        Nomogram for predicting overall survival in children with neuroblastoma based on SEER database

        Purpose: This study was performed to establish and validate a nomogram for predicting the overall survival in children with neuroblastoma. Methods: The latest clinical data of neuroblastoma in Surveillance, Epidemiology, and End Results (SEER) database was extracted from 2000 to 2016. The cases included were randomly divided into training and validation cohorts. The survival curves were drawn with a Kaplan-Meier estimator to investigate the influences of certain single factors on overall survival. Also, least absolute shrinkage and selection operator regression was applied to further select the prognostic variables for neuroblastoma. Additionally, receiver operating characteristic (ROC) curves and calibration curves were used to evaluate the accuracy of the nomogram. Results: In total, 1,262 patients were collected and 8 independent prognostic factors were achieved, including patients’ age, sex, race, tumor grade, radiotherapy, chemotherapy, tumor site, and tumor size. Then we constructed a nomogram by using the data of the training cohort with 886 cases. Subsequently, the nomogram was validated internally and externally with 886 and 376 cases, respectively. The internal validation revealed that the area under the curves (AUC) of ROC curves of 1-, 3-, and 5-year overall survival were 0.69, 0.78, and 0.81, respectively. Accordingly, the external validation also showed that the AUC of 1-, 3-, and 5-year overall survival were all ≥0.69. Both methods of validation demonstrated that the predictive calibration curves were consistent with standard curves. Conclusion: The nomogram possess the potential to be a new tool in predicting the survival rate of neuroblastoma patients.

        Experimental Study and Parameter Analysis of L-shaped Composite Column under Axial Loading

        A special-shaped column composed of concrete-filled steel tubes (SCFST column) is experimentally investigated in this paper. The SCFST column is composed of several small-sized concrete-filled square tubular columns (mono-columns) connected by connection plates. This kind of column is proposed because it can increase the usable space in residential buildings. The axial loading behavior of L-shaped SCFST columns is experimentally investigated. The failure mode, strain distribution, cooperation of mono-columns, bearing capacity, and the effect of a concrete core on column behavior were tested. As concluded, the SCFST column can be regarded as a useful kind of column. A finite element analysis model was proposed according to the experimental results, and the influence of connection plate size on bearing capacity was studied using the finite element model. Finally, proposed were the optimal width and thickness of connection plates for improving the design of the SCFST column.

        <P><B>Abstract</B></P> <P>Peroxynitrite (ONOO<SUP>−</SUP>) is one of the endogenous reactive oxygen species (ROS), which causes damage to a wide array of molecular components in the cells, including DNA and proteins, owing to its high oxidizing as well as nitrating properties. However, the precise pathogenic roles played by this substance in biological systems have not yet been elucidated completely owing to its short lifetime, high reactivity, low concentration and elusive nature in the <I>in vivo</I> applications. Thus, the development of more sensitive and selective techniques for detecting ONOO<SUP>−</SUP>, with high biocompatibilities, sensitivities, and site-specificities, is a significant goal.         Microstructures and Thermoelectric Properties of p-Type BixSb2-xTe3Thin Films with Various Compositions

        In the present study, p-type BixSb2-xTe3 thermoelectric thin films with various chemical compositions were fabricated by a magnetron co-sputtering process. The influence of Bi content (x) on the microstructures and thermoelectric properties of BixSb2-xTe3 films was investigated. It was found that the grain size of nanocrystalline BixSb2-xTe3 films decreased when increasing the Bi content from x = 0 to x = 0.57, which resulted in a concurrent decrease in the carrier mobility. Similarly, carrier concentration decreased with increasing Bi content,which caused an increase in the Seebeck coefficient. The largest room-temperature power factor reached 31.3 μWK−2cm−1 for the film with x = 0.45.

        Non-isothermal Crystallization Behavior, Rheological and Thermal Conductive Properties of Recycled Polyethylene Terephthalate/Polyethylene Blends

        In this study, we prepared series of recycled polyethylene terephthalate (RPET)/polyethylene (PE) blends using melt extrusion. The effect of RPET content on crystallization behavior and thermal conductive properties of the resultant blends were investigated using differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), etc. RPET was found to exert nucleating effect on the melt crystallization of PE. The Agari model presented fairly reasonable prediction of thermal conductivity as a function of RPET loading. The melt cooling process was predicted with an enthalpy transformation method (ETM), which is a well-established mean of evaluating the instantaneous heat conduction of crystalline polymers/composites, and the obtained curves were consistent with our experimental results. Besides, a four-parameter model (FPM) was adopted coupled with an in-situ temperature measurement in order to further disclose the solidification and crystallization kinetics of PE in the presence of RPET in the blends.

      • Expression and Functional Role of ALDH1 in Cervical Carcinoma Cells

        Tumor formation and growth is dictated by a very small number of tumor cells, called cancer stem cells, which are capable of self-renewal. The genesis of cancer stem cells and their resistance to conventional chemotherapy and radiotherapy via mechanisms such as multidrug resistance, quiescence, enhanced DNA repair abilities and anti-apoptotic mechanisms, make it imperative to develop methods to identify and use these cells as diagnostic or therapeutic targets. Aldehyde dehydrogenase 1 (ALDH1) is used as a cancer stem cell marker. In this study, we evaluated ALDH1 expression in CaSki, HeLa and SiHa cervical cancer cells using the Aldefluor method to isolate ALDH1-positive cells. We showed that higher ALDH1 expression correlated with significantly higher rates of cell proliferation, microsphere formation and migration. We also could demonstrate that SiHa-ALDH1-positive cells were significantly more tumorigenic compared to SiHa-ALDH1-negative cells. Similarly, SiHa cells overexpressing ALDH1 were significantly more tumorigenic and showed higher rates of cell proliferation and migration compared to SiHa cells where ALDH1 expression was knocked down using a lentivirus vector. Our data suggested that ALDH1 is a marker of cervical cancer stem cells and expand our understanding of its functional role.

      • Pretreatment Thrombocytosis as a Prognostic Factor in Women with Gynecologic Malignancies: a Meta-analysis

        Background: This study was performed to analyze the prognostic implications of pretreatment or preoperative thrombocytosis in women with gynecologic malignancies. Material and Methods: We surveyed 2 medical databases, PubMed and EMBASE, to identified all relevant studies. A total of 14 (n=3,490) that evaluated the link between thrombocytosis and 5-year survival were included. REVMAN version 5.1 was used for our analysis and publication bias was evaluated using the Begg's funnel plot and tested by STATA 11.0. Risk ratios (RRs) with 95% confidence intervals (CIs) generated by the random effect model were used to assess the strength of any association. Results: 709(20.3%) of the 3,490 patients exhibited thrombocytosis (platelet counts > $400{\times}10^9/L$) at primary diagnosis, and their mortality was 1.62-fold higher compared with the others (RR=1.62, 95%CI=[1.28-2.05], p<0.0001). Thrombocytosis failed to have a stronger effect on the survival of advanced patients of stages III to IV in our study (n=478, RR=1.29, 95% CI=[1.13-1.48], p=0.0003), nor in women with cervical cancer in stage IB (n=1371, RR=1.73, 95% CI=[1.71-2.58], p=0.007). In addition, when adjusted for different carcinoma, it was associated with worse prognosis for all except the ones with vulvar cancer (n=201, RR=0.43, 95% CI=[0.14-1.29], p=0.13). Conclusions: This meta-analysis indicated that thrombocytosis might be associated with a worse prognosis for patients with gynecologic malignancies but without specificity or sensitivity for the ones in advanced stage. When adjusted for different gynecologic malignancies, it showed a significant effect on survival of all except vulvar cancers.

        Establishment of a novel myocarditis mouse model based on cyclosporine A

        Background: Myocarditis is a myocardial injury that can easily cause adolescent death. Traditional research models of animal invasion with viral components, lipopolysaccharide (LPS) or porcine myocardial myosin, among others, have the shortcomings of potential biological safety hazards and high animal mortality. Objective: To explore the construction of a novel myocarditis model with cyclosporine A and the potential genes and pathways associated with it. Methods: BALB/c mice were used in this study, and cyclosporin A and LPS were injected into the peritoneal cavity of mice. The successful establishment of the model was assessed by detecting serum myocardial injury markers and inflammatory factors levels, HE, IHC staining, and RT-qPCR methods. Key genes were obtained using the GSE35182 dataset from the GEO database and validated with the RT-qPCR method. Results: We found that a large number of inflammatory cells infiltrated the myocardium of mice in each group of Cyclosporin A constructed model, while the expression of inflammatory factor indicators was increased, and this model has the characteristics of high degree of local inflammation in myocardial tissue, low mortality, and safe and non-toxic treatment. Using GSE35182 data, we selected 18 Hub genes and validated Hub genes in myocardial tissue with RT-qPCR and found that multiple signaling pathways such as Toll-likereceptor signaling pathway(TLRs), Rap1 signal pathway(Rap1), and Chemokine signaling pathway may be involved in the development of myocarditis. Conclusion: Cyclosporin A can construct a new myocarditis model, and TLRs, Chemokines and Rap1 signaling pathways may be the core pathways of myocarditis.

        Inhibition of BETs prevents heat shock-induced cell death via upregulating HSPs in SV40 large T antigen transfected cells

        Background: Heat shock response is a protected mechanism against environmental changes for the organism, which must be tightly regulated. Bromodomain and extra terminal-containing protein family (BETs) regulate numerous gene expression in many physiological and pathological conditions, including viral infection. SV40 is considered as a highly human disease-associated virus. Objective: We aimed to explore whether BETs play a role in heat shock in SV40 large T antigen transfected cells. Methods: SV40LTA was transfected in HeLa cells using the Lipofectamine 8000. BETs inhibitor JQ1 and I-BET-762 was employed to treat transfected cells and HEK-293 T cells. Heat shock treatment was performed to determine the effect of JQ1 and I-BET-762 on these cells. Western blot and quantitative RT-PCR were carried out to assess the expression of HSP70 and other HSPs. Results: We found that inhibition of BETs by JQ1 and I-BET-762 protects cells from heat shock-induced death in HEK293T cells. Both JQ1 and I-BET-762 induce the expression of HSPs and HSF1 in HEK-293 T cells. However, neither JQ1 nor I-BET-762 fail to induce the expression of HSPs in either HeLa or HBL-1 cells. When SV40 large T antigen was transfected into HeLa cells, the induction of HSP70 expressing and the protection of heat shock-induced cell death are reproduced by JQ1 and IBET treatment in these transfected cells. Conclusions: Inhibition of BETs by JQ1 and I-BET-762 prevents heat shock-induced cell death via upregulating HSPs in SV40 large T antigen transfected cells. Our data indicate a novel function of BETs in SV40 large T antigen transformed cells, affecting HSPs and HSF1 as well as its function on heat shock response.
