      • An Adaptive Cellular Genetic Algorithm Based on Selection Strategy for Test Sheet Generation

        Intelligent test sheet generation is a multi-objective constrained optimization problem. Genetic algorithm based on groups search strategy can provide a better solution for multi-objective optimization. Traditional genetic algorithm in test sheet generation process has many drawbacks, such as poor convergence, low fitness and high exposure times. To solve these problems, this paper proposes an adaptive cellular genetic algorithm based on selection strategy. Selection strategy can adaptively determine candidate test items set and the conceptual granularities according to the desired concept scope. Then, a new cellular population is formed by candidate test items. After evolution by the rule, genetic algorithms are executed. The experimental results show that the proposed algorithm gets rid of tests that do not meet the requirements which can reduce knowledge related errors, lower the exposure of tests, and increase the possibility of escape from local optima. In general, the algorithm proposed in this paper effectively improves the convergence speed as well as generates test papers more in line with people's demands.

        Energy Efficiency of Distributed Massive MIMO Systems

        In this paper, we investigate energy efficiency (EE) of thetraditional co-located and the distributed massive multiple-inputmultiple-output (MIMO) systems. First, we derive an approximateEE expression for both the idealistic and the realistic power consumptionmodels. Then an optimal energy-efficient remote accessunit (RAU) selection algorithm based on the distance between themobile stations (MSs) and the RAUs are developed to maximizethe EE for the downlink distributedmassive MIMO systems underthe realistic power consumption model. Numerical results showthat the EE of the distributedmassiveMIMOsystems is larger thanthe co-locatedmassiveMIMOsystems under both the idealistic andrealistic power consumptionmodels, and the optimal EE can be obtainedby the developed energy-efficient RAU selection algorithm.

        Potential application of ginseng in sepsis: Applications of ginseng in sepsis

        Sepsis and septic shock affect millions of people worldwide each year with high clinical mortality rates. At present, basic research on sepsis has emerged in an endless stream, but there are few effective clinical translation results. Ginseng, a medicinal and edible representative of Araliaceae plants, contains a variety of biologically active compounds including ginsenosides, alkaloids, glycosides, polysaccharides, and polypeptides. Neuromodulation, anticancer activity, blood lipid regulation, and antithrombotic activity have been linked to ginseng treatment. At present, basic and clinical research have suggested various applications of ginseng in sepsis. In view of the different effects of various ginseng components on the pathogenesis of sepsis, and in order to further understand and develop the possible value of ginseng in sepsis, this manuscript reviews the application of various components of ginseng in the treatment of sepsis in recent years.

        An Analytical Model and Its Application in Reliability Analysis of Formation Failure during Hydrate Production in Deep-sea Areas

        Methane hydrates (MHs) have potential economic and environmental significance. However, due to the sharp reduction in the mechanical properties of methane hydrate-bearing sediments (MHBS) caused by hydrate dissociation, the risk of formation failure in marine MHs exploitation is higher than that in conventional oil and gas exploitation. Formation failure in marine hydrate production probably leads to serious sand production or subsequent formation instability. A simplified and efficient analytical model is proposed at first for prediction of formation failure as well as the displacement around the wellbore during hydrate production, and then the reliability analysis for formation failure is performed by the Advanced First Order Second Moment Method based on the analytical solutions. This model considers the key factors affecting the formation failure, including 1) the partial coupling of multiple fields (the influence of pore pressure on mechanical field and the influence of hydrate dissociation on pore pressure, temperature and mechanical field), 2) the effect of gas/water or heat absorption caused by hydrate dissociation on pore pressure or temperature, 3) the change of mechanical properties of formation induced by hydrate dissociation and 4) the casing-formation interaction. The analytical solutions are verified and validated by numerical models. According to the analysis, the casing is sufficiently safe in the cross section due to its high stiffness, while the formation would be failure at the wellbore or dissociated front due to the stiffness/strength deterioration induced by hydrate dissociation. Hydrate dissociation leads to an increase in formation failure probability by approximately 20%. The probability of formation failure at the wellbore is about 30% higher than that at the dissociated front. The uncertainties in the internal friction angle and elastic modulus ratio have the greatest effect on formation failure probability.

        The molecular mechanism of propionate-regulating gluconeogenesis in bovine hepatocytes

        Objective: Cows that are nursing get around 80% of their glucose from liver gluconeogenesis. Propionate, a significant precursor of liver gluconeogenesis, can regulate the key genes involved in hepatic gluconeogenesis expression, but its precise effects on the activity of enzymes have not yet been fully elucidated. Therefore, the aim of this study was to investigate the effects of propionate on the activity, gene expression, and protein abundance of the key enzymes involved in the gluconeogenesis of dairy cow hepatocytes. Methods: The hepatocytes were cultured and treated with various concentrations of sodium propionate (0, 1.25, 2.50, 3.75, and 5.00 mM) for 12 h. Glucose content in the culture media was determined by an enzymatic coloring method. The activities of gluconeogenesis related enzymes were determined by enzyme linked immunosorbent assay kits, and the levels of gene expression and protein abundance of the enzymes were detected by real-time quantitative polymerase chain reaction and Western blot, respectively. Results: Propionate supplementation considerably increased the amount of glucose in the culture medium compared to the control (p<0.05); while there was no discernible difference among the various treatment concentrations (p>0.05). The activities of cytoplasmic phosphoenolpyruvate carboxylase (PEPCK1), mitochondrial phosphoenolpyruvate carboxylase (PEPCK2), pyruvate carboxylase (PC), and glucose-6-phosphatase (G6PC) were increased with the addition of 2.50 and 3.75 mM propionate; the gene expressions and protein abundances of PEPCK1, PEPCK2, PC, and G6PC were increased by 3.75 mM propionate addition. Conclusion: Propionate encouraged glucose synthesis in bovine hepatocytes, and 3.75 mM propionate directly increased the activities, gene expressions and protein abundances of PC, PEPCK1, PEPCK2, and G6PC in bovine hepatocytes, providing a theoretical basis of propionate-regulating gluconeogenesis in bovine hepatocytes.

        Pyrotinib Combined with Vinorelbine in Patients with Previously Treated HER2-Positive Metastatic Breast Cancer: A Multicenter, Single-arm, Prospective Study

        Purpose This study aims to evaluate the efficacy and safety of a new combination treatment of vinorelbine and pyrotinib in human epidermal growth factor receptor 2 (HER2)–positive metastatic breast cancer (MBC) and provide higher level evidence for clinical practice. Materials and Methods This was a prospective, single-arm, phase 2 trial conducted at three institutions in China. Patients with HER2-positive MBC, who had previously been treated with trastuzumab plus a taxane or trastuzumab plus pertuzumab combined with a chemotherapeutic agent, were enrolled between March 2020 and December 2021. All patients received pyrotinib 400 mg orally once daily plus vinorelbine 25 mg/m2 intravenously or 60-80 mg/m2 orally on day 1 and day 8 of 21-day cycle. The primary endpoint was progression-free survival (PFS), and the secondary endpoints included the objective response rate (ORR), disease control rate (DCR), overall survival, and safety. Results A total of 39 patients were enrolled. All patients had been pretreated with trastuzumab and 23.1% (n=9) of them had accepted trastuzumab plus pertuzumab. The median follow-up time was 16.3 months (95% confidence interval [CI], 5.3 to 27.2), and the median PFS was 6.4 months (95% CI, 4.0 to 8.8). The ORR was 43.6% (95% CI, 27.8% to 60.4%) and the DCR was 84.6% (95% CI, 69.5% to 94.1%). The median PFS of patients with versus without prior pertuzumab treatment was 4.6 and 8.3 months (p=0.017). The most common grade 3/4 adverse events were diarrhea (28.2%), neutrophil count decreased (15.4%), white blood cell count decreased (7.7%), vomiting (5.1%), and anemia (2.6%). Conclusion Pyrotinib plus vinorelbine showed promising efficacy and tolerable toxicity as second-line treatment in patients with HER2-positive MBC.
