        Association of polymorphisms in bone morphogenetic protein receptor-1B gene exon-9 with litter size in Dorset, Mongolian, and Small Tail Han ewes

        Objective: The present study was to investigate the association of polymorphisms in exon-9 of the bone morphogenetic protein receptor-1B (BMPR-1B) gene (C864T) with litter size in 240 Dorset, 232 Mongolian, and 124 Small Tail Han ewes. Methods: Blood samples were collected from 596 ewes and genomic DNA was extracted using the phenol: chloroform extraction method. The 304-bp amplified polymerase chain reaction product was analyzed for polymorphism by single-strand conformation polymorphism method. The genotypic frequency and allele frequency of BMPR-1B gene exon-9 were computed after sequence alignment. The χ2 independence test was used to analyze the association of genotypic frequency and litter size traits with in each ewe breed, where the phenotype was directly treated as category. Results: The results indicated two different banding patterns AA and AB for this fragment, with the most frequent genotype and allele of AA and A. Calculated Chi-square test for BMPR-1B gene exon-9 was found to be more than that of p value at the 5% level of significance, indicating that the population under study was in Hardy-Weinberg equilibrium for all ewes. The χ2 independence test analyses indicated litter size differences between genotypes was not the same for each breed. The 304-bp nucleotide sequence was subjected to BLAST analysis, and the C864T mutation significantly affected litter size in singletons, twins and multiples. The heterozygosity in exon-9 of BMPR-1B gene could increase litter size for all the studied ewes. Conclusion: Consequently, it appears that the polymorphism BMPR-1B gene exon-9 detected in this study may have potential use in marker assisted selection for litter size in Dorset, Mongolian, and Small Tail Han ewes.

      • MLH1 Polymorphisms and Cancer risk: a Meta-analysis Based on 33 Case-control Studies

        Objective: Cumulative evidence suggests that MLH1, the key component in the mismatch pathway, plays an important role in human cancers. Two potential functional polymorphisms (-93G>A and I219V) of MLH1 have been implicated in cancer risk. The aim of this meta-analysis was to summarize the evidence for associations. Methods: Eligible studies were identified by searching the electronic literature PubMed, ScienceDirect and Embase databases for relevant reports and bibliographies. Studies were included if of case-control design investigating MLH1 polymorphisms (-93G>A and I219V) and cancer risk with sufficient raw data for analysis. Odds ratios (OR) and 95% confidence intervals (95% CI) were used to evaluate the strength of associations. Results: Our meta-analysis from 33 published case-control studies showed the variant A allele of -93G>A polymorphism to be associated with increased risk in all genetic models (AA vs. GG: OR = 1.22, 95% CI: 1.03-1.44), especially among non-Asians (AA vs. GG: OR = 1.28, 95% CI: 1.04-1.58). For the I219V polymorphism, however, there was no main effect associated with overall cancer risk in any genetic model. Conclusions: The meta-analysis suggested that the MLH1 -93G>A polymorphism may be a biomarker of cancer susceptibility. Large sample association studies and assessment of gene-to-gene as well as gene-to-environment interactions are required to confirm these findings.

      • Up-regulating of RASD1 and Apoptosis of DU-145 Human Prostate Cancer Cells Induced by Formononetin in Vitro

        Prostate cancer is one of the most prevalent malignant cancers in men. The isoflavone formononetin is a main active component of red clover plants. In the present study, we assessed the effect of formononetin on human prostate cancer DU-145 cells in vitro, and elucidated posssible mechanisms. DU-145 cells were treated with different concentrations of formononetin and cell proliferation was assessed by MTT assay, cell apoptosis by Hoechst 33258 and flow cytometry, and protein levels of RASD1, Bcl-2 and Bax by Western blotting. The results showed that formononetin inhibited the proliferation of DU-145 cells in a dose-dependent manner. DU-145 cells treated with different concentrations of formononetin displayed obvious morphological changes of apoptosis under fluorescence microscopy. In addition, formononetin increased the proportion of early apoptotic DU-145 cells, down-regulated the protein levels of Bcl-2 and up-regulated those of RASD1 and Bax. The level of RASD1 reached its maximum at 48h post-treatment, and rapidly decreased thereafter. Together, we present evidence that formononetin triggered cell apoptosis through the mitochondrial apoptotic pathway by up-regulating RASD1.

        Study and Design of High Strength and Low Heat Generation Polymer Composites by Molecular Dynamics Simulation

        The molecular dynamics simulation was used to study and design a polymer composites system with excellent mechanical strength and less heat generation in a dynamic process. A series of system factors such as filler loading, surface modification onto filler, and network of cross-linking filler particles on mechanical and heat generation of polymer composites are systematically considered. It is found that the surface grafting onto fillers can restrain the heat generation of polymer composite in the dynamic process, while it shows less effect on the mechanical property. A network of cross-linking filler particles can be fabricated by a combination of grafting chains onto fillers. By filling such a network into the polymer, the mechanical and heat generation properties of polymer composites are significantly improved. Simulation results can help experimental fabrication of polymer composites with excellent mechanical and heat generation properties.

        Study on Crystallization Kinetics of Dynamically-Vulcanized PP/EPDM Blends

        Two types of β nucleating agents (β-NAs), aryl dicarboxylic acid amide (TMB-5) and diphenyl phthalate diamine (NT-C), were adopted to modify the polypropylene (PP)/ethylene propylene diene monomer (EPDM) blends, which were prepared by dynamic-vulcanization technology. Wide angle X-ray diffraction (WAXD) and differential scanning calorimetry (DSC) were used to study the crystallization kinetics of PP. Our results showed that the addition of β-NAs can considerably increase the crystallization temperature, and significantly decrease the spherulite size of β-PP (L300). The Jeziorny analysis showed there were ~82% and ~89% of relative crystallinity generated from the primary crystallization in the composites containing TMB-5 and NT-C, respectively. The crystallization half time (t0.5) showed that NT-C improved the overall crystallization rate more effectively than TMB-5. In addition, the peaks of the relative crystallization rate curves were shifted towards higher temperature by 14 and 9℃ with the addition of TMB-5 and NT-C, respectively.

        A Novel Kinetic Analysis of Crystallization of Polypropylene (PP) in Dynamically-vulcanized PP/Ethylene-propylene-diene Rubber (EPDM) Blends Using an In-situ Measurement Technique

        The effects of cooling medium temperatures and plastic/rubber ratios on solidification and crystallization kinetics of dynamically-vulcanized polypropylene/ethylene-propylene-diene rubber (PP/EPDM) blends were investigated with the aid of an in-situ measurement technique. The cooling medium temperature heavily influenced the solidification kinetics primarily due to a combination of latent heat liberated from the molten polymer and the heat transferred away via the metallic wall during the cooling period. Interestingly, the parameter C in three-parameter model was not only affected by the material properties, but also by the cooling condition, different from the previous literature. The crystallization kinetics analysis indicated that the effect of EPDM in the blends consisted of both nucleation-promoting effect (low EPDM loading) and steric effect (higher EPDM loading). The present kinetic analysis may be helpful to further studies on improving the product performances for industrial applications.

        In situ Copolymerized Toughened Polymethyl Methacrylate (PMMA) with Highly Transparency for Support Film of Polarizers

        In this study, a series of poly(methyl methacrylate) (PMMA) copolymer films were prepared via solutionpolymerization of methyl methacrylate (MMA) with butyl acrylate (BA) and lauryl methacrylate (LMA) as monomers. Mechanical properties, hydrophobic properties, and optical properties of the films were intensively investigated. The rheological results showed that the fluidity of the copolymer was considerably enhanced. When the monomer ratio ofMMA:BA:LMA was 100:30:10, the copolymer film S4 showed the best overall performance with perfect optical transparency maintained. The results of the dynamic mechanical and thermal analysis suggested that the glass transition temperature (Tg) moved towards lower temperature, with enhanced ductility of the PMMA films. A large number of yieldfolds and crazes appeared on the cross-sectional surface of copolymer films through morphological observations, displaying the obvious characteristics of toughness fracture and obeying the energy dissipation mechanism of cracks shearband. The present study provided a facile way of preparing PMMA films with high toughness and light transmittanceby appropriate selection of the monomers, which will be of practical significance for further studies on the replacementof triacetyl cellulose as a support film of polarizers.

        Evaluation of Non-isothermal Melt Crystallization Kinetics of the Poly(lactic acid)/Graphene Composites under Variant Cooling Circumstances via Solidification Analysis Approach

        The poly(lactic acid) (PLA)/graphene (GN) composites were prepared by solution blending. An in situ measurement technique was adopted to examine the non-isothermal crystallization of PLA/GN composites, which was different from the typical circumstances at constant cooling rate in DSC. The rheological properties of the samples were investigated, and the Carreau-A model was applied to non-linearly fit the rheological data. The non-isothermal crystallization kinetics and solidification behaviors at variant cooling rates were investigated using a modified version of Avrami equation by Jeziorny coupled with three-parameter model (TPM) and four-parameter model (FPM). Our findings showed that the viscosity of the PLA/GN composites dropped with increasing shear rate or GN content. Crystallization kinetic analysis suggested that GN served as an effective nucleating agent for PLA under non-isothermal melt crystallization conditions with variant cooling rate.

        Non-isothermal Crystallization Behavior, Rheological and Thermal Conductive Properties of Recycled Polyethylene Terephthalate/Polyethylene Blends

        In this study, we prepared series of recycled polyethylene terephthalate (RPET)/polyethylene (PE) blends using melt extrusion. The effect of RPET content on crystallization behavior and thermal conductive properties of the resultant blends were investigated using differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), etc. RPET was found to exert nucleating effect on the melt crystallization of PE. The Agari model presented fairly reasonable prediction of thermal conductivity as a function of RPET loading. The melt cooling process was predicted with an enthalpy transformation method (ETM), which is a well-established mean of evaluating the instantaneous heat conduction of crystalline polymers/composites, and the obtained curves were consistent with our experimental results. Besides, a four-parameter model (FPM) was adopted coupled with an in-situ temperature measurement in order to further disclose the solidification and crystallization kinetics of PE in the presence of RPET in the blends.
