effects. Methods We developed an Fe-curcumin nanozyme in which natural antioxidants coordinate with Fe3+ to form nanoparticles with excellent solubility for directing anti-inflammatory and ROS scavenging effects to treat EAU. Several experiments were used to detect the characteristics of nanozymes. EAU model rats were used to detect the abilities of decreasing autoinflammation and autoimmunity. PBMCs were used to detect the ability to inhibit cell proliferation. Results Free radical scavenging experiments showed that nanozymes decreased the level of free radicals at low concentrations. In vitro and in vivo experiments revealed that the group treated with Fe-curcumin nanozymes had lower inflammatory reactions and ROS levels than the control group, as reflected by the downregulated levels of several critical inflammatory cytokines, such as IFN-γ, IL-17, and TNF-α; decreased H2O2 release; inhibited proliferation of Th1 and Th17 cells; and alleviated pathological changes in the eye. Importantly, the Fe-curcumin nanozyme was detected in the retina using Prussian blue staining. Additionally, Fe-curcumin nanozyme is noncytotoxic when directing these biological activities. Conclusion This study has demonstrated the feasibility of using the Fe-curcumin nanozyme as a nanodrug to inhibit inflammatory reactions and scavenge ROS in the treatment of EAU, indicating that it may serve as a promising therapeutic agent in clinical treatment.

        The Retention Behavior of Ginsenosides in HPLC and Its Application to Quality Assessment of Radix Ginseng

        This study systematically investigated the retention behavior of seven neutral ginsenosides $Rg_1$, Re, Rf, $Rb_1$, $Rb_2$, Rc, Rd, and an acidic ginsenoside $R_0$, the major pharmacologically active components of Radix Ginseng with RP-HPLC. The effects of solvent, pH value, ionic strength of the mobile phase, and column temperature were investigated using an octadecylsiloxane-bonded silica gel column. Based on the ginsenosides' retention characteristics, the concentration of acetonitrile and the gradient of the mobile phase needed to maintain the baseline separation of the major neutral ginsenosides in Radix Ginseng were theoretically predicted. Furthermore, the ionic strength of mobile-phase necessary to achieve good resolution of the neutral ginsenosides and acidic ginsenosides was carefully investigated. According to the results of the quantitative analysis of ginsenosides in eight batches of ginseng samples from different sources, the developed HPLC technique may be a valuable tool for the quality assessment of Radix Ginseng.

        Genetic parameter estimation for reproductive traits in QingYu pigs and comparison of carcass and meat quality traits to Berkshire×QingYu crossbred pigs

        Objective: The QingYu pig is well known for its excellent meat quality attributes in Sichuan province, China. In order to improve its production efficiency, the determination of genetic factors contributing to quantifiable economic traits of livestock is important. Moreover, the cross-breeding of QingYu pigs with western breeds possessing strong growth attributes is an efficient way to improve the performance of this breed. Methods: Here, the genetic parameters of several important reproductive traits of QingYu pigs were estimated, include total number born (TNB), number born alive, litter birth weight, individual birth weight, number of piglets weaned, litter weaning weight, and individual weaning weight. The data was analyzed using the ASReml 3.0 software (NSW Inc., Sydney, Australia). Furthermore, the effects of crossing Berkshire with QingYu (BQ) pigs on carcass and meat quality traits, as well as the effects of slaughter weight on carcass and meat quality of BQ were characterized. Results: QingYu pigs exhibited superior reproductive traits. The TNB available to QingYu pigs was more than 8 per parity. The observed repeatability of the reproductive traits of the QingYu pigs was between 0.10 and 0.23. The significantly correlated genetic and phenotypic of reproduction traits were consistent. Interestingly, the BQ pigs exhibited improved carcass quality, with a significant increase in loin muscle area, lean percentage and reduction in sebum percentage. As a result, BQ had higher L45min, lower cooking scores, and lower drip loss. In addition, the loin muscle area, body length, and sebum percentage were significantly higher in 90 and 100 kg animals. Cooking loss showed a significant increase at 80 kg, and marbling increased significantly from 90 kg. Conclusion: The results of this study suggest that QingYu pigs exhibit excellent reproductive properties and heritability of these traits. Crossing with Berkshire is an efficient strategy to improve the carcass and meat quality of QingYu pigs for commercial operations. Furthermore, it appears as though the optimal slaughter weight of BQ pigs is at approximately 90 kg.

        WTAP-mediated m6A modification of FRZB triggers the inflammatory response via the Wnt signaling pathway in osteoarthritis

        Osteoarthritis (OA) is the most common form of arthritis. However, the exact pathogenesis remains unclear. Emerging evidence shows that N6-methyladenosine (m6A) modification may have an important role in OA pathogenesis. This study aimed to investigate the role of m6A writers and the underlying mechanisms in osteoarthritic cartilage. Among m6A methyltransferases, Wilms tumor 1-associated protein (WTAP) expression most significantly differed in clinical osteoarthritic cartilage. WTAP regulated extracellular matrix (ECM) degradation, inflammation and antioxidation in human chondrocytes. Mechanistically, the m6A modification and relative downstream targets in osteoarthritic cartilage were assessed by methylated RNA immunoprecipitation sequencing (MeRIP-seq) and RNA sequencing, which indicated that the expression of frizzled-related protein (FRZB), a secreted Wnt antagonist, was abnormally decreased and accompanied by high m6A modification in osteoarthritic cartilage. In vitro dysregulated WTAP had positive effects on β-catenin expression by targeting FRZB, which finally contributed to the cartilage injury phenotype in chondrocytes. Intra-articular injection of adeno-associated virus-WTAP alleviated OA progression in a mouse model, while this protective effect could be reversed by the application of a Wnt/β-catenin activator. In summary, this study revealed that WTAP-dependent RNA m6A modification contributed to Wnt/β-catenin pathway activation and OA progression through post-transcriptional regulation of FRZB mRNA, thus providing a potentially effective therapeutic strategy for OA treatment.
