        Anti-cariogenic Characteristics of Rubusoside

        Streptococcus mutans plays an important role in the development of dental caries in humans by synthesizing adhesive insoluble glucans from sucrose by mutansucrase activity. To explore the anti-cariogenic characteristics of rubusoside (Ru), a natural sweetener component in Rubus suavissimus S. Lee (Rosaceae), we investigated the inhibitory effect of Ru against the activity of mutansucrase and the growth of Streptococcus mutans. Ru (50 mM) showed 97% inhibitory activity against 0.1 U/mL mutansucrase of S. mutans with 500 mM sucrose. It showed competitive inhibition with a Ki value of 1.1 ± 0.2 mM and IC50 of 2.3 mM. Its inhibition activity was due to hydrophobic and hydrogen bonding interactions based on molecular docking analysis. Ru inhibited the growth of S. mutans as a bacteriostatic agent, with MIC and MBC values of 6 mM and 8 mM, respectively. In addition, Ru showed synergistic anti-bacterial activity when it was combined with curcumin. Therefore, Ru is a natural anti-cariogenic agent with antimutansucrase activity and antimicrobial activity against S. mutans.

        A newly developed capture-based sequencing panel for genomic assay of lung cancer

        Background The increase in genetic alterations targeted by specific chemotherapy in lung cancer has led to the need for universal use of more comprehensive genetic testing, which has highlighted the development of a lung cancer diagnostic panel using next-generation sequencing. Objective We developed a hybridization capture-based massively parallel sequencing assay named Friendly, Integrated, Research-based, Smart and Trustworthy (FIRST)-lung cancer panel (LCP), and evaluated its performance. Methods FIRST-LCP comprises 64 lung cancer-related genes to test for various kinds of genetic alterations including single nucleotide variations (SNVs), insertions and deletions (indels), copy number variations (CNVs), and structural variations. To assess the performance of FIRST-LCP, we compiled test sets using HapMap samples or tumor cell lines with disclosed genetic information, and also tested our clinical lung cancer samples whose genetic alterations were known by conventional methods. Results FIRST-LCP accomplished high sensitivity (99.4%) and specificity (100%) of the detection of SNVs. High precision was also achieved, with intra- or inter-run concordance rate of 0.99, respectively. FIRST-LCP detected indels and CNVs close to the expected allele frequency and magnitude, respectively. Tests with samples from lung cancer patients also identified all SNVs, indels and fusions. Conclusion Based on the current state of the art, continuous application of the panel design and analysis pipeline following up-to-date knowledge could ensure precision medicine for lung cancer patients.

        Genomic characterization of clonal evolution during oropharyngeal carcinogenesis driven by human papillomavirus 16

        Secondary prevention via earlier detection would afford the greatest chance for a cure in premalignant lesions. We investigated the exomic profiles of non-malignant and malignant changes in head and neck squamous cell carcinoma (HNSCC) and the genomic blueprint of human papillomavirus (HPV)-driven carcinogenesis in oropharyngeal squamous cell carcinoma (OPSCC). Whole-exome (WES) and whole-genome (WGS) sequencing were performed on peripheral blood and adjacent non-tumor and tumor specimens obtained from eight Korean HNSCC patients from 2013 to 2015. Next-generation sequencing yielded an average coverage of 94.3× for WES and 35.3× for WGS. In comparative genomic analysis of non-tumor and tumor tissue pairs, we were unable to identify common cancer-associated early mutations and copy number alterations (CNA) except in one pair. Interestingly, in this case, we observed that non-tumor tonsillar crypts adjacent to HPV-positive OPSCC appeared normal under a microscope; however, this tissue also showed weak p16 expression. WGS revealed the infection and integration of high-risk type HPV16 in this tissue as well as in the matched tumor. Furthermore, WES identified shared and tumor-specific genomic alterations for this pair. Clonal analysis enabled us to infer the process by which this transitional crypt epithelium (TrCE) evolved into a tumor; this evolution was accompanied by the subsequent accumulation of genomic alterations, including an ERBB3 mutation and large-scale CNAs, such as 3q27-qter amplification and 9p deletion. We suggest that HPV16-driven OPSCC carcinogenesis is a stepwise evolutionary process that is consistent with a multistep carcinogenesis model. Our results highlight the carcinogenic changes driven by HPV16 infection and provide a basis for the secondary prevention of OPSCC. [BMB Reports 2018; 51(11): 584-589]

        Roles of Vocabulary and Grammar Knowledge in L2 Reading Comprehension

        The present study sought to investigate the roles of vocabulary and grammar knowledge in second language reading in the case of Korean adolescent learners of English. To this end, 200 high school learners were asked to take three tests: a vocabulary knowledge test, a grammar knowledge test, and a reading comprehension test. The Korean version of the Vocabulary Levels Test was employed to measure the learners’ vocabulary knowledge, while grammar knowledge was measured with a 50-item multiple-choice grammar test designed by the researchers. The reading comprehension test consisted of four passages, which were each followed by five comprehension questions. The results of statistical analyses suggested that while the intersection of vocabulary and grammar accounted for most of the expected reading variance, unique properties of each of the two variables also played individual roles in explaining L2 reading comprehension. Though vocabulary seemed to have a slightly larger effect, vocabulary and grammar had similar unique effects on L2 reading. Therefore, it can be concluded that both breadth of vocabulary and grammar knowledge have equal positive effects on the reading of English passages by Korean adolescent learners of English. Theoretical implications and directions for further studies are discussed.

      • A genome-wide by PM<sub>10</sub> interaction study identifies novel loci for lung function near <i>BICD1</i> and <i>IL1RN-IL1F10</i> genes in Korean adults

        <P><B>Abstract</B></P> <P>Although several genome-wide interaction studies (GWIS) have been performed in specific European populations to understand the missing link between genetic and environmental factors for lung function, GWIS of Asian samples remain rare. Therefore, we performed a GWIS of exposure to air pollution to identify loci for lung function in Korean adult men. A total of 1826 adult men recruited from two health check-up centers were included in the analysis and the annual mean concentrations of ambient particulate matter with an aerodynamic diameter ≤10 μm (PM<SUB>10</SUB>) were used. In case of forced vital capacity (FVC), one SNP (rs12312730) that passed our genome-wide threshold of <I>p</I>int < 1 × 10–5 was detected in the intronic region of the <I>BICD1</I> gene on chromosome 12. In addition, we found two variants (rs6743376 and rs17042888) located near the <I>IL1RN-IL1F10</I> gene that were involved in the inflammatory response and associated with decreased FVC via interaction with PM<SUB>10</SUB> exposure. A stratified association analysis according to these SNP genotypes showed that PM<SUB>10</SUB> concentrations in subjects with one or two of the risk alleles, compared with those with the non-risk allele, were significantly correlated with a reduction in FVC. This pattern was replicated in another 892 Korean adult samples. The current study reports the first GWIS discovery in an Asian population: the <I>BICD1</I> and <I>IL1RN-IL1F10</I> genes may contribute to the decrease in FVC levels by interacting with PM<SUB>10</SUB> exposure.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Significant interactions between <I>BICD1 or IL1RN-IL1F10</I> and PM<SUB>10</SUB> for FVC were found. </LI> <LI> The several SNPs in these genes were more susceptible to FVC decline by PM<SUB>10</SUB>. </LI> <LI> For FVC, these interaction effects were reproducible in another sample. </LI> </UL> </P>

      • Highly Branched RuO<sub>2</sub> Nanoneedles on Electrospun TiO<sub>2</sub> Nanofibers as an Efficient Electrocatalytic Platform

        <P>Highly single-crystalline ruthenium dioxide (RuO<SUB>2</SUB>) nanoneedles were successfully grown on polycrystalline electrospun titanium dioxide (TiO<SUB>2</SUB>) nanofibers for the first time by a combination of thermal annealing and electrospinning from RuO<SUB>2</SUB> and TiO<SUB>2</SUB> precursors. Single-crystalline RuO<SUB>2</SUB> nanoneedles with relatively small dimensions and a high density on electrospun TiO<SUB>2</SUB> nanofibers are the key feature. The general electrochemical activities of RuO<SUB>2</SUB> nanoneedles–TiO<SUB>2</SUB> nanofibers and Ru(OH)<SUB>3</SUB>-TiO<SUB>2</SUB> nanofibers toward the reduction of [Fe(CN)<SUB>6</SUB>]<SUP>3–</SUP> were carefully examined by cyclic voltammetry carried out at various scan rates; the results indicated favorable charge-transfer kinetics of [Fe(CN)<SUB>6</SUB>]<SUP>3–</SUP> reduction via a diffusion-controlled process. Additionally, a test of the analytical performance of the RuO<SUB>2</SUB> nanoneedles–TiO<SUB>2</SUB> nanofibers for the detection of a biologically important molecule, hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>), indicated a high sensitivity (390.1 ± 14.9 μA mM<SUP>–1</SUP> cm<SUP>–2</SUP> for H<SUB>2</SUB>O<SUB>2</SUB> oxidation and 53.8 ± 1.07 μA mM<SUP>–1</SUP> cm<SUP>–2</SUP> for the reduction), a low detection limit (1 μM), and a wide linear range (1–1000 μM), indicating H<SUB>2</SUB>O<SUB>2</SUB> detection performance better than or comparable to that of other sensing systems.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/aamick/2015/aamick.2015.7.issue-28/acsami.5b03178/production/images/medium/am-2015-03178q_0005.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/am5b03178'>ACS Electronic Supporting Info</A></P>

        본 논문에서 Compressor 및 turbine 에서의 Blade failure등의 내부파손이 이를 둘러싸고 있는 케이스 내부에 머무르게 하는 엔진설계의 방법에 대한 이론 및 Simulation 등을 기술 하였다. 가장 무거운 부품 중에 하나인 케이스의 두께 최적화는, 항공기의 안정성뿐만 아니라 항공 효율을 높이기 위한 경량화의 목적을 위해서도 매우 중요한 설계목표 이다. 이러한 목적을 위하여 이론적 접근방법으로 에너지 밸런스 방법을 사용하였으며, 파손된 블레이드의 거동특성 및 영향성 평가를 위한 유한요소해석을 위하여 LS-DYNA가 사용 되어졌다. On the basis of the paper described herein, rotor blade failure in the compressor, gas generator turbine, and power turbine and the resulting internal damage is contained within the peripheral hardware and engine casings. For the safety reason, the blade containment was regulated by aviation authority. For reducing the weight of the case, a heaviest single component of a jet engine, the blade containment capability was analyzed by engine manufacturer. The procedure established for containment design involves an energy balance method based on the comparison of the kinetic energy of released blade and the strain energy of the containment zone.
