Fungal exopolysaccharides are important natural products having diverse biological functions. In this study, exopolysaccharides from Fomitopsis castanea mycelia (FEPS) were prepared, and the highest mushroom tyrosinase inhibitory activity was found. FEPS were prepared from cultivation broth by ethanol precipitation method. The extraction yield and protein concentration of FEPS were 213.1 mg/l and 0.03%, respectively. FEPS inhibited mushroom tyrosinase with the half maximal inhibitory concentration (IC<sub>50</sub>) of 16.5 mg/ml and dose-dependently inhibited cellular tyrosinase activity (63.9% at 50 μg/ml, and 83.3% at 100 μg/ml) in the cell-free extract of SK-MEL-5 human melanoma cell and α-melanocytestimulating hormone (α-MSH)-stimulated melanin formation in intact SK-MEL-5 human melanoma cell. The IC<sub>50</sub> of FEPS against NO production from RAW264.7 macrophage cells was 42.8 ± 0.64 μg/ml. By in vivo study using a zebrafish model, exposure of FEPS at 400 μg/ml to dechorionated zebrafish embryos for 18 h decreased the pigment density, compared to that without FEPS-treated control.
Identifying Typhoon Targeted Observations Sensitive Areas Using the Gradient Definition Based Method

Increasing additional observation in the typhoon targeted observation sensitive area can help providing more accurate initial field for numerical models, further to improve the typhoon forecast skill. The critical problem is how to identify the typhoon targeted observation sensitive area. Conditional nonlinear optimal perturbation (CNOP) has been proved to be an effective method. Generally, the CNOP is solved using adjoint-based method, which needs to utilize the adjoint models of the numerical models. However, the adjoint models for some numerical models have not been developed or only for some modules. The gradient definition based method is an adjoint-free method, which has been applied to solve the CNOP of Zebiak-Cane (ZC) model with 1080-dimensional solution space to study the optimal precursors of El Nino-Southern Oscillation (ENSO) event. It is very easy to realize, but the time efficiency will go down dramatically along with the rapidly increasing dimensions. In this paper, the gradient definition based method is applied to solve the CNOP ofMM5 model with more than 105-dimensional solution space to identify the typhoon targeted observation sensitive area. Compared to the adjoint-based method, the identified sensitive area and the benefits of the CNOPs are very similar for typhoon Matmo (2014) and Fitow (2013), and higher time efficiency can be achieved. Furthermore, the OSSEs’ results show that the sensitive area identified can be used to improve the forecast skill of typhoon Matmo and Fitow to different extent.
Listeriosis in a Pregnant Woman and a Neonate

Listeriosis is an infectious disease caused by Listeria monocytogenes. It’s rare in healthy adults, it could be fatal in pregnant women. Listeriosis in pregnant women causes fever and flu-like symptoms, but could lead to fatal consequences. However, because there are few cases of listeriosis reported in Korea, the symptoms are ambiguous and the standard diagnostic tool has yet to be determined, making accurate diagnosis difficult. We present a case of a 35-year-old pregnant woman who had fever and headache. Initially, we didn’t find the cause of fever. However, after delivery, L. monocytogenes was detected in the neonatal gastric aspirate culture, blood cultures. The placenta and amniotic membrane showed acute suppurative inflammation and chorioamnionitis. The mother recovered after antibiotic treatment and delivery, but in the case of the newborn baby, the patient died despite treatment. We report a case of listeriosis in a pregnant woman and a neonate with literature review.

<P><B>Abstract</B></P> <P>We evaluated the combined effect of high pressure (HP) and vinegar addition on control of <I>Clostridium perfringens</I> and quality in emulsion-type pork sausages. Sausages were manufactured with different levels of commercial white distilled vinegar made with jasmine tea extract (0, 1, and 2%) and sodium nitrite (0.02%, control). The sausages except for sodium nitrite treatment were subjected to HP at 500 MPa (four cycles and each for 3 min). After storage, the combined treatment of HP and vinegar showed the lowest number of <I>C</I>. <I>perfringens</I> vegetative cells (4.8-log CFU/g reduction) among the all treatments. The combined treatment also inhibited the growth of <I>C</I>. <I>perfringens</I> spores for five weeks (stored at 4 °C for two weeks followed by at 20 °C for three weeks). Among all treatment combinations, the best pH, water-holding capacity and chewiness, were found with the HP-vinegar sausage. In addition, vinegar treatment inhibited the lipid oxidation of sausage throughout the storage. Use of HP and vinegar addition inhibited growth of <I>C</I>. <I>perfringens</I> without observed adverse effects on quality of the emulsion-type sausage.</P> <P><B>Industrial relevance</B></P> <P>Currently, the use of natural additives and novel processing technologies for replacing sodium nitrite in meat products is gaining attention owing to increase in consumer demand for nitrite-free and safe meat products. The study showed that the addition of 1% (w/v) of vinegar and pressure treatment at 500 MPa (four cycles and each for 3 min) can replace sodium nitrite to inhibit growth of <I>C</I>. <I>perfringens</I> in emulsion-type sausages.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Vinegar supplemented sausages (0, 1, 2%) were subjected to HP (0.1, 500 MPa) and stored for 5 weeks. </LI> <LI> Vegetative cells and spores of <I>C. perfringens</I> in 1% vinegar-HP sausages were reduced by 4.8 and 2.8 log CFU/g, respectively. </LI> <LI> No significant quality changes were observed in the sausage with HP (500 MPa for 12 min) and vinegar (1%). </LI> <LI> HP and vinegar can be applied for safer nitrite-free sausage against <I>C</I>. <I>perfringens</I> with acceptable quality. </LI> </UL> </P>
Anti-cariogenic Characteristics of Rubusoside

Streptococcus mutans plays an important role in the development of dental caries in humans by synthesizing adhesive insoluble glucans from sucrose by mutansucrase activity. To explore the anti-cariogenic characteristics of rubusoside (Ru), a natural sweetener component in Rubus suavissimus S. Lee (Rosaceae), we investigated the inhibitory effect of Ru against the activity of mutansucrase and the growth of Streptococcus mutans. Ru (50 mM) showed 97% inhibitory activity against 0.1 U/mL mutansucrase of S. mutans with 500 mM sucrose. It showed competitive inhibition with a Ki value of 1.1 ± 0.2 mM and IC50 of 2.3 mM. Its inhibition activity was due to hydrophobic and hydrogen bonding interactions based on molecular docking analysis. Ru inhibited the growth of S. mutans as a bacteriostatic agent, with MIC and MBC values of 6 mM and 8 mM, respectively. In addition, Ru showed synergistic anti-bacterial activity when it was combined with curcumin. Therefore, Ru is a natural anti-cariogenic agent with antimutansucrase activity and antimicrobial activity against S. mutans.
